The acquisition of genotoxin-induced mutations in the mammalian germline is detrimental to the stable transfer of genomic information. In somatic cells, nucleotide excision repair (NER) is a major pathway to counteract the mutagenic effects of DNA damage. Two NER subpathways have been identified, global genome repair (GGR) and transcription-coupled repair (TCR). In contrast to somatic cells, little is known regarding the expression of these pathways in germ cells. To address this basic question, we have studied NER in rat spermatogenic cells in crude cell suspension, in enriched cell stages and within seminiferous tubules after exposure to UV or N-acetoxy-2-acetylaminofluorene. Surprisingly, repair in spermatogenic cells was inefficient in the genome overall and in transcriptionally active genes indicating non-functional GGR and TCR. In contrast, extracts from early/mid pachytene cells displayed dual incision activity in vitro as high as extracts from somatic cells, demonstrating that the proteins involved in incision are present and functional in premeiotic cells. However, incision activities of extracts from diplotene cells and round spermatids were low, indicating a stage-dependent expression of incision activity. We hypothesize that sequestering of NER proteins by mispaired regions in DNA involved in synapsis and recombination may underlie the lack of NER activity in premeiotic cells.

The quality of germ cell DNA is critical for the fate of the offspring, yet there is limited knowledge of the DNA repair capabilities of such cells. One of the main DNA repair pathways is base excision repair (BER) which is initiated by DNA glycosylases that excise damaged bases, followed by incision of the generated abasic (AP) sites. We have studied human and rat methylpurine-DNA glycosylase (MPG), uracil-DNA glycosylase (UNG), and the major AP endonuclease (HAP1/APEX) in male germ cells. Enzymatic activities and western analyses indicate that these enzymes are present in human and rat male germ cells in amounts that are at least as high as in somatic cells. Minor differences were observed between different cellular stages of rat spermatogenesis and spermiogenesis. Repair of methylated DNA was also studied at the cellular level using the Comet assay. The repair was highly efficient in both human and rat male germ cells, in primary spermatocytes as well as round spermatids, compared to rat mononuclear blood cells or hepatocytes. This efficient BER removes frequently occurring DNA lesions that arise spontaneously or via environmental agents, thereby minimising the number of potential mutations transferred to the next generation.

Salmonella sp. é um dos principais microrganismos causadores de surtos de enfermidades transmitidas por alimentos associados ao consumo de ovos e de alimentos formulados com este ingrediente. Ovos desidratados são largamente utilizados pelas indústrias de alimentos, por oferecer maior praticidade e maior padronização em relação ao produto \"in natura\". Apesar do processo tecnológico de desidratação do ovo incluir uma etapa de pasteurização, existe um risco de haver microrganismos sobreviventes, já que a pasteurização é feita em temperatura branda. Além disso, a pasteurização pode destruir os fatores intrínsecos antimicrobianos presentes na clara, possibilitando a multiplicação de microrganismos que sobreviveram ao processo de pasteurização ou que contaminaram o produto após a pasteurização. O controle da Aa do produto desidratado e o tempo de armazenamento são, portanto, fatores fundamentais para o controle da multiplicação de microrganismos indesejáveis. Nesse estudo, avaliou-se a cinética de multiplicação de Salmonella experimentalmente adicionada a ovo em pó Aa ajustada para 0,4, 0,6, 0,8 e 0,9, durante o armazenamento em quatro temperaturas: 8°C, 15°C, 25°C e 35°C. Os resultados indicaram que S. Enteritidis é capaz de sobreviver por longo tempo (pelo menos 56 dias) em ovo em pó com Aa próximo de 0,4 quando armazenado a 8°C, 15° e a 25°C. Essa sobrevivência é menor (até 28 dias) quando o armazenamento é feito a 35°C. No ovo em pó com Aa em tomo de 0,6 ou 0,8, S. Enteritidis sobrevive por menos tempo do que no produto com Aa de cerca de 0,4, independentemente da temperatura de armazenamento. No produto com Aa de cerca de 0,9, há grande multiplicação de S. Enteritidis quando o armazenamento é feito a 15°C, 25°C ou 35°C. Nesse produto, o armazenamento a 8°C impede a multiplicação do patógeno. Verificou-se também que Salmonella Radar, resistente a diversos antibióticos, apresentou o mesmo comportamento que S. Enteritidis nas amostras de ovo estudadas.

Cadwaladerite (Al(OH)2Cl∙4H2O) collected from Cerro Pintados, Chile described by Gordon in 1941 is designated as “doubtful” by the IMA. Material collected from the same locality in 2015 resembling the description of cadwaladerite gave a powder XRD pattern similar to lesukite (Al2(OH)5Cl∙2H2O). However, Gordon provided no X-ray data for his material from Cerro Pintados. In order to determine whether cadwaladerite and lesukite are the same mineral species, measurements were made on a suite of samples from various localities. A portion of the material collected by Gordon in 1941 was also obtained from the Mineralogical Museum of Harvard University. Type material of lesukite from a fumarolic environment at the Tolbachik Fissure in Kamchatka, Russia was obtained as well as lesukite from the Maria Mine, Chile (Arica Province) and a previously undescribed locality for lesukite (Barranaca del Sulfato, Mejillones Peninsula, Antofagasta Province). All samples are yellow to yellow-orange in colour and all exhibit small cubic crystals (up to 50µm), even Gordon’s cadwaladerite which was thought to be amorphous. The Chilean samples are all associated with halite and sometimes with anhydrite. These five samples were studied by SEM, FTIR, powder XRD, and Raman spectroscopy. A ratio of Al:Cl less than or equal to 1.3:1 was observed for all the samples, including measurements made on lesukite from the Russian locality Vergasova et al. studied in 1997, and determined to have a 2:1 ratio. SEM-EDS analyses also show all samples to have minor iron substitution, as well as copper substitution in two samples. FTIR spectra are very similar for all samples. Raman spectroscopy done on both samples collected in Cerro Pintados and the Russian lesukite gave similar spectra. Powder XRD analyses on all samples showed spectra identified to be lesukite, including Gordon’s cadwaladerite. Crystal cell parameters calculated from powder XRD ranged from 19.778Å to 19.878Å. Results using modern instrumental techniques confirm Gordon’s cadwaladerite, collected in 1939 and described in 1941, and lesukite are the same mineral species.

Religionskritik nach Stirner, die einer abgeklärten Aufklärung folgt, kann danach eigentlich nur zweierlei: Zum einen erklären, wie Der Mensch sich seine Religion und seine Götter erschafft (hat): das erledigt Gunnar Heinsohn. Und zum Zweiten aufzuzeigen: Welche funktionalen Äquivalente sich im Religiösen dem Menschen auftun – frei nach Feuerbach: Die Wahrheit der Religion ist das Bedürfnis nach ihr. Das freie Stirnersche Ich, das rational um die Nichtexistenz Gottes weiß, wählt sich seinen je eigenen Gott oder, ist es stark genug, kann es auch lassen.

Remineralization of organic matter in reactive marine sediments releases nutrients and dissolved organic matter (DOM) into the ocean. Here we focused on the molecular-level characterization of DOM by high-resolution Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS) in sediment pore waters and bottom waters from contrasting redox regimes in the northern Black Sea with particular emphasis on nitrogen-bearing compounds to derive an improved understanding of the molecular transformations involved in nitrogen release. The number of nitrogen-bearing molecules is generally higher in pore waters than in bottom waters. This suggests intensified degradation of nitrogen-bearing precursor molecules such as proteins in anoxic sediments: No significant difference was observed between sediments deposited under oxic vs anoxic conditions (average O/C ratios of 0.55) suggesting that the different organic matter quality induced by contrasting redox conditions does not impact protein diagenesis in the subseafloor. Compounds in the pore waters were on average larger, less oxygenated, and had a higher number of unsaturations. Applying a mathematical model, we could show that the assemblages of nitrogen-bearing molecular formulas are potential products of proteinaceous material that was transformed by the following reactions: (a) hydrolysis and deamination, both reducing the molecular size and nitrogen content of the products and intermediates; (b) oxidation and hydration of the intermediates; and (c) methylation and dehydration.
