A radiometric assay system has been used to study oxidation patterns of (U-14C) L-amino acids by drug-susceptible and drug-resistant mycobacteria. Drug-susceptible M. tuberculosis (H37Rv TMC 102 and Erdman) along with the drug-resistant organism M. tuberculosis (H37 Rv TMC 303), M. bovis, M. avium, M. intracellulare, M. kansasii and M. chelonei were used. The organisms were inoculated into a sterile reaction system with liquid 7H9 medium and one of the (U-14C) L-amino acids. Each organism displayed a different pattern of amino acid oxidation, but these patterns were not distinctive enough for identification of the organism. Complex amino acids such as proline, phenylalanine and tyrosine were of no use in identification of mycobacteria, since virtually all organisms failed to oxidize them. There was no combination of substrates able to separate susceptible from resistant organisms.

Hamster inoculated intraperitoneally with 1 x 10(7) parasites of L. donovani and L. major-like of the New World were studied in groups of 15, 30, 60 and 90 days of infection. The parasite load and density showed progressive increase with the evolution of the infection and was higher in the L. donovani groups than in the L. major-like groups. The L. major-like groups showed parasite density higher in the spleen than in the liver and was similar in both organs in L. donovani groups. The histopathology showed a diffuse marked hyperplasia and hypertrophy of the reticuloendothelial system with high parasitism in the L. donovani groups while there was focal involvement of these organs in L. major-like groups, forming nodules of macrophages that were scantly parasitised. The biological behaviour could be useful in the preliminary studies of Leishmania strain in regional laboratories and understanding the histopathology of lesions caused by different leishmania species.

Estudaram-se os aspectos histopatológicos relativos à evolução da infecção experimental produzida em Cebus apella (Primates: Cebidae) por Leishmania (V.) lainsoni, L. (V.) braziliensis e L. (L.) amazonensis. O exame microscópico de biópsias seqüênciais, obtidas dos animais a intervalos definidos de tempo (a primeira, à s 48 ou 72 horas após a inoculação, e as seguintes, a cada 30 dias), mostrou que o desenvolvimento das lesões, independentemente da espécie de Leishmania inoculada, passa por uma seqüência de etapas a nÃvel tecidual - 1) infiltrado inespecÃfico crônico; 2) nódulo macrofágico (com numerosos parasitas); 3) necrose das células parasitadas; 4) granuloma epitelióide; 5) absorção da área necrosada (à s vezes formando granuloma de corpo estranho); 6) infiltrado inespecÃfico crônico residual); e 7) cicatrização - que representaria a formação e a resolução das lesões. Discutiram-se também os prováveis mecanismos imunopatológicos que determinam esta seqüência de eventos e sua possÃvel semelhança com a evolução das lesões na leishmaniose tegumentar humana.

Se realizó E.L.I.S.A. con exoantÃgeno de Coccidioides immitis para la detectión de anticuerpos, en 67 sueros humanos diluidos 1/1000, 1/2000, 1/4000 y 1/8000. De los 18 sueros de enfermos de coccidioidomicosis comprobada por examen directo, cultivo y/o histologÃa, 5 fueron negativos, en otros 13 fueron positivos en una o varias diluciones. 3/26 sueros de personas sanas, coccidioidino positivas, fueron positivos en tÃtulos de 1/1000 y el resto no tuvo anticuerpos detectables. No presentaron reacciones positivas ninguno de los sueros controles de personas sanas, pero sà lo hicieron 4/8 pacientes con otras micosis. Se concluye que E.L.I.S.A. es útil para la detección de mÃnimas cantidades de anticuerpos o en sueros que no pueden ser procesados por fijación de complemento. No es recomendable el uso de la técnica en forma aislada por al presencia de reacciones cruzadas.

The effect of peel and seed removal, two commonly practiced procedures either at home or by the processing industry, on the physicochemical properties, bioactive compounds contents and antioxidant capacity of tomato fruits of four typical Portuguese cultivars (cereja, chucha, rama and redondo) were appraised. Both procedures caused significant nutritional and antioxidant activity losses in fruits of every cultivar. In general, peeling was more detrimental, since it caused a higher decrease in lycopene, bcarotene, ascorbic acid and phenolics contents (averages of 71%, 50%, 14%, and 32%, respectively) and significantly lowered the antioxidant capacity of the fruits (8% and 10%, using DPPH. and b-carotene linoleate model assays, correspondingly). Although seeds removal favored the increase of both color and sweetness, some bioactive compounds (11% of carotenoids and 24% of phenolics) as well as antioxidant capacity (5%) were loss. The studied cultivars were differently influenced by these procedures. The fruits most affected by peeling were those from redondo cultivar (-66% lycopene, -44% b-carotene, -26% ascorbic acid and -38% phenolics). Seeds removal, in turn, was more injurious for cereja tomatoes (-10% lycopene, -38% b-carotene, -25% ascorbic acid and -63% phenolics). Comparatively with the remaining ones, the rama fruits were less affected by the trimming procedures.

Similarly to other organisms, Vertebrates changed during the Cenozoic Era. Mammals are the only ones to change quickly; their well mineralised bones and teeth are often fossilised. They are highly diverse and even isolated teeth can be identified. They are thus a good tool for establishing a biochronological framewoork. Among Mammals, Rodents with a short lifetime evolve more quickly than the large Mammals. In Europe, the first elaborated zonation was investigated by the Regional Committee on Neogene Stratigraphy and issued as the MN Zonation (Mammals Neogene) by Mein, 1976. During the following years, progress in knowledge lead to new charts. The latest one resulted from collective work (de Bruijn et al., 1992). Bîochronology gives relative data; if we desire to have numerical age estimates, we must correlate these results with radiometrie data, marine biostratigraphîcal units or the Geomagnetic Polarity Time Scale. For Europe, these results are summarised by Steininger et al. (1989-1996) and Steininger (1999). After some recents developments on MN Zonation, here is discussed the succession of Neogene small mammals Portuguese localities. Fortunately these localities are in majority inbedded in marine context. Their assigment to MN Zones is proposed and correlations with the spanish Aragonian Scale (Daams el al., 1999) are also suggested. In fact, some differences appear between Portugal and Aragonian assemblages, probably for ecological reasons. Therefore, the MN zonation is always useful for short and long distance biochronological correlations.

Suspensões aquosas do pericarpo do fruto, da casca da raiz, das folhas e das sementes de Guaiacum officinale foram testadas como moluscicida, cercaricida e piscicida em diferentes concentrações. Em laboratório, a suspensão do pericarpo do fruto apresentou 100% de mortalidade a 100 ppm para desovas de B. glabrata, a 20 ppm para caramujos adultos de Biomphalaria glabrata, B. straminea e B. tenagophila, a 5 ppm para Lebistes reticulatus (peixes) e a 1 ppm para cercárias de Schistosoma mansoni. O extrato etanólico do pericarpo do fruto não foi ativo para caramujo adulto de B. glabrata. As doses letais para 90% dos caramujos adultos (DL90), após 24 horas de exposição, usando a suspensão do pericarpo do fruto foram de: 15 ppm para B. glabrata; 14 ppm para B. straminea e 18 ppm para B. tenagophila. As DL90 das suspensões das casca da raiz, sementes e folhas contra B. glabrata foram de 57, 33 e 15 ppm, respectivamente. No campo, coma suspensão do pericarpo do fruto a mortalidade de caramujos adultos de B. glabrata foi de 68% a 20 ppm e 100% a 40 ppm

La présente thèse doctorale propose l’étude de l’activité théâtrale développée dans les théâtres permanents construits en Amérique Portugaise au long du XVIIIe et dans la première décennie du XIXe siècle, en mettant l’accent sur les idéalisateurs des Casas da Ópera et les moyens de financement utilisés dans l’édification et l’entretien de celles-ci, le répertoire mis en scène, les artistes composant les compagnies théâtrales, les spectateurs et les possibilités de sociabilité établies dans l’espace théâtrale et les modèles architectoniques qui ont inspiré la construction des bâtiments théâtraux au cours de la période mentionnée. Ces cinq axes principaux soulignent l’approche interdisciplinaire qui a orienté nos études, dans un souci de contribution à une plus complète compréhension de notre sujet de recherche. The intensification of agricultural productivity is an important challenge worldwide. However, environmental stressors can provide challenges to this intensification. The progressive occurrence of the cyanotoxins cylindrospermopsin (CYN) and microcystin-LR (MC-LR) as a potential consequence of eutrophication and climate change is of increasing concern in the agricultural sector because it has been reported that these cyanotoxins exert harmful effects in crop plants. A proteomic-based approach has been shown to be a suitable tool for the detection and identification of the primary responses of organisms exposed to cyanotoxins. The aim of this study was to compare the leaf-proteome profiles of lettuce plants exposed to environmentally relevant concentrations of CYN and a MC-LR/CYN mixture. Lettuce plants were exposed to 1, 10, and 100 lg/l CYN and a MC-LR/CYN mixture for five days. The proteins of lettuce leaves were separated by twodimensional electrophoresis (2-DE), and those that were differentially abundant were then identified by matrix-assisted laser desorption/ionization time of flight-mass spectrometry (MALDI-TOF/TOF MS). The biological functions of the proteins that were most represented in both experiments were photosynthesis and carbon metabolism and stress/defense response. Proteins involved in protein synthesis and signal transduction were also highly observed in the MC-LR/CYN experiment. Although distinct protein abundance patterns were observed in both experiments, the effects appear to be concentration-dependent, and the effects of the mixture were clearly stronger than those of CYN alone. The obtained results highlight the putative tolerance of lettuce to CYN at concentrations up to 100 lg/l. Furthermore, the combination of CYN with MC-LR at low concentrations (1 lg/l) stimulated a significant increase in the fresh weight (fr. wt) of lettuce leaves and at the proteomic level resulted in the increase in abundance of a high number of proteins. In contrast, many proteins exhibited a decrease in abundance or were absent in the gels of the simultaneous exposure to 10 and 100 lg/l MC-LR/CYN. In the latter, also a significant decrease in the fr. wt of lettuce leaves was obtained. These findings provide important insights into the molecular mechanisms of the lettuce response to CYN and MC-LR/CYN and may contribute to the identification of potential protein markers of exposure and proteins that may confer tolerance to CYN and MC-LR/CYN. Furthermore, because lettuce is an important crop worldwide, this study may improve our understanding of the potential impact of these cyanotoxins on its quality traits (e.g., presence of allergenic proteins).
