This article investigates the researcher's work in the coproduction (or not) of complaint sequences in research interviews. Using a conversation analytic approach, we show how the interviewer's management of complaint sequences in a research setting is consequential for subsequent talk and thus directly affects the data generated. In the examples shown here, researchers sharing cocategorial incumbency with respondents may well provide spaces for research participants to formulate complaints. This article examines sequences of talk surrounding complaints to show how researchers generate complaints (or not) and handle unsafe complaints. Researchers are able to provoke specific types of accounts from respondents, whereas their respondents may actively resist the researchers' direction. For researchers using the interview as a method of data generation, examination of complaint sequences and how these appear in interview data provides insight into how interview talk is coproduced and managed within a socially situated setting.

1. A novel experimental method was developed to study negative physical and chemical effects of latex and cardiac glycosides on first-instar monarch butterfly larvae in their natural environment in north central Florida. Forceps were used to nibble through the petioles of leaves of the sandhill milkweed Asclepias humistrata, mimicking the behaviour of mature monarch larvae. This notching cut off the supply-of latex to the leaves without significantly reducing either their cardiac glycoside concentration or water content. 2. The mean cardiac glycoside concentration in larvae that fed on intact leaves was nearly two: and a half times greater than in larvae that fed on notched leaves. This was probably because more latex is present in the gut of the larvae that fed on the intact leaves. Supporting this is the fact that the mean concentration of cardiac glycosides in the latex was 34-47 times that in the leaves. 3. Wet weights, dry weights, and growth rates of first-instar larvae that fed on intact leaves over a 72-h period were less than half those of larvae that fed on notched leaves. 4. Mortality due to miring in the latex was 27% on the intact leaves compared with 2% on the notched leaves. 5. Latex, cardiac glycosides, and other as yet undetermined plant factors all have a negative effect on:first-instar larval survival. 6. Video-analyses indicated that ingestion of latex caused the larvae to become cataleptic and increased their chances of being mired on the leaf by the setting latex glue. Dysfunction resulting from latex ingestion may lead to the larvae falling off the plant and being killed by invertebrate predators. 7. The difficulty of neonate monarch larvae surviving on A. humistrata - one of the principal milkweed species fed on each spring as monarchs remigrate from Mexico into the southern U.S.A. - is evidence that a co-evolutionary arms race is operating in this plant-herbivore system.

El artículo analiza la evolución del regionalismo en América Latina desde los años noventa. Expone las opciones que se plantean a los países en sus políticas exteriores frente a las iniciativas de Estados Unidos y los cambios en el sistema internacional. Se expone la importancia funcional de las potencias emergentes (PEMS) y las regiones en la fase actual del capitalismo. Finalmente el artículo analiza las perspectivas del regionalismo en América Latina tanto en el sistema internacional como en la construcción de un modelo de gobernanza regional.

A produção de teoria em administração tende hoje a opor, pelo menos metodologicamente, ambientes acadêmicos e aqueles em que tal conhecimento se desenvolve sob motivação de mercado. Escrito do ponto de vista dos primeiros, este artigo é uma reflexão crítica sobre a base epistemológica que parece sustentar os segundos. De fato, trabalhando no limite entre simples senso comum e conhecimento tradicionalmente considerado científico, o método e o argumento usuais que alimentam diretamente empresas, seus consultores e um crescente número de leitores é a indução sobre a experiência - "caso após caso, se funcionou para outros, funcionará para você". Essa é, na verdade, uma questão epistemológica clássica que foi abordada de forma antológica por Karl R. Popper e constitui o centro do artigo. De suas conclusões sobre a lógica da pesquisa científica, podem fluir deduções interessantes tanto ao mundo da gerência quanto da academia, apresentadas na parte final do texto.

Nas classes e palestras de introdução à administração, desperta sempre a curiosidade do auditório lançar a pergunta: "O que vocês acham: Administração é ciência ou arte?" Estamos diante de um dictum interessante e estimulante à discussão. O artigo tem dois objetivos: desvendar a ambiguidade da pergunta - talvez uma das razões de sua sobrevivência - e, a partir dela, examinar a adequação de discutir-se "cientificidade" em nosso saber, questão importante para os rumos da academia nessa área. Para o primeiro objetivo, mais modesto, nos valeremos de análise pragmática e semântica da linguagem, para o segundo, que concentra a maior parte do texto, das teorias sobre a natureza da ciência e a pretendida distinção entre ciência e não ciência. Surgem, então, evidências de que o dictum encerra um mal-entendido.

Este artigo levanta e estabelece as relações entre as exigências organizacionais sobre o gerente moderno e o fenômeno workaholic, identificando algumas implicações na vida dos indivíduos. O artigo analisa as relações de influências mútuas entre contextos, teorias organizacionais, exigências sobre os gerentes e fenômeno workaholic. O foco de estudo é o gerente, aquele que ocupa cargo de comando, toma decisões e tem certo grau de responsabilidade acumulada, em configuração relativamente ampla do cenário nacional, porquanto a pesquisa de natureza qualitativa foi feita com oito gerentes de empresas de segmentos e ramos diferentes, atuando em várias regiões do Brasil.

Este artigo analisa o papel da consultoria organizacional externa (GVconsult/FGV) em dois projetos de desestatização previstos no Programa de Reforma do Estado de Pernambuco, no período 1999-2000: o da Centrais de Abastecimento de Pernambuco (Ceasa-PE) e o do Centro de Convenções (Cecon). O caminho metodológico foi a aproximação gradativa ao objeto de estudo, partindo de suposições iniciais e pontos críticos que serviram de orientação para a investigação. O trabalho de campo foi constituído por 31 entrevistas semi-estruturadas, em três rodadas, e os textos submetidos a uma análise semântico-pragmática da conversação (Mattos, 2005). A história do caso mostrou que a consultoria organizacional exerceu função técnica auxiliar ao governo de Pernambuco no Programa de Reforma do Estado, de acordo com diretrizes bem definidas e limitações impostas pela equipe do governo. Constatou-se também que, apesar de os conceitos da Reforma do Estado de Pernambuco terem origem nas concepções de Estado dos mentores acadêmicos da FGV e esposadas pelo governo federal (1995-98), não foram os consultores da GVconsult que as trouxeram, mas, sim, os formuladores das políticas públicas do governo de Pernambuco que as colheram diretamente do governo federal e da FGV.

Determinou-se a prevalência de toxoplasmose ovina em soros de 100 animais, provenientes de Uruguaiana, RS e abatidos em Bragança Paulista, SP, Brasil, através de reação de Sabin-Feldman (RSF). Considerando-se animais positivos aqueles com títulos > ou = 16, obtiveram-se 39% de soro-reagentes, com títulos e percentuais de soropositividade correspondentes a: 16 (66,7%), 64 (23%), 256 (2,6%), 1024 (5,1) e 4000 (2,6%).

OBJECTIVE: To assess the frequency of combination of antidepressants with other drugs and risk of drug interactions in the setting public hospital units in Brazil. METHODS: Prescriptions of all patients admitted to a public hospital from November 1996 to February 1997 were surveyed from the hospital's data processing center in São Paulo, Brazil. A manual search of case notes of all patients admitted to the psychiatric unit from January 1993 to December 1995 and all patients registered in the affective disorders outpatient clinic in December 1996 was carried out. Patients taking any antidepressant were identified and concomitant use of drugs was checked. By means of a software program (Micromedex®) drug interactions were identified. RESULTS: Out of 6,844 patients admitted to the hospital, 63 (0.9%) used antidepressants and 16 (25.3%) were at risk of drug interaction. Out of 311 patients in the psychiatric unit, 63 (20.2%) used antidepressants and 13 of them (20.6%) were at risk. Out of 87 patients in the affective disorders outpatient clinic, 43 (49.4%) took antidepressants and 7 (16.2%) were at risk. In general, the use of antidepressants was recorded in 169 patients and 36 (21.3%) were at risk of drug interactions. Twenty different forms of combinations at risk of drug interactions were identified: four were classified as mild, 15 moderate and one severe interaction. CONCLUSION: In the hospital general units the number of drug interactions per patient was higher than in the psychiatric unit; and prescription for depression was lower than expected.

Culex quinquefasciatus is a vector of human pathogens, including filarial nematodes and several viruses. Although its epidemiological relevance is known to vary across geographical regions, an understanding of its population genetic structure is still incipient. In light of this, we evaluated the genetic diversity of Cx. quinquefasciatus and Cx. pipiens x Cx. quinquefasciatus hybrids collected from nine localities in Brazil and one site in Argentina. We used mitochondrial genes cox1 and nd4, along with the coxA and wsp genes of the maternally-inherited Wolbachia endosymbiont. The nd4 fragment was invariant between samples, whilst cox1 exhibited four haplotypes that separated two types of Cx. quinquefasciatus, one clustered in southern Brazil. Low sequence diversity was generally observed, being discussed. Both Brazilian and Argentinian mosquitoes were infected with a single Wolbachia strain. As reported in previous studies with these populations, cox1 and nd4 diversity is not congruent with the population structure revealed by nuclear markers or alar morphology. Future Cx. quinquefasciatus research should, if possible, evaluate mtDNA diversity in light of other markers.

RESUMO: O vírus chikungunya (CHIKV) é um vírus de RNA, com invólucro, da família Togaviridae, transmitido por mosquitos Aedes spp. Distribuído por largas regiões de África e Ásia, causa grandes epidemias de artrite grave. A semelhança de sintomas com outras doenças como a dengue e a malária e a persistência de IgM específicas, dificultam o diagnóstico da infeção por CHIKV. A deteção no sangue de E3, uma glicoproteína viral secretada, a incluir num ensaio imunoenzimático poderá melhorar o diagnóstico nos países onde as técnicas de biologia molecular são de difícil acesso. Para testar a utilidade de E3 num ensaio de diagnóstico, esta deverá ser expressa em quantidade, purificada e usada para produção de anticorpos específicos. Para expressar E3 numa forma solúvel, suscetível de ser purificada num único passo cromatográfico sem proteases, recorreu-se à estratégia da fusão com o domínio de ligação à quitina (CBD)-inteína (IMPACT™ System, NEB). A sequência codificadora de E3 foi amplificada a partir de RNA viral, clonada em pTYB21 e expressa em E. coli como uma proteína de fusão insolúvel de 64 kDa. A expressão a 12ºC induzida por IPTG 0,1 mM aumentou a solubilidade de CBD-inteína-E3. A aplicação de lisados celulares em colunas de quitina originou a retenção de CBD-inteína-E3 na matriz. Porém, a autoclivagem da inteína na coluna, induzida com reagentes tiol, foi pouco eficiente e mesmo a proteína E3 separada não eluiu da coluna. E3 foi ainda expressa em E. coli com uma cauda de seis histidinas (E3[His]6) por clonagem no vetor pET28b(+). Lisados celulares aplicados em colunas de níquel permitiram a eluição de uma proteína de 9 kDa, compatível com a massa molecular estimada para E3[His]6, ainda que com outros contaminantes proteicos. A identidade da proteína de 9 kDa será confirmada pela indução de anticorpos com esta preparação e reatividade daqueles com células infetadas com CHIKV.----------------ABSTRACT: Chikungunya virus (CHIKV) is an enveloped, positive strand RNA virus belonging to the family Togaviridae. Transmitted by Aedes spp mosquitoes, CHIKV causes large epidemics of severe arthritogenic disease in Africa and Asia and represents a serious threat in countries where vectors are present. Symptoms similarity with other diseases, e.g. dengue and malaria, along with CHIKV IgM persistence turns accurate CHIKV diagnosis a difficult task in low-income countries. Detection of E3, a small secreted viral glycoprotein, to be included in an immunoenzymatic test was envisaged as a possible improvement in CHIKV diagnosis. To test the diagnostic value of E3, recombinant E3 should be expressed and purified to generate antibodies. In order to express CHIKV E3 in a soluble form amenable to purification by a single step affinity chromatography, the chitin binding domain (CBD)-intein fusion strategy without proteases (IMPACT™ System, NEB) was employed. The E3 coding sequence was amplified from viral RNA, cloned in pTYB21 and expressed in E. coli ER2566 as an insoluble 64 kDa CBD-intein-E3 fusion protein. Solubility was partially achieved by lowering the expression temperature to 12ºC and the inducer (IPTG) concentration to 0.1 mM. Clarified cell lysate loaded onto a chitin column allowed ligation of the fusion protein but the intein-mediated cleavage efficiency was low and E3 failed to elute from the column as demonstrated by SDS-PAGE. E3 was further expressed with a six histidine tag, E3[His]6, employing the pET System (Novagen). E3[His]6 was expressed in E. coli Rosetta (30ºC, 0.4 mM IPTG) as a 9 kDa protein. Soluble cell extracts in 20-40 mM imidazole, applied onto a nickel column and eluted with 500 mM imidazole yielded a protein preparation enriched in the 9kDa protein. The 9 kDa will be used as antigen to generate antibodies that upon reaction with CHIKV infected cells will confirm its identity.

INTRODUCTION: Approximately 30% of hepatitis C virus (HCV) monoinfected patients present persistently normal alanine aminotransferase (ALT) levels. Most of these patients have a slow progression of liver fibrosis. Studies have demonstrated the rate of liver fibrosis progression in hepatitis C virus-human immunodeficiency virus (HCV-HIV) coinfected patients is faster than in patients infected only by HCV. Few studies have evaluated the histological features of chronic hepatitis C in HIV-infected patients with normal ALT levels. METHODS: HCV-HIV coinfected patients (HCV-RNA and anti-HIV positive) with known time of HCV infection (intravenous drugs users) were selected. Patients with hepatitis B surface antigen (HBsAg) positive or hepatitis C treatment before liver biopsy were excluded. Patients were considered to have a normal ALT levels if they had at least 3 normal determinations in the previous 6 months prior to liver biopsy. All patients were submitted to liver biopsy and METAVIR scale was used. RESULTS: Of 50 studied patients 40 (80%) were males. All patients were treated with antiretroviral therapy. The ALT levels were normal in 13 (26%) patients. HCV-HIV co-infected patients with normal ALT levels had presented means of the liver fibrosis stages (0.77±0.44 versus 1.86±1.38; p<0.001) periportal inflammatory activity (0.62±0.77 versus 2.24±1.35; p<0.001) and liver fibrosis progression rate (0.058±0.043 fibrosis unit/year versus 0.118±0.102 fibrosis unit/year) significantly lower as compared to those with elevated ALT. CONCLUSIONS: HCV-HIV coinfected patients with persistently normal ALTs showed slower progression of liver fibrosis. In these patients the development of liver cirrhosis is improbable.
