El pasado día 4 de abril tuvimos la oportunidad de probar un nuevo T6.160 AutoCommand en tierras de Cantalejo, Segovia. Se han realizado dos tipos de tarea con el tractor: preparación del lecho de siembra con una grada rotativa y transporte de un remolque cargado. La parcela tenía una textura fuertemente arenosa típica de la zona y se encontraba en un estado de tempero óptimo. En las distintas pruebas se ha evaluado el modo automático frente al manual. Veamos sus resultados.

Este estudio presenta una comparativa entre un LIDAR modelo LMS-111 (Sick Ltd.) y una cámara de profundidad de uso doméstico: Kinect (Microsoft Corporation), orientada a determinar las condiciones de uso de uno y otro sensor, así como sus ventajas e inconvenientes cuando son empleados en condiciones de campo, en una explotación agrícola. Para ello se realizaron diversos ensayos en una parcela experimental del CSIC-CAR de Arganda del Rey, España. Para los ensayos ambos sensores fueron instalados en un tractor operado remotamente diseñado y construido en el marco del proyecto europeo RHEA. Dicho tractor realizó dos recorridos diferentes: el primero se efectuó en paralelo a un muro y el segundo paralelo a una hilera de olivos. El primer ensayo se realizó con el propósito de cuantificar la uniformidad de las mediciones de ambos sensores y el segundo para validar los resultados en un cultivo real. Los recorridos se realizaron empleando cuatro marchas diferentes, con el objetivo de determinar si los diferentes regímenes de operación del motor influyen sobre la precisión de los sensores. Los resultados muestran que el LIDAR posee un mayor alcance máximo de medición, pero una resolución menor frente a Kinect, muestran además que el LIDAR puede ser operado a cualquier hora del día y condición meteorológica, mientras que Kinect, no puede operar en exteriores, salvo en horas del día con baja intensidad lumínica. Por otra parte la gran desventaja del LIDAR es su coste, 30 veces más alto que Kinect.

El pasado día 17 de junio de 2014 tuvimos la oportunidad de realizar ensayos en campo a un tractor Same Virtus 120, en las instalaciones que la empresa posee junto a Toledo. La prueba de laboreo se realizó con un cultivador Kverneland CLC Evo con nueve brazos amortiguados con ballestas, rejas rectas y 2,4 m de anchura.También realizamos una prueba de transporte de un remolque con cebada, y evaluamos la maniobrabilidad y frenada del tractor.

A novel slow push asteroid deflection strategy has been recently proposed in which an Earth threatening asteroid can be deflected by exploiting the momentum transmitted by a collimated beam of quasi-neutral plasma impinging against the asteroid surface. The beam can be generated with state-of-the art ion engines from a hovering spacecraft with no need for physical attachment or gravitational interaction with the celestial body. The spacecraft, placed at a distance of a few asteroid diameters, would need an ion thruster pointed at the asteroid surface as well as a second propulsion system to compensate for the ion engine reaction and keep the distance between the asteroid and the shepherd satellite constant throughout the deflection phase. A comparison in terms of required spacecraft mass per total imparted deflection impulse shows that the method outperforms the gravity tractor concept by more than one order of magnitude for asteroids up to about 200 m diameter. The two methods would yield comparable performance for asteroids larger than about 2 km

Cover crops in Mediterranean vineyards are scarcely used due to water competition between the cover crop and the grapevine; however, bare soil management through tillage or herbicides tends to have negative effects on the soil over time (organic matter decrease, soil structure and soil fertility degradation, compaction, etc). The objective of this study was to understand how soil management affects soil fertility, compaction and infiltration over time. To this end, two bare soil techniques were compared, tillage (TT) and total herbicide (HT) with two cover crops; annual cereal (CT) and annual grass (AGT), established for 8 years. CT treatment showed the highest organic matter content, having the biggest amount of biomass incorporated into the soil. The annual adventitious vegetation in TT treatment (568 kg dry matter ha-1) that was incorporated into the soil, kept the organic matter content higher than HT levels and close to AGT level, in spite of the greater aboveground annual biomass production of this treatment (3632 kg dry matter ha-1) whereas only its roots were incorporated into the soil. TT presented the highest bulk density under the tractor track lines and a greatest resistance to penetration (at 0.2 m depth). AGT presented bulk density values (upper 0.4 m) lower than TT and penetration resistance in CT lower (at 0.20 m depth) than TT too. The theoretical basis for evaluating shear strength in rock joints is presented and used to derive an equation that governs the relationship between tangential and normal stress on the joint during situations of slippage between the joint faces. The dependent variables include geometric dilatancy, the instantaneous friction angle, and a parameter that considers joint surface roughness. The effect roughness is studied, and the aforementioned formula is used to analyse joints under different conditions. A mathematical expression is deduced that explains Barton's value for the joint roughness coefficient (JRC) according to the roughness geometry. In particular, when the Hoek and Brown failure criterion is used for a rock in the contact with the surface roughness plane, it is possible to determine the shear strength of the joint as a function of the relationship between the uniaxial compressive strength of the wall with the normal stress acting on the wall. En esta ocasión el tractor ensayado en el Campus de New Holland (Peñarrubias del Pirón) pertenece a la nueva serie T8 de la marca, en la que se ha introducido la transmisión continuamente variable Auto Command, dando lugar a los tractores más potentes y de más porte con CVT. El estado del suelo, después de varios días de lluvia y nieve (hasta 50 mm en los 3 días previos al ensayo, 23 de enero), condicionó el ensayo, obligándonos a centrar la jornada en la tarea de transporte, para la que New Holland señala que el T8.350 Auto Command está especialmente equipado y diseñado

El sector español de la biomasa es tremendamente amplio y heterogéneo, al ser un sector muy vinculado a otros sectores productivos profundamente arraigados en nuestro país, como son el agrícola, el forestal y el ganadero. La cercanía a estos sectores, y en general al medio rural, lo convierten en un sector que despierta gran interés entre los agricultores, los propietarios forestales, los ganaderos y entre los empresarios que gestionan explotaciones agroindustriales, agroganaderas, alimentarias y forestales, entre otras. Se trata de un valioso tejido productivo fuertemente vinculado al medio rural y con un significativo potencial tractor en términos de actividad económica y creación de empleo estable. Las biomasas que se generan en todos los ámbitos mencionados pueden ser valorizadas energéticamente, con lo que en el caso de tratarse de residuos (agrícolas, forestales, ganaderos, industriales, etc.) esta transformación en energía permite convertir un residuo en un recurso, en este caso energético, que de no ser valorizado sería potencialmente contaminante si fuera abandonado o enterrado, o bien podría quemarse incontroladamente provocando la destrucción de grandes masas forestales, hábitats, y cobrándose incluso vidas humanas.

Strong positive Darwinian selection acts on two sperm fertilization proteins, lysin and 18-kDa protein, from abalone (Haliotis). To understand the phylogenetic context for this dramatic molecular evolution, we obtained sequences of mitochondrial cytochrome c oxidase subunit I (mtCOI), and genomic sequences of lysin, 18-kDa, and a G protein subunit. Based on mtDNA differentiation, four north Pacific abalone species diverged within the past 2 million years (Myr), and remaining north Pacific species diverged over a period of 4–20 Myr. Between-species nonsynonymous differences in lysin and 18-kDa exons exceed nucleotide differences in introns by 3.5- to 24-fold. Remarkably, in some comparisons nonsynonymous substitutions in lysin and 18-kDa genes exceed synonymous substitutions in mtCOI. Lysin and 18-kDa intron/exon segments were sequenced from multiple red abalone individuals collected over a 1,200-km range. Only two nucleotide changes and two sites of slippage variation were detected in a total of >29,000 nucleotides surveyed. However, polymorphism in mtCOI and a G protein intron was found in this species. This finding suggests that positive selection swept one lysin allele and one 18-kDa allele to fixation. Similarities between mtCOI and lysin gene trees indicate that rapid adaptive evolution of lysin has occurred consistently through the history of the group. Comparisons with mtCOI molecular clock calibrations suggest that nonsynonymous substitutions accumulate 2–50 times faster in lysin and 18-kDa genes than in rapidly evolving mammalian genes.

Rearrangements between tandem sequence homologies of various lengths are a major source of genomic change and can be deleterious to the organism. These rearrangements can result in either deletion or duplication of genetic material flanked by direct sequence repeats. Molecular genetic analysis of repetitive sequence instability in Escherichia coli has provided several clues to the underlying mechanisms of these rearrangements. We present evidence for three mechanisms of RecA-independent sequence rearrangements: simple replication slippage, sister-chromosome exchange-associated slippage, and single-strand annealing. We discuss the constraints of these mechanisms and contrast their properties with RecA-dependent homologous recombination. Replication plays a critical role in the two slipped misalignment mechanisms, and difficulties in replication appear to trigger rearrangements via all these mechanisms.

The whole genome sequence (1.83 Mbp) of Haemophilus influenzae strain Rd was searched to identify tandem oligonucleotide repeat sequences. Loss or gain of one or more nucleotide repeats through a recombination-independent slippage mechanism is known to mediate phase variation of surface molecules of pathogenic bacteria, including H. influenzae. This facilitates evasion of host defenses and adaptation to the varying microenvironments of the host. We reasoned that iterative nucleotides could identify novel genes relevant to microbe-host interactions. Our search of the Rd genome sequence identified 9 novel loci with multiple (range 6-36, mean 22) tandem tetranucleotide repeats. All were found to be located within putative open reading frames and included homologues of hemoglobin-binding proteins of Neisseria, a glycosyltransferase (IgtC gene product) of Neisseria, and an adhesin of Yersinia. These tetranucleotide repeat sequences were also shown to be present in two other epidemiologically different H. influenzae type b strains, although the number and distribution of repeats was different. Further characterization of the IgtC gene showed that it was involved in phenotypic switching of a lipopolysaccharide epitope and that this variable expression was associated with changes in the number of tetranucleotide repeats. Mutation of IgtC resulted in attenuated virulence of H. influenzae in an infant rat model of invasive infection. These data indicate the rapidity, economy, and completeness with which whole genome sequences can be used to investigate the biology of pathogenic bacteria.

A functional methyl-directed mismatch repair pathway in Escherichia coli prevents the formation of deletions between 101-bp tandem repeats with 4% sequence divergence. Deletions between perfectly homologous repeats are unaffected. Deletion in both cases occurs independently of the homologous recombination gene, recA. Because the methyl-directed mismatch repair pathway detects and excises one strand of a mispaired duplex, an intermediate for RecA-independent deletion of tandem repeats must therefore be a heteroduplex formed between strands of each repeat. We find that MutH endonuclease, which in vivo incises specifically the newly replicated strand of DNA, and the Dam methylase, the source of this strand-discrimination, are required absolutely for the exclusion of "homeologous" (imperfectly homologous) tandem deletion. This supports the idea that the heteroduplex intermediate for deletion occurs during or shortly after DNA replication in the context of hemi-methylation. Our findings confirm a "replication slippage" model for deletion formation whereby the displacement and misalignment of the nascent strand relative to the repeated sequence in the template strand accomplishes the deletion.

Formation of deletions by recombination between short direct repeats is thought to involve either a break-join or a copy-choice process. The key step of the latter is slippage of the replication machinery between the repeats. We report that the main replicase of Escherichia coli, DNA polymerase III holoenzyme, slips between two direct repeats of 27 bp that flank an inverted repeat of approximately equal 300bp. Slippage was detected in vitro, on a single-stranded DNA template, in a primer extension assay. It requires the presence of a short (8 bp) G+C-rich sequence at the base of a hairpin that can form by annealing of the inverted repeats. It is stimulated by (i) high salt concentration, which might stabilize the hairpin, and (ii) two proteins that ensure the processivity of the DNA polymerase III holoenzyme: the single-stranded DNA binding protein and the beta subunit of the polymerase. Slippage is rather efficient under optimal reaction conditions because it can take place on >50% of template molecules. This observation supports the copy-choice model for recombination between short direct repeats.

Genomic double-strand breaks (DSBs) are key intermediates in recombination reactions of living organisms. We studied the repair of genomic DSBs by homologous sequences in plants. Tobacco plants containing a site for the highly specific restriction enzyme I-Sce I were cotransformed with Agrobacterium strains carrying sequences homologous to the transgene locus and, separately, containing the gene coding for the enzyme. We show that the induction of a DSB can increase the frequency of homologous recombination at a specific locus by up to two orders of magnitude. Analysis of the recombination products demonstrates that a DSB can be repaired via homologous recombination by at least two different but related pathways. In the major pathway, homologies on both sides of the DSB are used, analogous to the conservative DSB repair model originally proposed for meiotic recombination in yeast. Homologous recombination of the minor pathway is restricted to one side of the DSB as described by the nonconservative one-sided invasion model. The sequence of the recombination partners was absolutely conserved in two cases, whereas in a third case, a deletion of 14 bp had occurred, probably due to DNA polymerase slippage during the copy process. The induction of DSB breaks to enhance homologous recombination can be applied for a variety of approaches of plant genome manipulation.

Several human neurological disorders are associated with proteins containing abnormally long runs of glutamine residues. Strikingly, most of these proteins contain two or more additional long runs of amino acids other than glutamine. We screened the current human, mouse, Drosophila, yeast, and Escherichia coli protein sequence data bases and identified all proteins containing multiple long homopeptides. This search found multiple long homopeptides in about 12% of Drosophila proteins but in only about 1.7% of human, mouse, and yeast proteins and none among E. coli proteins. Most of these sequences show other unusual sequence features, including multiple charge clusters and excessive counts of homopeptides of length > or = two amino acid residues. Intriguingly, a large majority of the identified Drosophila proteins are essential developmental proteins and, in particular, most play a role in central nervous system development. Almost half of the human and mouse proteins identified are homeotic homologs. The role of long homopeptides in fine-tuning protein conformation for multiple functional activities is discussed. The relative contributions of strand slippage and of dynamic mutation are also addressed. Several new experiments are proposed.
