The objective of this work was to determine the transcript profile of tomato plants (Lycopersicon esculentum Mill.), during Fusarium oxysporum f. sp. lycopersici infection and after foliar application of salicylic acid. The suppression subtractive hybridization (SSH) technique was used to generate a cDNA library enriched for transcripts differentially expressed. A total of 307 clones was identified in two subtractive libraries, which allowed the isolation of several defense-related genes that play roles in different mechanisms of plant resistance to phytopathogens. Genes with unknown roles were also isolated from the two libraries, which indicates the possibility of identifying new genes not yet reported in studies of stress/defense response. The SSH technique is effective for identification of resistance genes activated by salicylic acid and F. oxysporum f. sp. lycopersici infection. Not only the application of this technique enables a cost effective isolation of differentially expressed sequences, but also it allows the identification of novel sequences in tomato from a relative small number of sequences.

The Watershed Improvement Board is an independent, self-governing body which awards grants for water quality improvement in the state. Eligible applicants include soil and water conservation districts, local watershed improvement committees, public water supply utilities, counties, county conservation boards and cities. These grants are funded by the Watershed Improvement Fund. Funding for these grants comes from annual appropriations and funds from the Animal Agriculture Compliance Fund Penalties. The Board awarded ten grants totalling $2,307,554 this year. In addition to providing environmental benefits, these implementation projects help stimulate economic activity and create jobs through the purchase of local goods and services. Additional grants will be awarded this spring.

BACKGROUND: Synthesis of the Staphylococcus aureus peptidoglycan pentaglycine interpeptide bridge is catalyzed by the nonribosomal peptidyl transferases FemX, FemA and FemB. Inactivation of the femAB operon reduces the interpeptide to a monoglycine, leading to a poorly crosslinked peptidoglycan. femAB mutants show a reduced growth rate and are hypersusceptible to virtually all antibiotics, including methicillin, making FemAB a potential target to restore beta-lactam susceptibility in methicillin-resistant S. aureus (MRSA). Cis-complementation with wild type femAB only restores synthesis of the pentaglycine interpeptide and methicillin resistance, but the growth rate remains low. This study characterizes the adaptations that ensured survival of the cells after femAB inactivation. RESULTS: In addition to slow growth, the cis-complemented femAB mutant showed temperature sensitivity and a higher methicillin resistance than the wild type. Transcriptional profiling paired with reporter metabolite analysis revealed multiple changes in the global transcriptome. A number of transporters for sugars, glycerol, and glycine betaine, some of which could serve as osmoprotectants, were upregulated. Striking differences were found in the transcription of several genes involved in nitrogen metabolism and the arginine-deiminase pathway, an alternative for ATP production. In addition, microarray data indicated enhanced expression of virulence factors that correlated with premature expression of the global regulators sae, sarA, and agr. CONCLUSION: Survival under conditions preventing normal cell wall formation triggered complex adaptations that incurred a fitness cost, showing the remarkable flexibility of S. aureus to circumvent cell wall damage. Potential FemAB inhibitors would have to be used in combination with other antibiotics to prevent selection of resistant survivors.

Pursuant to Iowa Code section 307.20, the biodiesel fuel revolving fund (Fund) was created and is to be used to purchase biodiesel fuel for use in the Department of Transportation’s vehicles. The act directed that the Fund receive money from the sale of EPAct credits banked by the DOT on the effective date of the act, monies appropriated by the General Assembly, and any other monies obtained or accepted by the DOT for deposit in the Fund. This report is of the expenditures made from the Fund during FY 2015.

Introduction: Evidence suggests that citrullinated fibrin(ogen) may be a potential in vivo target of anticitrullinated protein/peptide antibodies (ACPA) in rheumatoid arthritis (RA). We compared the diagnostic yield of three enzyme-linked immunosorbent assay (ELISA) tests by using chimeric fibrin/filaggrin citrullinated synthetic peptides (CFFCP1, CFFCP2, CFFCP3) with a commercial CCP2-based test in RA and analyzed their prognostic values in early RA. Methods: Samples from 307 blood donors and patients with RA (322), psoriatic arthritis (133), systemic lupus erythematosus (119), and hepatitis C infection (84) were assayed by using CFFCP- and CCP2-based tests. Autoantibodies also were analyzed at baseline and during a 2-year follow-up in 98 early RA patients to determine their prognostic value. Results: With cutoffs giving 98% specificity for RA versus blood donors, the sensitivity was 72.1% for CFFCP1, 78.0% for CFFCP2, 71.4% for CFFCP3, and 73.9% for CCP2, with positive predictive values greater than 97% in all cases. CFFCP sensitivity in RA increased to 80.4% without losing specificity when positivity was considered as any positive anti-CFFCP status. Specificity of the three CFFCP tests versus other rheumatic populations was high (> 90%) and similar to those for the CCP2. In early RA, CFFCP1 best identified patients with a poor radiographic outcome. Radiographic progression was faster in the small subgroup of CCP2-negative and CFFCP1-positive patients than in those negative for both autoantibodies. CFFCP antibodies decreased after 1 year, but without any correlation with changes in disease activity. Conclusions: CFFCP-based assays are highly sensitive and specific for RA. Early RA patients with anti-CFFCP1 antibodies, including CCP2-negative patients, show greater radiographic progression.

Systemic administration of cyclosporine A (CsA) is commonly used in the treatment of local ophthalmic conditions involving cytokines, such as corneal graft rejection, autoimmune uveitis and dry eye syndrome. Local administration is expected to avoid the various side effects associated with systemic delivery. However, the currently available systems using oils to deliver CsA topically are poorly tolerated and provide a low bioavailability. These difficulties may be overcome through formulations aimed at improving CsA water solubility (e.g. cyclodextrins), or those designed to facilitate tissue drug penetration using penetration enhancers. The use of colloidal carriers (micelles, emulsions, liposomes and nanoparticles) as well as the approach using hydrosoluble prodrugs of CsA have shown promising results. Solid devices such as shields and particles of collagen have been investigated to enhance retention time on the eye surface. Some of these topical formulations have shown efficacy in the treatment of extraocular diseases but were inefficient at reaching intraocular targets. Microspheres, implants and liposomes have been developed to be directly administered subconjunctivally or intravitreally in order to enhance CsA concentration in the vitreous. Although progress has been made, there is still room for improvement in CsA ocular application, as none of these formulations is ideal.

O objetivo deste trabalho foi avaliar o progresso da ferrugem asiática (Phakopsora pachyrhizi) em genótipos de soja destinados à produção de silagem, sob condições naturais de infecção. Avaliou-se a severidade da ferrugem ao longo do tempo e a capacidade combinatória de cinco cultivares e 20 progênies F3 de soja. A maior área abaixo da curva de progresso da ferrugem (AACPF) foi de 2.116, para as progênies originadas do cruzamento entre as cultivares UFVS 2003 x UFV16, e a menor de 1.307, para UFV16 x Tucunaré. Houve efeito significativo das capacidades geral e específica de combinação, para a AACPF entre os genitores e as progênies, bem como para os cruzamentos recíprocos. Esses resultados indicam que a resistência à ferrugem asiática da soja pode ser influenciada por componentes maternos.

[spa]La costumbre de enrolar a combatientes del anfiteatro (arenarii) como guardia personal en el marco de luchas faccionales sobrevivió durante el siglo IV. Incluso si en la mayor parte de casos este fenómeno se explica por razones políticas, conocemos dos ejemplos en los que los candidatos a obispo recurrieron a gladiadores para triunfar sobre sus oponentes. El primer ejemplo está relacionado con la elección de Silvano (en el 305 o en el 307/308), quien llegó a hacerse elegir obispo de Cirta (actual Constantine)gracias a la ayuda del arenarius Muto. Esta ayuda quedó bien demostrada por el testimonio de diversas personas durante el proceso seguido contra Silvano en el 320, en el que se reconoció su culpabilidad y se le condenó al exilio. El segundo ejemplo es el del papa Dámaso (366), elegido obispo de Roma en la iglesia de Lucina el mismo día en el que el diácono Ursino era escogido en la basílica de Julio. En el conflicto siguiente, Dámaso convocó a arenarii, quadrigari y fossores, quienes acudieron en su ayuda fuertemente armados. Éstos atacaron la basílica en la que se habían refugiado los partidarios de Ursino y el resultado del asalto fue de 137 muertos. [eng]The habit of engaging fighters of the amphitheatre (arenarii) as a personal guard in the factional struggles survived during the 4th century. Although in most cases it was about political causes, we know two examples where the candidates to be bishops turned to gladiators in order to overcome their opponents. The first example is that of election of Silvanus (305 or 307-308), who got the bishopric of Cirta (the modern Constantine) thanks to the help of the arenarius Mutus. This help was proved by the testimony of several witnesses during the trial which was followed against Silvanus in 320, where his guilt was admitted and he was condemned to the exile. The second exemple is that of the the Pope Damasus (366), elected bishop of Rome in the church of Lucina in the same day as the deacon Ursinus was elected in the basilica of Julius. In the conflict that was to follow, Damasus called arenarii, quadrigarii and fossores who came to help him heavily armed. They attacked the basilica where the supporters of Ursinus had taken refuge and as a result of the assault 137 people died.

Introduction et But de l'étude. - La plupart des études d'évaluation de la nutrition en réanimation concernent les patients ventilés bénéficiant d'un support nutritionnel artificiel. La nutrition per os qui concerne les patients les moins graves ou sortis de la phase critique est peu étudiée. Cette étude observationnelle, menée dans le cadre d'un travail de Bachelor en collaboration avec les Hôpitaux Universitaires de Genève, a pour objectif de réaliser un état des lieux des pratiques nutritionnelles actuelles, tous patients confondus, dans un service de réanimation médico-chirurgicale, et de mesurer la couverture des besoins énergétiques en fonction du type de nutrition administré ou consommé. Matériel et Méthodes. - Inclusion de tous les patients adultes hospitalisés pour plus de 24 h en réanimation durant 6 semaines consécutives, suivis jusqu'à la sortie du service mais pendant 14 jours au maximum. Les apports caloriques entéraux et parentéraux ont été récoltés dans le dossier patient informatisé. Les apports per os ont été estimés sur la base de la composition nutritionnelle standard des repas servis et le report infirmier des quantités consommées. Les suppléments nutritifs oraux ont été inclus dans les apports per os. Les solutés glucosés et les dilutions de médicaments n'ont pas été comptabilisés. La cible calorique a été fixée à 30 kcal/kg pour les hommes et 25 kcal/kg pour les femmes, sur la base du poids mesuré ou anamnestique ou sur le poids correspondant à un BMI de 22,5 en cas d'obésité. Résultats. - Six cent quatre-vingt douze journées-patients ont été analysées, issues de 114 patients dont le score SAPS II moyen est de 43. Toutes journées confondues (sans tenir compte du début ou fin de séjour) : 44 % (n = 307) des journées sont sous NE exclusive, 9 % (n = 64) sous NP exclusive et 7 % (n = 47) combinent ces deux types d'apports. La nutrition per os exclusive représente 17 % (n = 116) des journées et 23 % (n = 158) ne comportent aucun apport nutritionnel. Lorsque l'on s'intéresse au pour centage d'atteinte de la cible calorique selon le type de nutrition, il se répartit comme tel (médiane ± espace interquartile) : NE : 83 ± 44 % ; NP : 84 ± 75 % ; combiné : 95 ± 39 % ; per os : 28 ± 33 %. La nutrition per os se retrouve en fin de séjour mais pas uniquement puisque seules un peu plus de la moitié des journées de nutrition per os ont lieu lors du dernier (35 %) ou de l'avant dernier (28 %) jour du séjour. L'atteinte de la cible calorique par la nutrition per os exclusive évolue peu au long du séjour. Conclusion. - La nutrition per os représente le mode d'alimentation d'un nombre important de journées dans cette observation. La très faible couverture des besoins caloriques lors de ces journées, tout au long du séjour, classe ces patients dans une catégorie à risque nutritionnel important. Une évaluation plus précise des apports protéino-énergétique devrait être réalisée afin de quantifier plus exactement les déficits et un suivi de l'évolution de ces patients apporterait un éclairage important.

Late Variscan volcanic activity is documented in the Late Carboniferous Salvan-Dorenaz sedimentary basin and in the neighboring basement units of the Aiguilles-Rouges and Mont-Blanc crystalline massifs (Western Alps). Precise U/Pb isotopic dating, zircon morphology and geochemical analyses indicate that volcanism occurred during short-lived pulses and that coexisting crustal and mantle sources were involved in the production of melts. Volcanic and subvolcanic products were emplaced along major N-S to NNE-SSW transtensional fracture zones, similar to the ones that governed intense basement exhumation and that favored the formation and filling of the Late Carboniferous Salvan-Dorenaz continental basin. In the Aiguilles-Rouges massif, dacitic flows outcropping at the base of the Salvan-Dorenaz basin erupted at 308 +/- 3 Ma; they represent the surface equivalent of the nearby Vallorcine peraluminous granite and associated rhyolitic dykes (311 +/- 17 Ma). In the Mont Blanc massif, calc-alkaline rhyolitic dykes were emplaced simultaneously (307 +/- 2 Ma) at shallow crustal levels, but they derive from deeper magma sources denoting enhanced mantellic activity. Recently identified tuffs and volcaniclastic layers embedded at different levels of the Salvan-Dorenaz stratigraphic record testify a 295 +3/-4 Ma old episode of highly explosive volcanism from distant volcanic centers, possibly located in the Aar-Gotthard massifs (Central Alps). Their zircon typology is highly heterogeneous. documenting wall-rock contamination of the melts and/or admixture of crustal sediments, whereas consistent subpopulations point to high-temperature magmas of deep-seated origin and alkaline affinity. The dated volcanic layers from the Salvan-Dorenaz basin set the beginning of the detrital sedimentation at 308 +/- 3 Ma and constrain the deposition of 1.5-1.7 km thick of elastic sediments within a time span of 10-15 Ma. These results infer minimum, long-term subsidence rates during basin evolution in the order of >0.1 mm/a, while in the surrounding basement units estimated exhumation rates are in the range of 1 mm/a. All dated rocks contain inherited zircon populations about 350, 450 or 600 Ma old.

Background. The time passed since the infection of a human immunodeficiency virus (HIV)-infected individual (the age of infection) is an important but often only poorly known quantity. We assessed whether the fraction of ambiguous nucleotides obtained from bulk sequencing as done for genotypic resistance testing can serve as a proxy of this parameter. Methods. We correlated the age of infection and the fraction of ambiguous nucleotides in partial pol sequences of HIV-1 sampled before initiation of antiretroviral therapy (ART). Three groups of Swiss HIV Cohort Study participants were analyzed, for whom the age of infection was estimated on the basis of Bayesian back calculation (n = 3,307), seroconversion (n = 366), or diagnoses of primary HIV infection (n = 130). In addition, we studied 124 patients for whom longitudinal genotypic resistance testing was performed while they were still ART-naive. Results. We found that the fraction of ambiguous nucleotides increased with the age of infection with a rate of .2% per year within the first 8 years but thereafter with a decreasing rate. We show that this pattern is consistent with population-genetic models for realistic parameters. Finally, we show that, in this highly representative population, a fraction of ambiguous nucleotides of >.5% provides strong evidence against a recent infection event < 1 year prior to sampling (negative predictive value, 98.7%). Conclusions. These findings show that the fraction of ambiguous nucleotides is a useful marker for the age of infection.

Surgery is the cornerstone of ovarian cancer treatment and maximal cytoreduction is important. In the early 1980’s primary surgical treatment of ovarian cancer was performed in over 80 hospitals in Finland. The significance of the operative volume of the hospital, of the training of the surgeons and of centralization of surgical treatment has been widely discussed. The aim of the present study was to evaluate the outcome of surgical treatment of ovarian cancer in different hospital categories retrospectively and prospectively, and to analyze if any differences are reflected in survival. The retrospective study included 3851 ovarian cancer patients operated between 1983 and 1994 in Finland. The data was analyzed according to hospital category (university, central, and other) and by quartiles of the hospital operative volume. The results showed that patients operated in the highest operative volume hospitals had the best relative survival. When stratifying the analysis by the period of diagnosis (1983-1988 and 1989-1994), the university hospitals improved their performance the most. The prospective part of the thesis was initiated in 1999 and included 307 patients with invasive ovarian cancer and 65 patients with an ovarian borderline tumor. The baseline and 5-year surveys used a questionnaire that was filled in by the operating surgeons. For analysis of the 5-year followup data, the hospitals were divided into three categories (<10, 10-20, or >20 patients operated in 1999). The effect of the surgical volume was analyzed also as a continuous variable (1-47 operations per year). In university hospitals, pelvic lymphadenectomy was performed in 88 %, and para-aortic lymphadenectomy in 73 %, of the patients with stage I disease. The corresponding figures ranged from 11 % to 21 % in the other hospitals. For stage III ovarian cancer patients operated by gynecological oncologists, the estimated odds ratio for no macroscopic residual tumor was 3.0 times higher (95 % CI 1.2-7.5) than for those operated by general gynecologists. In the university and other hospitals 82% of the patients received platinum-based chemotherapy. Platinum + taxane combination was given to 63 % of the patients in the university and in 49 % in the other hospitals (p = 0.0763). Only a minority of the patients with tumors of borderline malignancy were staged according to recommendations, most often multiple peritoneal biopsies and omentectomy were neglected. FIGO stage, patient age, and residual tumor were independent prognostic factors of cancer-specific 5-year survival. A higher hospital operative volume was also a significant prognostic factor for better cancer-specific survival (p = 0.036) and disease-free survival (p = 0.048). In conclusion, ovarian cancer patients operated in high-volume university hospitals were more often optimally debulked and had a significantly better cancer-specific survival than patients operated in other hospitals. These results favor centralization of primary surgical treatment of ovarian cancer.
