Drastic biodiversity declines have raised concerns about the deterioration of ecosystem functions and have motivated much recent research on the relationship between species diversity and ecosystem functioning. A functional trait framework has been proposed to improve the mechanistic understanding of this relationship, but this has rarely been tested for organisms other than plants. We analysed eight datasets, including five animal groups, to examine how well a trait-based approach, compared with a more traditional taxonomic approach, predicts seven ecosystem functions below- and above-ground. Trait-based indices consistently provided greater explanatory power than species richness or abundance. The frequency distributions of single or multiple traits in the community were the best predictors of ecosystem functioning. This implies that the ecosystem functions we investigated were underpinned by the combination of trait identities (i.e. single-trait indices) and trait complementarity (i.e. multi-trait indices) in the communities. Our study provides new insights into the general mechanisms that link biodiversity to ecosystem functioning in natural animal communities and suggests that the observed responses were due to the identity and dominance patterns of the trait composition rather than the number or abundance of species per se.

In mammalian cells, fusion between early endocytic vesicles has been shown to require the ubiquitous intracellular fusion factors N-ethylmaleimide-sensitive factor (NSF) and alpha-SNAP, as well as a factor specific for early endosomes, the small GTPase Rab5 [1-3]. We have previously demonstrated an additional requirement for phosphatidylinositol 3-kinase (PI 3-kinase) activity [4]. The membrane association of early endosomal antigen 1 (EEA1), a specific marker of early endosomes [5,6], has recently been shown to be similarly dependent on PI 3-kinase activity [7], and we therefore postulated that it might be involved in endosome fusion. Here, we present evidence that EEA1 has an important role in determining the efficiency of endosome fusion in vitro. Both the carboxy-terminal domain of EEA1 (residues 1098-1411) and specific antibodies against EEA1 inhibited endosome fusion when included in an in vitro assay. Furthermore, depletion of EEA1, both from the membrane fraction used in the assay by washing with salt and from the cytosol using an EEA1-specific antibody, resulted in inhibition of endosome fusion. The involvement of EEA1 in endosome fusion accounts for the sensitivity of the endosome fusion assay to inhibitors of PI 3-kinase.

Inbreeding depression is most pronounced for traits closely associated with fitness. The traditional explanation is that natural selection eliminates deleterious mutations with additive or dominant effects more effectively than recessive mutations, leading to directional dominance for traits subject to strong directional selection. Here we report the unexpected finding that, in the butterfly Bicyclus anynana, male sterility contributes disproportionately to inbreeding depression for fitness (complete sterility in about half the sons from brother-sister matings), while female fertility is insensitive to inbreeding. The contrast between the sexes for functionally equivalent traits is inconsistent with standard selection arguments, and suggests that trait-specific developmental properties and cryptic selection play crucial roles in shaping genetic architecture. There is evidence that spermatogenesis is less developmentally stable than oogenesis, though the unusually high male fertility load in B. anynana additionally suggests the operation of complex selection maintaining male sterility recessives. Analysis of the precise causes of inbreeding depression will be needed to generate a model that reliably explains variation in directional dominance and reconciles the gap between observed and expected genetic loads carried by populations. This challenging evolutionary puzzle should stimulate work on the occurrence and causes of sex differences in fertility load.

Rational catalyst design is one of the most fundamental goals in heterogeneous catalysis. Herein, we briefly review our previous design work, and then introduce a general optimization framework, which converts catalyst design into an optimization problem. Furthermore, an example is given using the gradient ascent method to show how this framework can be used for rational catalyst design. This framework may be applied to other design schemes.

This study examines the potential of next-generation sequencing based ‘genotyping-by-sequencing’ (GBS) of microsatellite loci for rapid and cost-effective genotyping in large-scale population genetic studies. The recovery of individual genotypes from large sequence pools was achieved by PCR-incorporated combinatorial barcoding using universal primers. Three experimental conditions were employed to explore the possibility of using this approach with existing and novel multiplex marker panels and weighted amplicon mixture. The GBS approach was validated against microsatellite data generated by capillary electrophoresis. GBS allows access to the underlying nucleotide sequences that can reveal homoplasy, even in large datasets and facilitates cross laboratory transfer. GBS of microsatellites, using individual combinatorial barcoding, is potentially faster and cheaper than current microsatellite approaches and offers better and more data.

The author states his opinion that the most valuable contribution of the July 2007 issue is Gene Amdahl's new paper, which he thinks will probably become as popular as the Amdahl paper reprinted in the same issue since it provides much needed clarifications, and most notably includes the original 'law' formula as presented by Amdahl four decades ago.

Summary form only given, as follows. In Vol. 12, no. 3 (Summer 2007), page 9, bottom of the left column, in 'Computer Architecture and Amdahl??s Law' by Gene Amdahl, the claim about invalidating Amdahl??s Law in 1988 came from a team at Sandia National Laboratories, and not Los Alamos. The correct text should read: "Several years later I was informed of a proof that Amdahl's Law was invalidated by someone at Sandia National Laboratories, where a number of computers interconnected as an Ncube by communication lines, but with each computer also connected to I/O devices for loading the operating system, initial data, and results." On page 20 of the same issue, in the second sentence of the diagram explanation note by Justin Rattner, the percentage figures for the sequential and the system coordination parts of the workload were interchanged. The correct version of this sentence should read: "assuming a fixed sized problem, Amdahl speculated that most programs would require at least 10% of the computation to be sequential (only one instruction executing at a time), with overhead due to interprocessor coordination averaging 25%."

A genetic algorithm used to design radio-frequency binary-weighted differential switched capacitor arrays (RFDSCAs) is presented in this article. The algorithm provides a set of circuits all having the same maximum performance. This article also describes the design, implementation, and measurements results of a 0.25 lm BiCMOS 3-bit RFDSCA. The experimental results show that the circuit presents the expected performance up to 40 GHz. The similarity between the evolutionary solutions, circuit simulations, and measured results indicates that the genetic synthesis method is a very useful tool for designing optimum performance RFDSCAs.

Wireless networks have joined to the sports venues, offering to the public a set of facilities, such as the access to email, news, and also to use the social networking, uploading their photos. New challenges have emerged to provide Wi-Fi in this densely populated stadiums, such as increasing capacity and coverage. In this article, an access point antenna array to cover a sector of a stadium is presented. Its structure, designed in a low cost material allows to reduce the total manufacturing costs, an important factor due to the large number of antennas required in these venues. The material characteristic, the broad bandwidth of operation (300 MHz), along with to the low side lobe levels, important to reduce interference between sectors, makes this antenna well-positioned for wireless communications in these particular locals. Introdução: A gravidez e a prática da atividade física (AF) são temas ainda pouco abordados na nossa sociedade. Muitas mulheres sofrem de ansiedade no período pré-natal e não conhecem os benefícios que a prática da AF tem sobre a mesma. Estudos demonstram que a AF está associada à diminuição da ansiedade, contudo poucos avaliam este efeito na gravidez. Objetivo(s): Os objetivos deste estudo foram classificar o nível de AF das grávidas; avaliar os sintomas de ansiedade e analisar a associação entre os níveis de ansiedade e a AF das gestantes. Métodos: Trata-se de um estudo longitudinal prospetivo, realizado numa amostra de 56 grávidas com idades entre os 18 e os 37 anos (29,14 ±4,641 anos). As participantes foram avaliadas em dois momentos, T1 (10-12 semanas gestacionais) e T2 (20-22 semanas gestacionais), tendo sido mensurada a AF com acelerómetros GTX3 e a ansiedade com a escala de Zung. Resultados: A média da AF leve e moderada não apresentou diferenças entre os dois momentos de avaliação; contudo, verificou-se uma tendência para a AF leve diminuir do primeiro para o segundo trimestre (1283,88 ± 530,37 vs 1098,16 ± 489,72 counts/min total semana; p=0,459) e a AF moderada aumentar (2470,62 ± 404,50 vs 2528,55 ± 493,05 counts/min total semana; p=0,459). A maioria das grávidas não cumpriu as recomendações da ACSM nos dois momentos de avaliação (T1: 72,1% e T2: 73,2%) (p=0,06). Não se verificaram diferenças no estado de ansiedade das grávidas de T1 para T2. 10,7% das grávidas apresentaram ansiedade em T1 e 7,1% em T2 (p=0,063). Cumprir ou não as recomendações da ACSM não esteve associado ao estado de ansiedade. Conclusão: Neste estudo verificámos que a maioria das gravidas não cumpre as recomendações da AF segundo ACSM, os níveis de ansiedade não diferiram ao longo da gravidez, verificámos ainda que o cumprimento das recomendações da AF não está associado aos níveis de ansiedade.

One of the major factors threatening chimpanzees (Pan troglodytes verus) in Guinea-Bissau is habitat fragmentation. Such fragmentation may cause changes in symbiont dynamics resulting in increased susceptibility to infection, changes in host specificity and virulence. We monitored gastrointestinal symbiotic fauna of three chimpanzee subpopulations living within Cantanhez National Park (CNP) in Guinea Bissau in the areas with different levels of anthropogenic fragmentation. Using standard coproscopical methods (merthiolate-iodine formalin concentration and Sheather's flotation) we examined 102 fecal samples and identified at least 13 different symbiotic genera (Troglodytella abrassarti, Troglocorys cava, Blastocystis spp., Entamoeba spp., Iodamoeba butschlii, Giardia intestinalis, Chilomastix mesnili, Bertiella sp., Probstmayria gombensis, unidentified strongylids, Strongyloides stercoralis, Strongyloides fuelleborni, and Trichuris sp.). The symbiotic fauna of the CNP chimpanzees is comparable to that reported for other wild chimpanzee populations, although CNP chimpanzees have a higher prevalence of Trichuris sp. Symbiont richness was higher in chimpanzee subpopulations living in fragmented forests compared to the community inhabiting continuous forest area. We reported significantly higher prevalence of G. intestinalis in chimpanzees from fragmented areas, which could be attributed to increased contact with humans and livestock.

In mammalian circadian clockwork, the CLOCK-BMAL1 complex binds to DNA enhancers of target genes and drives circadian oscillation of transcription. Here we identified 7,978 CLOCK-binding sites in mouse liver by chromatin immunoprecipitation-sequencing (ChIP-Seq), and a newly developed bioinformatics method, motif centrality analysis of ChIP-Seq (MOCCS), revealed a genome-wide distribution of previously unappreciated noncanonical E-boxes targeted by CLOCK. In vitro promoter assays showed that CACGNG, CACGTT, and CATG(T/C)G are functional CLOCK-binding motifs. Furthermore, we extensively revealed rhythmically expressed genes by poly(A)-tailed RNA-Seq and identified 1,629 CLOCK target genes within 11,926 genes expressed in the liver. Our analysis also revealed rhythmically expressed genes that have no apparent CLOCK-binding site, indicating the importance of indirect transcriptional and posttranscriptional regulations. Indirect transcriptional regulation is represented by rhythmic expression of CLOCK-regulated transcription factors, such as Krüppel-like factors (KLFs). Indirect posttranscriptional regulation involves rhythmic microRNAs that were identified by small-RNA-Seq. Collectively, CLOCK-dependent direct transactivation through multiple E-boxes and indirect regulations polyphonically orchestrate dynamic circadian outputs.
