The Jiyang superdepression is one of the richest hydrocarbon accumulations in the Bohai Bay basin, eastern China. Comprehensive seismic methods have been used in buried hill exploration in Jiyang to describe these fractured reservoirs better. Accurate seismic stratigraphic demarcation and variable-velocity mapping were applied to reveal the inner structure of the buried hills and determine the nature of the structural traps more precisely. Based on the analysis of rock properties and the characteristics of well-developed buried hill reservoirs, we have successfully linked the geology and seismic response by applying seismic forward technology. Log-constrained inversion, absorption coefficient analysis and tectonic forward-inversion with FMI loggings were applied to analyse and evaluate the buried hill reservoirs and gave satisfying results. The reservoir prediction was successful, which confirmed that the comprehensive utilization of these methods can be helpful in the exploration of buried hill reservoirs.

Based on the monthly average SST and 850 hPa monthly average wind data, the seasonal, interannual and long-term variations in the eastern Indian Ocean warm pool (EIWP) and its relationship to the Indian Ocean Dipole (IOD), and its response to the wind over the Indian Ocean are analyzed in this study. The results show that the distribution range, boundary and area of the EIWP exhibited obviously seasonal and interannual variations associated with the ENSO cycles. Further analysis suggests that the EIWP had obvious long-term trend in its bound edge and area, which indicated the EIWP migrated westwards by about 14 longitudes for its west edge, southwards by about 5 latitudes for its south edge and increased by 3.52x10(6) km(2) for its area, respectively, from 1950 to 2002. The correlation and composite analyses show that the anomalous westward and northward displacements of the EIWP caused by the easterly wind anomaly and the southerly wind anomaly over the eastern equatorial Indian Ocean played an important and direct role in the formation of the IOD.

Chromosome identification is an essential step in genomic research, which so far has not been possible in oysters. We tested bacteriophage P1 clones for chromosomal identification in the eastern oyster Crassostrea virginica, using fluorescence in situ hybridization (FISH). P1 clones were labeled with digoxigenin-11-dUTP using nick translation. Hybridization was detected with fluorescein-isothiocyanate-labeled anti-digoxigenin antibodies and amplified with 2 layers of antibodies. Nine of the 21 P1 clones tested produced clear and consistent FISH signals when Cot-1 DNA was used as a blocking agent against repetitive sequences. Karyotypic analysis and cohybridization positively assigned the 9 P1 clones to 7 chromosomes. The remaining 3 chromosomes can be separated by size and arm ratio. Five of the 9 P1 clones were sequenced at both ends, providing sequence-tagged sites that can be used to integrate linkage and cytogenetic maps. One sequence is part of the bone morphogenetic protein type 1b receptor, a member of the transforming growth factor superfamily, and mapped to the telomeric region of the long arm of chromosome 2. This study shows that large-insert clones such as P1 are useful as chromosome-specific FISH probes and for gene mapping in oysters.

Chromosomal location of the 5S ribosomal RNA gene was studied in the eastern oyster, Crassostrea virginica Gmelin. using fluorescence in situ hybridization (FISH). Metaphase chromosomes were obtained from early embryos, and the FISH probe was made by PCR (polymerase chain reaction) amplification of the 5S rRNA gene and labeled by incorporation of digoxigenin-1 1-dUTP during PCR. Hybridization was detected with fluorescein-labeled antidigoxigenin antibodies. Two pairs of FISH signals were observed on metaphase chromosomes. Karyotypic analysis showed that the 5S rRNA gene cluster is interstitially located on short arms of chromosomes 5 and 6. On chromosome 5, the 5S rRNA genes were located immediately next to the centromere, whereas on chromosome 6, they were located approximately half way between the telomere and the centromere. Chromosomes of C. virginica are difficult to identify because of their similarities in size and arm ratio, and the chromosomal location of 5S rRNA genes provides unambiguous identification of chromosomes 5 and 6. Previous studies have mapped the major rRNA gene cluster (18S-5.8S-28S) to chromosome 2. and this study shows that the 5S rRNA gene cluster is not linked to the major rRNA genes and duplicated during evolution.

We measured ecosystem CO2 fluxes for an alpine shrubland on the north-eastern Tibetan Plateau, Qinghai, China. The study is to understand (1) the seasonal variation of CO2 flux and (2) how environmental factors affect the seasonality of CO2 exchange in the alpine ecosystem. Daytime ecosystem respiration was extrapolated from the relationship between temperature and nighttime CO2 fluxes under high turbulent conditions.Seasonal patterns of gross ecosystem production, ecosystem respiration and net ecosystem CO2 exchange followed highly the seasonal change of aboveground biomass in the alpine shrubland. The net ecosystem CO2 exchange was mainly controlled by the variation of photosynthetic photon flux density, while the ecosystem respiration was closely correlated to the soil temperature at 5-cm depth. Integrated values of gross ecosystem production, ecosystem respiration and net ecosystem CO2 exchange for the period from November 1, 2002 to October 31 2003 were estimated to be 1418, 1155 and 222 g CO2 m(-2) yr(-1), respectively.

The photosynthetic pathway of plant species collected at Menyuan, Henan, and Maduo sites, east of Tibetan Plateau, China, during the growing season were studied using stable carbon isotopes in leaves. The 232 samples leaves analyzed belonged to 161 species, 30 families, and 94 genera. The delta(13)C values (from -24.6 to -29.2 %o) indicated that all the considered species had a photosynthetic C-3 pathway. The absence of plant species with C-4 photosynthetic pathway might be due to the extremely low air temperature characterizing the Tibetan Plateau. The average delta(13)C value was significantly (p < 0.05) different between annuals and perennials at the three considered study sites. Hence the longer-lived species had greater water-use efficiency (WUE) than shorter-lived species, that is, longer-lived species are better adapted to the extreme environmental conditions of the Tibetan Plateau.

Ligularia, a highly diversified genus in the eastern Qinghai-Tibet Plateau and adjacent areas, was chosen as a suitable subject in which to study speciation patterns in this 'hot spot' area at the chromosomal level. Chromosome numbers and karyotypes were studied in 23 populations of 14 species, most of which are endemic to this area. The basic number x = 29 was confirmed for all species. Ligularia virgaurea was found to have diploid and triploid cytotypes, 2n = 58 and 87. Other species are only diploid, with 2n = 58. The karyotypes of all populations within any species, and all species spanning most sections and covering most of the morphological range in Ligularia, are very similar to each other, belonging to type 2A according to Stebbin's classification. This karyotype was also found in its close allies, e.g. Cremanthodium, Ligulariopsis, Parasenecio, and Sinacalia. Aneuploid reduction of chromosome number from 2n = 60 to 58 and karyotypic variation was found in Ligularia and its allies. Such a chromosomal pattern with few polyploids infers that variation of karyotype structure at the diploid level seems to be the predominant feature of chromosomal evolution in this group and sympatric speciation via hybridization and polyploidization has played a minor role in its species diversity. The presumed pair relationships of intercontinental vicariad species in the Podophyllum group (Sinopodophyllum hexandrum vs. Podophyllum pelatum and Diphylleia grayi vs. D. cymosa) were recently, considered to be paraphyletic. In the present paper, the trnL-F and ITS gene sequences of the representatives were used to examine the sister relationships of these two vicariad species. A heuristic parsimony analysis based on the trnLF data identified Diphylleia as the basal clade of the other three genera, but provided poor resolution of their inter-relationships. High sequence divergence was found in the ITS data. ITS1 region, more variable but parsimonyuninformative. has no phylogenetic value, Sequence divergence of the ITS2 region provided abundant, phylogenetically informative variable characters. Analysis of ITS2 sequences confirmeda sister relationship between the presumable vicariad species, in spite of a low bootstrap support for Sinopodophyllum hexandrum vs. Podophyllum pelatum. The combined ITS2 and trnL-F data enforced a sister relationship between Sinopodophyllum hexandrum and Podophyllum pelatum with an elevated bootstrap support of 100%. Based on molecular phylogeny, the morphological evolution of this group was discussed. The self-pollination might have evolved from cross-fertilization two times in this group. The different pollination and seed dispersal systems of Sinopodophyllum hexandrum and Podophlyllum pelatum resulted from their adaptations to different ecological habitats. The divergence time of Sinopodophyllum hexandrum-Podophyllum pelatum is estimated to be 6.52+/-1.89 myr based on the ITS divergence. The divergence of this species pair predated or co-occurred with the recent uplift of the Himalayas 4-3 myr during the late Miocene and the formation of the alpine habitats. Sinopodophyllum hexandrum developed a host of specialized characters in its subsequent adaptation to the arid alpine surroundings. The present study confirmed the different patterns of species relationship between Asian-North American disjuncts. The isolation of plant elements between North America and eastern Asia must have been a gradual process, resulting in the different phylogenetic patterns and divergence times of the disjuncts.
