Atualmente, o assunto de segurança em fundações tem despertado a atenção de diversos pesquisadores. Muitos entendem ser ultrapassada a utilização, pura e simples, de fatores de segurança em obras geotécnicas. O tratamento determinístico nos projetos de fundações não corresponde às variações e incertezas geradas em todo o processo de investigação do subsolo e processo executivo da fundação, sendo ainda o projeto de fundações um fim de linha de uma longa fila de disciplinas que a precedem e que geram também incertezas nas informações de esforços de suas estruturas e solicitações nas fundações. Porém, são as fundações as primeiras a serem executadas em qualquer obra civil. Este aspecto tem levado à necessidade crescente de verificar também a probabilidade de ruína da fundação por meio da chamada análise de confiabilidade. Com o objetivo de contribuir com a discussão sobre o emprego da análise de confiabilidade, as orientações da norma e o aprimoramento de modelos de cálculo, esta dissertação apresenta, através da aplicação em um estudo de caso, os diferentes aspectos de segurança abordados em projetos de fundações. Procura-se destacar dois tipos básicos de incertezas: aquelas inerentes à variabilidade espacial das propriedades do solo e as decorrentes do modelo de cálculo. Nesta pesquisa são procedidas inicialmente as extrapolações das curvas das provas de carga, que resultarão nos valores da função de Verossimilhança, para a atualização por Bayes. Numa segunda etapa se procede à estimativa da capacidade de carga a priori por alguns métodos consagrados e amplamente utilizados no Brasil. A atualização da estimativa da capacidade de carga é, a seguir, realizada através da metodologia de Bayes, sendo esta uma aplicação de conceitos da probabilidade condicional. A ideia de se proceder a estas duas alternativas de análise à priori foi norteada por duas motivações distintas: i) contribuir para a prática, pelo procedimento de combinação de informações num caso real bem documentado, conforme proposto por Vrouwenvelder (1992) e ii) motivar discussão de opiniões polêmicas de engenheiros de fundações brasileiros em relação à adoção de um fator de segurança inferior a 2 no caso de provas de carga especificadas na fase de projeto.

This paper proposes an HMM-based approach to generating emotional intonation patterns. A set of models were built to represent syllable-length intonation units. In a classification framework, the models were able to detect a sequence of intonation units from raw fundamental frequency values. Using the models in a generative framework, we were able to synthesize smooth and natural sounding pitch contours. As a case study for emotional intonation generation, Maximum Likelihood Linear Regression (MLLR) adaptation was used to transform the neutral model parameters with a small amount of happy and sad speech data. Perceptual tests showed that listeners could identify the speech with the sad intonation 80% of the time. On the other hand, listeners formed a bimodal distribution in their ability to detect the system generated happy intontation and on average listeners were able to detect happy intonation only 46% of the time. © Springer-Verlag Berlin Heidelberg 2005.

Southern bluefin tuna (SBT) (Thunnus maccoyii) growth rates are estimated from tag-return data associated with two time periods, the 1960s and 1980s. The traditional von Bertalanffy growth model (VBG) and a two-phase VBG model were fitted to the data by maximum likelihood. The traditional VBG model did not provide an adequate representation of growth in SBT, and the two-phase VBG yielded a significantly better fit. The results indicated that significant change occurs in the pattern of growth in relation to a VBG curve during the juvenile stages of the SBT life cycle, which may be related to the transition from a tightly schooling fish that spends substantial time in near and surface shore waters to one that is found primarily in more offshore and deeper waters. The results suggest that more complex growth models should be considered for other tunas and for other species that show a marked change in habitat use with age. The likelihood surface for the two-phase VBG model was found to be bimodal and some implications of this are investigated. Significant and substantial differences were found in the growth for fish spawned in the 1960s and in the 1980s, such that after age four there is a difference of about one year in the expected age of a fish of similar length which persists over the size range for which meaningful recapture data are available. This difference may be a density-dependent response as a consequence of the marked reduction in the SBT population. Given the key role that estimates of growth have in most stock assessments, the results indicate that there is a need both for the regular monitoring of growth rates and for provisions for changes in growth over time (possibly related to changes in abundance) in the stock assessment models used for SBT and other species.

As an endangered animal group, musk deer (genus Moschus) are not only a great concern of wildlife conservation, but also of special interest to evolutionary studies due to long-standing arguments on the taxonomic and phylogenetic associations in this group. Using museum samples, we sequenced complete mitochondrial cytochrome b genes (1140 bp) of all suggested species of musk deer in order to reconstruct their phylogenetic history through molecular information. Our results showed that the cytochrome b gene tree is rather robust and concurred for all the algorithms employed (parsimony, maximum likelihood, and distance methods). Further, the relative rate test indicated a constant sequence substitution rate among all the species, permitting the dating of divergence events by molecular clock. According to the molecular topology, M. moschiferus branched off the earliest from a common ancestor of musk deer (about 700,000 years ago); then followed the bifurcation forming the M. berezouskii lineage and the lineage clustering M. fuscus, M. chrysogaster, and M. leucogaster (around 370,000 years before present), interestingly the most recent speciation event in musk deer happened rather recently (140,000 years ago), which might have resulted from the diversified habitats and geographic barriers in southwest China caused by gigantic movements of the Qinghai-Tibetan Plateau in history. Combining the data of current distributions, fossil records, and molecular data of this study, we suggest that the historical dispersion of musk deer might be from north to south in China. Additionally, in our further analyses involving other pecora species, musk deer was strongly supported as a monophyletic group and a valid family in Artiodactyla, closely related to Cervidae. For many realistic scenarios, there are multiple factors that affect the clean speech signal. In this work approaches to handling two such factors, speaker and background noise differences, simultaneously are described. A new adaptation scheme is proposed. Here the acoustic models are first adapted to the target speaker via an MLLR transform. This is followed by adaptation to the target noise environment via model-based vector Taylor series (VTS) compensation. These speaker and noise transforms are jointly estimated, using maximum likelihood. Experiments on the AURORA4 task demonstrate that this adaptation scheme provides improved performance over VTS-based noise adaptation. In addition, this framework enables the speech and noise to be factorised, allowing the speaker transform estimated in one noise condition to be successfully used in a different noise condition. © 2011 IEEE.

Recently there has been interest in structured discriminative models for speech recognition. In these models sentence posteriors are directly modelled, given a set of features extracted from the observation sequence, and hypothesised word sequence. In previous work these discriminative models have been combined with features derived from generative models for noise-robust speech recognition for continuous digits. This paper extends this work to medium to large vocabulary tasks. The form of the score-space extracted using the generative models, and parameter tying of the discriminative model, are both discussed. Update formulae for both conditional maximum likelihood and minimum Bayes' risk training are described. Experimental results are presented on small and medium to large vocabulary noise-corrupted speech recognition tasks: AURORA 2 and 4. © 2011 IEEE.

采用PCR 方法从4 个灵长目物种中获得了泌乳刺激素(Prolactin, PRL)基因的第2~5 外显子的序 列, 结合GenBank 中已有的序列, 计算了PRL 基因在灵长目中的氨基酸替换速率. 对异义替换速率和 同义替换速率进行比较的结果显示, 在灵长目进化的过程中未发现任何一个枝系有正选择作用的迹象. 此外, 用最大似然(Maximum-Likelihood, ML)法没有检验到任何一个受到正选择作用的位点; 在快速 进化时期内发生的所有32 个氨基酸替换中, 只有2 个位点包含在功能重要的40 个氨基酸之内, 提示所 发生的氨基酸替换主要集中在功能上并不是很重要的位点上. 此外, 在PRL 基因的快速进化过程中, PRL 的部分功能被胎盘泌乳刺激素(placental lactogen, PL)所替代. 基于以上原因, 推测发生在灵长目 PRL 基因的快速进化很可能是选择压力的放松, 而不是正选择作用的结果.

We investigated the phylogenetic relationships among most Chinese species of lizards in the genus Phrynocephalus (118 individuals, collected from 56 populations of 14 well-defined species and several unidentified specimens) using four mitochondrial gene fragments (12S rRNA, 16S rRNA, cytochrome b, and ND4-tRNA(LEU)). The partition-homogeneity tests indicated that the combined dataset was homogeneous, and maximum-parsimony (MP), neighbor-joining (NJ), maximum-likelihood (ML) and Bayesian (BI) analyses were performed on this combined dataset (49 haplotypes including outgroups for 2058 bp in total). The maximum-parsimony analysis resulted in 24 equally parsimonious trees, and their strict consensus tree shows that there are two major clades representing the Chinese Phrynocephalus species: the viviparous group (Clade A) and the oviparous group (Clade B). The trees derived from Bayesian, ML. and NJ analyses were topologically identical to the MP analysis except for the position of P. mystaceus. All analyses left the nodes for the oviparous group, the most basal clade within the oviparous group, and P. mystaceus unresolved. The phylogenies further suggest that the monophyly of the viviparous species may have resulted from vicariance, while recent dispersal may have been important in generating the pattern of variation among the oviparous species. In spite of several classification attempts among taxa of the genus Lepus, phylogenetic relationships still remain poorly understood. Here, we present molecular genetic evidence that may resolve some of the current incongruities in the phylogeny of the leporids. The complete mitochondrial cytb, 12S genes, and parts of ND4 and control region fragments were sequenced to examine phylogenetic relationships among Chinese hare taxa and other leporids throughout the World using maximum parsimony, maximum likelihood, and Bayesian phylogenetic reconstruction approaches. Using reconstructed phylogenies, we observed that the Chinese hare is not a single monophyletic group as originally thought. Instead, the data infers that the genus Lepus is monophyletic with three unique species groups: North American, Eurasian, and African. Ancestral area analysis indicated that ancestral Lepus arose in North America and then dispersed into Eurasia via the Bering Land Bridge eventually extending to Africa. Brooks Parsimony analysis showed that dispersal events followed by subsequent speciation have occurred in other geographic areas as well and resulted in the rapid radiation and speciation of Lepus. A Bayesian relaxed molecular clock approach based on the continuous autocorrelation of evolutionary rates along branches estimated the divergence time between the three major groups within Lepus. The genus appears to have arisen approximately 10.76 MYA (+/- 0.86 MYA), with most speciation events occurring during the Pliocene epoch (5.65 +/- 1.15 MYA similar to 1.12 +/- 10.47 MYA). In the present study, we obtained exon 2-5 of prolactin (PRL) gene from four primate species by PCR and sequencing. Adding other genes available in GenBank, we calculate amino acid substitution rates for prolactin gene in primate. Comparison of nonsynonymous substitution rate to synonymous substitution rate ratios shows no evidence of positive selection for any lineage of primate prolactin gene. According to this and the facts that (i) no sites under positive selection are inferred by using maximum-likelihood method; (ii) among 32 amino acid replacement that occurred along the rapid evolutionary phase, only two are included in the 40 functionally important residues, indicating that amino acid replacement tends to occur in those functionally unimportant residues; (iii) partial of prolactin function is replaced by placental lactogen in primate at the rapid evolutionary phase of prolactin gene, we thus deem that it is relaxation of purifying selection to some extent rather than positive selection that enforces the rapid evolution of primate prolactin gene.

The vomeronasal receptor 1 (V1R) are believed to be pheromone receptors in rodents. Here we used computational methods to identify 95 and 62 new putative V1R genes from the draft rat and mouse genome sequence, respectively. The rat V1R repertoire consists of 11 subfamilies, 10 of which are shared with the mouse, while rat appears to lack the H and I subfamilies found in mouse and possesses one unique subfamily (M). The estimations of the relative divergence times suggest that many subfamilies originated after the split of rodents and primates. The analysis also reveals that these clusters underwent an expansion very close to the split of mouse and rat. In addition, maximum likelihood analysis showed that the nonsynonymous and synonymous rate ratio for most of these clusters was much higher than one, suggesting the role of positive selection in the diversification of these duplicated V1R genes. Because V1R are thought to mediate the process of signal transduction in response to pheromone detection, we speculate that the V1R genes have evolved under positive Darwinian selection to maintain the ability to discriminate between large and complex pheromonal mixtures.

More than 10 species within the freshwater fish genus Sinoncyclocbeilus adapt to caves and show different degrees of degeneration of eyes and pigmentation. Therefore, this genus can be useful for studying evolutionary developmental mechanisms, role of natural selection and adaptation in cave animals. To better understand these processes, it is indispensable to have background knowledge about phylogenetic relationships of surface and cave species within this genus. To investigate phylogenetic relationships among species within this genus, we determined nucleotide sequences of complete mitochondrial cytochrome b gene (1140 bp) and partial ND4 gene (1032 bp) of 31 recognized ingroup species and one outgroup species Barbodes laticeps. Phylogenetic trees were reconstructed using maximum parsimony. Bayesian, and maximum likelihood analyses. Our phylogenetic results showed that all species except for two surface species S. jii and S. macrolepis clustered as five major monophyletic clades (I, II, III, IV, and V) with strong supports. S. jii was the most basal species in all analyses, but the position of S. macrolepis was not resolved. The cave species were polyphyletic and occurred in these five major clades. Our results indicate that adaptation to cave environments has occurred multiple times during the evolutionary history of Sinocyclocheilus. The branching orders among the clades I, II, III, and IV were not resolved, and this might be due to early rapid radiation in Sinocyclocheilus. All species distributed in Yunnan except for S. rhinocerous and S. hyalinus formed a strongly supported monophyletic group (clade V), probably reflecting their common origins. This result suggested that the diversification of Sinocyclocheilus in Yunnan may correlate with the uplifting of Yunnan Plateau. © 2005 Published by Elsevier Inc.

Fringillidae is a large and diverse family of Passeriformes. So far, however, Fringillidae relationships deduced from morphological features and by a number of molecular approaches have remained unproven. Recently, much attention has been attracted to mitochondrial tRNA genes, whose sequence and secondary structural characteristics have shown to be useful for Acrodont Lizards and deep-branch phylogenetic studies. In order to identify useful phylogenetic markers and test Fringillidae relationships, we have sequenced three major clusters of mitochondrial tRNA genes from 15 Fringillidae, taxa. A coincident tree, with coturnix as outgroup, was obtained through Maximum-likelihood method using combined dataset of 11 mitochondrial tRNA gene sequences. The result was similar to that through Neighbor-joining but different from Maximum-parsimony methods. Phylogenetic trees constructed with stem-region sequences of 11 genes had many different topologies and lower confidence than with total sequences. On the other hand, some secondary structural characteristics may provide phylogenetic information on relatively short internal branches at under-genus level. In summary, our data indicate that mitochondrial tRNA genes can achieve high confidence on Fringillidae phylogeny at subfamily level, and stem-region sequences may be suitable only at above-family level. Secondary structural characteristics may also be useful to resolve phylogenetic relationship between different genera of Fringillidae with good performance.
