The yeast peptide-methionine sulfoxide reductase (MsrA) was overexpressed in a Saccharomyces cerevisiae null mutant of msrA by using a high-copy plasmid harboring the msrA gene and its promoter. The resulting strain had about 25-fold higher MsrA activity than its parent strain. When exposed to either hydrogen peroxide, paraquat, or 2,2′-azobis-(2-amidinopropane) dihydrochloride treatment, the MsrA overexpressed strain grew better, had lower free and protein-bound methionine sulfoxide and had a better survival rate under these conditions than did the msrA mutant and its parent strain. Substitution of methionine with methionine sulfoxide in a medium lacking hydrogen peroxide had little effect on the growth pattern, which suggests that the oxidation of free methionine in the growth medium was not the main cause of growth inhibition of the msrA mutant. Ultraviolet A radiation did not result in obvious differences in survival rates among the three strains. An enhanced resistance to hydrogen peroxide treatment was shown in human T lymphocyte cells (Molt-4) that were stably transfected with the bovine msrA and exposed to hydrogen peroxide. The survival rate of the transfected strain was much better than its parent strain when grown in the presence of hydrogen peroxide. These results support the proposition that the msrA gene is involved in the resistance of yeast and mammalian cells to oxidative stress.

Analyzing the pathways by which retinoic acid (RA) induces promyelocytic leukemia/retinoic acid receptor α (PML/RARα) catabolism in acute promyelocytic leukemia (APL), we found that, in addition to caspase-mediated PML/RARα cleavage, RA triggers degradation of both PML/RARα and RARα. Similarly, in non-APL cells, RA directly targeted RARα and RARα fusions to the proteasome degradation pathway. Activation of either RARα or RXRα by specific agonists induced degradation of both proteins. Conversely, a mutation in RARα that abolishes heterodimer formation and DNA binding, blocked both RARα and RXRα degradation. Mutations in the RARα DNA-binding domain or AF-2 transcriptional activation region also impaired RARα catabolism. Hence, our results link transcriptional activation to receptor catabolism and suggest that transcriptional up-regulation of nuclear receptors by their ligands may be a feedback mechanism allowing sustained target-gene activation.

To identify genes involved in macrophage development, we used the differential display technique and compared the gene expression profiles for human myeloid HL-60 leukemia cell lines susceptible and resistant to macrophage maturation. We identified a gene coding for a protein kinase, protein kinase X (PRKX), which was expressed in the maturation-susceptible, but not in the resistant, cell line. The expression of the PRKX gene was found to be induced during monocyte, macrophage, and granulocyte maturation of HL-60 cells. We also studied the expression of the PRKX gene in 12 different human tissues and transformed cell lines and found that, among these tissues and cell types, the PRKX gene is expressed only in blood. Among the blood cell lineages, the PRKX gene is specifically expressed in macrophages and granulocytes. Antisense inhibition of PRKX expression blocked terminal development in both the leukemic HL-60 cells and normal peripheral blood monocytes, implying that PRKX is a key mediator of macrophage and granulocyte maturation. Using the HL-60 cell variant deficient in protein kinase C-β (PKC-β) and several stable PKC-β transfectants, we found that PRKX gene expression is under control of PKC-β; hence PRKX is likely to act downstream of this PKC isozyme in the same signal transduction pathway leading to macrophage maturation.

O presente trabalho teve por finalidade investigar a posição ocupada pela mulher dita de meia-idade nos meios de comunicação social. Para tanto, foram selecionadas cerca de quinze Revistas Femininas, editadas pela Bloch e Abril Cultural, tendo sido as fotos e ilustrações apresentadas em tais veículos de comunicação, submetidas ã Análise de Conteúdo, para a qual se contou com uma equipe formada por quatorze juízes, selecionados aleatoriamente. Os estímulos apresentados foram classificados em três categorias, quais sejam, "mulher jovem", "mulher de meia-idade" e "mulher idosa", e os resultados obtidos submetidos a uma análise lógica e estatística. Efetuou-se um estudo sobre a condição da mulher, segundo as dimensões biológica, social e psicológica, considerados aspectos relativos à mítica feminina, além de um-breve histórico sobre a situação da mulher no contexto social do País. Foram, também, observados dados referentes ã Psicologia do Consumidor. A partir dos resultados obtidos constatou-se que a incidência de estímulos classificados como sendo mulheres de meia-idade não foi significativa, alcançando, a figura da mulher jovem, a quase totalidade dos estímulos apresentados em todas as revistas examinadas, mantendo-se elevada mesmo naquelas dirigidas, segundo as Editoras, primordialmente, às mulheres de mais idade. Observou-se, também, uma tendência acentuada a se classificar na categoria "jovem" figuras femininas que se apresentassem como sexualmente atraentes, fisicamente belas e, ainda, a de artistas populares - esta, mesmo naqueles casos em que, reconhecidamente, se sabia possuidoras de idade superior aos 43 anos. Desse modo, concluiu-se que a figura feminina correspondente à mulher considerada de meia-idade, inexiste como forma de apelo social, o que contribuiu, sobremaneira, para tornar ainda mais conflitivo um período já crítico em si mesmo, como o é o climatério.
