Lunacloud is a cloud service provider with offices in Portugal, Spain, France and UK that focus on delivering reliable, elastic and low cost cloud Infrastructure as a Service (IaaS) solutions. The company currently relies on a proprietary IaaS platform - the Parallels Automation for Cloud Infrastructure (PACI) - and wishes to expand and integrate other IaaS solutions seamlessly, namely open source solutions. This is the challenge addressed in this thesis. This proposal, which was fostered by Eurocloud Portugal Association, contributes to the promotion of interoperability and standardisation in Cloud Computing. The goal is to investigate, propose and develop an interoperable open source solution with standard interfaces for the integrated management of IaaS Cloud Computing resources based on new as well as existing abstraction libraries or frameworks. The solution should provide bothWeb and application programming interfaces. The research conducted consisted of two surveys covering existing open source IaaS platforms and PACI (features and API) and open source IaaS abstraction solutions. The first study was focussed on the characteristics of most popular open source IaaS platforms, namely OpenNebula, OpenStack, CloudStack and Eucalyptus, as well as PACI and included a thorough inventory of the provided Application Programming Interfaces (API), i.e., offered operations, followed by a comparison of these platforms in order to establish their similarities and dissimilarities. The second study on existing open source interoperability solutions included the analysis of existing abstraction libraries and frameworks and their comparison. The approach proposed and adopted, which was supported on the conclusions of the carried surveys, reuses an existing open source abstraction solution – the Apache Deltacloud framework. Deltacloud relies on the development of software driver modules to interface with different IaaS platforms, officially provides and supports drivers to sixteen IaaS platform, including OpenNebula and OpenStack, and allows the development of new provider drivers. The latter functionality was used to develop a new Deltacloud driver for PACI. Furthermore, Deltacloud provides a Web dashboard and REpresentational State Transfer (REST) API interfaces. To evaluate the adopted solution, a test bed integrating OpenNebula, Open- Stack and PACI nodes was assembled and deployed. The tests conducted involved time elapsed and data payload measurements via the Deltacloud framework as well as via the pre-existing IaaS platform API. The Deltacloud framework behaved as expected, i.e., introduced additional delays, but no substantial overheads. Both the Web and the REST interfaces were tested and showed identical measurements. The developed interoperable solution for the seamless integration and provision of IaaS resources from PACI, OpenNebula and OpenStack IaaS platforms fulfils the specified requirements, i.e., provides Lunacloud with the ability to expand the range of adopted IaaS platforms and offers a Web dashboard and REST API for the integrated management. The contributions of this work include the surveys and comparisons made, the selection of the abstraction framework and, last, but not the least, the PACI driver developed.

Thirty six cases of acute disseminated paracoccidioidomycosis in 3 to 12 year-old children, natives of the state of Rio de Janeiro, were seen in the period 1981-1996. All patients were residents in the rural region of 15 counties, scattered on the Southwestern part of this state. The rural region of two neighboring counties, where 16 cases (44.4%) occurred, was visited. It exhibited the environmental conditions that are considered favorable to the survival of P. brasiliensis. The most important of these conditions, abundant watercourses and autochthonous forest, are distributed on well defined and limited areas, in which the dwellings are also localized. Probably, a careful epidemiological study of forthcoming cases of the disease in children may facilitate the search for the micro-niche of the fungus.

Tendo em conta a popularidade que as comunicações Wi-Fi têm na atualidade em vários dispositivos como computadores portáteis, telemóveis ou tablets, sendo estes utilizados praticamente por qualquer pessoa, surgiu a ideia de utilizar esta tecnologia de baixo custo e isenta de licenciamento num cenário de comunicações marítimas. Neste contexto, esta permite fornecer o acesso à Internet em banda larga a grupos de embarcações, que atualmente recorrem a tecnologias de elevado custo (satélite) e/ou de banda estreita (rádios VHF). Com o acesso em banda larga, os proprietários poderão utilizar aplicações informáticas de interesse à atividade de negócio ou de lazer, até então só disponíveis junto à costa onde existe cobertura celular. Nesta tese pretende-se fazer um estudo teórico e prático sobre o alcance e respetivo desempenho de comunicações de banda larga em ambiente marítimo, utilizando parte da gama de frequências dos 5,8 GHz, isenta de licença, e a norma IEEE 802.11n. Para se utilizar equipamento produzido em massa a operar nessa gama, existem duas normas disponíveis, a IEEE 802.11a e a IEEE 802.11n. Optou-se pelo IEEE 802.11n pois os esquemas de codificação ao nível físico permitem débitos mais elevados e MIMO. Para a realização dos testes experimentais, foi necessário elaborar um protótipo de comunicação ponto a ponto, constituído por dois nós de comunicação. Um deles foi instalado numa embarcação de pesca em colaboração com a Associação Propeixe e o outro no Edifício Transparente, no Porto, em colaboração com a entidade gestora do edifício e a Associação Porto Digital. Tanto quanto se conhece é o primeiro teste de comunicações Wi-Fi realizado nestas condições a nível mundial. Os objetivos do trabalho foram atingidos. Foi possível estabelecer comunicações Wi-Fi na banda dos 5,8 GHz até cerca de 7 km com débito médio mínimo de 1 Mbit/s. O ambiente de testes desenvolvido e os resultados obtidos servirão de base para futuros trabalhos de investigação na área das comunicações marítimas.

Com as variações e instabilidade dos preços do petróleo, assim como as políticas europeias para adoção de estratégias para o desenvolvimento sustentável, têm levado à procura de forma crescente de novas tecnologias e fontes de energia alternativas. Neste contexto, tem-se assistido a políticas energéticas que estimulam o aumento da produção e a utilização do gás natural, visto que é considerado uma fonte de energia limpa. O crescimento do mercado do gás natural implica um reforço significativo das redes de transporte deste combustível, quer ao nível do armazenamento e fornecimento, quer ao nível dos gasodutos e da sua gestão. O investimento em gasodutos de transporte implica grandes investimentos, que poderiam não ser remunerados da forma esperada, sendo um dos motivos para que exista em Portugal cinco distritos se veem privados deste tipo de infraestruturas. O transporte de gás natural acarreta custos elevados para os consumidores, tanto maiores quanto maior forem as quantidades de gás transacionadas e quanto maior for o percurso pelo gás natural percorrido. Assim assume especial importância a realização de um despacho de gás natural: quais as cargas que cada unidade de fornecimento de gás irá alimentar, qual a quantidade de gás natural que cada UFGs deve injetar na rede, qual o menor percurso possível para o fazer, o tipo de transporte que será utilizado? Estas questões são abordadas na presente dissertação, por forma a minimizar a função custo de transporte, diminuindo assim as perdas na rede de alta pressão e os custos de transporte que serão suportados pelos consumidores. A rede de testes adotada foi a rede nacional de transporte, constituída por 18 nós de consumos, e os tipos de transporte considerados, foram o transporte por gasoduto físico e o transporte através de gasoduto virtual – rotas de transporte rodoviário de gás natural liquefeito. Foram criados diversos cenários, baseados em períodos de inverno e verão, os diferentes cenários abrangeram de forma distinta as variáveis de forma a analisar os impactos que estas variáveis teriam no custo relativo ao transporte de gás natural. Para dar suporte ao modelo de despacho económico, foi desenvolvida uma aplicação computacional – Despacho_GN com o objetivo de despachar as quantidades de gás natural que cada UFG deveria injetar na rede, assim como apresentar os custos acumulados relativos ao transporte. Com o apoio desta aplicação foram testados diversos cenários, sendo apresentados os respectivos resultados. A metodologia elaborada para a criação de um despacho através da aplicação “Despacho_GN” demonstrou ser eficiente na obtenção das soluções, mostrando ser suficientemente rápida para realizar as simulações em poucos segundos. A dissertação proporciona uma contribuição para a exploração de problemas relacionados com o despacho de gás natural, e sugere perspectivas futuras de investigação e desenvolvimento.

Background: Prostate cancer (PCa), a highly incident and heterogeneous malignancy, mostly affects men from developed countries. Increased knowledge of the biological mechanisms underlying PCa onset and progression are critical for improved clinical management. MicroRNAs (miRNAs) deregulation is common in human cancers, and understanding how it impacts in PCa is of major importance. MiRNAs are mostly downregulated in cancer, although some are overexpressed, playing a critical role in tumor initiation and progression. We aimed to identify miRNAs overexpressed in PCa and subsequently determine its impact in tumorigenesis. Results: MicroRNA expression profiling in primary PCa and morphological normal prostate (MNPT) tissues identified 17 miRNAs significantly overexpressed in PCa. Expression of three miRNAs, not previously associated with PCa, was subsequently assessed in large independent sets of primary tumors, in which miR-182 and miR-375 were validated, but not miR-32. Significantly higher expression levels of miR-375 were depicted in patients with higher Gleason score and more advanced pathological stage, as well as with regional lymph nodes metastases. Forced expression of miR-375 in PC-3 cells, which display the lowest miR-375 levels among PCa cell lines, increased apoptosis and reduced invasion ability and cell viability. Intriguingly, in 22Rv1 cells, which displayed the highest miR-375 expression, knockdown experiments also attenuated the malignant phenotype. Gene ontology analysis implicated miR-375 in several key pathways deregulated in PCa, including cell cycle and cell differentiation. Moreover, CCND2 was identified as putative miR-375 target in PCa, confirmed by luciferase assay. Conclusions: A dual role for miR-375 in prostate cancer progression is suggested, highlighting the importance of cellular context on microRNA targeting.

Este documento descreve um modelo de tolerância a falhas para sistemas de tempo-real distribuídos. A sugestão deste modelo tem como propósito a apresentação de uma solu-ção fiável, flexível e adaptável às necessidades dos sistemas de tempo-real distribuídos. A tolerância a falhas é um aspeto extremamente importante na construção de sistemas de tempo-real e a sua aplicação traz inúmeros benefícios. Um design orientado para a to-lerância a falhas contribui para um melhor desempenho do sistema através do melhora-mento de aspetos chave como a segurança, a confiabilidade e a disponibilidade dos sis-temas. O trabalho desenvolvido centra-se na prevenção, deteção e tolerância a falhas de tipo ló-gicas (software) e físicas (hardware) e assenta numa arquitetura maioritariamente basea-da no tempo, conjugada com técnicas de redundância. O modelo preocupa-se com a efi-ciência e os custos de execução. Para isso utilizam-se também técnicas tradicionais de to-lerância a falhas, como a redundância e a migração, no sentido de não prejudicar o tempo de execução do serviço, ou seja, diminuindo o tempo de recuperação das réplicas, em ca-so de ocorrência de falhas. Neste trabalho são propostas heurísticas de baixa complexida-de para tempo-de-execução, a fim de se determinar para onde replicar os componentes que constituem o software de tempo-real e de negociá-los num mecanismo de coordena-ção por licitações. Este trabalho adapta e estende alguns algoritmos que fornecem solu-ções ainda que interrompidos. Estes algoritmos são referidos em trabalhos de investiga-ção relacionados, e são utilizados para formação de coligações entre nós coadjuvantes. O modelo proposto colmata as falhas através de técnicas de replicação ativa, tanto virtual como física, com blocos de execução concorrentes. Tenta-se melhorar ou manter a sua qualidade produzida, praticamente sem introduzir overhead de informação significativo no sistema. O modelo certifica-se que as máquinas escolhidas, para as quais os agentes migrarão, melhoram iterativamente os níveis de qualidade de serviço fornecida aos com-ponentes, em função das disponibilidades das respetivas máquinas. Caso a nova configu-ração de qualidade seja rentável para a qualidade geral do serviço, é feito um esforço no sentido de receber novos componentes em detrimento da qualidade dos já hospedados localmente. Os nós que cooperam na coligação maximizam o número de execuções para-lelas entre componentes paralelos que compõem o serviço, com o intuito de reduzir atra-sos de execução. O desenvolvimento desta tese conduziu ao modelo proposto e aos resultados apresenta-dos e foi genuinamente suportado por levantamentos bibliográficos de trabalhos de in-vestigação e desenvolvimento, literaturas e preliminares matemáticos. O trabalho tem também como base uma lista de referências bibliográficas.

The involvement of the gastrointestinal tract in the co-infection of HIV and Leishmania is rarely reported. We report the case of an HIV-infected adult man co-infected with a disseminated form of leishmaniasis involving the liver, lymph nodes, spleen and, as a feature reported for the first time in the English literature, the pancreas. Light microscopy showed amastigote forms of Leishmania in pancreatic macrophages and immunohistochemical staining revealed antigens for Leishmania and also for HIV p24. Microscopic and ultrastructural analysis revealed severe acinar atrophy, decreased zymogen granules in the acinar cytoplasm and also nuclear abnormalities such as pyknosis, hyperchromatism and thickened chromatin. These findings might correspond to the histologic pattern of protein-energy malnutrition in the pancreas as shown in our previous study in pancreas with AIDS and no Leishmania. In this particular case, the protein-energy malnutrition may be due to cirrhosis, or, Leishmania or HIV infection or all mixed. We believe that this case represents the morphologic substratum of the protein energy malnutrition in pancreas induced by the HIV infection. Further studies are needed to elucidate these issues.

In recent years, vehicular cloud computing (VCC) has emerged as a new technology which is being used in wide range of applications in the area of multimedia-based healthcare applications. In VCC, vehicles act as the intelligent machines which can be used to collect and transfer the healthcare data to the local, or global sites for storage, and computation purposes, as vehicles are having comparatively limited storage and computation power for handling the multimedia files. However, due to the dynamic changes in topology, and lack of centralized monitoring points, this information can be altered, or misused. These security breaches can result in disastrous consequences such as-loss of life or financial frauds. Therefore, to address these issues, a learning automata-assisted distributive intrusion detection system is designed based on clustering. Although there exist a number of applications where the proposed scheme can be applied but, we have taken multimedia-based healthcare application for illustration of the proposed scheme. In the proposed scheme, learning automata (LA) are assumed to be stationed on the vehicles which take clustering decisions intelligently and select one of the members of the group as a cluster-head. The cluster-heads then assist in efficient storage and dissemination of information through a cloud-based infrastructure. To secure the proposed scheme from malicious activities, standard cryptographic technique is used in which the auotmaton learns from the environment and takes adaptive decisions for identification of any malicious activity in the network. A reward and penalty is given by the stochastic environment where an automaton performs its actions so that it updates its action probability vector after getting the reinforcement signal from the environment. The proposed scheme was evaluated using extensive simulations on ns-2 with SUMO. The results obtained indicate that the proposed scheme yields an improvement of 10 % in detection rate of malicious nodes when compared with the existing schemes.

IEEE 802.11 is one of the most well-established and widely used standard for wireless LAN. Its Medium Access control (MAC) layer assumes that the devices adhere to the standard’s rules and timers to assure fair access and sharing of the medium. However, wireless cards driver flexibility and configurability make it possible for selfish misbehaving nodes to take advantages over the other well-behaving nodes. The existence of selfish nodes degrades the QoS for the other devices in the network and may increase their energy consumption. In this paper we propose a green solution for selfish misbehavior detection in IEEE 802.11-based wireless networks. The proposed scheme works in two phases: Global phase which detects whether the network contains selfish nodes or not, and Local phase which identifies which node or nodes within the network are selfish. Usually, the network must be frequently examined for selfish nodes during its operation since any node may act selfishly. Our solution is green in the sense that it saves the network resources as it avoids wasting the nodes energy by examining all the individual nodes of being selfish when it is not necessary. The proposed detection algorithm is evaluated using extensive OPNET simulations. The results show that the Global network metric clearly indicates the existence of a selfish node while the Local nodes metric successfully identified the selfish node(s). We also provide mathematical analysis for the selfish misbehaving and derived formulas for the successful channel access probability.

Multi-standard mobile devices are allowing users to enjoy higher data rates with ubiquitous connectivity. However, the benefits gained from multiple interfaces come at an expense—that being higher energy consumption in an era where mobile devices need to be energy compliant. One promising solution is the usage of short-range cooperative communication as an overlay for infrastructure-based networks taking advantage of its context information. However, the node discovery mechanism, which is pivotal to the bearer establishment process, still represents a major burden in terms of the total energy budget. In this paper, we propose a technology agnostic approach towards enhancing the MAC energy ratings by presenting a context-aware node discovery (CANDi) algorithm, which provides a priori knowledge towards the node discovery mechanism by allowing it to search nodes in the near vicinity at the ‘right time and at the right place’. We describe the different beacons required for establishing the cooperation, as well as the context information required, including battery level, modes, location and so on. CANDi uses the long-range network (WiMAX and WiFi) to distribute the context information about cooperative clusters (Ultra-wideband-based) in the vicinity. The searching nodes can use this context in locating the cooperative clusters/nodes, which facilitates the establishing of short-range connections. Analytical and simulation results are obtained, and the energy saving gains are further demonstrated in the laboratory using a customised testbed. CANDi saves up to 50% energy during the node discovery process, while the demonstrative testbed shows up to 75% savings in the total energy budget, thus validating the algorithm, as well as providing viable evidence to support the usage of short-range cooperative communications for energy savings.

A total of 323 patients with lymphadenopathy were selected in Belém, Brazil, between January 1996 and December 2001, and screened for the presence of human herpesvirus-6 (HHV-6) IgM- and- IgG antibodies by enzyme-linked immunosorbent assay (ELISA). When seroprevalence is analyzed by gender, similar rates are found for female (60.6%) and male (55.7%) individuals. Seventy-seven (23.8%) patients were HHV-6-IgM-and- IgG-positive (IgM+ subgroup), with positivity rates of 29.7% and 17.7% (p = 0.0007) for female- and male individuals, respectively. Sera from a subgroup (n = 120) of these subjects, with high HHV-6 antibody levels (either IgM+ or IgG+ reactivities), were subsequently processed for the presence of HHV-6 DNA by polymerase chain reaction (PCR)/nested PCR. Active infections (IgM+ and/or IgG+ high levels specific antibodies plus detection of viral DNA) were diagnosed in 20/77 (20.0%) and 8/43 (18.6%); subgroup of the 120 individuals suspected of having HHV-6 suggestive recent infection. All (n = 28) cases of active infection were found to be associated with HHV-6 variant-A (HHV-6A), as detectable by PCR/nested PCR, using variant-specific primer that amplify regions of 195 base pairs (bp) (HHV-6A) and 423 bp (HHV-6B). Rates of HHV-6 DNA detection between female and male patients were similar (p > 0.05) in the IgM+ and IgG+ groups: 20.4% versus 35.7% and 25.0% versus 13.0%, respectively. HHV-6 DNA was detected across < 5 through 41-50-year age-groups for patients whose serum samples were IgM+, with rates ranging from 7.7% (female subjects aged < 5 years) to 80.0% (male, 11-20 years). Among patients whose serological status was IgG+, HHV-6 DNA was detected in < 5, 6-10, 21-30 and > 50 age-groups at rates that ranged from 15.4% (male, < 5 years of age) to 100.0% (female aged 11-20 years). Swelling cervical lymph nodes were the most common sign, accounting for 9 (32.0%) cases in each gender group. Among patients (n = 28) with active infection by HHV-6A variant, duration of symptoms lasted 1-5 days in 35.7% of subjects, whereas in 64.3% of them the disease lasted 6-20 days. Our data suggest that it is worth seeking for HHV-6 infection whenever a patient (infant or adult) presents with lymphadenopathy as a prominent symptom in the course of an acute febrile illness.
