Textile Technology: The sun-blocking properties of a textile are enhanced when a dye, pigment, delustrant, or ultraviolet absorber finish is present that absorbs ultraviolet radiation and blocks its transmission through a fabric to the skin. For this reason, dyed fabrics provide better sun protection than bleached fabrics. Since naturally-colored cottons contain pigments that produce shades ranging from light green to tan and brown, it seemed reasonable to postulate that they would provide better sun protection than conventional bleached cotton, and that natural pigments might prove more durable to laundering and light exposure than dyes, but there is no published research on the ultraviolet transmission values for naturally-pigmented cottons. The purpose of this study was to determine the ultraviolet protection (UPF) values of naturally-pigmented cotton in three shades (green, tan, and brown), and the effect of light exposure and laundering on the sun-blocking properties of naturally-pigmented cotton. Naturally-pigmented cotton specimens were exposed to xenon light and accelerated laundering, ultraviolet transmission values measured, and UPF values calculated following light exposure and laundering. The naturally-pigmented cottons exhibited significantly higher UPF values than conventional cotton (bleached or unbleached). Although xenon light exposure and laundering caused some fading, the UPF values of naturally-pigmented cotton continue to be sufficiently high so that all three shades continue to provide good sun protection after the equivalent of 5 home launderings and 80 American Association of Textile Chemists and Colorists fading units (AFUs) of xenon light exposure.

The excited-state dynamics of free-base meso-tetrakis(sulfonatophenyl) porphyrin J-aggregates obtained by the Z-scan technique in femto- and picosecond time scales, along with UV-Vis spectroscopy and flash photolysis is reported. Besides obtaining the S-1 state lifetime, the discrimination between internal conversion and intersystem crossing nonradiative processes from that state was also possible, and their rates and respective quantum yields were found. The aggregates present reverse saturable absorption at 532 nm for both singlet and triplet excited states. The data shown is important for several applications such as optical limiting, photodynamic therapy and others. (C) 2011 Elsevier B.V. All rights reserved.

The quantification of ammonia (NH3) losses from sugarcane straw fertilized with urea can be performed with collectors that recover the NH3 in acid-treated absorbers. Thus, the use of an open NH3 collector with a polytetrafluoroethylene (PTFE)-wrapped absorber is an interesting option since its cost is low, handling easy and microclimatic conditions irrelevant. The aim of this study was to evaluate the efficiency of an open collector for quantifying NH3-N volatilized from urea applied over the sugarcane straw. The experiment was carried out in a sugarcane field located near Piracicaba, Sao Paulo, Brazil. The NH3-N losses were estimated using a semi-open static collector calibrated with N-15 (reference method) and an open collector with an absorber wrapped in PTFE film. Urea was applied to the soil surface in treatments corresponding to rates of 50, 100, 150 and 200 kg ha(-1) N. Applying urea-N fertilizer on sugarcane straw resulted in losses NH3-N up to 24 % of the applied rate. The amount of volatile NH3-N measured in the open and the semi-open static collector did not differ. The effectiveness of the collection system varied non-linearly, with an average value of 58.4 % for the range of 100 to 200 kg ha(-1) of urea-N. The open collector showed significant potential for use; however, further research is needed to verify the suitability of the proposed method.

Pathogenic Leptospira is the etiological agent of leptospirosis, a life-threatening disease that affects populations worldwide. Surface proteins have the potential to promote several activities, including adhesion. This work aimed to study the leptospiral coding sequence (CDS) LIC11087, genome annotated as hypothetical outer membrane protein. The LIC11087 gene was cloned and expressed in Escherichia coil BL21 (DE3) strain by using the expression vector pAE. The recombinant protein tagged with N-terminal 6XHis was purified by metal-charged chromatography and characterized by circular dichroism (CD) spectroscopy. The recombinant protein has the ability to mediate attachment to the extracellular matrix (ECM) components, laminin and plasma fibronectin, and was named Lsa30 (Leptospiral surface adhesin of 30 kDa). Lsa30 binds to laminin and to plasma fibronectin in a dose-dependent and saturable manner, with dissociation equilibrium constants (K-D) of 292 +/- 24 nM and 157 +/- 35 nM, respectively. Moreover, the Lsa30 is a plasminogen (PLC) receptor, capable of generating plasmin, in the presence of activator. This protein may interfere with the complement cascade by interacting with C4bp regulator. The Lsa30 is probably a new surface protein of Leptospira as revealed by immunofluorescence assays with living organisms and the reactivity with antibodies present in serum samples of experimentally infected hamsters. Thus, Lsa30 is a novel versatile protein that may play a role in mediating adhesion and may help pathogenic Leptospira to overcome tissue barriers and to escape the immune system. Background: Leptospirosis is considered a re-emerging infectious disease caused by pathogenic spirochaetes of the genus Leptospira. Pathogenic leptospires have the ability to survive and disseminate to multiple organs after penetrating the host. Leptospires were shown to express surface proteins that interact with the extracellular matrix (ECM) and to plasminogen (PLG). This study examined the interaction of two putative leptospiral proteins with laminin, collagen Type I, collagen Type IV, cellular fibronectin, plasma fibronectin, PLG, factor H and C4bp. Results: We show that two leptospiral proteins encoded by LIC11834 and LIC12253 genes interact with laminin in a dose - dependent and saturable mode, with dissociation equilibrium constants (K-D) of 367.5 and 415.4 nM, respectively. These proteins were named Lsa33 and Lsa25 (Leptospiral surface adhesin) for LIC11834 and LIC12253, respectively. Metaperiodate - treated laminin reduced Lsa25 - laminin interaction, suggesting that sugar moieties of this ligand participate in this interaction. The Lsa33 is also PLG - binding receptor, with a K-D of 23.53 nM, capable of generating plasmin in the presence of an activator. Although in a weak manner, both proteins interact with C4bp, a regulator of complement classical route. In silico analysis together with proteinase K and immunoflorescence data suggest that these proteins might be surface exposed. Moreover, the recombinant proteins partially inhibited leptospiral adherence to immobilized laminin and PLG. Conclusions: We believe that these multifunctional proteins have the potential to participate in the interaction of leptospires to hosts by mediating adhesion and by helping the bacteria to escape the immune system and to overcome tissue barriers. To our knowledge, Lsa33 is the first leptospiral protein described to date with the capability of binding laminin, PLG and C4bp in vitro.

TSSA (trypomastigote small surface antigen) is a polymorphic mucin-like molecule displayed on the surface of Trypanosoma cruzi trypomastigote forms. To evaluate its functional properties, we undertook comparative biochemical and genetic approaches on isoforms present in parasite stocks from extant evolutionary lineages (CL Brener and Sylvio X-10). We show that CL Brener TSSA, but not the Sylvio X-10 counterpart, exhibits dose-dependent and saturable binding towards non-macrophagic cell lines. This binding triggers Ca2+-based signalling responses in the target cell while providing an anchor for the invading parasite. Accordingly, exogenous addition of either TSSA-derived peptides or specific antibodies significantly inhibits invasion of CL Brener, but not Sylvio X-10, trypomastigotes. Non-infective epimastigote forms, which do not express detectable levels of TSSA, were stably transfected with TSSA cDNA from either parasite stock. Although both transfectants produced a surface-associated mucin-like TSSA product, epimastigotes expressing CL Brener TSSA showed a similar to 2-fold increase in their attachment to mammalian cells. Overall, these findings indicate that CL Brener TSSA functions as a parasite adhesin, engaging surface receptor(s) and inducing signalling pathways on the host cell as a prerequisite for parasite internalization. More importantly, the contrasting functional features of TSSA isoforms provide one appealing mechanism underlying the differential infectivity of T. cruzi stocks.

In this study we investigate the singlet excited state absorption of lutetium bisphthalocyanine (LuPc2) over a wide spectral range. It was observed distinct nonlinear absorption behaviors; saturable (SA) and reverse saturable absorption (RSA). The RSA effect was observed below 640 and above 680 nm, while SA occurs around the Q-band region, located around 660 nm. To describe the main singlet-singlet transitions, we employed the rate equation model considering the simplified three-energy level diagram. Our results reveal a ratio between excited and ground state absorption smaller than 0.05 at the Q-band region, and of approximately 4 for the other regions. (C) 2012 Elsevier B.V. The motivation for the work presented in this thesis is to retrieve profile information for the atmospheric trace constituents nitrogen dioxide (NO2) and ozone (O3) in the lower troposphere from remote sensing measurements. The remote sensing technique used, referred to as Multiple AXis Differential Optical Absorption Spectroscopy (MAX-DOAS), is a recent technique that represents a significant advance on the well-established DOAS, especially for what it concerns the study of tropospheric trace consituents. NO2 is an important trace gas in the lower troposphere due to the fact that it is involved in the production of tropospheric ozone; ozone and nitrogen dioxide are key factors in determining the quality of air with consequences, for example, on human health and the growth of vegetation. To understand the NO2 and ozone chemistry in more detail not only the concentrations at ground but also the acquisition of the vertical distribution is necessary. In fact, the budget of nitrogen oxides and ozone in the atmosphere is determined both by local emissions and non-local chemical and dynamical processes (i.e. diffusion and transport at various scales) that greatly impact on their vertical and temporal distribution: thus a tool to resolve the vertical profile information is really important. Useful measurement techniques for atmospheric trace species should fulfill at least two main requirements. First, they must be sufficiently sensitive to detect the species under consideration at their ambient concentration levels. Second, they must be specific, which means that the results of the measurement of a particular species must be neither positively nor negatively influenced by any other trace species simultaneously present in the probed volume of air. Air monitoring by spectroscopic techniques has proven to be a very useful tool to fulfill these desirable requirements as well as a number of other important properties. During the last decades, many such instruments have been developed which are based on the absorption properties of the constituents in various regions of the electromagnetic spectrum, ranging from the far infrared to the ultraviolet. Among them, Differential Optical Absorption Spectroscopy (DOAS) has played an important role. DOAS is an established remote sensing technique for atmospheric trace gases probing, which identifies and quantifies the trace gases in the atmosphere taking advantage of their molecular absorption structures in the near UV and visible wavelengths of the electromagnetic spectrum (from 0.25 μm to 0.75 μm). Passive DOAS, in particular, can detect the presence of a trace gas in terms of its integrated concentration over the atmospheric path from the sun to the receiver (the so called slant column density). The receiver can be located at ground, as well as on board an aircraft or a satellite platform. Passive DOAS has, therefore, a flexible measurement configuration that allows multiple applications. The ability to properly interpret passive DOAS measurements of atmospheric constituents depends crucially on how well the optical path of light collected by the system is understood. This is because the final product of DOAS is the concentration of a particular species integrated along the path that radiation covers in the atmosphere. This path is not known a priori and can only be evaluated by Radiative Transfer Models (RTMs). These models are used to calculate the so called vertical column density of a given trace gas, which is obtained by dividing the measured slant column density to the so called air mass factor, which is used to quantify the enhancement of the light path length within the absorber layers. In the case of the standard DOAS set-up, in which radiation is collected along the vertical direction (zenith-sky DOAS), calculations of the air mass factor have been made using “simple” single scattering radiative transfer models. This configuration has its highest sensitivity in the stratosphere, in particular during twilight. This is the result of the large enhancement in stratospheric light path at dawn and dusk combined with a relatively short tropospheric path. In order to increase the sensitivity of the instrument towards tropospheric signals, measurements with the telescope pointing the horizon (offaxis DOAS) have to be performed. In this circumstances, the light path in the lower layers can become very long and necessitate the use of radiative transfer models including multiple scattering, the full treatment of atmospheric sphericity and refraction. In this thesis, a recent development in the well-established DOAS technique is described, referred to as Multiple AXis Differential Optical Absorption Spectroscopy (MAX-DOAS). The MAX-DOAS consists in the simultaneous use of several off-axis directions near the horizon: using this configuration, not only the sensitivity to tropospheric trace gases is greatly improved, but vertical profile information can also be retrieved by combining the simultaneous off-axis measurements with sophisticated RTM calculations and inversion techniques. In particular there is a need for a RTM which is capable of dealing with all the processes intervening along the light path, supporting all DOAS geometries used, and treating multiple scattering events with varying phase functions involved. To achieve these multiple goals a statistical approach based on the Monte Carlo technique should be used. A Monte Carlo RTM generates an ensemble of random photon paths between the light source and the detector, and uses these paths to reconstruct a remote sensing measurement. Within the present study, the Monte Carlo radiative transfer model PROMSAR (PROcessing of Multi-Scattered Atmospheric Radiation) has been developed and used to correctly interpret the slant column densities obtained from MAX-DOAS measurements. In order to derive the vertical concentration profile of a trace gas from its slant column measurement, the AMF is only one part in the quantitative retrieval process. One indispensable requirement is a robust approach to invert the measurements and obtain the unknown concentrations, the air mass factors being known. For this purpose, in the present thesis, we have used the Chahine relaxation method. Ground-based Multiple AXis DOAS, combined with appropriate radiative transfer models and inversion techniques, is a promising tool for atmospheric studies in the lower troposphere and boundary layer, including the retrieval of profile information with a good degree of vertical resolution. This thesis has presented an application of this powerful comprehensive tool for the study of a preserved natural Mediterranean area (the Castel Porziano Estate, located 20 km South-West of Rome) where pollution is transported from remote sources. Application of this tool in densely populated or industrial areas is beginning to look particularly fruitful and represents an important subject for future studies.

Im Rahmen der vorliegenden Arbeit wurden zum ersten Mal kalorimetrische Tieftemperatur-Detektoren in der Beschleuniger-Massenspektrometrie (Accelerator Mass Spectrometry AMS), einer Standard-Methode zur Bestimmung kleinster Isotopenverhältnisse, eingesetzt, um das Isotopenverhältnis von 236U zu 238U zu bestimmen. Das Uran-Isotop 236U entsteht in der Neutroneneinfang-Reaktion 235U(n,gamma)236U und kann daher als Monitor-Nuklid für Neutronenflüsse verwendet werden. Die Detektoren bestehen aus einem Saphir-Absorber, auf den ein supraleitender Aluminium-Film aufgedampft ist, der als Thermistor dient. Ein energetisches Schwerion deponiert seine kinetische Energie als Wärme im Absorber, dessen Temperaturänderung durch die Widerstandsänderung des Supraleiters nachgewiesen wird. Mit solchen Detektoren konnte in vorhergehenden Experimenten bei GSI in einem Energiebereich von E = 5 - 300 MeV/amu für eine Vielzahl von Ionen von Neon bis Uran eine relative Energieauflösung von (1 - 4) E-3 erreicht werden. Der für die Beschleuniger-Massenspektrometrie typische Energiebereich liegt bei E = 0.1 - 1 MeV/amu. Im ersten Schritt wurde daher die systematische Untersuchung der Detektoreigenschaften auf diesen Energiebereich ausgedehnt. Diese Untersuchungen sowie die AMS-Messungen wurden am Tandem-Beschleuniger VERA des Instituts für Isotopenforschung und Kernphysik der Universität Wien durchgeführt. In einem Energiebereich von 10 - 60 MeV konnte für verschiedene Ionen (13C, 197Au, 238U) zunächst eine relative Energieauflösung von DeltaE/E = 7 E-3 erreicht werden. Dies übertrifft die Auflösung konventioneller Ionisations-Detektoren um ca. eine Größenordnung. Durch eine Verbesserung thermischer und elektronischer Rauschbeiträge konnte in einem zweiten Experiment für Uran der Energie 17 MeV die Auflösung auf DeltaE/E = 4.6 E-3 verbessert werden. Die Energie-Response des Detektors war linear über den gesamten beobachteten Energiebereich und unabhängig von der Ionenmasse; bis auf ein Niveau von 0.1 % wurde kein Pulshöhendefekt beobachtet. Diese Ergebnisse zeigen, daß solche Detektoren ein wertvolles Werkzeug in der Schwerionenphysik im Bereich relativ niedriger Ionenenergien darstellen. Mit der erreichten Energieauflösung war es möglich, für mehrere Proben aus natürlichem Uran das Isotopenverhältnis 236U/238U zu bestimmen: Um einen Material-Standard für Uran in der AMS zu etablieren, wurde das Isotopenverhältnis 236U/238U für zwei Proben aus der Mine ''K.u.K. Joachimsthal'' möglichst präzise bestimmt. Die Ergebnisse in der vorliegenden Arbeit stimmen gut mit früheren Messungen überein, die mit einem konventionellen Detektorsystem durchgeführt wurden. Sowohl der statistische als auch der systematische Fehler konnten deutlich reduziert werden. Für eine weitere Probe, extrahiert aus dem Wasser einer Uran-haltigen Quelle in Bad Gastein, wurde ein Isotopenverhältnis von 6.1 E-12 gemessen. Dies stellt das kleinste bislang für 236U/238U gemessene Isotopenverhältnis dar und bedeutet eine Steigerung der Sensitivität um eine Größenordnung. Die erreichte Energieauflösung ermöglicht es außerdem, die Detektoren zur direkten Massenidentifikation von schweren Ionen mittels einer kombinierten Energie-Flugzeit-Messung einzusetzen. In ersten Test-Messungen im Rahmen der vorliegenden Arbeit wurde eine Massenauflösung von DeltaM/M = (8.5 - 11.0) E-3 erreicht. In einem ersten Test für den Einsatz dieser Detektoren zum Nachweis sog. ''superschwerer Elemente (Z >= 112)'' erlaubte der große dynamische Bereich, die Reaktionsprodukte und ihre nachfolgenden Alpha-Zerfälle mit hoher Energieauflösung simultan und zeitaufgelöst nachzuweisen.

Photovoltaic (PV) solar panels generally produce electricity in the 6% to 16% efficiency range, the rest being dissipated in thermal losses. To recover this amount, hybrid photovoltaic thermal systems (PVT) have been devised. These are devices that simultaneously convert solar energy into electricity and heat. It is thus interesting to study the PVT system globally from different point of views in order to evaluate advantages and disadvantages of this technology and its possible uses. In particular in Chapter II, the development of the PVT absorber numerical optimization by a genetic algorithm has been carried out analyzing different internal channel profiles in order to find a right compromise between performance and technical and economical feasibility. Therefore in Chapter III ,thanks to a mobile structure built into the university lab, it has been compared experimentally electrical and thermal output power from PVT panels with separated photovoltaic and solar thermal productions. Collecting a lot of experimental data based on different seasonal conditions (ambient temperature,irradiation, wind...),the aim of this mobile structure has been to evaluate average both thermal and electrical increasing and decreasing efficiency values obtained respect to separate productions through the year. In Chapter IV , new PVT and solar thermal equation based models in steady state conditions have been developed by software Dymola that uses Modelica language. This permits ,in a simplified way respect to previous system modelling softwares, to model and evaluate different concepts about PVT panel regarding its structure before prototyping and measuring it. Chapter V concerns instead the definition of PVT boundary conditions into a HVAC system . This was made trough year simulations by software Polysun in order to finally assess the best solar assisted integrated structure thanks to F_save(solar saving energy)factor. Finally, Chapter VI presents the conclusion and the perspectives of this PhD work.

Wir haben die linearen und nichtlinearen optischen Eigenschaften von dünnen Schichten und planaren Wellenleitern aus mehreren konjugierten Polymeren (MEH-PPV und P3AT) und Polymeren mit -Elektronen Systemen in der Seitenkette (PVK und PS) untersucht und verglichen. PVK und PS haben relativ kleine Werte des nichtlinearen Brechungsindex n2 bei 532 nm, nämlich (1,2 ± 0,5)10-14 cm2/W und (2,6 ± 0,5) 10-14 cm2/W.rnWir haben die linearen optischen Konstanten von mehreren P3ATs untersucht, insbesondere den Einfluss der Regioregularität und Kettenlänge der Alkylsubstituenten. Wir haben das am besten geeignete Polymere für Wellenleiter Anwendungen identifiziert, welches P3BT-ra genannt ist. Wir haben die linearen optischen Eigenschaften dünner Schichten des P3BT-ra untersucht, die mit Spincoating aus verschiedenen Lösungsmitteln mit unterschiedlichen Siedetemperaturen präparieret wurden. Wir haben festgestellt, dass P3BT-ra Filme aus Toluol-Lösungen die am besten geeigneten Wellenleiter für die intensitätsabhängigen Prismen-Kopplungs Experimente sind, weil diese geringe Wellenleiterdämpfungsverluste bei = 1064 nm haben. rnWir haben die Dispersionen des Wellenleiterdämfungsverlustes gw, des nichtlinearen Brechungsindex n2 und des nichtlinearen Absorptionskoeffizienten 2 von Wellenleitern aus P3BT-ra im Bereich von 700 - 1500 nm gemessen. Wir haben große Werte des nichtlinearen Brechungsindex bis 1,5x10-13 cm2/W bei 1150 nm beobachtet. Wir haben gefunden, dass die Gütenkriterien (“figures of merit“) für rein optische Schalter im Wellenlängebereich 1050 - 1200 nm erfüllt sind. Die Gütekriterien von P3BT-ra gehören zu den besten der bisher bekannten Werte von konjugierten Polymeren.rnWir haben gefunden, dass P3BT-ra ein vielversprechender Kandidat für integriert-optische Schalter ist, weil es eine gute Kombination aus großer Nichtlinearität dritter Ordnung, geringen Wellenleiterdämpfungverlusten und ausreichender Photostabilität zeigt. rnWir haben einen Vergleich der gemessenen Dispersion von gw, n2 und 2 mit der Theorie durchgeführt. Durch Kurvenanpassung der Dispersion von gw haben wir gefunden, dass Rayleigh-Streuung der dominierende Dämpfungsmechanismus in MEH-PPV und P3BT-ra Wellenleitern ist. Ein quantenmechanischer Ansatz wurde zur Berechnung der nichtlinearen Suszeptibilität dritter Ordnung (3) verwendet, um die gemessenen Spektren von n2 und 2 von P3BT-ra und MEH-PPV zu simulieren. Dies kann erklären, dass sättigbare Absorption und Zwei-Photonen Absorption die hauptsächlichen Effekte sind, welche die Dispersion von n2 und 2 verursachen. rn

Kalorimetrische Tieftemperatur-Detektoren (Calorimetric Low Temperature Detectors, CLTDs) wurden erstmals in Messungen zur Bestimmung des spezifischen Energieverlustes (dE/dx) niederenergetischer Schwerionen beim Durchgang durch Materie eingesetzt. Die Messungen wurden im Energiebereich unterhalb des Bragg-Peaks, mit 0.1 - 1.4 MeV/u 238U-Ionen in Kohlenstoff und Gold sowie mit 0.05 - 1.0 MeV/u 131Xe-Ionen in Kohlenstoff, Nickel und Gold, durchgeführt. Die Kombination der CLTDs mit einem Flugzeitdetektor ermöglichte dabei, kontinuierliche dE/dx-Kurven über größere Energiebereiche hinweg simultan zu bestimmen. Im Vergleich zu herkömmlichen Meßsystemen, die Ionisationsdetektoren zur Energiemessung verwenden, erlaubten die höhere Energieauflösung und -linearität der CLTDs eine Verringerung der Kalibrierungsfehler sowie eine Erweiterung des zugänglichen Energiebereiches der dE/dx-Messungen in Richtung niedriger Energien. Die gewonnen Daten können zur Anpassung theoretischer und semi-empirischer Modelle und somit zu einer Erhöhung der Präzision bei der Vorhersage spezifischer Energieverluste schwerer Ionen beitragen. Neben der experimentellen Bestimmung neuer Daten wurden das alternative Detektionsprinzip der CLTDs, die Vorteile dieser Detektoren bezüglich Energieauflösung und -linearität sowie der modulare Aufbau des CLTD-Arrays aus mehreren Einzeldetektoren genutzt, um diese Art von Messung auf potentielle systematische Unsicherheiten zu untersuchen. Unter anderem wurden hierbei unerwartete Channeling-Effekte beim Durchgang der Ionen durch dünne polykristalline Absorberfolien beobachtet. Die koinzidenten Energie- und Flugzeitmessungen (E-ToF) wurden weiterhin genutzt, um das Auflösungsvermögen des Detektor-Systems bei der direkten in-flight Massenbestimmung langsamer und sehr schwerer Ionen zu bestimmen. Durch die exzellente Energieauflösung der CLTDs konnten hierbei Massenauflösungen von Delta-m(FWHM) = 1.3 - 2.5 u für 0.1 - 0.6 MeV/u 238U-Ionen erreicht werden. In einer E-ToF-Messung mit Ionisationsdetektoren sind solche Werte in diesem Energie- und Massenbereich aufgrund der Limitierung der Energieauflösung durch statistische Schwankungen von Verlustprozessen beim Teilchennachweis nicht erreichbar.
