The FK is a two-stage optical concentrator for CPV, composed by a Fresnel lens working as POE and a refractive element working as SOE. Both elements perform Köhler integration, for uniform irradiance purposes. The FK has demonstrated that compares very well with other Fresnel-based concentrator optics. Recent on-sun measurements carried out on an FK mono-module prototype have already shown outstanding results, achieving electrical efficiencies over 34%. Further optimization of optical design together with application of AR coating on SOE will predictably lead to efficiencies over 35%.

---ABSTRACT---Unravel how the brain works is one of the main challenges faced by current science. A field of study which has aroused great expectations and interest is the analysis of the cortical structure from a morphological point of view, so that a molecular level simulation of the brain is achieved. This is expected to deepen the study of many neurological and pathological diseases. This project seeks the study of the soma and spines from the theoretical neuromorphology point of view. In the state of the art it is common that when it comes to analyze the morphological characteristics of a three dimension neuron the soma is ignored or, in the best case, it is replaced by a simple sphere. In fact, the concept of soma is abstract because there is not a robust and strict definition on exactly where it ends and dendrites begin. In this project a mathematical definition is reached for the first time to determine what a soma is. With the aim to simulate somas the atributes applied in the state of the art are studied. These properties, generic in nature, do not specify a unique morphology. It is why it was proposed a method to group local and global morphology properties. In arrangement of the characteristics it was proceed with the categorization of the celular body into diferent classes by using a new subtype of dynamic Bayesian network adapted to space. From the result the existance of different classes of somas and diferences among pyramidal somas from distinct brain layers are discovered. From the mathematical model virtual somas were simulated for the first time. Some morphologies of spines have been attributed to certain cognitive behaviours. For this reason it is interesting to rule the existent classes and to relate them with their functions in the brain activity. The most extended classification (Peters y Kaiserman-Abramof, 1970) presents an ambiguous and subjective definition that relies on the interpretation of each individual and consequently it is arguable. This study was based on the set of descriptors extracted from a local topological analysis technique for 3D representations. On these data it was tried to reach the most suitable set of classes to group the spines as well as to describe each cluster by unambiguous rules. From these results, the existance of a continuum of spines and the properties that characterize each spine subtype were discussed .

This paper explores the possibility of using the Moire-Fourier deflectometry for measuring the local heat transfer coefficient inside small confined flows (micro-channels) and their relevance for checking theoretical models. This optical technique, supplemented with a digital image processing method of fringes, is applied for studying the local heat transfer over a backward facing step. The experimental results are compared with numerical results obtained from a commercial code, which has been contrasted with relevant solutions from the literature and bulk fluid temperature measurements at the inlet and outlet sections. In order to show the possibilities of the experimental technique, the influence of assuming an adiabatic wall on the numerical heat-transfer model is examined and the degree of agreement is discussed. As a result, the paper shows that the proposed Moiré-Fourier technique is a simple experimental setup suitable for temperature measurements with an accuracy similar to the thermocouples but with a spatial resolution near 0.01 mm.Moiré-Fourier deflectometry for local heat transfer measurement over a backward-facing step

ABSTRACT. Aiming to strengthen knowledge in Telecommunications Systems, is intended to design a network for advanced services broadcasting, including digital TV, broadband internet and telephony, by using FTTH technology. This network will be entirely passive optic, and high bandwidth, PON type (Passive Optical Network) so that the transmitter beam is broadcasting to multiple optical fibers, branching out in different ways or joining them in the opposite. First, it is necessary to perform a theoretical study on the characteristics of the optical fiber, along with the properties of the active and passive elements that interact with it, to understand the technology that is based and offer a final solution according needs that arise. After an overview of the optical fiber technology, we proceed to study the characteristics and topologies access networks based on that, together with the requirements to face the network design and project management. System objectives will be determined, since the complexity of the network depends on factors such as the size and scope of it, or bandwidth. Later, the network will be designing in downstream architecture to deliver a single optical fiber to each household within the network area. All necessary specifications to define the network (power, modulation and fiber types) will be determined, as instruments and other means to operate it. Although FTTH network design includes many more aspects of those studied in this project, it was taken as objective the network design and planning corresponding to a section of a sparsely populated area where the facility was taken as feasible thus obviating small details and focus on the deployment of optical fiber. This is intended to provide a solution of household telecommunications infrastructure suitable for future needs involving the growing demand for services such as broadband internet or processing storage and cloud applications. The optical fiber has much to offer to telecommunications, and it is increasingly common to find fiber optic networks as part of a telecommunications service, both to individuals and businesses.

Workflows are increasingly used to manage and share scientific computations and methods. Workflow tools can be used to design, validate, execute and visualize scientific workflows and their execution results. Other tools manage workflow libraries or mine their contents. There has been a lot of recent work on workflow system integration as well as common workflow interlinguas, but the interoperability among workflow systems remains a challenge. Ideally, these tools would form a workflow ecosystem such that it should be possible to create a workflow with a tool, execute it with another, visualize it with another, and use yet another tool to mine a repository of such workflows or their executions. In this paper, we describe our approach to create a workflow ecosystem through the use of standard models for provenance (OPM and W3C PROV) and extensions (P-PLAN and OPMW) to represent workflows. The ecosystem integrates different workflow tools with diverse functions (workflow generation, execution, browsing, mining, and visualization) created by a variety of research groups. This is, to our knowledge, the first time that such a variety of workflow systems and functions are integrated.

ABSTRACT The progressive internationalization of Spanish universities has transformed these organizations into plurilingual scenarios. Spanish lives in them sharing the stage with other languages, especially English, as a means of access and transmission of expert knowledge. This requires a process of academic literacy in foreign language that host universities should assume, in order to safeguard students from failures in international programs. At the moment, in Spain, undergraduate or graduate programs do not set filters with minimum language requirements to gain access, except for some universities that merely require certificates of general Spanish. There are no Spanish language public admission exams, or post-enrollment tests of their own, to assess the communicative language ability of foreign students in academic contexts. In this dissertation, we start from the hypothesis that those tests that certify the student degree of mastery of the Spanish language do not serve the purpose of discriminating against students capable of successfully pursuing university programs. To prove it, a specific test that measures the ability to use Spanish in academic contexts was developed. This language test focused on the tasks associated with the most common genre, which revealed prominent in a first exploratory phase of the investigation, related to the use of Spanish as a means of instruction: university lectures. Once piloted, the test was administered along with others designed to assess the degree of mastery of the language in general contexts. Contrast results of these measurements and various analyzes of the data showed evidence that this type of test measures a specific construct: the ability to use Spanish in academic contexts.

ABSTRACT This study, which lies within the field of operations research in airports, considers the optimisation of the sequence for clearing snow from stretches of the manoeuvring area of an airport, known as RM-AM. This issue involves the optimisation of limited resources to remove snow from taxiways and runways thereby leaving them in an acceptable condition for operating aircraft. The airfield is divided into subsets of significant stretches for the purpose of operations and target times are established during which these are open to aircraft traffic. The study contains several mathematical models each with different functions, such as the end time of the process, the sum of the end times of each stretch, and gap between the estimated and the real end times. During this process, we introduce different operating restrictions on partial fulfilment of the operational targets as applied to zones of special interest, or relating to the operation of the snow-clearing machines. The problem is solved by optimisation based on linear programming. Computational tests are carried out on five distinct models of the manoeuvring area, which cover increasingly complex situations and larger areas. The different algorithms are then compared to one other. Having defined the mathematical model for the optimisation, we then set out a structured methodology to deal with any type of manoeuvring area. In other words, we define an operational strategy. The airfield is divided into subsets of significant stretches for the purpose of operations and target times are set at which these are to be open to aircraft traffic. The manoeuvring area is also divided into zones, with the condition that the subsets of significant stretches lie within just one of these zones. The problem is solved by an iterative optimisation process based on linear programming applied successively to the zones that make up the manoeuvring area during each iteration. The impact of the results on DMAN, AMAN and TP processes is analysed for their integration into the calculation of TSAT and EBIT. The method is particularized for the case of the manoeuvring area of Adolfo Suarez Madrid - Barajas Airport.

The study of development has relied primarily on the isolation of mutations in genes with specific functions in development and on the comparison of their expression patterns in normal and mutant phenotypes. Comparative evolutionary analyses can complement these approaches. Phylogenetic analyses of Sonic hedgehog (Shh) and Hoxd-10 genes from 18 cyprinid fish species closely related to the zebrafish provide novel insights into the functional constraints acting on Shh. Our results confirm and extend those gained from expression and crystalline structure analyses of this gene. Unexpectedly, exon 1 of Shh is found to be almost invariant even in third codon positions among these morphologically divergent species suggesting that this exon encodes for a functionally important domain of the hedgehog protein. This is surprising because the main functional domain of Shh had been thought to be that encoded by exon 2. Comparisons of Shh and Hoxd-10 gene sequences and of resulting gene trees document higher evolutionary constraints on the former than on the latter. This might be indicative of more general evolutionary patterns in networks of developmental regulatory genes interacting in a hierarchical fashion. The presence of four members of the hedgehog gene family in cyprinid fishes was documented and their homologies to known hedgehog genes in other vertebrates were established.

Like most proteins, complex RNA molecules often are modular objects made up of distinct structural and functional domains. The component domains of a protein can associate in alternative combinations to form molecules with different functions. These observations raise the possibility that complex RNAs also can be assembled from preexisting structural and functional domains. To test this hypothesis, an in vitro evolution procedure was used to isolate a previously undescribed class of complex ligase ribozymes, starting from a pool of 1016 different RNA molecules that contained a constant region derived from a large structural domain that occurs within self-splicing group I ribozymes. Attached to this constant region were three hypervariable regions, totaling 85 nucleotides, that gave rise to the catalytic motif within the evolved catalysts. The ligase ribozymes catalyze formation of a 3′,5′-phosphodiester linkage between adjacent template-bound oligonucleotides, one bearing a 3′ hydroxyl and the other a 5′ triphosphate. Ligation occurs in the context of a Watson–Crick duplex, with a catalytic rate of 0.26 min−1 under optimal conditions. The constant region is essential for catalytic activity and appears to retain the tertiary structure of the group I ribozyme. This work demonstrates that complex RNA molecules, like their protein counterparts, can share common structural domains while exhibiting distinct catalytic functions.

The Cdc7p protein kinase is essential for the G1/S transition and initiation of DNA replication during the cell division cycle in Saccharomyces cerevisiae. Cdc7p appears to be an evolutionarily conserved protein, since a homolog Hsk1 has been isolated from Schizosaccharomyces pombe. Here, we report the isolation of a human cDNA, HsCdc7, whose product is closely related in sequence to Cdc7p and Hsk1. The HsCdc7 cDNA encodes a protein of 574 amino acids with predicted size of 64 kDa. HsCdc7 contains the conserved subdomains common to all protein-serine/threonine kinases and three “kinase inserts” that are characteristic of Cdc7p and Hsk1. Immune complexes of HsCdc7 from cell lysates were able to phosphorylate histone H1 in vitro. Indirect immunofluorescence staining demonstrated that HsCdc7 protein was predominantly localized in the nucleus. Although the expression levels of HsCdc7 appeared to be constant throughout the cell cycle, the protein kinase activity of HsCdc7 increased during S phase of the cell cycle at approximately the same time as that of Cdk2. These results, together with the functions of Cdc7p in yeast, suggest that HsCdc7 may phosphorylate critical substrate(s) that regulate the G1/S phase transition and/or DNA replication in mammalian cells.

Dendritic cells (DC) have been thought to represent a family of closely related cells with similar functions and developmental pathways. The best-characterized precursors are the epidermal Langerhans cells, which migrate to lymphoid organs and become activated DC in response to inflammatory stimuli. Here, we demonstrate that a large subset of DC in the T cell-dependent areas of human lymphoid organs are nonactivated cells and belong to a separate lineage that can be identified by high levels of the interleukin 3 receptor α chain (IL-3Rαhi). The CD34+IL-3Rαhi DC progenitors are of myeloid origin and are distinct from those that give rise to Langerhans cells in vitro. The IL-3Rαhi DC furthermore appear to migrate to lymphoid organs independently of inflammatory stimuli or foreign antigens. Thus, DC are heterogeneous with regard to function and ontogeny.

Cdc48p from Saccharomyces cerevisiae and its highly conserved mammalian homologue VCP (valosin-containing protein) are ATPases with essential functions in cell division and homotypic fusion of endoplasmic reticulum vesicles. Both are mainly attached to the endoplasmic reticulum, but relocalize in a cell cycle-dependent manner: Cdc48p enters the nucleus during late G1; VCP aggregates at the centrosome during mitosis. The nuclear import signal sequence of Cdc48p was localized near the amino terminus and its function demonstrated by mutagenesis. The nuclear import is regulated by a cell cycle-dependent phosphorylation of a tyrosine residue near the carboxy terminus. Two-hybrid studies indicate that the phosphorylation results in a conformational change of the protein, exposing the nuclear import signal sequence previously masked by a stretch of acidic residues.

PDZ and LIM domains are modular protein interaction motifs present in proteins with diverse functions. Enigma is representative of a family of proteins composed of a series of conserved PDZ and LIM domains. The LIM domains of Enigma and its most related family member, Enigma homology protein, bind to protein kinases, whereas the PDZ domains of Enigma and family member actin-associated LIM protein bind to actin filaments. Enigma localizes to actin filaments in fibroblasts via its PDZ domain, and actin-associated LIM protein binds to and colocalizes with the actin-binding protein α-actinin-2 at Z lines in skeletal muscle. We show that Enigma is present at the Z line in skeletal muscle and that the PDZ domain of Enigma binds to a skeletal muscle target, the actin-binding protein tropomyosin (skeletal β-TM). The interaction between Enigma and skeletal β-TM was specific for the PDZ domain of Enigma, was abolished by mutations in the PDZ domain, and required the PDZ-binding consensus sequence (Thr-Ser-Leu) at the extreme carboxyl terminus of skeletal β-TM. Enigma interacted with isoforms of tropomyosin expressed in C2C12 myotubes and formed an immunoprecipitable complex with skeletal β-TM in transfected cells. The association of Enigma with skeletal β-TM suggests a role for Enigma as an adapter protein that directs LIM-binding proteins to actin filaments of muscle cells.

Clusters of orthologous groups [COGs; Tatusov, R. L., Koonin, E. V. & Lipman, D. J. (1997) Science 278, 631–637] were identified for a set of 13 completely sequenced herpesviruses. Each COG represented a family of gene products conserved across several herpes genomes. These families were defined without using an arbitrary threshold criterion based on sequence similarity. The COG technique was modified so that variable stringency in COG construction was possible. High stringencies identify a core set of highly conserved genes. Varying COG stringency reveals differences in the degree of conservation between functional classes of genes. The COG data were used to construct whole-genome phylogenetic trees based on gene content. These trees agree well with trees based on other methods and are robust when tested by bootstrap analysis. The COG data also were used to construct a reciprocal tree that clustered genes with similar phylogenetic profiles. This clustering may give clues to genes with related functions or with related histories of acquisition and loss during herpesvirus evolution.

Neuropeptides are implicated in many tumors, breast cancer (BC) included. Preprotachykinin-I (PPT-I) encodes multiple neuropeptides with pleiotropic functions such as neurotransmission, immune/hematopoietic modulation, angiogenesis, and mitogenesis. PPT-I is constitutively expressed in some tumors. In this study, we investigated a role for PPT-I and its receptors, neurokinin-1 (NK-1) and NK-2, in BC by using quantitative reverse transcription–PCR, ELISA, and in situ hybridization. Compared with normal mammary epithelial cells (n = 2) and benign breast biopsies (n = 21), BC cell lines (n = 7) and malignant breast biopsies (n = 25) showed increased expression of PPT-I and NK-1. NK-2 levels were high in normal and malignant cells. Specific NK-1 and NK-2 antagonists inhibited BC cell proliferation, suggesting autocrine and/or intercrine stimulation of BC cells by PPT-I peptides. NK-2 showed no effect on the proliferation of normal cells but mediated the proliferation of BC cells. Cytosolic extracts from malignant BC cells enhanced PPT-I translation whereas extracts from normal mammary epithelial cells caused no change. These enhancing effects may be protein-specific because a similar increase was observed for IL-6 translation and no effect was observed for IL-1α and stem cell factor. The data suggest that PPT-I peptides and their receptors may be important in BC development. Considering that PPT-I peptides are hematopoietic modulators, these results could be extended to understand early integration of BC cells in the bone marrow, a preferred site of metastasis. Molecular signaling transduced by PPT-I peptides and the mechanism that enhances translation of PPT-I mRNA could lead to innovative strategies for BC treatments and metastasis.
