Monitoring of BCR-ABL transcripts has become established practice in the management of chronic myeloid leukemia. However, nucleic acid amplification techniques are prone to variations which limit the reliability of real-time quantitative PCR (RQ-PCR) for clinical decision making, highlighting the need for standardization of assays and reporting of minimal residual disease (MRD) data. We evaluated a lyophilized preparation of a leukemic cell line (K562) as a potential quality control reagent. This was found to be relatively stable, yielding comparable respective levels of ABL, GUS and BCR-ABL transcripts as determined by RQ-PCR before and after accelerated degradation experiments as well as following 5 years storage at -20 degrees C. Vials of freeze-dried cells were sent at ambient temperature to 22 laboratories on four continents, with RQ-PCR analyses detecting BCR-ABL transcripts at levels comparable to those observed in primary patient samples. Our results suggest that freeze-dried cells can be used as quality control reagents with a range of analytical instrumentations and could enable the development of urgently needed international standards simulating clinically relevant levels of MRD.

OBJECTIVE: To study the visual acuity and astigmatism of persons undergoing cataract extraction by local surgeons in rural China. METHODS: Visual acuity, keratometry, and refraction were measured 10 to 14 months postoperatively for all cataract cases during 4 months in Sanrao, China. RESULTS: Among 313 eligible subjects, 242 (77%) could be contacted, of whom 176 (73%) were examined. Of those who were examined, mean +/- SD age was 69.3 +/- 10.5 years, 66.5% were female, 35 had been operated on bilaterally at Sanrao, and 85.2% had a preoperative presenting visual acuity of 6/60 or worse. Presenting and best-corrected postoperative acuity in the eye that was operated on were 6/18 or better in 83.4% and 95.7%, respectively. Among 27 fellow eyes operated on elsewhere, 40.7% had a presenting acuity of 6/18 or better and 40.7% were blind (P < .001). Mean +/- SD postoperative astigmatism did not differ between 211 eyes that were operated on (-1.13 +/- 0.84 diopters) and 109 eyes that were not (-1.13 +/- 1.17 diopters; P = .27). Presence of operative complications (8.5%) and older age were associated with worse vision; bilateral surgery was associated with better vision. CONCLUSIONS: These results confirm the effectiveness of skill transfer in this setting, with superior outcomes to most studies in rural Asia and to eyes in this cohort operated on at other facilities.

Esta dissertação de mestrado desenvolveu-se no âmbito do projecto “Arqueologia” das Aprendizagens no concelho de Alandroal, que teve, como objectivo primordial, a identificação e caracterização do universo de aprendizagens institucionais disponíveis e concretizadas pela comunidade do território em estudo (Freguesia de Capelins (Santo António) /Alandroal), na década 1997 – 2007. Foram inquiridas, nesta freguesia, 16 instituições e identificadas 30 aprendizagens institucionais, que tiveram maior ocorrência nos anos de 2004, 2005 e 2006. Embora seja uma freguesia essencialmente rural, a área de actividade da maioria das instituições identificadas inscreve-se no sector do comércio por grosso e a retalho e de reparação de veículos automóveis e motociclos. São, na sua maioria, instituições de natureza privada e reveladoras de organização, no que respeita à sua organização. No entanto, o relacionamento interinstitucional foi pouco dinâmico, existindo um fraco trabalho em rede. Quando existiram parcerias, estas ocorreram extra freguesia e, até mesmo, extra concelho. Seguindo uma análise quantitativa dos dados extraídos da aplicação dos questionários, com recurso a análise estatística, conclui-se que os processos de aprendizagem das instituições inquiridas da freguesia de Capelins apresentam características centradas nas próprias instituições promotoras dessas mesmas aprendizagens, assentes na formação do pessoal, com carácter formal e não formal e relacionaram-se com as áreas de administração pública e defesa, alojamento e restauração e ainda com a área do comércio por grosso e a retalho; ABSTRACT: This dissertation developed under the project "Archaeology" of Learning in the county of Alandroal, which had as its primary objective, the identification and characterization of the universe of available institutional learning and implemented by the community of the area under study (Parish of capelin (San Antonio) / Alandroal), in the decade from 1997 to 2007. Were surveyed in this parish, and 16 institutions identified 30 institutional learning, which have a higher occurrence in the years 2004, 2005 and 2006. Although it is an essentially rural, the parish area of activity of most institutions identified in the sector of wholesale and retail trade and repair of motor vehicles and motorcycles. Are, for the most part, private institutions and revealing of organization, with regard to its organization. However, the interinstitutional relationship was weak and there is a little dynamic networking. When partnered, this extra parish occurred and even extra municipality. Following a quantitative analysis of data taken from the application of questionnaires, using statistical analysis, it is concluded that the learning processes of the institutions surveyed in the parish of Capelins characteristics focused on promoting institutions of those learnings, based on staff training, formal and non-formal nature and related to the areas of public administration and defense, hotels and restaurants and also the area of wholesale and retail trade.

Este estudo procurou conhecer a realidade do insucesso/abandono escolares no Concelho de Alandroal, no período de 1997-2007, identificando as causas das duas problemáticas e as medidas educativas de resolução, a partir de uma amostra de 41 sujeitos. Pretendemos identificar e caracterizar os episódios de insucesso e abandono escolares protagonizados por um grupo de alunos que iniciaram o seu percurso escolar em 1996/1997, no Concelho de Alandroal. No corpus teórico deste estudo, procedemos à revisão da literatura e à análise de normativos legais que abordam e regulamentam esta temática, de maneira a olhar para ela, de forma reflexiva. Ao nível do corpus empírico, utilizámos uma metodologia quantitativa/qualitativa, no sentido de obtermos o melhor conhecimento e interpretação desta realidade. Os resultados permitem concluir que existe uma relação entre as causas individuais, familiares, escolares e do meio envolvente que levaram ao insucesso e abandono escolares. Sublinhou-se a importância da implementação de medidas educativas adequadas e a sua relação com as instituições do concelho de Alandroal que levaram parte dos sujeitos a concluir o ensino obrigatório com sucesso; ABSTRACT: This study aimed to know the reality of failure / dropping out of school in the Municipality of Alandroal in the period 1997-2007, identifying the causes of both problems and educational measures of resolution, from a sample of 41 subjects. We intend to identify and characterize the episodes of school failure and dropping out staged by a group of students who started their schooling in 1996/1997, Municipality of Alandroal. In the theoretical corpus of this study, we carried out the literature review and analysis of legal norms that regulate and address this issue in order to look at her reflectively. In terms of empirical corpus, we used a method quantitative / qualitative, in order to obtain a better understanding and interpretation of reality. The results indicate that there is a relationship between the causes of individual, family, school and environment that led to failure and early school leaving. It underlined the importance of implementing appropriate educational measures and their relationship with the institutions of the county of Alandroal that led the subjects to complete the compulsory education successfully.

Teacher commitment has been found to be a critical predictor of teachers’ work performance, absenteeism, retention, burnout and turnover, as well as having an important influence on students’ motivation, achievement, attitudes towards learning and being at school (Firestone (1996). Educational Administration Quarterly, 32(2), 209–235; Graham (1996). Journal of Physical Education, Recreation and Dance, 67(1), 45–47; Louis (1998). It is also a necessary ingredient to the successful implementation, adaptation or resistance reform agendas. Surprisingly, however, the relationship between teachers’ motivation, efficacy, job satisfaction and commitment, and between commitment and the quality of their work has not been the subject of extensive research. Some literature presents commitment as a feature of being and behaving as a professional (Helsby, Knight, McCulloch, Saunders, & Warburton (1997). A report to participants on the professional cultures of Teachers Research Project, Lancaster University, January). Others suggest that it fluctuates according to personal, institutional and policy contexts (Louis (1998). School Effectiveness and School Improvement, 9(1), 1–27) and identify different dimensions of commitment which interact and fluctuate (Tyree (1996). Journal of Educational Research, 89(5), 295–304). Others claim that teachers’ commitment tends to decrease progressively over the course of the teaching career (Fraser, Draper, & Taylor (1998). Evaluation and Research in Education, 12 (2), 61–71; Huberman (1993). The lives of teachers. London: Cassell). In this research, experienced teachers in England and Australia were interviewed about their understandings of commitment. The data suggest that commitment may be better understood as a nested phenomena at the centre of which is a set of core, relatively permanent values based upon personal beliefs, images of self, role and identity which are subject to challenge by change which is socio-politically constructed.

An important feature of UK housing policy has been the promotion of consortia between local authorities, private developers and housing associations in order to develop mixed tenure estates to meet a wide range of housing needs. Central to this approach has been a focus on the management of neighbourhoods, based on the assumption that high densities and the inter-mixing of tenure exacerbates the potential for incivility and anti-social behaviour and exerts a disproportionate impact on residents' quality of life. Landlord strategies are therefore based on a need to address such issues at an early stage in the development. In some cases community-based, third sector organisations are established in order to manage community assets and to provide a community development service to residents. In others, a common response is to appoint caretakers and wardens to tackle social and environmental problems before they escalate and undermine residents’ quality of life. A number of innovative developments have promoted such neighbourhood governance approaches to housing practice by applying community development methods to address potential management problems. In the process, there is an increasing trend towards strategies that shape behaviour, govern ethical conduct, promote aesthetic standards and determine resident and landlord expectations. These processes can be related to the wider concept of governmentality whereby residents are encouraged to become actively engaged in managing their own environments, based on the assumption that this produces more cohesive, integrated communities and projects positive images. Evidence is emerging from a number of countries that increasingly integrated and mutually supportive roles and relationships between public, private and third sector agencies are transforming neighbourhood governance in similar ways. This paper will review the evidence for this trend towards community governance in mixed housing developments by drawing on a series of UK case studies prepared for two national agencies in 2007. It will review in particular the contractual arrangements with different tenures, identify codes and guidelines promoting 'good neighbour' behaviour and discuss the role of community development trusts and other neighbourhood organisations in providing facilities and services, designed to generate a well integrated community. The second part of the paper will review evidence from the USA and Australia to see how far there is a convergence in this respect in advanced economies. The paper will conclude by discussing the extent to which housing management practice is changing, particularly in areas of mixed development, whether there is a convergence in practice between different countries and how far these trends are supported by theories of governmentality.

This paper presents a genetic algorithm for the Resource Constrained Project Scheduling Problem (RCPSP). The chromosome representation of the problem is based on random keys. The schedule is constructed using a heuristic priority rule in which the priorities of the activities are defined by the genetic algorithm. The heuristic generates parameterized active schedules. The approach was tested on a set of standard problems taken from the literature and compared with other approaches. The computational results validate the effectiveness of the proposed algorithm.

This paper presents the development of a fish-like robot called Bro-Fish. Bro-Fish aims to be an educational toy dedicated to teaching mechanics, programming and the physics of floating objects to youngsters. The underlying intention is to awaken the interest of children for technology, especially biomimetic (biologically inspired) approaches, in order to promote sustainability and raise the level of ecological awareness. The main focus of this project was to create a robot with carangiform locomotion and controllable swimming, providing the opportunity to customize parts and experiment with the physics of floating objects. Therefore, the locomotion principles of fishes and mechanisms developed in related projects were analysed. Inspired by this background knowledge, a prototype was designed and implemented. The main achievement is the new tail mechanism that propels the robot. The tail resembles the undulation motion of fish bodies and is actuated in an innovative way, triggered by an elegant movement of a rotating helicoidal. First experimental tests revealed the potential of the proposed methodology to effectively generate forward propulsion.

The evolution of the electrical grid into a smart grid, allowing user production, storage and exchange of energy, remote control of appliances, and in general optimizations over how the energy is managed and consumed, is also an evolution into a complex Information and Communication Technology (ICT) system. With the goal of promoting an integrated and interoperable smart grid, a number of organizations all over the world started uncoordinated standardization activities, which caused the emergence of a large number of incompatible architectures and standards. There are now new standardization activities which have the goal of organizing existing standards and produce best practices to choose the right approach(es) to be employed in specific smart grid designs. This paper follows the lead of NIST and ETSI/CEN/CENELEC approaches in trying to provide taxonomy of existing solutions; our contribution reviews and relates current ICT state-of-the-art, with the objective of forecasting future trends based on the orientation of current efforts and on relationships between them. The resulting taxonomy provides guidelines for further studies of the architectures, and highlights how the standards in the last mile of the smart grid are converging to common solutions to improve ICT infrastructure interoperability.

This study deals with investigating the groundwater quality for irrigation purpose, the vulnerability of the aquifer system to pollution and also the aquifer potential for sustainable water resources development in Kobo Valley development project. The groundwater quality is evaluated up on predicting the best possible distribution of hydrogeochemicals using geostatistical method and comparing them with the water quality guidelines given for the purpose of irrigation. The hydro geochemical parameters considered are SAR, EC, TDS, Cl-, Na+, Ca++, SO4 2- and HCO3 -. The spatial variability map reveals that these parameters falls under safe, moderate and severe or increasing problems. In order to present it clearly, the aggregated Water Quality Index (WQI) map is constructed using Weighted Arithmetic Mean method. It is found that Kobo-Gerbi sub basin is suffered from bad water quality for the irrigation purpose. Waja Golesha sub-basin has moderate and Hormat Golena is the better sub basin in terms of water quality. The groundwater vulnerability assessment of the study area is made using the GOD rating system. It is found that the whole area is experiencing moderate to high risk of vulnerability and it is a good warning for proper management of the resource. The high risks of vulnerability are noticed in Hormat Golena and Waja Golesha sub basins. The aquifer potential of the study area is obtained using weighted overlay analysis and 73.3% of the total area is a good site for future water well development. The rest 26.7% of the area is not considered as a good site for spotting groundwater wells. Most of this area fall under Kobo-Gerbi sub basin.

Transplantation of insulin secreting cells is regarded as a possible treatment for type 1 diabetes. One major difficulty in this approach is, however, that the transplanted cells are exposed to the patient's inflammatory and autoimmune environment, which originally destroyed their own beta-cells. Therefore, even if a good source of insulin-secreting cells can be identified for transplantation therapy, these cells need to be protected against these destructive influences. The aim of this project was to evaluate, using a clonal mouse beta-cell line, whether genetic engineering of protective genes could be a viable option to allow these cells to survive when transplanted into autoimmune diabetic mice. We demonstrated that transfer of the Bcl-2 anti-apoptotic gene and of several genes specifically interfering with cytokines intracellular signalling pathways, greatly improved resistance of the cells to inflammatory stresses in vitro. We further showed that these modifications did not interfere with the capacity of these cells to correct hyperglycaemia for several months in syngeneic or allogeneic streptozocin-diabetic mice. However, these cells were not protected against autoimmune destruction when transplanted into type 1 diabetic NOD mice. This suggests that in addition to inflammatory attacks by cytokines, autoimmunity very efficiently kills the transplanted cells, indicating that multiple protective mechanisms are required for efficient transplantation of insulin-secreting cells to treat type 1 diabetes.
