BACKGROUND: In the alpha subclass of proteobacteria iron homeostasis is controlled by diverse iron responsive regulators. Caulobacter crescentus, an important freshwater α-proteobacterium, uses the ferric uptake repressor (Fur) for such purpose. However, the impact of the iron availability on the C. crescentus transcriptome and an overall perspective of the regulatory networks involved remain unknown. RESULTS: In this work we report the identification of iron-responsive and Fur-regulated genes in C. crescentus using microarray-based global transcriptional analyses. We identified 42 genes that were strongly upregulated both by mutation of fur and by iron limitation condition. Among them, there are genes involved in iron uptake (four TonB-dependent receptor gene clusters, and feoAB), riboflavin biosynthesis and genes encoding hypothetical proteins. Most of these genes are associated with predicted Fur binding sites, implicating them as direct targets of Fur-mediated repression. These data were validated by β-galactosidase and EMSA assays for two operons encoding putative transporters. The role of Fur as a positive regulator is also evident, given that 27 genes were downregulated both by mutation of fur and under low-iron condition. As expected, this group includes many genes involved in energy metabolism, mostly iron-using enzymes. Surprisingly, included in this group are also TonB-dependent receptors genes and the genes fixK, fixT and ftrB encoding an oxygen signaling network required for growth during hypoxia. Bioinformatics analyses suggest that positive regulation by Fur is mainly indirect. In addition to the Fur modulon, iron limitation altered expression of 113 more genes, including induction of genes involved in Fe-S cluster assembly, oxidative stress and heat shock response, as well as repression of genes implicated in amino acid metabolism, chemotaxis and motility. CONCLUSIONS: Using a global transcriptional approach, we determined the C. crescentus iron stimulon. Many but not all of iron responsive genes were directly or indirectly controlled by Fur. The iron limitation stimulon overlaps with other regulatory systems, such as the RpoH and FixK regulons. Altogether, our results showed that adaptation of C. crescentus to iron limitation not only involves increasing the transcription of iron-acquisition systems and decreasing the production of iron-using proteins, but also includes novel genes and regulatory mechanisms

Phospholipase C (PLC) has been known to be a key effector protein in signal transduction pathway for cell proliferation and differentiation. Studies on signalling through the insulin/IGF-1 receptors in muscle differentiation have revealed that PLCγ1 is involved during this process and that both mRNA and protein levels were increased during myogenesis. Based on increasing signal transduction pathways that required both PLCγ1 and PKCε, we investigated its role in insulin stimulation of skeletal muscle differentiation. The precise effects of insulin on specific PKC isoforms are as yet unknown. Insulin stimulation produced a gradual increase in PKCε expression and activation of PKCε through skeletal muscle differentiation. By immunoprecipitation we have demonstrated that endogenous PLCγ1 and PKCε belong to the same immunocomplex that increase during through myogenic differentiation. Furthermore, the SH domain of PLCγ1 is involved in the protein complex and that its confine to the Golgi membrane. PLCγ1 has been involved in cyclin D3 up-regulation. By overexpression and silencing approach we have evidenced that PKCε modulate the espression of cyclin D3; the kinase dead form of PKCε doesn’t maintain the same ability. Using a reporter hGH vector we proved that PKCε acts at transcriptional level by affecting the -37 region of cyclin D3 promoter, as has been described previous for PLCγ1. In summary this data proved the involvement of PKCε in the regulation of cyclin D3 expression, together with PLCγ1.

The quality of astronomical sites is the first step to be considered to have the best performances from the telescopes. In particular, the efficiency of large telescopes in UV, IR, radio etc. is critically dependent on atmospheric transparency. It is well known that the random optical effects induced on the light propagation by turbulent atmosphere also limit telescope’s performances. Nowadays, clear appears the importance to correlate the main atmospheric physical parameters with the optical quality reachable by large aperture telescopes. The sky quality evaluation improved with the introduction of new techniques, new instrumentations and with the understanding of the link between the meteorological (or synoptical parameters and the observational conditions thanks to the application of the theories of electromagnetic waves propagation in turbulent medias: what we actually call astroclimatology. At the present the site campaigns are evolved and are performed using the classical scheme of optical seeing properties, meteorological parameters, sky transparency, sky darkness and cloudiness. New concept are added and are related to the geophysical properties such as seismicity, microseismicity, local variability of the climate, atmospheric conditions related to the ground optical turbulence and ground wind regimes, aerosol presence, use of satellite data. The purpose of this project is to provide reliable methods to analyze the atmospheric properties that affect ground-based optical astronomical observations and to correlate them with the main atmospheric parameters generating turbulence and affecting the photometric accuracy. The first part of the research concerns the analysis and interpretation of longand short-time scale meteorological data at two of the most important astronomical sites located in very different environments: the Paranal Observatory in the Atacama Desert (Chile), and the Observatorio del Roque de Los Muchachos(ORM) located in La Palma (Canary Islands, Spain). The optical properties of airborne dust at ORM have been investigated collecting outdoor data using a ground-based dust monitor. Because of its dryness, Paranal is a suitable observatory for near-IR observations, thus the extinction properties in the spectral range 1.00-2.30 um have been investigated using an empirical method. Furthermore, this PhD research has been developed using several turbulence profilers in the selection of the site for the European Extremely Large Telescope(E-ELT). During the campaigns the properties of the turbulence at different heights at Paranal and in the sites located in northern Chile and Argentina have been studied. This given the possibility to characterize the surface layer turbulence at Paranal and its connection with local meteorological conditions.

In der vorliegenden Arbeit wurde das Imprinting von Genen der Chromosomenregion 11p15.5 des Menschen und des orthologen murinen Abschnitts 7F5 untersucht. Bei der Analyse der humanen Gene H19, IGF2 und KCNQ1OT1 stand deren Regulation durch differentiell methylierte Regionen (DMR) und die Identifizierung von Methylierungsfehlern bei Patienten mit Verdacht auf Beckwith-Wiedemann Syndrom (BWS) im Vordergrund. Hierzu wurden unmethylierte Cytosinnukleotide durch Bisulfitbehandlung in Uracilnukleotide umgewandelt und PCR-amplifizierte DNA-Fragmente sequenziert. Die elterliche Herkunft der Allele wurde mit Hilfe von Einzelnukleotidpolymorphismen (SNP) bestimmt. Während in der H19-Promotorregion in Lymphozyten eine nur tendenziell allelspezifische Methylierung festgestellt werden konnte, wurde im B1-Repeat der H19/IGF2-Region in allen Kontroll- und 20 Patienten-DNAs eine spezifische Methylierung des väterlichen Allels nachgewiesen. In E. coli dient L-Tartrat als Elektronenakzeptor während des anaeroben Wachstums und wird schließlich zu Succinat umgesetzt. Der sekundäre Carrier TtdT (YgjE) von E. coli ist ein Antiporter, der die Aufnahme von L-Tartrat im elektroneutralen Austausch gegen intrazelluläres Succinat katalysiert. TtdT besitzt eine hohe Substratspezifität und katalysiert den Transport von L-Tartrat und Succinat, nicht aber von meso- und D-Tartrat. Das Gen ttdT (ygjE) bildet mit den Genen ttdA und ttdB, welche für die L-Tartratdehydratase kodieren, ein Operon. Das benachbarte Gen ttdR (ygiP) kodiert für TtdR (YgiP), einen Tartrat-spezifischen Regulator vom LysR-Typ. TtdR reguliert die L-Tartratfermentation direkt durch Induktion des ttdABT-Operons und durch Autoregulation. TtdR stellt damit den Tartrat-spezifischen Regulator dar, der auf die Expression des ttdR ttdABT-Genclusters spezialisiert ist. Dagegen reguliert DcuSR, das Zweikomponentensystem für C4-Dicarboxylate, die L-Tartratfermentation indirekt durch die Regulation der Gene für die Fumaratatmung. YfaV und YeaV sind weitere potentielle Tartrattransporter. YfaV katalysiert vermutlich den Transport von C4-Dicarboxylaten, einschließlich Tartrat, unter aeroben und anaeroben Bedingungen. YeaV wird nur in Anwesenheit von L- und meso-Tartrat und unter aeroben Bedingungen gebildet. Die yeaUVWX-Gene unterliegen der trankriptionellen Regulation durch YeaT, dessen Gen yeaT vor yeaU liegt. YeaT ist wie TtdR ein Tartrat-spezifischer Regulator und besitzt eine signifikante Ähnlichkeit zu TtdR.

IL-33 is a novel member of the IL-1 family and ligand for the IL-1 receptor-related protein, ST2. Recent evidence suggests that the IL-33/ST2 axis plays a critical role in several autoimmune and inflammatory disorders; however, its role in inflammatory bowel disease (IBD) has not been clearly defined. We characterized IL-33 and ST2 expression and modulation following conventional anti-TNF therapy in Crohn’s disease and ulcerative colitis (UC) patients, and investigated the role of IL-33 in SAMP1/YitFc (SAMP) mice, a mixed Th1/Th2 model of IBD. Our results showed a specific increase of mucosal IL-33 in active UC, localized primarily to intestinal epithelial cells (IEC) and colonic inflammatory infiltrates. Importantly, increased expression of full-length IL-33, representing the most bioactive form, was detected in UC epithelium, while elevated levels of cleaved IL-33 were present in IBD serum. ST2 isoforms were differentially modulated in UC epithelium and sST2, a soluble decoy receptor with anti-inflammatory properties, was also elevated in IBD serum. Infliximab (anti-TNF) treatment of UC decreased circulating IL-33 and increased sST2, while stimulation of HT-29 IEC confirmed IL-33 and sST2 regulation by TNF. Similarly, IL-33 significantly increased and correlated with disease severity, and potently induced IL-5, IL-6 and IL-17 from mucosal immune cells in SAMP mice. Taken together, the IL-33/ST2 system plays an important role in IBD and experimental colitis, is modulated by anti-TNF therapy, and may represent a specific biomarker for active UC.

This thesis is focused on the study of techniques that allow to have reliable transmission of multimedia content in streaming and broadcasting applications, targeting in particular video content. The design of efficient error-control mechanisms, to enhance video transmission systems reliability, has been addressed considering cross-layer and multi-layer/multi-dimensional channel coding techniques to cope with bit errors as well as packet erasures. Mechanisms for unequal time interleaving have been designed as a viable solution to reduce the impact of errors and erasures by acting on the time diversity of the data flow, thus enhancing robustness against correlated channel impairments. In order to account for the nature of the factors which affect the physical layer channel in the evaluation of FEC schemes performances, an ad-hoc error-event modeling has been devised. In addition, the impact of error correction/protection techniques on the quality perceived by the consumers of video services applications and techniques for objective/subjective quality evaluation have been studied. The applicability and value of the proposed techniques have been tested by considering practical constraints and requirements of real system implementations.

Die Ursachen für die Entstehung von Lungentumoren sind vielseitig. Aus geschädigtem Drüsengewebe der Lunge kann sich die Tumorart des Adenokarzinoms entwickeln, welches zu den malignen Krebserkrankungen gehört und somit nach Etablierung eines Primärtumors metastasieren kann. Es wurde vielfach gezeigt, daß das Immunsystem bei der Bekämpfung eines mutierten Gewebes im fortschreitenden Verlauf des Tumorwachstums an Effektivität verliert. Die dahinter stehenden Mechanismen sind noch nicht ganz verstanden. Eine mögliche Ursache könnte eine fehlerhafte Regulation der Immunabwehr sein. Das Zytokin, welches bei dieser Regulation eine wichtige Rolle spielt, ist das Interleukin-2 (IL-2). Dieses aktiviert immunkompetente Zellen und gewährleistet deren Fortbestand während der Immunreaktion. In der vorliegenden Arbeit ist in einem murinen Modell von Bronchioadenokarzinom die Regulation von CD4+ T-Zellen durch IL-2 untersucht worden, beziehungsweise inwieweit eine Einflußnahme auf diese Regulation zur Verbesserung der Tumorabwehr beitragen kann. Die alpha-Kette des IL-2 Rezeptorkomplexes (CD25) ist neben dem Transkriptionsfaktor Foxp3 ein gängiger Marker für die Population der so genannten regulatorischen T-Zellen. Regulatorische T-Zellen treten im Tumorgewebe in erhöhtem Maße auf und inhibieren die gegen den Tumor gerichtete Effektorfunktion anderer Immunzellen. Durch intranasale Applikation eines anti-CD25 Antikörpers sollte, im speziellen bei den regulatorischen T-Zellen, das CD25 Molekül blockiert werden, um auf diese Weise die hochaffine Signalgebung zu unterbinden und die regulatorischen T-Zellen intratumoral zu depletieren. Es konnte gezeigt werden, daß die Blockade des IL-2 Rezeptors nicht zur Reduktion des Tumorwachstums beitrug. Trotz Applikation des Antikörpers waren die regulatorischen T-Zellen signifikant erhöht. Lediglich die Produktion des Zytokins Tumornekrosisfaktor-alpha (TNF-alpha) wurde durch die Zugabe des Antikörpers gesteigert, was aber keine Verbesserung der Tumorabwehr bewirkte. Als Alternative zur Blockade des IL-2 Rezeptors wurden verschiedene Dosen von rekombinantem IL-2 ebenfalls intranasal appliziert, um die T-Zell Populationen zusätzlich zu stimulieren. In diesem Fall war bei hohen Dosierungen eine Regression des Tumors zu erreichen. Die Regression ist auf eine erhöhte, durch das IL-2 aktivierte Produktion des Zytokins Interferon-gamma (IFN-gamma) zurückzuführen. Jedoch wurde sowohl bei der Blockade des IL-2 Rezeptors, als auch bei der Stimulation durch IL-2 ersichtlich, daß im Zusammenhang mit Adenokarzinom dem Zytokin TNF-alpha eine besondere Position zugedacht werden muß. Es ist bekannt, daß TNF-alpha in verschiedenen experimentellen Tumor-Modellen unterschiedliche Funktionen besitzt. Die Deletion des TNFs, hier dargestellt mittels TNF-knockout Mäusen, hatte eine kurative Wirkung. Die TNF-knockout Mäuse wiesen fast kein Tumorwachstum auf, die CD4+ T-Zellen aus den knockout Mäusen zeigten eine im Vergleich zum Wildtyp mehrfach höhere Produktion von IFN-gamma, bei gleichzeitiger Reduktion der regulatorischen T-Zellen. Es kann vermutet werden, daß TNF-alpha in dem verwendeten Adenokarzinom-Modell eine tumorunterstützende Wirkung hat. Dahingehend wäre die Neutralisierung der TNF-Signalgebung bei zusätzlicher Stimulation mit IL-2 als wirksamer Therapieansatz in Betracht zu ziehen.

Zusammenfassung:rnrnDas Ziel der Arbeit bestand darin mehr über die Funktion des T-Box Transkriptionsfaktors Omb zu erfahren. Dm omb ist der nächste Verwandte zu Hs Tbx2/3, die wegen ihrer Rolle bei verschiedenen Krebsarten für die Entwicklung neuer Therapien bedeutsam sind. rnIn drei, von Herrn Pflugfelder hergestellten, omb Allelen l(1)omb282, l(1)omb12, l(1)omb15 wurden neue Mutationen kartiert. Dabei handelt es sich um zwei missense-Mutationen und eine Stopmutation. Sie betreffen Aminosäurereste, die in allen T-Box Proteinen konserviert sind und daher vermutlich lebenswichtige Proteinabschnitte betreffen. In EMSA Versuchen konnte gezeigt werden, dass die missense-Mutationen die DNA-Bindung des Omb-T Proteins verhindern.rnFür die Suche nach Omb Zielgenen wurden Gene und phylogenetisch konservierte TBE-Genabschnitte auf ihre Regulation durch Omb getestet. Dabei wurde das Expressionsmuster von Genen mitels in situ und das Muster von enhancer getriebener β-Gal Expression histochemisch oder durch Immunfärbung von wildtypischen und l(1)omb15 Larven des dritten Stadiums verglichen. rnUpstream der mirr Transkriptionseinheit wurde ein cis-regulatorisches TBE-Fragment identifiziert, das ein Aktivitätsmuster in Flügelimaginalscheiben zeigte, welches dem von Mirr nahe kommt. Sowohl ein Omb Verlust als auch die Mutation der TBE Sequenz führten zu einer ähnlichen ektopischen Aktivierung des Fragments, was auf eine Abhängigkeit von Omb hinweist. rnIn der intronischen Sequenz von inv wurde ebenfalls ein TBE-Fragment entdeckt, das eine β-Gal-Aktivität in Flügelscheiben des späten L3 Stadiums anterior der A/P Grenze zeigte. Diese Expression könnte sich mit der späten für en/inv beschriebenen Expression (Blair, 1992) decken. Immunfärbungen bestätigten, dass der Verlust dieser Aktivität in omb0 tatsächlich durch den Verlust von Omb hervorgerufen wird und nicht durch eine Entwicklungsverzögerung der Larven verursacht wird.rnSchließlich wurde durch die Reparatur von TBX Expressionsvektoren eine Konstruktreihe (Legler, 2010) fertiggestellt, mit deren Hilfe die Auswirkungen einer Überexpression auf die Zellmotilität in Drosophila untersucht werden kann. Das soll helfen den Einfluss von TBX Proteinen auf die Invasivität von Krebszellen zu verstehen.rn

Triple negative breast cancer (TNBC) is a very aggressive tumor subtype characterized by the lack of expression of estrogen receptor 1 (ESR1), due in the most of cases to an increased expression of DNA methyltransferases (DNMTs) and hypermethylation in CpG islands, resulting in gene silencing. Furthermore, in ESR1- negative breast cancers, androgen receptor (AR) is highly expressed and some studies suggest that it can drive tumor progression and might represent a therapeutic target. A correlation between microRNAs, small non-coding RNAs that regulate gene expression, and DNMTs was investigated in a TNBC cell line to restore a normal methylation pattern of ESR1, leading to its re-expression and conferring again sensitivity to selective estrogen receptor modulators (SERMs). miR-148A and miR-29B were found to be involved in the reduction of the expression of DNMT1 and DNMT3A and in a slight increase of ESR1 expression, but not at protein level. Then, we found a down-regulation of AR by miRs-7, -9, -27a, -27b, -29a, -29b, -29c, -127-3p, -127-5p and -376 at 48h post transfection and an up-regulation by miR-15a and miR-16 at every time considered. We concomitantly investigated a possible increase of Tamoxifen, Herceptin and Metformin sensitivity after AR silencing in MDA-MB 453 and T-47D cell lines. Cells seemed more sensitive when silenced for AR only in MDA-MB-453 at 24h post Tamoxifen treatment. Studies on Metformin have basically confirmed an increase of drug sensitivity due to AR silencing in both cell lines. Analysis of Herceptin showed how MDA-MB 453 samples silenced for AR have a slight decrease in the percentage of proliferating cells, demonstrating a possible increase in the response to treatment. These preliminary data provide the basis for further study of the modulation of the expression of AR by microRNAs and it will be interesting to understand the molecular mechanisms underlying these interactions.

In a prior bioinformatic analysis by Hüyseyin Binbas, potential Tbx targets sequences in wing-related genes have been identified. Guided by this information, enhancer trap/reporter lacZ insertions were characterized by X-gal staining first in wildtype and then in l(1)omb imaginal discs.rnIn several lines I observed an increase in reporter expression in a l(1)omb mutant background. Since Omb is assumed to function predominantly as a transcriptional repressor, this may indicate direct regulation. Repression by Omb was observed e.g. for brk and tkv. These genes are negatively regulated by Dpp, while omb is induced by Dpp. Omb which mediates the effects of Dpp on proliferation could, thus, also mediate the Dpp effect on patterning of the wing disc. However, brk and tkv were not completely derepressed in l(1)omb indicating that Dpp represses these genes also by an Omb-independent mechanism.rnMore frequently I observed loss of reporter expression in an l(1)omb mutant background. In these cases, regulation by Omb presumably is indirect. For example, STAT92E-lacZ expression in the wildtype eye was symmetrically expressed at the dorsal and ventral margins. In l(1)omb, ventral expression was selectively lost. Loss of omb is known to cause ventral overproliferation of the eye by activation of the Jak/STAT pathway. STAT92E expression is negatively regulated by Jak/STAT signaling suggesting that loss of omb activates Jak/STAT further upstream in the pathway.rnRegional overproliferation of eye and wing in the l(1)omb mutant background proved a complicating issue in the search for Omb targets. This effect made it difficult to decide whether an expanded reporter expression pattern was due to tissue expansion or reporter gene derepression. For instance hth-lacZ appeared to expand along the ventral eye disc margin in l(1)omb. Without addtional experiments it cannot be concluded whether this is due to de-repression or to activation in association with the proliferative state. Parallel to my experiments, evidence accumulated in our laboratory that loss of omb may attenuate Wg and Hegehog signaling. Since these diffusible proteins are the main patterning molecules in the wing imaginal disc, with dpp being downstream of Hh, many of the observed effects could be secondary to reduced Wg and Hh activity. Examples are ab-lacZ, Dll-lacZ and vgBE-lacZ (reduced expression on the dorso-ventral boundary) and inv-lacZ (late larval expression in the anterior wing disc compartment is lost) or sal-lacZ. Epistasis experiment will be required to clarifiy these issues.rnFurthermore, loss of omb appeared to induce cell fate changes. It was reported previously that in an omb null mutant, the dorsal determinant apterous (ap) is ectopically expressed in the ventral compartment (an effect I did not observe with the strongly hypomorphic l(1)omb15, indicating strong dose dependence). Ventral repression of ap is maintained by epigenetic mechanisms. The patchy and variable nature of ectopic expression of ap or grn-1.1-lacZ points to an effect of omb on epigenetic stability.rnIn the second part of my thesis, an analysis of Omb expression in the Drosophila embryonic ventral nervous system was performed. Omb was found co-expressed with Eve in the medial aCC and RP2 motorneurons as well as the fpCC interneuron and the mediolateral CQ neurons. Additionally, Omb was detected in the Eg positive NB7-3 GW serotonergic motoneuron and the N2-4 neurons. Omb was not found in Repo positive glial cells. During embryonic stage 14, Omb showed some coepression with Dpn or Pros. At the embryonic stage 16, Omb was expressed in minor subset of Mid and Wg positive cells.

Graphene, the thinnest two-dimensional material possible, is considered as a realistic candidate for the numerous applications in electronic, energy storage and conversion devices due to its unique properties, such as high optical transmittance, high conductivity, excellent chemical and thermal stability. However, the electronic and chemical properties of graphene are highly dependent on their preparation methods. Therefore, the development of novel chemical exfoliation process which aims at high yield synthesis of high quality graphene while maintaining good solution processability is of great concern. This thesis focuses on the solution production of high-quality graphene by wet-chemical exfoliation methods and addresses the applications of the chemically exfoliated graphene in organic electronics and energy storage devices.rnPlatinum is the most commonly used catalysts for fuel cells but they suffered from sluggish electron transfer kinetics. On the other hand, heteroatom doped graphene is known to enhance not only electrical conductivity but also long term operation stability. In this regard, a simple synthetic method is developed for the nitrogen doped graphene (NG) preparation. Moreover, iron (Fe) can be incorporated into the synthetic process. As-prepared NG with and without Fe shows excellent catalytic activity and stability compared to that of Pt based catalysts.rnHigh electrical conductivity is one of the most important requirements for the application of graphene in electronic devices. Therefore, for the fabrication of electrically conductive graphene films, a novel methane plasma assisted reduction of GO is developed. The high electrical conductivity of plasma reduced GO films revealed an excellent electrochemical performance in terms of high power and energy densities when used as an electrode in the micro-supercapacitors.rnAlthough, GO can be prepared in bulk scale, large amount of defect density and low electrical conductivity are major drawbacks. To overcome the intrinsic limitation of poor quality of GO and/or reduced GO, a novel protocol is extablished for mass production of high-quality graphene by means of electrochemical exfoliation of graphite. The prepared graphene shows high electrical conductivity, low defect density and good solution processability. Furthermore, when used as electrodes in organic field-effect transistors and/or in supercapacitors, the electrochemically exfoliated graphene shows excellent device performances. The low cost and environment friendly production of such high-quality graphene is of great importance for future generation electronics and energy storage devices. rn

Groundwater represents the most important raw material. Germany struggles to maintain the best water quality possible by providing advanced monitoring systems and legal measures to prevent further pollution. In areas involved in the intensive growing of plantations, one of the major contamination factors derives from nitrate. The aim of this master thesis is the characterisation of the Water Protection Area of Bremen (Germany). Denitrification is a natural process, representing the best means of natural reduction of the hazardous nitrate ion, which is dangerous both for human health and for the development of eutrophication. The study has been possible thanks to the collaboration with the University of Bremen, the Geological Service of Bremen (GDfB) and Peter Spiedt (Water Supply Company of Bremen). It will be defined whether nitrate amounts in the groundwater still overcome the threshold legally imposed, and state if the denitrification process takes place, thanks to new samples collected in 2015 and their integration with historical data. Gas samples have been gathered to test them with the “N2/Ar method”, which is able to estimate the denitrification rate quantitatively. Analyses stated the effective occurrence of the reaction, nevertheless showing that it only affects the chemical of the deep aquifers and not shallow ones. Temporal trends concentrations of nitrate have shown that no real improvement took place in the past years. It will be commented that despite the denitrification being responsible for an efficacious lowering in the nitrate ion, it needs reactive materials to take place. Since the latter are finite elements, it is not an endless process. It is thus believed that is clearly necessary to adopt a better attitude in order to maintain the best chemical qualities possible in such an important area, providing drinking water.

The Gram-positive bacteria Enterococcus hirae, Lactococcus lactis, and Bacillus subtilis have received wide attention in the study of copper homeostasis. Consequently, copper extrusion by ATPases, gene regulation by copper, and intracellular copper chaperoning are understood in some detail. This has provided profound insight into basic principles of how organisms handle copper. It also emerged that many bacterial species may not require copper for life, making copper homeostatic systems pure defense mechanisms. Structural work on copper homeostatic proteins has given insight into copper coordination and bonding and has started to give molecular insight into copper handling in biological systems. Finally, recent biochemical work has shed new light on the mechanism of copper toxicity, which may not primarily be mediated by reactive oxygen radicals.

Reprogramming of gene expression contributes to structural and functional adaptation of muscle tissue in response to altered use. The aim of this study was to investigate mechanisms for observed improvements in leg extension strength, gain in relative thigh muscle mass and loss of body and thigh fat content in response to eccentric and conventional strength training in elderly men (n = 14) and women (n = 14; average age of the men and women: 80.1 ± 3.7 years) by means of structural and molecular analyses. Biopsies were collected from m. vastus lateralis in the resting state before and after 12 weeks of training with two weekly resistance exercise sessions (RET) or eccentric ergometer sessions (EET). Gene expression was analyzed using custom-designed low-density PCR arrays. Muscle ultrastructure was evaluated using EM morphometry. Gain in thigh muscle mass was paralleled by an increase in muscle fiber cross-sectional area (hypertrophy) with RET but not with EET, where muscle growth is likely occurring by the addition of sarcomeres in series or by hyperplasia. The expression of transcripts encoding factors involved in muscle growth, repair and remodeling (e.g., IGF-1, HGF, MYOG, MYH3) was increased to a larger extent after EET than RET. MicroRNA 1 expression was decreased independent of the training modality, and was paralleled by an increased expression of IGF-1 representing a potential target. IGF-1 is a potent promoter of muscle growth, and its regulation by microRNA 1 may have contributed to the gain of muscle mass observed in our subjects. EET depressed genes encoding mitochondrial and metabolic transcripts. The changes of several metabolic and mitochondrial transcripts correlated significantly with changes in mitochondrial volume density. Intramyocellular lipid content was decreased after EET concomitantly with total body fat. Changes in intramyocellular lipid content correlated with changes in body fat content with both RET and EET. In the elderly, RET and EET lead to distinct molecular and structural adaptations which might contribute to the observed small quantitative differences in functional tests and body composition parameters. EET seems to be particularly convenient for the elderly with regard to improvements in body composition and strength but at the expense of reducing muscular oxidative capacity.
