No presente trabalho foram avaliados os benefícios da previsão de vazão afluente de curto e longo prazo, na operação de um reservatório com dois usos conflitantes: geração de energia e controle de cheias. A simulação da operação do reservatório foi realizada com base em dois tipos de modelos para avaliar os benefícios da previsão. Um modelo de operação sem previsão e outro com previsão de vazão afluente, este último desenvolvido no presente trabalho. Regras de operação simples, na forma de curvas-guia lineares, foram utilizadas nos casos de operação com e sem previsão de vazões afluentes. As curvas-guia foram otimizadas através de uma técnica de parametrização, simulação e otimização utilizando um algoritmo evolutivo semelhante a um algoritmo genético. Como base para as análises foram utilizados dados relativos ao reservatório de Três Marias, no Rio São Francisco, principalmente pela disponibilidade de previsões reais de vazão de curto prazo a partir de um trabalho prévio. Essas previsões reais de vazão foram calculadas através de um modelo hidrológico distribuído que utiliza como dados de entrada, previsões de chuva do modelo atmosférico regional ETA. Para avaliar o potencial benefício das previsões de vazão na operação do reservatório, foram realizados testes considerando às vazões afluentes observadas como “previsões perfeitas de vazão”. Os resultados com previsões perfeitas de vazão mostram que pode haver um benefício relativo (incremento na geração de energia) de aproximadamente 8% (cerca de 4,77 milhões de dólares anuais), se forem utilizadas previsões de vazão de longo prazo com dois meses de antecedência, e se a operação for planejada com essa mesma antecedência. A operação baseada em previsões de prazos ou horizontes mais curtos apresenta benefícios inferiores, mas ainda assim significativos. Por exemplo, a previsão perfeita com freqüência semanal e horizonte de 12 dias pode trazer um benefício de aproximadamente 4,45% (cerca de 2,75 milhões de dólares anuais). Esses benefícios foram obtidos com o mesmo desempenho no controle de cheias. Posteriormente, foram realizados testes utilizando as previsões reais de vazão. Os benefícios obtidos com as previsões reais de curto prazo são inferiores aos benefícios obtidos com as previsões perfeitas de curto prazo, como era esperado. Entretanto, com as previsões reais de vazão, foram obtidos benefícios superiores a 50% dos que seriam esperados com a previsão perfeita (vazões observadas). Os resultados obtidos são promissores e mostram que há vantagens evidentes na utilização de previsões de chuva para se obter previsões de vazão na operação de reservatórios com usos múltiplos, quando também é associada à otimização sistêmica de um aproveitamento hidrelétrico.

Os microsatélites, também chamados STRs (Short Tandem Repeat), são pequenas sequências de DNA que consistem numa sequência de repetições de um motivo que varia de um a seis pares de bases. Existem em quase todos os cromossomas humanos e podem situar-se nos exões ou nos intrões. Estes últimos são altamente polimórficos e são por isso utilizados na identificação de indivíduos em testes de paternidade e também em estudos de genética de populações. A combinação dos vários genótipos possíveis faz com que cada indivíduo possua um perfil único, que permite a sua identificação. Existem também microsatélites associados a exões ou a regiões promotoras dos genes, normalmente repetições trinucleotídicas CGG/CCG ou CAG/CTG, associados a doenças neurodegenerativas como a síndrome do X-frágil e a doença de Huntington. Neste trabalho caracterizaram-se geneticamente várias populações humanas dos arquipélagos da Madeira, Açores e Cabo Verde. A partir do estudo dos microsatélites do cromossoma Y, foram definidas idades de coalescência que permitiram concluir que as cópias do gene DAZ situado no cromossoma Y são o resultado de um processo evolutivo estando a sua evolução associada a alguns haplogrupos. Verificou-se também a ocorrência de possíveis mutações nos SNPs que definem os haplogrupos, através da comparação dos microsatélites do cromossoma Y dentro de cada haplogrupo, especialmente no haplogrupo E3b. Verificou-se existir uma associação entre o número de repetições CAG e GGC do gene Receptor de Androgénios (AR), situado no cromossoma X, e a infertilidade especialmente quando combinados os dois polimorfismos, parecendo haver um efeito protector dos alelos maiores e alguma susceptibilidade para os alelos menores. Quando se estudou o número de repetições GGC do gene FMR1 em doentes com suspeita de síndrome de X-frágil observaram-se diferenças significativas quando comparadas com a população em geral e com um grupo de sobredotados. Essa diferença deveu-se principalmente à presença do alelo 29 em quase todos os indivíduos do primeiro grupo o que por si só não constitui um factor de risco mas poderá ser uma indicação da associação deste alelo com outra mutação no mesmo gene que possa ser responsável por este fenótipo.

Com a realização deste trabalho pretendeu-se estabelecer o perfil urinário de níveis de biomarcadores do stress oxidativo (5-HMU; UAc; MDA; 8-OHdG) em indivíduos saudáveis (grupo controlo) comparando com o de doenças cardiovasculares (grupo CVD) de modo a avaliar o seu potencial como possíveis biomarcadores da possibilidade de ocorrência de CVD. A extração dos compostos alvos foi realizada por recurso a uma nova técnica extrativa - microextração com adsorvente empacotado em seringa (MEPS) controlada digitalmente (eVol). A análise dos biomarcadores foi efetuada por cromatografia líquida de ultra eficiência (UHPLC) utilizando como coluna analítica a HSS T3 (100 mm × 2,1 mm, 1,7 μm de tamanho da partícula) e com um sistema deteção de fotodiodos (PDA). Otimizaram-se os parâmetros experimentais com influência no processo extrativo, nomeadamente no que se refere ao tipo de adsorvente, á influência do pH, ao volume de amostra, ao número de ciclos extrativos, lavagem e ao volume de eluição. Foram ensaiadas diferentes condições experimentais e selecionadas as que corresponderam a uma maior eficiência extrativa, expressa pela área total relativa dos analitos e reprodutibilidade. Os melhores resultados foram obtidos usando como adsorvente C8, o pH da amostra ajustado a 6, o adsorvente foi carregado com 5x50 μL de amostra e a eluição com 1x50 μL de 0,01% ácido formico e 3x50 μL de 20% metanol. Para a separação cromatográfica dos analitos usou-se uma fase móvel binária (0,01% ácido fórmico:20% metanol), em modo isocrático e um fluxo de 250 μL min-1. O método analítico foi validado em termos de seletividade, linearidade, limite de deteção (LOD), limite de quantificação (LOQ), efeito matriz, exatidão e precisão (intra e interdias) e aplicada a determinação de biomarcadores alvo nos dois grupos estudados, obtiveram-se bons resultados em termos seletividade e linearidade (R2>0,9906), os valores de LOD e LOQ obtidos foram baixos, variando entre 0,00005 - 0,72 μg mL-1 e 0,00023 – 2,31 μg mL-1 respetivamente. Os resultados da percentagem de recuperação (91,06 – 123,02 %), precisão intra-dia (0,95 – 8,34 %), precisão inter-dia (4,58 -6,33 %) e o efeito de matriz (60,11 – 110,29 %) deste método foram satisfatórios. A aplicação da metodologia validada aos dois grupos em estudo permitiu concluir que as concentrações de UAc e MDA entre os dois grupos, contrariamente ao 5-HMU e ao 8- OhdG cujas concentrações são estatisticamente diferentes entre o grupo controlam e o grupo CVD.

This study aimed to describe the spatial and temporal patterns of variation in trophic state and its relation with the structure and dynamics of planktonic community of a large reservoir located in semi-arid tropical region of Northeast Brazil. The reservoir Armando Ribeiro Gonçalves is the biggest reservoir of the Rio Grande do Norte State and is responsible for about 53% of all surface water accumulated in the State. The samples of water and plankton were taken monthly in 10 points distributed throughout the longitudinal axis of the reservoir and over a full hydrological cycle. The samples were collected to determine concentrations of total phosphorus, total nitrogen, chlorophyll a and suspended solids (fixed and volatile) and for determining the composition and abundance of phytoplanktonic and zooplanktonic species. During the study period, the reservoir was characterized as eutrophic and there was no trend of increasing the trophic state of the reservoir in the period of drought. The concentrations of total phosphorus and suspended fixed solids decreased towards the dam while the N:P ratios increased in the same direction due to the reduction in the phosphorus concentrations and relative constancy in the nitrogen concentrations. The N:P ratios observed were indicative of greater limitation by phosphorus than by nitrogen. However, as concentrations of both nutrients were high and the water transparency was very low, with secchi depth usually lower than one meter, it seems likely that the planktonic primary production of the reservoir is more limited by the availability of light than the availability of nutrients. High nutrient concentrations coupled with low availability of light may explain the continuing dominance of filamentous cyanobacteria such as Cylindrospermopsis raciborskii in the plankton of the reservoir.These cyanobacteria are potentially toxic and pose a serious environmental problem because it compromises the water quality for public supply, recreation and fishing when present in high densities as in this study. The mesozooplankton of the reservoir was dominated by the calanoid Notodiaptomus cearensis and the cladoceran Diaphanosoma spinulosum. In general, the structure of zooplankton community seems to be particularly influenced by the spatial variation of cyanobacteria. The results of the regression analyses show that both the chlorophyll a concentrations and the cyanobacteria biovolume were more strongly correlated with the nitrogen than with phosphorus and that the water transparency was more strongly correlated with algal biomass than with other sources of turbidity. The maximum load of phosphorus to attain the maximum permissible concentration of total P in the reservoir was estimated in 63.2 tonnes P/ year. The current external P load to the reservoir is estimated in 324 tonnes P / year and must be severely reduced to improve the water quality for water supply and allow the implementation of aquaculture projects that could contribute to the socio-economic development of the region

The aims of this study were: i) assessing the trophic state of the Mendubim reservoir (semi-arid, Rio Grande do Norte, Brazil; 05° 38 99,0 S 36°55 98,0 W) based on chlorophyll-a, total phosphorus and nitrogen concentrations and water transparency; ii) relating the patterns of temporal variation of zooplankton and phytoplankton to the trophic state of the reservoir and iii) investigating the carrying capacity of the reservoir for cage fish farming. The samplingwas done monthly from July 2006 to July 2007 in three stations at the reservoir: next to the dam (barrage), in the central region and in the mouth of the main tributary. The abiotic and biotic variables analyzed were: Secchi depth, volatiles and fixed suspended solids, chlorophyll-a, total phosphorus and nitrogen, TN:TP ratio and mesozooplankton and phytoplankton composition and biomass. The results showed that the reservoir can be considered as mesotrophic with mean concentrations of total nitrogen, phosphorus and chlorophyll-a equal to 1711, 1 μg.L-1, 30,8 μg.L-1 and 5,62 μg.L-1 respectively. The Cyanophyceae class was the most representative in terms of density, with the presence of potentially toxic species such as Microcystis aeruginosa, Planktothrix planctonica, Cylindrospermopsis raciborskii, Aphanizomenon sp. ,Aphanocapsa delicatissima and Pseudanabaena acicularis. Among the zooplankton, the genus Notodiaptomus presented the largest biomass values. Overall, our results show that the light limitation should explain the weak relationship between chlorophyll-a and total phosphorus and nitrogen concentrations. We concluded that the water of Mendubim reservoir is suitable for intensive fish cage aquaculture. Based on the carrying capacity calculations for this reservoir, we found that the maximum sustainable yield of tilapias in cages in the reservoir is 126 ton per year assuming a factor of food conversion of 1.5: 1.0 and a phosphorus content in the fish food of 1%

Top-down (grazing) and bottom-up (nutrient, light) controls are important in freshwater ecosystems regulation. Relative importance of these factors could change in space and time, but in tropical lakes bottom-up regulation has to been appointed as more influent. Present study aimed to test the hypothesis that phytoplankton growths rate in Armando Ribeiro reservoir, a huge eutrophic reservoir in semi-arid region of Rio Grande do Norte state, is more limited by nutrient available then zooplankton grazing pressure. Bioassay was conduced monthly from September (2008) to August (2009) manipulating two levels of nutrients (with/without addition) and two level of grazers (with/without removal). Experimental design was factorial 2X2 with four treatments (X5), (i) control with water and zooplankton from natural spot ( C ), (ii) with nutrient addition ( +NP ), (iii) with zooplankton remove ( -Z ) and (iv) with zooplankton remove and nutrient addition ( -Z+NP ). For bioassay confection transparent plastic bottles (500ml) was incubate for 4 or 5 days in two different depths, Secchi`s depth (high luminosity) and 3 times Secchi`s depth (low luminosity). Water samples were collected from each bottle in begins and after incubates period for chlorophyll a concentration analysis and zoopalnktonic organisms density. Phytoplankton growths rates were calculated. Bifactorial ANOVA was performance to test if had a significant effect (p<0,005) of nutrient addition and grazers remove as well a significant interaction between factors on phytoplankton growths rates. Effect magnitude was calculated the relative importance of each process. Results show that phytoplankton growth was in generally stimulated by nutrient addition, as while zooplankton remove rarely stimulated phytoplankton growth. Some significant interactions happening between nutrient additions and grazers remove on phytoplankton growth. In conclusion this study suggests that in studied reservoir phytoplankton growth is more controlled by ascendent factors than descendent

Currently a resource more and more used by the petroleum industry to increase the efficiency of steam flood mechanism is the addition of solvents. The process can be understood as a combination of a thermal method (steam injection) with a miscible method (solvent injection), promoting, thus, the reduction of interfacial tensions and oil viscosity. The use of solvent alone tends to be limited because of its high cost. When co-injected with steam, the vaporized solvent condenses in the cooler regions of the reservoir and mixes with the oil, creating a zone of low viscosity between the steam and the heavy oil. The mobility of the displaced fluid is then improved, resulting in an increase of oil recovery. To better understand this improved oil recovery method, a numerical study of the process was done contemplating the effects of some operational parameters (distance between wells, injection steam rate, kind of solvent and injected solvent volume)on the accumulated production of oil, recovery factor and oil-steam rate. Semisynthetic models were used in this study but reservoir data can be extrapolated for practical applications situations on Potiguar Basin. Simulations were performed in STARS (CMG, 2007.11). It was found that injected solvent volumes increased oil recovery and oil rates. Further the majority of the injected solvent was produced and can be recycled

The application of thermal methods, to increase the recovery of heavy oil in mature fields through drainage with multilateral and horizontal wells, has been thoroughly studied, theorically, experimentally, testing new tools and methods. The continuous injection of steam, through a steam injector well and a horizontal producer well in order to improve horizontal sweep of the fluid reservoir, it is an efficient method. Starting from an heterogeneous model, geologically characterized, modeling geostatistics, set history and identification of the best path of permeability, with seismic 3D, has been dubbed a studying model. It was studied horizontal wells in various directions in relation to the steam and the channel of higher permeability, in eight different depths. Into in the same area were studied, the sensitivity of the trajectories of horizontal wells, according to the depth of navigation. With the purpose of obtaining the highest output of oil to a particular flow, quality, temperature and time for the injection of steam. The wells studied showed a significant improvement in the cumulative oil recovery in one of the paths by promoting an alternative to application in mature fields or under development fields with heavy oil

Water injection is the most widely used method for supplementary recovery in many oil fields due to various reasons, like the fact that water is an effective displacing agent of low viscosity oils, the water injection projects are relatively simple to establish and the water availability at a relatively low cost. For design of water injection projects is necessary to do reservoir studies in order to define the various parameters needed to increase the effectiveness of the method. For this kind of study can be used several mathematical models classified into two general categories: analytical or numerical. The present work aims to do a comparative analysis between the results presented by flow lines simulator and conventional finite differences simulator; both types of simulators are based on numerical methods designed to model light oil reservoirs subjected to water injection. Therefore, it was defined two reservoir models: the first one was a heterogeneous model whose petrophysical properties vary along the reservoir and the other one was created using average petrophysical properties obtained from the first model. Comparisons were done considering that the results of these two models were always in the same operational conditions. Then some rock and fluid parameters have been changed in both models and again the results were compared. From the factorial design, that was done to study the sensitivity analysis of reservoir parameters, a few cases were chosen to study the role of water injection rate and the vertical position of wells perforations in production forecast. It was observed that the results from the two simulators are quite similar in most of the cases; differences were found only in those cases where there was an increase in gas solubility ratio of the model. Thus, it was concluded that in flow simulation of reservoirs analogous of those now studied, mainly when the gas solubility ratio is low, the conventional finite differences simulator may be replaced by flow lines simulator the production forecast is compatible but the computational processing time is lower.
