OBJETIVO: Estudar o padrão de suscetibilidade do Aedes aegypti ao inseticida organofosforado temefós. MÉTODOS: Amostras de larvas de Ae. aegypti foram obtidas com armadilhas para oviposição, em oito cidades do Distrito Federal, nos anos 2000 e 2001. As larvas foram submetidas à dose diagnóstica de 0,012 mg/l de temefós, segundo metodologia padronizada pela Organização Mundial da Saúde. As populações de campo foram testadas em paralelo com a cepa de referência Rockefeller e a cepa DIVAL, do insetário da Diretoria de Vigilância Ambiental. A concentração e a pureza das soluções de temefós foram analisadas por cromatografia gasosa. Os cálculos de correlação foram determinados pelo programa StatView - SAS Institute Inc., versão 5. Utilizou-se o teste t de Student para verificar diferenças de suscetibilidade, com níveis de significância, alfa=0,05. RESULTADOS: Em 2000, as populações de larvas de Ae. aegypti nas cidades de Taguatinga, Guará e Núcleo Bandeirante apresentaram-se resistentes ao temefós, com mortalidade de larvas entre 54,1 e 63,4%. As populações do Gama, Planaltina e Sobradinho apresentaram alterações nos níveis de suscetibilidade (mortalidade de 83,6 a 92,8%). A população de Ceilândia foi a única suscetível, com 98% de mortalidade. Em 2001, todas as populações testadas mostraram-se resistentes (44,4 a 66,4% de mortalidade). Nenhuma correlação significativa foi encontrada entre a suscetibilidade das populações e a distância entre essas cidades ou a quantidade de inseticida aplicado nos anos anteriores ao estudo. CONCLUSÕES: Os níveis de suscetibilidade do Ae. aegypti ao temefós vêm se alterando no Distrito Federal. É essencial a continuidade de programas de monitoramento da resistência desse vetor aos inseticidas para se garantir a eficiência dos programas de controle e a proteção da saúde humana.

OBJECTIVE: To estimate the basic reproduction number (R0) of dengue fever including both imported and autochthonous cases. METHODS: The study was conducted based on epidemiological data of the 2003 dengue epidemic in Brasília, Brazil. The basic reproduction number is estimated from the epidemic curve, fitting linearly the increase of initial cases. Aiming at simulating an epidemic with both autochthonous and imported cases, a "susceptible-infectious-resistant" compartmental model was designed, in which the imported cases were considered as an external forcing. The ratio between R0 of imported versus autochthonous cases was used as an estimator of real R0. RESULTS: The comparison of both reproduction numbers (only autochthonous versus all cases) showed that considering all cases as autochthonous yielded a R0 above one, although the real R0 was below one. The same results were seen when the method was applied on simulated epidemics with fixed R0. This method was also compared to some previous proposed methods by other authors and showed that the latter underestimated R0 values. CONCLUSIONS: It was shown that the inclusion of both imported and autochthonous cases is crucial for the modeling of the epidemic dynamics, and thus provides critical information for decision makers in charge of prevention and control of this disease.

Os autores descrevem a ocorrência de epidemias causadas pelo vírus Oropouche (ORO) nos Estados do Maranhão (MA) e Goiás (GO) em 1988. 36 amostras de vírus foram obtidas a partir da inoculação do sangue de 120 pacientes em camundongos recém nascidos. A doença foi caracterizada por febre, cefaléia, dores musculares, articulares, fotofobia, dor retro ocular, náuseas e tontura. 128 das 197 pessoas examinadas em Porto Franco, MA, tinham anticorpos inibidores da hemaglutinação (IH) para o agente e, em 106 foram detectados anticorpos IgM por MAC ELISA. Todos os grupos etários foram infectados, embora a incidência tenha sido mais elevada entre aqueles com 10 a 19 anos de idade. Quanto ao sexo, a infecção ocorreu igualmente em ambos os sexos. Recorrência dos sintomas foi observada em 56% dos casos positivos estudados. A inoculação em camundongos Swiss recém nascidos de 3.624 Culicoides paraensis (Ceratopogonidae) e 1.970 Culex (Culex) quinquefasciatus (Culicidae), coletados em Porto Franco-MA, resultou em um único isolamento do vírus ORO a partir dos Culicoides. Essa é a primeira descrição de casos confirmados de infecção pelo vírus Oropouche nos Estados do Maranhão e Goiás, Brasil.

Os autores revêem os aspectos ecoepidemiológicos apresentados pelos virus da encefalite de St. Louis (SLE), encefalites equinas Leste (EEE), Oeste (WEE) e Venezuelana [subtipos III, Mucambo (MUC) e IV, Pixuna (PIX)], decorrentes dos estudos realizados em diversas áreas da Região Amazônica brasileira, especialmente ao longo das rodovias e projetos de desenvolvimento. Esses vírus são amplamente distribuídos na Amazônia e pelo menos quatro deles, EEE, WEE, MUC e SLE já demonstraram ser patógenos do homem. O diagnóstico da doença humana foi feito por sorologia, sendo que de MUC e SLE obteve-se também isolamento viral. O vírus PIX, parece ser o menos prevalente e foi isolado em poucas oportunidades. Virtualmente se desconhecem os vetores do PIX e WEE. As aves silvestres constituem os hospedeiros principais de todos esses vírus, exceto do MUC, para o qual constituem os roedores. O quadro clínico apresentado pelos pacientes infectados na Amazônia é discutido, comparando-o ao apresentado em outras áreas, especialmente nos EUA, onde periodicamente SLE, EEE e WEE causam surtos de doença humana. Nenhuma epidemia foi até o presente detectada, embora em 1960 uma epizootia em eqüinos causada pelo EEE tenha sido registrada em Bragança, Pará, onde em um rebanho de 500 animais ocorreu uma letalidade de 5%. Quatro outras pequenas epizootias determinadas pelo SLE ocorreram nas florestas adjacentes a Belém, envolvendo aves silvestres e animais sentinelas.

Registramos a ocorrência de epidemia de dengue causada pelo sorotipo 2 (DEN 2) na cidade de Araguaina, estado do Tocantins (TO) situado no Brasil central. Quatrocentos indíviduos de 74 famílias, residentes nos bairros S. João, Araguaina Sul e Neblina foram entrevistados e sangrados, independentemente de terem adoecido ou não. Os soros tanto de adultos quanto de crianças de ambos os sexos foram usados para pesquisa de anticorpos inibidores da hemaglutinação (IH) e IgM através de ensaio imunoenzimático (MAC ELISA). Nas casas onde haviam doentes no momento do inquérito, sangue total também foi colhido para tentativa de isolamento de vírus. O quadro clínico apresentado pelos pacientes foi caracterizado por febre, cefaléia, mialgias, artralgias e exantema do tipo máculo-papular não pruriginoso. A infecção foi mais frequente em mulheres (33.9%) do que nos homens (23.8%), ocorrendo em todas as faixas etárias, inclusive em crianças com menos de um ano de idade, bem como em maiores de 70 anos. Um total de 1105 mosquitos (56 fêmeas e 45 machos de Culex quinquefasciatus e 567 fêmeas e 437 machos de Aedes aegypti) foram obtidos a partir de larvas coletadas em Araguaina. As fêmeas de Ae. aegypti obtidas das larvas fizeram repasto sangüíneo em 8 pacientes febris. O diagnóstico laboratorial foi feito por isolamento de vírus (cultura de células de Aedes albopictus, clone C6/36) e por sorologia (IH e MAC ELISA). Foram isoladas 5 amostras de DEN 2 de pacientes febris e tipadas por imunofluorescência indireta usando anticorpos monoclonais de dengue. Nenhuma amostra viral foi isolada de mosquitos. Outrossim, comprovou-se infecção em 111 pessoas sangradas, o que revelou um índice de positividade de 27.75% (111 em 400), sendo que 66.2% das famílias estudadas apresentaram pelo menos um indivíduo positivo. Ocorreram ainda, 26.1% de infecções assintomáti-cas. Por outro lado, a correlação de positividade entre os dois testes usados (IH e MAC ELISA) foi de 94.6%. Estimamos que ocorreram aproximadamente 83.250 casos da doença, entre 15 de março a 31 de maio de 1991. Esta é a primeira epidemia de DEN 2 em um estado da Amazônia Brasileira, portanto em área endêmica de febre amarela, e a primeira evidência da interiorização do DEN 2, até então restrito ao Rio de Janeiro.

We evaluated the antiviral activity of the marine alga, Ulva fasciata, collected from Rasa beach and Forno beach, Búzios, Rio de Janeiro, Brazil on the replication of human metapneumovirus (HMPV). The algae extracts were prepared using three different methodologies to compare the activity of different groups of chemical composites obtained through these different methodologies. Four out of the six extracts inhibited nearly 100% of viral replication. The results demonstrated that the majority of the extracts (five out of six) possess virucidal activity and therefore have the ability to interact with the extracellular viral particles and prevent the infection. On the other hand, only two extracts (from Forno beach, obtained by maceration and maceration of the decoction) were able to interact with cell receptors, hindering the viral entry. Finally, only the extract of algae collected at Forno beach, obtained by maceration presented intracellular activity. To our knowledge, this is a pioneer study on antiviral activity of marine algae against HMPV. It is also the first on antiviral activity against HMPV ever done in Brazil. The study also shows the effect of different environment factors and different chemical procedures used to obtain the extract on its biological properties.

Dengue is present in the Federal District since 1991 and virological studies of the vector began in 1998. Two strains of DEN1 were isolated from 9 pools of female Aedes aegypti (78 mosq.), collected in April in Gama county, where the Breteau index was 5.4, and 32 autochtonous human cases were notified.

An HIV seroprevalence and molecular study was conducted among 935 subjects: 723 female commercial sex workers, 92 men who have sex with men and 120 HIV-positive volunteers. The reported injection drug use rates were 0.7% in female commercial sex workers and 3% in men who have sex with men. Sexually transmitted infections were reported in 265 (37%) of the female commercial sex workers and 38 (41%) of the men who have sex with men. A total of 20 (2.8%) female commercial sex workers and 12 (13%) men who have sex with men became HIV infected during the study period. A history of sexually transmitted infection increased the risk of subsequent HIV infection twofold (adjusted odds ratio of 2.5) among the female commercial sex workers, while cocaine use had an adjusted odds ratios of 6.61 among men who have sex with men. From 130 samples, and based on heteroduplex mobility assaying for the env gene, with sequencing of part of pol and/or full genomes, subtype B was the predominant subtype identified (66%); followed by subtype F (22%) and subtype C (4%). Recombinant CRF12-BF strains were identified in 6% and CRF17_BF was identified in 2%.

INTRODUCTION: Although various studies have been published regarding the treatment of chronic hepatitis C (CHC) with peginterferon (Peg-IFN) and ribavirin, little is known regarding the real impact of gender on the characteristics that influence the effectiveness and safety of antiviral treatment for CHC patients. The objective of this study was to evaluate the influence of gender on HCV treatment outcomes. METHODS: A retrospective analytical study was conducted among selected carriers of CHC genotype 1, who were treated with Peg-IFN α-2b at a dose of 1.5 μg/kg or Peg-IFN α-2a at a dose of 180 μg/week plus a ribavirin dose of 1,000-1,250 mg/day, according to weight, between 2001 and 2007. RESULTS: Among 181 patients undergoing treatment, the mean age was 46.4 ± 11.0 years and 46% were women. At baseline, 32% of the patients had advanced fibrosis (F3-F4 Scheuer), and 83% of the subjects had viral load > 400,000 IU/ml, without significant difference between the genders (p = 0.428 and p = 0.452, respectively). When compared with men, women had higher incidence of many adverse events such as anemia (p < 0.001) and higher need for dose reduction, for both Peg-IFN (p = 0.004) and ribavirin (p = 0.006). However, the rate of sustained virological response (SVR) did not differ between the genders: 45% (female) vs 41% (male); p=0.464. CONCLUSIONS: This study suggests that women and men react differently to combined therapy, especially in relation to the incidence of adverse events and the need for dose modification. Nevertheless, these differences do not influence the SVR rate.

One apparent growth opportunity for Compal, the leading producer of juices and nectars, is to expand its commercial reach to new occasions of consumption. This report carefully analyzes the opportunities related to an expansion to the main meal occasion and introduces a complete marketing and communications plan for a possible new main meal juice, Compal à Mesa. The product concept represents a rather premium positioning for the main meal occasion, including new flavor mixes that are targeted at different occasions of meals. Many potentially harmful pesticides for both human health and the environment are used in Brazilian Amazon. However, no scientific datum on pesticide usage is presently available for this region. Consequently, it is difficult to assess which substances arc used and in which quantities. As an important premise for future work on pesticide contamination in the county of Santarém (State of Pará, Brazil), a survey was conducted in order to qualify and quantify the use of some pesticides in this region. This investigation was made between January and March 1997 and August and October 1998 and revealed use of several organophosphates, synthetic pyrethroids and carbamates insecticides. Furthermore, many herbicides and fungicides were listed. These pesticides are used for agriculture, domestic, and sanitary programs. This paper also provides a first estimation of quantities of some insecticides commonly used in agriculture (chlorpyrifos, malathion, metamidophos and methyl-parathion). The annual consumption for these four compounds is estimated at ca. 1 910 kg. Organophosphate insecticide consumption in the county of Santarém seems to be lower than the Brazilian average in terms of «per capita» and «per agricultural area» consumptions. Nevertheless, this county uses toxic substances on sensitive environments such as floodplains (várzeas), making relevant a thorough study on the potential contamination of this environment and its biota.

Trace levels of three organophosphate insecticides (OPI) were detected in eight fish species from the region of Santarém, State of Pará, Brazil. Individual concentrations of OPI in fish ranged from less than the detection limit to 2,1 ppb. Mean concentrations of chlorpyrifos, malathion, and methyl-parathion were 0,3 ± 0,3, 0,1 ± 0,1, and 0,3 ± 0,3 ppb, respectively. Pellona flavipinnis, the largest and fattest piscivorous species analyzed, was the most contaminated. Since an inhabitant of this Amazonian region consumes 220 g of fish per day on average, ingested doses of chlorpyrifos, malathion, and methyl-parathion may reach up to 308, 220, and 462 ng·d-1, respectively. Compared to acceptable daily intakes (ADI), quantities of OPI absorbed via fish consumption on a daily basis are far below deleterious levels. We estimated that even considering the highest OPI contents detected, the average daily fish consumption of anadult of 60 kg would have to increase by ca. 1 950, 5 450, and 2 600 times to reach ADI of chlorpyrifos, malathion, and methyl-parathion, respectively. Neither fish diet nor fish lipid content enabled us to completely explain the interspecific differences observed.

This study reports the ability of one hyperthermophile and two thermophilic microorganisms to grow anaerobically by the reduction of chlorate and perchlorate. Physiological, genomic and proteome analyses suggest that the Crenarchaeon Aeropyrum pernix reduces perchlorate with a periplasmic enzyme related to nitrate reductases, but that it lacks a functional chlorite-disproportionating enzyme (Cld) to complete the pathway. A. pernix, previously described as a strictly aerobic microorganism, seems to rely on the chemical reactivity of reduced sulfur compounds with chlorite, a mechanism previously reported for perchlorate-reducing Archaeoglobus fulgidus. The chemical oxidation of thiosulfate (in excessive amounts present in the medium) and the reduction of chlorite result in the release of sulfate and chloride, which are the products of a biotic-abiotic perchlorate reduction pathway in A. pernix. The apparent absence of Cld in two other perchlorate-reducing microorganisms, Carboxydothermus hydrogenoformans and Moorella glycerini strain NMP, and their dependence on sulfide for perchlorate reduction is consistent with observations made on A. fulgidus. Our findings suggest that microbial perchlorate reduction at high temperature differs notably from the physiology of perchlorate- and chlorate-reducing mesophiles and that it is characterized by the lack of a chlorite dismutase and is enabled by a combination of biotic and abiotic reactions.
