Nitrogen-fixing bacterial isolate from the intercellular spaces of tomato root cortical cells was studied for the location of nif genes on the chromosomal or plasmid DNA. The bacterial isolate showed two plasmids of approximate molecular sizes of 220 and 120 kb. Klebsiella pneumoniae nif HDK probe hybridized with the chromosomal DNA and not with the plasmid DNA thereby showing that nif genes are localised on the chromosomal DNA.

The present paper develops a family of explicit algorithms for rotational dynamics and presents their comparison with several existing methods. For rotational motion the configuration space is a non-linear manifold, not a Euclidean vector space. As a consequence the rotation vector and its time derivatives correspond to different tangent spaces of rotation manifold at different time instants. This renders the usual integration algorithms for Euclidean space inapplicable for rotation. In the present algorithms this problem is circumvented by relating the equation of motion to a particular tangent space. It has been accomplished with the help of already existing relation between rotation increments which belongs to two different tangent spaces. The suggested method could in principle make any integration algorithm on Euclidean space, applicable to rotation. However, the present paper is restricted only within explicit Runge-Kutta enabled to handle rotation. The algorithms developed here are explicit and hence computationally cheaper than implicit methods. Moreover, they appear to have much higher local accuracy and hence accurate in predicting any constants of motion for reasonably longer time. The numerical results for solutions as well as constants of motion, indicate superior performance by most of our algorithms, when compared to some of the currently known algorithms, namely ALGO-C1, STW, LIEMID[EA], MCG, SUBCYC-M.

In this paper we discuss the recent progresses in spectral finite element modeling of complex structures and its application in real-time structural health monitoring system based on sensor-actuator network and near real-time computation of Damage Force Indicator (DFI) vector. A waveguide network formalism is developed by mapping the original variational problem into the variational problem involving product spaces of 1D waveguides. Numerical convergence is studied using a h()-refinement scheme, where is the wavelength of interest. Computational issues towards successful implementation of this method with SHM system are discussed.

Hepatic cell culture on a three-dimensional (3D) matrix or as a hepatosphere appears to be a promising in vitro biomimetic system for liver tissue engineering applications. In this study, we have combined the concept of a 3D scaffold and a spheroid culture to develop an in vitro model to engineer liver tissue for drug screening. We have evaluated the potential of poly(ethylene glycol)-alginate-gelatin (PAG) cryogel matrix for in vitro culture of human liver cell lines. The synthesized cryogel matrix has a flow rate of 7 mL/min and water uptake capacity of 94% that enables easy nutrient transportation in the in vitro cell culture. Youngs modulus of 2.4 kPa and viscoelastic property determine the soft and elastic nature of synthesized cryogel. Biocompatibility of PAG cryogel was evaluated through MTT assay of HepG2 and Huh-7 cells on matrices. The proliferation and functionality of the liver cells were enhanced by culturing hepatic cells as spheroids (hepatospheres) on the PAG cryogel using temperature-reversible soluble-insoluble polymer, poly(N-isopropylacrylamide) (PNIPAAm). Pore size of the cryogel above 100 mu m modulated spheroid size that can prevent hypoxia condition within the spheroid culture. Both the hepatic cells have shown a significant difference (P < 0.05) in terms of cell number and functionality when cultured with PNIPAAm. After 10 days of culture using 0.05% PNIPAAm, the cell number increased by 11- and 7-fold in case of HepG2 and Huh-7 cells, respectively. Similarly, after 10 days of hepatic spheroids culture on PAG cryogel, the albumin production, urea secretion, and CYP450 activity were significantly higher in case of culture with PNIPAAm. The developed tissue mass on the PAG cryogel in the presence of PNIPAAm possess polarity, which was confirmed using F-actin staining and by presence of intercellular bile canalicular lumen. The developed cryogel matrix supports liver cells proliferation and functionality and therefore can be used for in vitro and in vivo drug testing.

Campylobacter jejuni is a prevalent cause of food-borne diarrhoeal illness in humans. Understanding of the physiological and metabolic capabilities of the organism is limited. We report a detailed analysis of the C. jejuni growth cycle in batch culture. Combined transcriptomic, phenotypic and metabolic analysis demonstrates a highly dynamic 'stationary phase', characterized by a peak in motility, numerous gene expression changes and substrate switching, despite transcript changes that indicate a metabolic downshift upon the onset of stationary phase. Video tracking of bacterial motility identifies peak activity during stationary phase. Amino acid analysis of culture supernatants shows a preferential order of amino acid utilization. Proton NMR (1H-NMR) highlights an acetate switch mechanism whereby bacteria change from acetate excretion to acetate uptake, most probably in response to depletion of other substrates. Acetate production requires pta (Cj0688) and ackA (Cj0689), although the acs homologue (Cj1537c) is not required. Insertion mutants in Cj0688 and Cj0689 maintain viability less well during the stationary and decline phases of the growth cycle than wild-type C. jejuni, suggesting that these genes, and the acetate pathway, are important for survival.

One of the goals of Knowledge Exchange (www.knowledge-exchange.info) is to help partners to share knowledge and expertise and facilitate the build of expert networks. In the area of digitisation, including non-textual and 3-D-digitisation, a first step is to provide a snapshot of current activities and challenges in the KE partner countries. This paper is a synthesis of the information gathered in a questionnaire that was sent to 15 infrastructure institutions, e.g. libraries and also funders, within the five partner countries of Knowledge Exchange: Denmark (DK), Finland (FIN), Germany (GER), the Netherlands (NL) and the United Kingdom (UK). The paper is based on the answers provided by 6 respondents from four countries: DK o Danish Agency of Culture (Henrik Jarl Hansen) o State and University Library (Tonny Skovgård Jensen) GER o German research foundation, DFG (Franziska Regner) NL o Royal Library (Hildelies Balk) o Leiden University Library (Saskia van Bergen) UK o Jisc (Paola Marchionni, Peter Findlay) The absence of Finnish responses may be due to Finland participating in the recent Enumerate Core Survey II that also addressed digitisation. We have included some of the outcomes of this survey to present a richer picture.

The work presented here represents an 18-month study to examine the relationship between environmental conditions, bacterial load in the water and bacteria levels in tissue macrophages of a range of clinically healthy freshwater fish species, farmed in a range of culture systems in Thailand and Vietnam. Preliminary assessment was made of the clinical significance of the macrophage bacterial load. The aim of this work was to improve production in fresh-water aquaculture through the control of clinical bacterial disease and subclinical infection, and to identify management practices most effective in promoting fish health. We propose a highly efficient content-lossless compression scheme for Chinese document images. The scheme combines morphologic analysis with pattern matching to cluster patterns. In order to achieve the error maps with minimal error numbers, the morphologic analysis is applied to decomposing and recomposing the Chinese character patterns. In the pattern matching, the criteria are adapted to the characteristics of Chinese characters. Since small-size components sometimes can be inserted into the blank spaces of large-size components, we can achieve small-size pattern library images. Arithmetic coding is applied to the final compression. Our method achieves much better compression performance than most alternative methods, and assures content-lossless reconstruction. The aim of this paper is to investigate to what extent the known theory of subdifferentiability and generic differentiability of convex functions defined on open sets can be carried out in the context of convex functions defined on not necessarily open sets. Among the main results obtained I would like to mention a Kenderov type theorem (the subdifferential at a generic point is contained in a sphere), a generic Gâteaux differentiability result in Banach spaces of class S and a generic Fréchet differentiability result in Asplund spaces. At least two methods can be used to prove these results: first, a direct one, and second, a more general one, based on the theory of monotone operators. Since this last theory was previously developed essentially for monotone operators defined on open sets, it was necessary to extend it to the context of monotone operators defined on a larger class of sets, our "quasi open" sets. This is done in Chapter III. As a matter of fact, most of these results have an even more general nature and have roots in the theory of minimal usco maps, as shown in Chapter II.

Understanding the roles of microorganisms in environmental settings by linking phylogenetic identity to metabolic function is a key challenge in delineating their broad-scale impact and functional diversity throughout the biosphere. This work addresses and extends such questions in the context of marine methane seeps, which represent globally relevant conduits for an important greenhouse gas. Through the application and development of a range of culture-independent tools, novel habitats for methanotrophic microbial communities were identified, established settings were characterized in new ways, and potential past conditions amenable to methane-based metabolism were proposed. Biomass abundance and metabolic activity measures – both catabolic and anabolic – demonstrated that authigenic carbonates associated with seep environments retain methanotrophic activity, not only within high-flow seep settings but also in adjacent locations exhibiting no visual evidence of chemosynthetic communities. Across this newly extended habitat, microbial diversity surveys revealed archaeal assemblages that were shaped primarily by seepage activity level and bacterial assemblages influenced more substantially by physical substrate type. In order to reliably measure methane consumption rates in these and other methanotrophic settings, a novel method was developed that traces deuterium atoms from the methane substrate into aqueous medium and uses empirically established scaling factors linked to radiotracer rate techniques to arrive at absolute methane consumption values. Stable isotope probing metaproteomic investigations exposed an array of functional diversity both within and beyond methane oxidation- and sulfate reduction-linked metabolisms, identifying components of each proposed enzyme in both pathways. A core set of commonly occurring unannotated protein products was identified as promising targets for future biochemical investigation. Physicochemical and energetic principles governing anaerobic methane oxidation were incorporated into a reaction transport model that was applied to putative settings on ancient Mars. Many conditions enabled exergonic model reactions, marking the metabolism and its attendant biomarkers as potentially promising targets for future astrobiological investigations. This set of inter-related investigations targeting methane metabolism extends the known and potential habitat of methanotrophic microbial communities and provides a more detailed understanding of their activity and functional diversity.

Nas comunidades, a transmissão se dá via oral. No samba não é diferente. Desde a fundação das escolas de samba, crianças e adolescentes participam ativamente, junto com as suas famílias, das atividades dessas escolas, inclusive do desfile carnavalesco. Essas crianças e adolescentes têm, há décadas, espaços próprios nas escolas de samba: a ala das crianças e mais recentemente escolinhas de mestre-sala e porta-bandeira, e participação em baterias mirins, trazendo perspectiva de profissionalização e de renovação nas próprias escolas e por fim, a criação, a partir de 1980, das escolas de samba mirins, que atualmente abrem o carnaval do Rio de Janeiro. Hoje há 16 escolas, agregadas em uma associação específica, majoritariamente derivadas das escolas mães, que trazem nos desfiles mais de vinte e cinco mil crianças e adolescentes na sexta-feira que antecede o Carnaval. As Escolas de Samba Mirins tentam inserir-se nas políticas sociais para a juventude, principalmente a pobre, para a promoção da cidadania e a revitalização do sentido de comunidade. Fundadas nas áreas mais antigas do Rio de Janeiro, principalmente a área de planejamento três os subúrbios onde se concentram, estas escolas de samba mirins mantém estreito laço com sua vizinhança, estimulando a sociabilidade, as relações intergeracionais e a construção da confiança, fundamental para o surgimento da eficácia coletiva e do desenvolvimento do capital social nestes espaços. Além disso, suprem a ausência de áreas de lazer e equipamentos culturais destes espaços, fortalecendo os laços com os vizinhos e amigos e evitando, de alguma maneira, que o tráfico de drogas violento fragmente ainda mais a vida social e cultural da região. Nesse sentido, as escolas de samba mirins contribuem para a valorização da cultura carioca e se constituem enquanto proposta para promoção da saúde e prevenção da violência, principalmente a gerada pelo tráfico de drogas e a repressão policial contra este tráfico com um caráter desagregador nas vizinhanças onde essas escolas se organizaram originalmente.

The advent of molecular biology has had a dramatic impact on all aspects of biology, not least applied microbial ecology. Microbiological testing of water has traditionally depended largely on culture techniques. Growing understanding that only a small proportion of microbial species are culturable, and that many microorganisms may attain a viable but non-culturable state, has promoted the development of novel approaches to monitoring pathogens in the environment. This has been paralleled by an increased awareness of the surprising genetic diversity of natural microbial populations. By targeting gene sequences that are specific for particular microorganisms, for example genes that encode diagnostic enzymes, or species-specific domains of conserved genes such as 16S ribosomal RNA coding sequences (rrn genes), the problems of culture can be avoided. Technical developments, notably in the area of in vitro amplification of DNA using the polymerase chain reaction (PCR), now permit routine detection and identification of specific microorganisms, even when present in very low numbers. Although the techniques of molecular biology have provided some very powerful tools for environmental microbiology, it should not be forgotten that these have their own drawbacks and biases in sampling. For example, molecular techniques are dependent on efficient lysis and recovery of nucleic acids from both vegetative forms and spores of microbial species that may differ radically when growing in the laboratory compared with the natural environment. Furthermore, PCR amplification can introduce its own bias depending on the nature of the oligonucleotide primers utilised. However, despite these potential caveats, it seems likely that a molecular biological approach, particularly with its potential for automation, will provide the mainstay of diagnostic technology for the foreseeable future.

A análise de arquivos jornalísticos para a formação do conceito de historicidade em torno das obras Memórias de um rato de hotel (1912), de João do Rio, e Bandoleiros (1985), de João Gilberto Noll, foi a nossa primeira contribuição para a formação do objeto autobiografia de ficção, ou autoficção, como também denominamos. A partir desses arquivos de memórias, relativos a dois contextos finisseculares, o XIX e o XX respectivamente, pudemos compreender a historicidade como matriz do nosso objeto autoficção, permeada pelo que Foucault chama de efetividade cotidiana. Essa efetividade do cotidiano é costurada pelo fio da oralidade, que se refere ao elenco das atividades humanas no todo social, tendo como principal característica a ação comunicativa entre os sujeitos. Assim, ligamos a oralidade à historicidade em duas perspectivas complementares: os ditos e os escritos. A primeira diz respeito aos processos da comunicação humana e suas trocas simbólicas, que são projetadas na cultura: os ditos. A segunda se refere ao produto das representações literais do sujeito, grafadas no dorso impresso da memória: os escritos. A memória é a chave de acesso à escrita do si, que se distingue do que chamamos de autoficção, porque nessa última prevalecem as experiências do tempo presente para o futuro. Memórias de um rato de hotel é a escrita de um Eu-autor, contando do cárcere as suas experiências de gatuno no contexto da belle époque carioca. Por meio da memória, ele reconstrói o contexto finissecular, enfatizando a degradação urbana e a decadência ética da burguesia em ascensão, nos tempos da recém-inaugurada República. Na autoficção Bandoleiros, temos o relato vertiginoso de um Eu-narrador, contando do seus fracassos literários e conjugais. Ele é um escritor decadente, transitando nos espaços degradados da pós-modernidade, retirando dessa perambulação o material vivo de seu livro Sol macabro. Nessa autoficção, Noll registra as impressões sobre a realidade dos 1980, focalizando as subjetividades agônicas à margem do capitalismo tardio, num trânsito indômito entre Porto Alegre, Rio de Janeiro e os Estados Unidos. Na nossa tese, compreendemos a realidade como a matéria plástica da autobiografia de ficção. A experiência mundana do escritor é também forte aliada na composição de uma estética subjetiva, ou subjestética, que está para além do auto-retrato narcísico da autobiografia, em suas formas tradicionais

A presente pesquisa que ora se apresenta trata das representações sociais da comunidade cabo-verdiana residente no Rio de Janeiro sobre diversos objetos que estão implicados no processo de construção de suas identidades, no entrecruzamento das culturas e identidades cabo-verdianas e brasileiras. Nesse sentido, realizou-se um estudo comparativo entre três grupos de cabo-verdianos residentes no Rio de Janeiro. As entrevistas realizadas com 20 estudantes, 20 imigrantes e 10 brasileiros filhos de cabo-verdianos residentes no Brasil permitiram identificar conteúdos mais específicos, diversificados e detalhados das representações formadas por eles acerca do Brasil e do povo brasileiro, bem como das suas próprias relações com os países de origem e de acolhimento, bem como com os seus respectivos povos, o que, em última análise, consubstancia as identidades sociais próprias que terão reconstruído durante a sua permanência aquém do ou melhor, de parte do Oceano Atlântico. A teoria das Representações Sociais, desenvolvida por Serge Moscovici (1961) se constituiu em uma valiosa sustentação teórica para o presente estudo. Por meio da pesquisa comparativa das representações sociais dos distintos grupos de cabo-verdianos no Rio de Janeiro acerca de variados aspectos dos seus contextos sócio-culturais de origem e de acolhimento, foi possível compreender o complexo processo de construção, reconstrução e atualização das suas respectivas identidades. É isso que se acredita ter aqui demonstrado no que se refere às trajetórias dos estudantes e imigrantes cabo-verdianos, bem como às histórias de vida dos filhos destes, no Brasil e, em especial, no Rio de Janeiro. É nessa perspectiva que os cabo-verdianos no Rio de Janeiro tomados tanto como sujeitos quanto como objetos de representação, na presente pesquisa têm construído um conhecimento ao mesmo tempo, prático e reflexivo da sua inserção nos contextos brasileiro e carioca, através do contato face a face com a sociedade receptora e, no caso de dois desses grupos (estudantes e imigrantes), a partir das representações que já haviam elaborado no país de origem, sob a influência dos meios de comunicação de massa. Assim, com base na hibridização cultural e identitária dos cabo-verdianos, procurou-se compreender de forma mais ampla e circunstanciada o processo de (re) construção das identidades dos estudantes, imigrantes e dos descendentes destes no Rio de Janeiro. Tais mudanças mostraram-se mais visíveis nos convívios sociais em que os grupos cabo-verdianos participam em diferentes espaços sociais desta cidade, nas próprias residências, nas sedes de associações, as quais participam, não só cabo-verdianos, mas, também, alguns brasileiros e outros africanos. Nestes convívios, notam-se diferenças de relacionamento entre eles, estruturam-se em pequenos grupos, ocorrendo uma intensificação das aproximações identitárias e representacionais. As mudanças que foram observadas superficialmente entre os estudantes são, por conseguinte, mais intensas entre os cabo-verdianos imigrantes, em virtude do maior tempo e freqüência cotidiana da comunicação e da troca de experiências com mais variados estratos da população brasileira, o que se aproxima a uma autêntica fusão cultural. Não obstante, constatou-se que, nesse processo, a comunidade imigrante perdeu alguns elementos importantes da cultura cabo-verdiana, o principal dos quais foi a língua crioula. A perda do crioulo representa uma descontinuidade na reprodução da identidade cabo-verdiana pelos imigrantes no Brasil. Nesse sentido, os brasileiros filhos de pais cabo-verdianos não tiveram acesso a esse poderoso instrumento de comunicação e de preservação da cultura cabo-verdiana. Esses cabo-verdianos imigrantes consideram-se bem sucedidos e avaliam positivamente a sua trajetória de vida enquanto imigrantes no Brasil. De igual modo, os estudantes se consideram realizados, devido à oportunidade de estudar no Brasil, o que representa a concretização de um desejo coletivo, uma vez que a instrução escolar é amplamente valorizada no país de origem.
