Este artigo revisita a Escola Inglesa - da obra seminal de Hedley Bull em 1977, passando pelos esforços de Adam Watsom nos anos 80, e culminando com o repaginamento de Barry Buzan e Richard Little na década de 90 - resgatando sua ontologia histórica e esboçando um programa de pesquisa que a concilie com o da historiografia francesa. Conclui-se que pode ser detectado um sistema de causalidades múltiplas em nível sistêmico, com forças profundas materiais e psicosociais operando sob os auspícios do conceito de ordem, tão crucial a esta escola.

The Portuguese arrival in India marked the beginning of the age of modern globalization that has been sustained until very recently in conditions that were favourable for the western domination of the globe. Adam Smith analyzed the origin of the wealth of nations in pre-Gamic and post-Gamic eras. It is presently experiencing tremors that were predicted by Andre Gunder Frank in his ReOrient, which the bulk of the western sociologists and economists sought to ignore. For Gunder Frank the past five centuries saw an ephemeral rise of the West, exploring and exploiting the pre-existent markets of China and India. He saw these making a comeback.

OBJECTIVE: The Integrated Management of Childhood Illness is a strategy designed to address major causes of child mortality. The aim of this study was to assess the impact of the strategy on the quality of child health care provided at primary facilities. METHODS: Child health quality of care and costs were compared in four states in Northeastern Brazil, in 2001. There were studied 48 health facilities considered to have had stable strategy implementation at least two years before the start of study, with 48 matched comparison facilities in the same states. A single measure of correct management of sick children was used to assess care provided to all sick children. Costs included all resources at the national, state, local and facility levels associated with child health care. RESULTS: Facilities providing strategy-based care had significantly better management of sick children at no additional cost to municipalities relative to the comparison municipalities. At strategy facilities 72% of children were correctly managed compared with 56% in comparison facilities (p=0.001). The cost per child managed correctly was US$13.20 versus US$21.05 in the strategy and comparison municipalities, respectively, after standardization for population size. CONCLUSIONS: The strategy improves the efficiency of primary facilities in Northeastern Brazil. It leads to better health outcomes at no extra cost.

RESUMO Nos últimos anos têm sido identificados vários factores de risco para asma brônquica em crianças com sibilância recorrente, não se encontrando clara a importância da avaliação funcional respiratória nestas crianças. De igual modo, têm sido documentados resultados contraditórios na avaliação imunológica das populações de células reguladoras bem como na expressão de polimorfismos para a asma. O objectivo deste estudo consistiu na avaliação e comparação de parâmetros de avaliação funcional respiratória, imunológica e de polimorfismos genéticos em crianças entre 8 e 20 meses de idade, com três ou mais episódios de sibilância (n=50), sem qualquer terapêutica anti-inflamatória prévia, diagnosticados por um médico, com e sem factores de risco para asma brônquica (história de asma parental ou história pessoal de eczema ou pelo menos dois dos seguintes: história pessoal de rinite alérgica, sibilância fora do contexto infeccioso e contagem de eosinófilos no sangue periférico > 4%), comparados com um grupo controlo (n=30). Nestas crianças foram efectuadas provas de função respiratória em volume corrente e em volume aumentado através de técnicas de compressão torácica, avaliação de populações celulares por citometria de fluxo, expressão de citocinas por mARN em culturas de células estimuladas com PMA (leitura às 24 horas) e com extractos de ácaros do pó doméstico (leitura ao 7º dia) e expressão de polimorfismos para alguns genes associados a asma (ADAM 33, DPP10, GPRA). Na comparação entre as crianças com sibilância recorrente em relação ao grupo controlo foram observadas reduções significativas nos Z-scores para FVC (diferença média [95% IC]: -0,7 [-1,2; -0,1], p=0,01), FEV0.5 (-1,0 [-1,5; -0,5], p=0,0001), FEF75 (-0,6 [-1,0; -0,2], p=0,0001) e FEF25-75 (-0,8 [-1,2; -0,4], p=0,0001), bem como valores significativamente mais baixos para a quantificação do número absoluto de CD4+CD25forte (-47,9 [-89,6; -6,1], p=0,03), do número absoluto e percentual de CD4+CD25+CTLA-4 (p=0,0001) e da expressão de CTLA-4 (p=0,03) e IFN-

DNA amplification techniques are being used increasingly in clinical laboratories to confirm the identity of medically important bacteria. A PCR-based identification method has been in use in our centre for 10 years for Burkholderia pseudomallei and was used to confirm the identity of bacteria isolated from cases of melioidosis in Ceará since 2003. This particular method has been used as a reference standard for less discriminatory methods. In this study we evaluated three PCR-based methods of B. pseudomallei identification and used DNA sequencing to resolve discrepancies between PCR-based results and phenotypic identification methods. The established semi-nested PCR protocol for B. pseudomallei 16-23s spacer region produced a consistent negative result for one of our 100 test isolates (BCC #99), but correctly identified all 71 other B. pseudomallei isolates tested. Anomalous sequence variation was detected at the inner, reverse primer binding site for this method. PCR methods were developed for detection of two other B. pseudomallei bacterial metabolic genes. The conventional lpxO PCR protocol had a sensitivity of 0.89 and a specificity of 1.00, while a real-time lpxO protocol performed even better with sensitivity and specificity of 1.00, and 1.00. This method identified all B. pseudomallei isolates including the PCR-negative discrepant isolate. The phaC PCR protocol detected the gene in all B. pseudomallei and all but three B. cepacia isolates, making this method unsuitable for PCR-based identification of B. pseudomallei. This experience with PCR-based B. pseudomallei identification methods indicates that single PCR targets should be used with caution for identification of these bacteria, and need to be interpreted alongside phenotypic and alternative molecular methods such as gene sequencing.

Until this day, the most efficient Cu(In,Ga)Se2 thin film solar cells have been prepared using a rather complex growth process often referred to as three-stage or multistage. This family of processes is mainly characterized by a first step deposited with only In, Ga and Se flux to form a first layer. Cu is added in a second step until the film becomes slightly Cu-rich, where-after the film is converted to its final Cu-poor composition by a third stage, again with no or very little addition of Cu. In this paper, a comparison between solar cells prepared with the three-stage process and a one-stage/in-line process with the same composition, thickness, and solar cell stack is made. The one-stage process is easier to be used in an industrial scale and do not have Cu-rich transitions. The samples were analyzed using glow discharge optical emission spectroscopy, scanning electron microscopy, X-ray diffraction, current–voltage-temperature, capacitance-voltage, external quantum efficiency, transmission/reflection, and photoluminescence. It was concluded that in spite of differences in the texturing, morphology and Ga gradient, the electrical performance of the two types of samples is quite similar as demonstrated by the similar J–V behavior, quantum spectral response, and the estimated recombination losses.

Understanding how the brain works will require tools capable of measuring neuron elec-trical activity at a network scale. However, considerable progress is still necessary to reliably increase the number of neurons that are recorded and identified simultaneously with existing mi-croelectrode arrays. This project aims to evaluate how different materials can modify the effi-ciency of signal transfer from the neural tissue to the electrode. Therefore, various coating materials (gold, PEDOT, tungsten oxide and carbon nano-tubes) are characterized in terms of their underlying electrochemical processes and recording ef-ficacy. Iridium electrodes (177-706 μm2) are coated using galvanostatic deposition under different charge densities. By performing electrochemical impedance spectroscopy in phosphate buffered saline it is determined that the impedance modulus at 1 kHz depends on the coating material and decreased up to a maximum of two orders of magnitude for PEDOT (from 1 MΩ to 25 kΩ). The electrodes are furthermore characterized by cyclic voltammetry showing that charge storage capacity is im-proved by one order of magnitude reaching a maximum of 84.1 mC/cm2 for the PEDOT: gold nanoparticles composite (38 times the capacity of the pristine). Neural recording of spontaneous activity within the cortex was performed in anesthetized rodents to evaluate electrode coating performance.

The presence and importance of the sea as a factor that has helped shape the history of England since at least the Roman invasions of 55-54 BC (less successful, incidentally, than most of Caesar’s other military ventures ...) need no particular urging or demonstration. Nonetheless, a bird’s-eye view would necessarily survey the waves of invasions and settlements that, one after the other, came dashing over the centuries upon England’s shores; not to mention the requested invasion of 1688, Angles and Saxons, Scandinavians, Normans, they all crossed the whale’s path and cast anchor in England’s green and pleasant land. In the course of this retrospective voyage through the oceans of History, one would inevitably stop at the so-called ‘Discoveries’ of the 15th-16th centuries, meet their navigators, sailors and pirates extolled by Richard Hakluyt (1553?-1616), face an anonymous crowd of merchants and witness the huge expansion of trade, largely to the benefit of the ‘discovering’ countries as prescribed by the economic Gospel Adam Smith (1723-90) would later baptize as “mercantilism”.

Esta dissertação de mestrado, voltada para a área da Análise do Discurso, propõe abordar diferentes perspectivas de estudo acerca da argumentação. Tendo também como base teórica a Análise textual dos discursos proposta por Jean-Michel Adam, este trabalho procura analisar um corpus constituído de dois gêneros de texto – a entrevista política e o discurso de posse presidencial – em duas variedades de língua, nomeadamente o Português Brasileiro e o Português Europeu. O trabalho consiste em atestar alguns recursos verbais com finalidade persuasiva, bem como os elementos contextuais possivelmente verificáveis através dos enunciados produzidos pelas respectivas personagens políticas. Uma vez que os textos abordados serão descritos numa perspectiva linguístico-discursiva, pretende-se, ainda, verificar nas construções dos enunciados, a responsabilidade enunciativa (as vozes do texto), bem como a forma de argumentar retoricamente, construindo o que já na Antiguidade Clássica eram denominados de ethos e pathos.

Understanding how the brain works has been one of the greatest goals of mankind. This desire fuels the scientific community to pursue novel techniques able to acquire the complex information produced by the brain at any given moment. The Electrocorticography (ECoG) is one of those techniques. By placing conductive electrodes over the dura, or directly over the cortex, and measuring the electric potential variation, one can acquire information regarding the activation of those areas. In this work, transparent ECoGs, (TrECoGs) are fabricated through thin film deposition of the Transparent Conductive Oxides (TCOs) Indium-Zinc-Oxide (IZO) and Gallium-Zinc-Oxide (GZO). Five distinct devices have been fabricated via shadow masking and photolithography. The data acquired and presented in this work validates the TrECoGs fabricated as efficient devices for recording brain activity. The best results were obtained for the GZO- based TrECoG, which presented an average impedance of 36 kΩ at 1 kHz for 500 μm diameter electrodes, a transmittance close to 90% for the visible spectrum and a clear capability to detect brain signal variations. The IZO based devices also presented high transmittance levels (90%), but with higher impedances, which ranged from 40 kΩ to 100 kΩ.

Fully comprehending brain function, as the scale of neural networks, will only be possi-ble with the development of tools by micro and nanofabrication. Regarding specifically silicon microelectrodes arrays, a significant improvement in long-term performance of these implants is essential. This project aims to create a silicon microelectrode coating that provides high-quality electrical recordings, while limiting the inflammatory response of chronic implants. To this purpose, a combined chitosan and gold nanoparticles coating was produced allied with electrodes modification by electrodeposition with PEDOT/PSS in order to reduce the im-pedance at 1kHz. Using a dip-coating mechanism, the silicon probe was coated and then charac-terized both morphologically and electrochemically, with focus on the stability of post-surgery performance in anesthetized rodents. Since not only the inflammatory response analysis is vital, the electrodes recording degradation over time was also studied. The produced film presented a thickness of approximately 50 μm that led to an increase of impedance of less than 20 kΩ in average. On a 3 week chronic implant, the impedance in-crease on the coated probe was of 641 kΩ, compared with 2.4 MΩ obtained for the uncoated probe. The inflammatory response was also significantly reduced due to the biocompatible film as proved by histological tests.

The unique proprieties exhibited by nanoscale particles compared to their macro size counterparts allow for the creation of novel neural activity manipula-tion procedures. In this sense, gold nanoparticles (AuNPs) can be used to stimu-late the electrical activity of neuron by converting light into heat. During this dissertation, AuNPs are synthesized by the citrate reduction method, resulting in a hydrodynamic diameter of approximately 16 nm and an absorbance peak of 530 nm. A system to control a 532 nm laser and measure the temperature variation was custom built from scratch specifically for this project. Temperature is then measured with recourse to a thermocouple and through changes in impedance. The built system had in consideration the necessities pre-sented by in vivo tests. Trials were performed by measuring the temperature rise of colloidal AuNP solutions, having the temperature variation reached a maximum of ap-proximately 18 ºC relative to control trials; successfully showing that light is ef-fectively transduced into heat when AuNPs are present. This novel approach enables an alternative to optogenetics, which require the animal to be genetically modified in order to allow neuron stimulation.
