Cells and cell-free solutions of the culture filtrate of the bacterial symbiont, Xenorhabdus nematophila taken from the entomopathogenic nematode Steinernema carpocapsae in aqueous broth suspensions were lethal to larvae of the diamondback moth Plutella xylostella. Their application on leaves of Chinese cabbage indicated that the cells can penetrate into the insects in the absence of the nematode vector. Cell-free solutions containing metabolites were also proved as effective as bacterial cells suspension. The application of aqueous suspensions of cells of X. nematophila or solutions containing its toxic metabolites to the leaves represents a possible new strategy for controlling insect pests on foliage.

Larvae of the pine beauty moth Panolis flammae (Denis & Schiffermuller) were reared in sleeve cages on five different seed origins (provenances) of pole stage Pinus contorta in the field in each of four years from 1985 to 1988. Survival varied significantly between the years. In those years when survival was high, significant differences between tree provenance were not found. However, between provenance significant differences were found in larval weight and stage of development. In the years when survival was low, the results seen in good years were reversed. Significant differences attributable to provenance were found but these were not reflected in significant differences between larval weight or development. In addition, there was a significant correlation between the proportion surviving and larval weight, which was not the case in those years where larval survival was high. The results are discussed in light of the pest status of P, flammea in Britain and in view of current silvicultural policies. The use of trees resistant to insect attack as part of an integrated pest management programme is highlighted and the need to coordinate laboratory and field studies so as to control for environmental variation discussed.

Polyommatus bellargus is a priority species of butterfly in the UK as a result of its scarcity and the rate of population decline over the last few years. In the UK, the species is associated with chalk grassland on hot, south-facing slopes suitable for the growth of the food plant Hippocrepis comosa. Shooting game birds is a popular pastime in the UK. Over 40 million game birds, principally Phasianus colchicus and Alectoris rufa, are bred and released into the countryside each year for shooting interests. There is a concern that the release of such a large number of non-native birds has an adverse effect on native wildlife. A study was carried out over a period of 3 years out to examine whether there was any evidence that A. rufa released into chalk grassland habitat negatively affects populations of P. bellargus. A comparison was made between sites where large numbers of A. rufa were released versus sites where no, or few, birds were released. The study involved the construction of exclosures in these sites to allow an examination of the number of butterflies emerging from H. comosa when the birds were excluded versus when the birds had free range across the area. Where birds were present the on-site vegetation was shorter than where they were absent indicating that the birds were definitely influencing habitat structure. However, the evidence that A. rufa was negatively influencing the number of adult butterflies emerging was not strong, although there was a largely non-significant tendency for higher butterfly emergence when the birds were excluded or absent.

The courtship behavior of the navel orangeworm, Amyelois transitella, was examined in a wind tunnel. Sixty nine courtship sequences were analyzed and successful sequences divided into two categories: rapid courtship sequences, which involved few breaks in contact, short or no periods of male/female chasing and lasted <10 s between initial contact and mating; and prolonged courtship sequences, which involved many breaks in contact, extended periods of male/female chasing and lasted >10 s. Fifty six (81%) courtships were successful (50.7% rapid courtship and 30.4% prolonged courtship); the remaining 13 (18.8%) sequences were failed courtships. Of failed courtships, 9 (13.0%) were due to males losing contact with females during courtship chases and 4 (5.8%) due to females flying away immediately after male contact. Of all courtship sequences involving a break in contact during a chase, 38.5% resulted in an unsuccessful mating attempt. These findings contrast with previous studies of the courtship behavior of the navel orangeworm, potentially indicating that the type of bioassay used to study courtship may have a large effect on the behavioral sequences displayed. We evaluate several diagnostic techniques for the analysis of sequences of behavioral transitions.

Lonomia obliqua caterpillar bristle extract induces hemolysis in vitro on washed human and rat erythrocytes, in either the absence or presence of exogenous lecithin. In the former condition, phospholipases A(2) are key enzymes involved in hemolysis. However, the mechanism whereby this extract causes direct hemolysis is not known. Thus, the aim of this study was to investigate the hemolytic mechanism of the crude extract of the caterpillar L obliqua on human erythrocytes in the absence of lecithin. The extract significantly increased the erythrocyte osmotic fragility and promoted the removal of glycophorins A and C, and band 3 from the erythrocyte membrane. The use of Ca(2+) and Mg(2+) ions significantly potentiated glycoprotein removal, remarkably of erythrocyte band 3. The composition of fatty acids was analyzed by HPLC in both L obliqua caterpillar bristle extract and human erythrocyte membranes incubated with the extract. The levels of unsaturated fatty acids were remarkably augmented in erythrocytes incubated with the extract than in control erythrocytes, modifying thereby the saturated/unsaturated fatty acid ratio. Altogether, evidence is provided here that the interplay of at least three mechanisms of action accounts for the direct activity of the bristle extract on erythrocyte membrane, leading to hemolysis: the removal of glycoproteins and band 3; the insertion of fatty acids; and the action of phospholipases. Such mechanisms might affect erythrocyte flexibility and deformability, which may induce hemolysis by increasing erythrocyte fragility. However, whether the direct hemolytic activity of L obliqua caterpillar is the major cause of intravascular hemolysis during envenomation still needs further investigation. (C) 2010 Elsevier Ltd. Insect digestive chymotrypsins are present in a large variety of insect orders but their substrate specificity still remains unclear. Ewer insect chymotrypsins from 3 different insect orders (Dictyoptera, Coleoptera and two Lepidoptera) were isolated using affinity chromatography. Enzymes presented molecular masses in the range of 20 to 31 kDa and pH optima in the range of 7.5 to 10.0. Kinetic characterization. using different, colorimetric and fluorescent substrates indicated that insect chymotrypsins differ from, bovine chymotrypsin in their primary specificity toward small substrates (like N-benzoyl-L-Tyr p-nitroanilide) rather than on their preference for large substrates (exemplified by Succynil-Ala-Ala-Pro-Phe P-nitroanilide). Chloromethyl ketones (TPCK, N-alpha-tosyl-L-Phe chloromethyl ketone and Z-GGF-CK, N-carbobenzoxy-Gly-Gly-phe-CK) inactivated all chymotrypsins legated. Inactivation rates follow apparent first-order kinetics with variable second order rates (TPCK, 42 to 130 M(-1)s(-1); Z-GGF-CK, 150 to 450 M(-1)s(-1) that may be remarkably low for S. frugiperda chymotrypsin (TPCK, 6 M(-1)s(-1); Z-GGF-CK, 6.1 M(-1) s(-1)). Homology modelling and sequence alignment showed that. in lepidopteran chymotrypsins, differences in the amino acid residues in the neighborhood of the catalytic His 57 may affect its pKa, value. This is Proposed as the cause of the decrease in His 57 reactivity toward chloromethyl ketones. Such amino acid replacement in the active site is proposed. to be an adaptation to the presence of dietary ketones. (C) 2009 Wiley Periodicals, Inc.

The characterization and identification of proteolytic bacteria from the gut of the velvetbean caterpillar (Anticarsia gemmatalis) were the objectives of this study. Twelve aerobic and anaerobic isolates of proteolytic bacteria were obtained from the caterpillar gut in calcium caseinate agar. The number of colony forming units (CFUs) of proteolytic bacteria was higher when the bacteria were extracted from caterpillars reared on artificial diet rather than on soybean leaves (1.73 +/- 0.35 X 10(3) and 0.55 +/- 0.22 X 10(3) CFU/mg gut, respectively). The isolated bacteria were divided into five distinct groups, according to their polymerase chain reaction restriction fragment-length polymorphism profiles. After molecular analysis, biochemical tests and fatty acid profile determination, the bacteria were identified as Bacillus subtilis, Bacillus cereus, Enterococcus gallinarum, Enterococcus mundtii, and Staphylococcus xylosus. Bacterial proteolytic activity was assessed through in vitro colorimetric assays for (general) proteases, serine proteases, and cysteine proteases. The isolated bacteria were able of hydrolyzing all tested substrates, except Staphylococcus xylosus, which did not exhibit serine protease activity. This study provides support for the hypothesis that gut proteases from velvetbean caterpillar are not exclusively secreted by the insect cells but also by their symbiotic gut bacteria. The proteolytic activity from gut symbionts of the velvetbean caterpillar is suggestive of their potential role minimizing the potentially harmful consequences of protease inhibitors from some of this insect host plants, such as soybean, with implications for the management of this insect pest species.

Neste estudo, foram abordadas as implicações da condição nutricional e características físicas de Passiflora suberosa e Passiflora misera para a performance, comportamento de escolha pelo adulto, digestão e nutrição das larvas de H. erato phyllis. Inicialmente, avaliaram-se aspectos do desenvolvimento e foram realizadas medidas morfométricas de Passiflora suberosa e Passiflora misera cultivadas em três níveis de nitrogênio no solo. Avaliou-se o crescimento, a área foliar, o comprimento dos internódios, a dureza, a espessura, e a pilosidade, bem como analisou-se o conteúdo de água, de macro e micronutrientes presentes nos tecidos das respectivas plantas de ambas espécies. A seguir verificou-se a influência destas plantas na performance de Heliconius erato phyllis. Para isto, avaliou-se a sobrevivência, o tempo de desenvolvimento das larvas, o tamanho das pupas e dos adultos. Também, testou-se o comportamento de escolha dos adultos para oviposição, frente aos ramos de tais passifloráceas. Aspectos da ecologia nutricional das larvas foram avaliados, de forma comparativa, frente a P. suberosa e P. misera, através de índices nutricionais. Determinou-se: 1) o índice de consumo; 2) a taxa de crescimento relativo; 3) a digestibilidade aparente 4) a eficiência na conversão do alimento ingerido e 5) a eficiência na conversão do alimento digerido e convertido em biomassa. Ainda, 6) foi traçado um perfil de macro e micro nutrientes de P. suberosa e P. misera das plantas cultivadas e utilizadas para a criação das larvas, e de duas populações naturais. Em nível de digestão, determinou-se: 1) a quantidade de alimento ingerido, processado e retido 2) o tamanho dos fragmentos vegetais encontrados no intestino anterior e posterior; e ainda, 3) possíveis modificações morfológicas dos fragmentos, no decorrer da passagem destes através do tubo digestivo. O crescimento, a área foliar, e o comprimento dos internódios, de ambas passifloráceas, foram influenciados pelos níveis de nitrogênio. As plantas sem adição deste mineral apresentaram menores medidas em todos estes parâmetros. Foi detectada maior dureza e menor conteúdo de água nas primeiras folhas de P. suberosa cultivadas sem adição de nitrogênio. P. misera cultivada nesta condição também apresentou menor conteúdo de água, no entanto não houve maior dureza nas folhas. P.misera apresentou maior espessura nas primeiras e quintas folhas cultivadas com mais nitrogênio. A maioria dos macro e micronutrientes foram acumulados em maior concentração nas plantas com maior adição de nitrogênio no solo. O maior tempo no desenvolvimento e o menor tamanho alcançado pelos adultos foram causados pela menor porcentagem de nitrogênio e água contidos nos tecidos das plantas sem adição deste mineral. As larvas alimentadas com ramos de P. suberosa nesta condição, também enfrentaram a maior dureza da folha nos primeiros instares. As modificações morfológicas e nutricionais de ambas passifloráceas produziram efeitos na performance e na preferência de H. erato phyllis. A maior taxa de crescimento das larvas alimentadas com P. misera foi explicada pelo maior consumo e pela maior digestibilidade. Conseqüentemente, houve maior eficiência na conversão do alimento ingerido em substância corpórea. Não houve rompimentos das células da epiderme, nem dos fragmentos de P. misera nem dos de P. suberosa, retirados de ambas regiões intestinais. A maior permanência no intestino e o maior contato com o meio digestivo possibilitaram maior aproveitamento dos nutrientes de P. misera, em relação à P. suberosa. Isto ocorreu devido ao menor tamanho com que P. misera foi cortada pelas larvas, e pela menor espessura da folha. Quanto aos nutrientes, com exceção do fósforo e do manganês, os demais foram encontrados em maior concentração em P. suberosa. Dessa forma, as vantagens obtidas pelas larvas devem-se à maior eficiência destas no aproveitamento dos nutrientes contidos em P. misera, e não a um conteúdo nutricional superior nesta espécie.
