Owing to their important roles in biogeochemical cycles, phytoplankton functional types (PFTs) have been the aim of an increasing number of ocean color algorithms. Yet, none of the existing methods are based on phytoplankton carbon (C) biomass, which is a fundamental biogeochemical and ecological variable and the "unit of accounting" in Earth system models. We present a novel bio-optical algorithm to retrieve size-partitioned phytoplankton carbon from ocean color satellite data. The algorithm is based on existing methods to estimate particle volume from a power-law particle size distribution (PSD). Volume is converted to carbon concentrations using a compilation of allometric relationships. We quantify absolute and fractional biomass in three PFTs based on size - picophytoplankton (0.5-2 µm in diameter), nanophytoplankton (2-20 µm) and microphytoplankton (20-50 µm). The mean spatial distributions of total phytoplankton C biomass and individual PFTs, derived from global SeaWiFS monthly ocean color data, are consistent with current understanding of oceanic ecosystems, i.e., oligotrophic regions are characterized by low biomass and dominance of picoplankton, whereas eutrophic regions have high biomass to which nanoplankton and microplankton contribute relatively larger fractions. Global climatological, spatially integrated phytoplankton carbon biomass standing stock estimates using our PSD-based approach yield - 0.25 Gt of C, consistent with analogous estimates from two other ocean color algorithms and several state-of-the-art Earth system models. Satisfactory in situ closure observed between PSD and POC measurements lends support to the theoretical basis of the PSD-based algorithm. Uncertainty budget analyses indicate that absolute carbon concentration uncertainties are driven by the PSD parameter No which determines particle number concentration to first order, while uncertainties in PFTs' fractional contributions to total C biomass are mostly due to the allometric coefficients. The C algorithm presented here, which is not empirically constrained a priori, partitions biomass in size classes and introduces improvement over the assumptions of the other approaches. However, the range of phytoplankton C biomass spatial variability globally is larger than estimated by any other models considered here, which suggests an empirical correction to the No parameter is needed, based on PSD validation statistics. These corrected absolute carbon biomass concentrations validate well against in situ POC observations.

Background and Objectives: Mobility limitations are a prevalent issue in older adult populations, and an important determinant of disability and mortality. Neighborhood conditions are key determinants of mobility and perception of safety may be one such determinant. Women have more mobility limitations than men, a phenomenon known as the gender mobility gap. The objective of this work was to validate a measure of perception of safety, examine the relationship between neighborhood perception of safety and mobility limitations in seniors, and explore if these effects vary by gender. Methods: This study was cross-sectional, using questionnaire data collected from community-dwelling older adults from four sites in Canada, Colombia, and Brazil. The exposure variable was the neighborhood aggregated Perception of Safety (PoS) scale, derived from the Physical and Social Disorder (PSD) scale by Sampson and Raudenbush. Its construct validity was verified using factor analyses and correlation with similar measures. The Mobility Assessment Tool – short form (MAT-sf), a video-based measure validated cross-culturally in the studied populations, was used to assess mobility limitations. Based on theoretical models, covariates were included in the analysis, both at the neighborhood level (SES, social capital, and built environment) and the individual level (age, gender, education, income, chronic illnesses, depression, cognitive function, BMI, and social participation). Multilevel modeling was used in order to account for neighborhood clustering. Gender specific analyses were carried out. SAS and M-plus were used in this study. Results: PoS was validated across all sites. It loaded in a single factor, after excluding two items, with a Cronbach α value of approximately 0.86. Mobility limitations were present in 22.08% of the sample, 16.32% among men and 27.41% among women. Neighborhood perception of safety was significantly associated with mobility limitations when controlling for all covariates, with an OR of 0.84 (CI 95%: 0.73-0.96), indicating lower odds of having mobility limitations as neighborhood perception of safety improves. Gender did not affect this relationship despite women being more likely to have mobility limitations and live in neighborhoods with poor perception of safety. Conclusion: Neighborhood perception of safety affected the prevalence of mobility limitations in older adults in the studied population.

A presente dissertação é relativa aos assistentes sociais que exerceram funções de deputados em Portugal Continental, na Assembleia Nacional e na Assembleia da República Portuguesa. Procedeu-se à sua caracterização e à análise dos seus principais discursos parlamentares. A pertinência da investigação é justificada pela inexistência de trabalhos acerca da temática escolhida e pela reduzida produção académica no que respeita à vasta dimensão política do Serviço Social em Portugal. A investigação alicerçou-se numa pesquisa bibliográfica sobre os temas em foco, e documental, na análise das fichas biográficas dos deputados identificados e das transcrições textuais das sessões da Assembleia Nacional e da Assembleia da República Portuguesa, onde constam os discursos destes deputados. Estas fontes primárias estão acessíveis no sítio oficial da Assembleia da República (www.parlamento.pt). Pretende-se com este estudo abordar o lugar atribuído à Assembleia Nacional e à Assembleia da República Portuguesa nos respetivos regimes de ditadura e democracia e identificar e caracterizar académica, profissional e politicamente os assistentes sociais que exerceram estas funções. Objetiva-se ainda a análise das principais intervenções destes deputados no contexto da Assembleia Nacional e da Assembleia da República Portuguesa. Apurou-se que o número de deputadas à Assembleia Nacional e à Assembleia da República Portuguesa não variou significativamente nos regimes políticos abordados. Foram identificadas cinco deputadas à Assembleia Nacional, da V à XI legislatura (de 1949 a 1974 – 25 anos), sendo que a primeira deputada identificada exerceu funções ao longo da V e VI legislaturas da Assembleia Nacional. Após a queda do regime ditatorial e depois de 21 anos em democracia, assistentes sociais voltaram a exercer funções de deputados. Entre 1995 e 2011 exerceram funções seis deputados à Assembleia da República Portuguesa. Um deles desenvolveu atividades ao longo da X e XI legislaturas. Os resultados da investigação mostram que a maioria destes deputados são mulheres, havendo apenas um homem. Relativamente à formação académica dos deputados constatou-se que antes de 1974 o estabelecimento que formou mais assistentes sociais deputadas foi o Instituto de Serviço Social de Lisboa (3), seguido do Instituto Superior de Ciências Sociais e Políticas Ultramarinas (1) e do Instituto Superior de Serviço Social Pio XII, em Luanda (1). Os deputados à Assembleia da República formaram-se no Instituto Superior de Serviço Social de Coimbra (2); no Instituto Superior Bissaya Barreto, Coimbra (1); no Instituto Superior de Serviço Social do Porto (1); no Instituto Superior de Serviço Social de Lisboa (1) e no Instituto Superior de Serviço Social Pio XII, Luanda (1). As deputadas que exerceram funções durante o Estado Novo pertenciam à União Nacional e Acção Nacional Popular, sendo que uma delas pertencia à Ala Liberal da ANP. Quanto aos deputados que exerceram funções no período democrático percebe-se que todos eram vinculados a partidos do bloco central e da direita: uma deputada do PSD, uma do CDS e quatro deputados do PS. No que respeita às intervenções parlamentares dos deputados identificados, observou-se que as deputadas à Assembleia Nacional apresentaram e fundamentaram propostas de medidas de política social apoiando-se na sua experiência profissional, principalmente em debates em que o tema foi a família ou a pobreza. Os deputados à Assembleia da República direcionaram os seus discursos para o apoio e/ou complemento das iniciativas propostas pelo governo, pois quatro destes deputados exerceram funções enquanto o partido a que pertenciam estava na liderança do governo. Face á vastidão da temática abordada, a presente dissertação constitui-se como um primeiro contributo para a produção de bibliografia no âmbito da dimensão política do Serviço Social, reconhecendo-se a necessidade de aprofundar mais o tema. / This thesis is on social workers who acted as deputies in Portugal, on the National Assembly and the Assembly of the Portuguese Republic. Characterization and analysis of its key parliamentary speeches was the chosen procedure. The relevance of the research is justified by the lack of previous work on the chosen theme and the reduced academic outputs with respect to the vast political dimension of social work in Portugal. Research has its foundations in a literature research on the topics in focus, and also documentary, by the analysis of the biographical records of Members discussed and the verbatim transcripts of the sessions of the National Assembly and the Assembly of the Portuguese Republic, which contains the speeches of such Members. These primary sources include legislative sessions and are available on the Assembly of the Republic (www.parlamento.pt) official website. The study aims to address the place assigned to the National Assembly and the Assembly of the Portuguese Republic, regarding the respective regimes of dictatorship and democracy and to identify and characterize academically, professionally and politically, the social workers who have exercised these functions. Furthermore on the objective is the analysis of the main interventions in the context of such Members of the National Assembly and the Assembly of the Portuguese Republic. It was found that the number of deputies to the National Assembly and the Assembly of the Portuguese Republic did not significantly vary in both political regimes. Five deputies to the National Assembly were identified on the legislatures V to XI (1949-1974 - 25 years), with the first identified lady holding positions along the V and VI legislatures of the National Assembly. After the fall of the dictatorship and after 21 years of democracy, social workers returned to act as deputies. Between 1995 and 2011 held office six members of the Assembly of the Portuguese Republic. One developed activities along the X and XI legislatures of the Assembly of the Portuguese Republic. Research results show that the majority of these are women, and only one man. Regarding the academic training of the deputies, it was found that before 1974 the establishment that formed most of the social workers was, the Institute of Social Service of Lisbon (3), followed by the School of Social Science and Policy Overseas (1) and the Higher Institute of Social Service Pius XII in Luanda (1). Members of Parliament formed in the Institute of Social Service of Coimbra (2), the Higher Institute Bissaya Barreto, Coimbra (1), the Institute of Social Service of Porto (1), the Higher Institute of Social service of Lisbon (1) and the Institute of Social service Pius XII, Luanda(1). Deputies who held office during the New state belonged to the National Union and National People's Action parties, one of which belonged to the Liberal Wing of the ANP. As for the deputies who held office in the democratic period one realizes that all parties were bound to the central block and right wing: A Member of the PSD, one of CDS and four members of the PS. Regarding the parliamentary speeches of identified Members, it was noted that the National Assembly deputies presented and substantiated proposals for social policy measures relying on their professional experience, primarily on the debates in which the theme was family or poverty. Members of Parliament have directed their speeches to support and / or complement the initiatives proposed by the government, since four of these deputies exercise their functions while the party they belonged was in governmental leadership. Given the vastness of the topic addressed in this thesis, it constitutes itself as a first contribution to the production of literature within the political dimension of social work, recognizing the need to go deeper into the subject.

El trato de las personas en situación de discapacidad (PsD) ha ido variando en la historia de la humanidad desde el infanticidio en la antigüedad hasta la actualidad donde se considera a la persona como un sujeto de derecho que debe ser respetado, no discriminado y con igualdad de oportunidades. El Modelo Multidimensional de la discapacidad (AAIDD, 2011) promueve las habilidades conceptuales de lectura, escritura y matemática considerándolas destrezas fundamentales para la plena inclusión de las personas en la comunidad, y a que les permiten acceder al conocimiento, comunicarse, manejarse en la vida diaria y lograr la inclusión social. ¿Pero que pasa cuando las personas no acceden a la lectoescritura o sus habilidades son tan básicas que no le permiten beneficiarse de ellas en la sociedad? Atendiendo a estas inquietudes nace el interés por estudiar las competencias de las PsD, siendo el objetivo de esta investigación “Analizar las competencias en Comprensión Lectora, Producción de de Textos y Manejo de la Lengua en estudiantes con Discapacidad Intelectual insertos en escuelas especiales de la Región de Los Lagos de Chile”.

This paper discusses the advantages of using a combined environmagnetic and geochemical approach to the provenance and characterization of distal IRDs occurring during the Last Glacial Period in core CI12PC3 from the Galicia Interior Basin (GIB). Six Heinrich layers (HL1-6) have been identified in the area in base to the detection of distinct populations of exotic magnetic mineral assemblages alien to the local/regional sedimentation environment. Their extension has been determined by Ca/Sr and Si/Sr ratios and their provenance by 143Nd/144Nd and 87Sr/86Sr isotopic ratios and FORCs. The sedimentary expression of HL is characterized by the presence of distal Ice Rafted Detritus (IRD). Distal IRD magnetic signatures in the GIB consist of (i) an increase of one order of magnitude in the peak amplitude of magnetic susceptibility from background values, (ii) a general coarsening of the magnetic grain size in a mineral assemblage dominated by titano-magnetites, (iii) FORC distributions pushing towards the coarse MD or PSD component, and (iv) thermomagnetic curves depicting the occurrence of several magnetite phases. These four features are very different from the fine-grained biogenic magnetic assemblages characterized by the combination of lower MS and higher coercivity values that dominate the predominant mixtures of the non-interacting SSD and PSD components in the non-IRD influenced background sedimentation. Our results show that the last 70.000 yr of sedimentation in the GIB were controlled by the relative contribution of local detrital material derived from the Iberian Variscan Chain and IRD alien material from the iceberg melting during the Heinrich Events. They also show two main IRD provenance fields: Europe and Canada. And that the later is more important for for HL1, HL2, HL4 and HL5. FORCs analysis complemented the isotopic information and provided a very unique information, indicating that glacial flour may not always have the same provenance as IRD and that ice-melted derived suspended sediment has its own dynamics and may reach further and/or persists longer than IRD.

Purpose. To identify stroke survivors with symptoms of poststroke depression and the extent of psychiatry needs and care they have received while on physiotherapy rehabilitation. Participants. Fifty stroke survivors (22 females and 28 males) at the outpatient unit of Physiotherapy Department, University of Nigeria Teaching Hospital, Enugu, who gave their informed consent, were randomly selected. Their age range and mean age were 26–66 years and 54.76 ± 8.79 years, respectively. Method. A multiple case study of 50 stroke survivors for symptoms of poststroke depression was done with Beck’s Depression Inventory, mini mental status examination tool, and Modified Motor Assessment Scale. The tests were performed independently by the participants except otherwise stated and scored on a scale of 0–6. Data were analyzed using -test for proportional significance and chi-square test for determining relationship between variables, at p < 0.05. Results. Twenty-one (42.0%) stroke survivors had symptoms of PSD, which was significantly dependent on duration of stroke ( = 21.680, df = 6, and p = 0.001), yet none of the participants had a psychiatry review. Conclusions. Symptoms of PSD may be common in cold compared to new cases of stroke and may need psychiatry care while on physiotherapy rehabilitation.

Le béton conventionnel (BC) a de nombreux problèmes tels que la corrosion de l’acier d'armature et les faibles résistances des constructions en béton. Par conséquent, la plupart des structures fabriquées avec du BC exigent une maintenance fréquent. Le béton fibré à ultra-hautes performances (BFUP) peut être conçu pour éliminer certaines des faiblesses caractéristiques du BC. Le BFUP est défini à travers le monde comme un béton ayant des propriétés mécaniques, de ductilité et de durabilité supérieures. Le BFUP classique comprend entre 800 kg/m³ et 1000 kg/m³ de ciment, de 25 à 35% massique (%m) de fumée de silice (FS), de 0 à 40%m de poudre de quartz (PQ) et 110-140%m de sable de quartz (SQ) (les pourcentages massiques sont basés sur la masse totale en ciment des mélanges). Le BFUP contient des fibres d'acier pour améliorer sa ductilité et sa résistance aux efforts de traction. Les quantités importantes de ciment utilisées pour produire un BFUP affectent non seulement les coûts de production et la consommation de ressources naturelles comme le calcaire, l'argile, le charbon et l'énergie électrique, mais affectent également négativement les dommages sur l'environnement en raison de la production substantielle de gaz à effet de serre dont le gas carbonique (CO[indice inférieur 2]). Par ailleurs, la distribution granulométrique du ciment présente des vides microscopiques qui peuvent être remplis avec des matières plus fines telles que la FS. Par contre, une grande quantité de FS est nécessaire pour combler ces vides uniquement avec de la FS (25 à 30%m du ciment) ce qui engendre des coûts élevés puisqu’il s’agit d’une ressource limitée. Aussi, la FS diminue de manière significative l’ouvrabilité des BFUP en raison de sa surface spécifique Blaine élevée. L’utilisation du PQ et du SQ est également coûteuse et consomme des ressources naturelles importantes. D’ailleurs, les PQ et SQ sont considérés comme des obstacles pour l’utilisation des BFUP à grande échelle dans le marché du béton, car ils ne parviennent pas à satisfaire les exigences environnementales. D’ailleurs, un rapport d'Environnement Canada stipule que le quartz provoque des dommages environnementaux immédiats et à long terme en raison de son effet biologique. Le BFUP est généralement vendu sur le marché comme un produit préemballé, ce qui limite les modifications de conception par l'utilisateur. Il est normalement transporté sur de longues distances, contrairement aux composantes des BC. Ceci contribue également à la génération de gaz à effet de serre et conduit à un coût plus élevé du produit final. Par conséquent, il existe le besoin de développer d’autres matériaux disponibles localement ayant des fonctions similaires pour remplacer partiellement ou totalement la fumée de silice, le sable de quartz ou la poudre de quartz, et donc de réduire la teneur en ciment dans BFUP, tout en ayant des propriétés comparables ou meilleures. De grandes quantités de déchets verre ne peuvent pas être recyclées en raison de leur fragilité, de leur couleur, ou des coûts élevés de recyclage. La plupart des déchets de verre vont dans les sites d'enfouissement, ce qui est indésirable puisqu’il s’agit d’un matériau non biodégradable et donc moins respectueux de l'environnement. Au cours des dernières années, des études ont été réalisées afin d’utiliser des déchets de verre comme ajout cimentaire alternatif (ACA) ou comme granulats ultrafins dans le béton, en fonction de la distribution granulométrique et de la composition chimique de ceux-ci. Cette thèse présente un nouveau type de béton écologique à base de déchets de verre à ultra-hautes performances (BEVUP) développé à l'Université de Sherbrooke. Les bétons ont été conçus à l’aide de déchets verre de particules de tailles variées et de l’optimisation granulaire de la des matrices granulaires et cimentaires. Les BEVUP peuvent être conçus avec une quantité réduite de ciment (400 à 800 kg/m³), de FS (50 à 220 kg/m³), de PQ (0 à 400 kg/m³), et de SQ (0-1200 kg/m³), tout en intégrant divers produits de déchets de verre: du sable de verre (SV) (0-1200 kg/m³) ayant un diamètre moyen (d[indice inférieur 50]) de 275 µm, une grande quantité de poudre de verre (PV) (200-700 kg/m³) ayant un d50 de 11 µm, une teneur modérée de poudre de verre fine (PVF) (50-200 kg/m³) avec d[indice inférieur] 50 de 3,8 µm. Le BEVUP contient également des fibres d'acier (pour augmenter la résistance à la traction et améliorer la ductilité), du superplastifiants (10-60 kg/m³) ainsi qu’un rapport eau-liant (E/L) aussi bas que celui de BFUP. Le remplacement du ciment et des particules de FS avec des particules de verre non-absorbantes et lisse améliore la rhéologie des BEVUP. De plus, l’utilisation de la PVF en remplacement de la FS réduit la surface spécifique totale nette d’un mélange de FS et de PVF. Puisque la surface spécifique nette des particules diminue, la quantité d’eau nécessaire pour lubrifier les surfaces des particules est moindre, ce qui permet d’obtenir un affaissement supérieur pour un même E/L. Aussi, l'utilisation de déchets de verre dans le béton abaisse la chaleur cumulative d'hydratation, ce qui contribue à minimiser le retrait de fissuration potentiel. En fonction de la composition des BEVUP et de la température de cure, ce type de béton peut atteindre des résistances à la compression allant de 130 à 230 MPa, des résistances à la flexion supérieures à 20 MPa, des résistances à la traction supérieure à 10 MPa et un module d'élasticité supérieur à 40 GPa. Les performances mécaniques de BEVUP sont améliorées grâce à la réactivité du verre amorphe, à l'optimisation granulométrique et la densification des mélanges. Les produits de déchets de verre dans les BEVUP ont un comportement pouzzolanique et réagissent avec la portlandite générée par l'hydratation du ciment. Cependant, ceci n’est pas le cas avec le sable de quartz ni la poudre de quartz dans le BFUP classique, qui réagissent à la température élevée de 400 °C. L'addition des déchets de verre améliore la densification de l'interface entre les particules. Les particules de déchets de verre ont une grande rigidité, ce qui augmente le module d'élasticité du béton. Le BEVUP a également une très bonne durabilité. Sa porosité capillaire est très faible, et le matériau est extrêmement résistant à la pénétration d’ions chlorure (≈ 8 coulombs). Sa résistance à l'abrasion (indice de pertes volumiques) est inférieure à 1,3. Le BEVUP ne subit pratiquement aucune détérioration aux cycles de gel-dégel, même après 1000 cycles. Après une évaluation des BEVUP en laboratoire, une mise à l'échelle a été réalisée avec un malaxeur de béton industriel et une validation en chantier avec de la construction de deux passerelles. Les propriétés mécaniques supérieures des BEVUP a permis de concevoir les passerelles avec des sections réduites d’environ de 60% par rapport aux sections faites de BC. Le BEVUP offre plusieurs avantages économiques et environnementaux. Il réduit le coût de production et l’empreinte carbone des structures construites de béton fibré à ultra-hautes performances (BFUP) classique, en utilisant des matériaux disponibles localement. Il réduit les émissions de CO[indice inférieur 2] associées à la production de clinkers de ciment (50% de remplacement du ciment) et utilise efficacement les ressources naturelles. De plus, la production de BEVUP permet de réduire les quantités de déchets de verre stockés ou mis en décharge qui causent des problèmes environnementaux et pourrait permettre de sauver des millions de dollars qui pourraient être dépensés dans le traitement de ces déchets. Enfin, il offre une solution alternative aux entreprises de construction dans la production de BFUP à moindre coût.

Memory storage in the brain involves adjustment of the strength of existing synapses and formation of new neural networks. A key process underlying memory formation is synaptic plasticity, the ability of excitatory synapses to strengthen or weaken their connections in response to patterns of activity between their connected neurons. Synaptic plasticity is governed by the precise pattern of Ca²⁺ influx through postsynaptic N-methyl-D-aspartate-type glutamate receptors (NMDARs), which can lead to the activation of the small GTPases Ras and Rap. Differential activation of Ras and Rap acts to modulate synaptic strength by promoting the insertion or removal of 2-amino-3-(3-hydroxy-5-methyl-isoxazol-4-yl)propanoic acid receptors (AMPARs) from the synapse. Synaptic GTPase activating protein (synGAP) regulates AMPAR levels by catalyzing the inactivation of GTP-bound (active) Ras or Rap. synGAP is positioned in close proximity to the cytoplasmic tail regions of the NMDAR through its association with the PDZ domains of PSD-95. SynGAP’s activity is regulated by the prominent postsynaptic protein kinase, Ca²⁺/calmodulin-dependent protein kinase II (CaMKII) and cyclin-dependent kinase 5 (CDK5), a known binding partner of CaMKII. Modulation of synGAP’s activity by phosphorylation may alter the ratio of active Ras to Rap in spines, thus pushing the spine towards the insertion or removal of AMPARs, subsequently strengthening or weakening the synapse. To date, all biochemical studies of the regulation of synGAP activity by protein kinases have utilized impure preparations of membrane bound synGAP. Here we have clarified the effects of phosphorylation of synGAP on its Ras and Rap GAP activities by preparing and utilizing purified, soluble recombinant synGAP, Ras, Rap, CaMKII, CDK5, PLK2, and CaM. Using mass spectrometry, we have confirmed the presence of previously identified CaMKII and CDK5 sites in synGAP, and have identified novel sites of phosphorylation by CaMKII, CDK5, and PLK2. We have shown that the net effect of phosphorylation of synGAP by CaMKII, CDK5, and PLK2 is an increase in its GAP activity toward HRas and Rap1. In contrast, there is no effect on its GAP activity toward Rap2. Additionally, by assaying the GAP activity of phosphomimetic synGAP mutants, we have been able to hypothesize the effects of CDK5 phosphorylation at specific sites in synGAP. In the course of this work, we also found, unexpectedly, that synGAP is itself a Ca²⁺/CaM binding protein. While Ca²⁺/CaM binding does not directly affect synGAP activity, it causes a conformational change in synGAP that increases the rate of its phosphorylation and exposes additional phosphorylation sites that are inaccessible in the absence of Ca²⁺/CaM.
The postsynaptic density (PSD) is an electron-dense region in excitatory postsynaptic neurons that contains a high concentration of glutamate receptors, cytoskeletal proteins, and associated signaling enzymes. Within the PSD, three major classes of scaffolding molecules function to organize signaling enzymes and glutamate receptors. PDZ domains present in the Shank and PSD-95 scaffolds families serve to physically link AMPARs and NMDARs to signaling molecules in the PSD. Because of the specificity and high affinity of PDZ domains for their ligands, I reasoned that these interacting pairs could provide the core components of an affinity chromatography system, including affinity resins, affinity tags, and elution agents. I show that affinity columns containing the PDZ domains of PSD-95 can be used to purify active PDZ domain-binding proteins to very high purity in a single step. Five heterologously expressed neuronal proteins containing endogenous PDZ domain ligands (NMDAR GluN2B subunit Tail, synGAP, neuronal nitric oxide synthase PDZ domain, cysteine rich interactor of PDZ three and cypin) were purified using PDZ domain resin, with synthetic peptides having the sequences of cognate PDZ domain ligands used as elution agents. I also show that conjugation of PDZ domain-related affinity tags to Proteins Of Interest (POIs) that do not contain endogenous PDZ domains or ligands does not alter protein activity and enables purification of the POIs on PDZ domain-related affinity resins.

Le sujet de cette thèse est la caractérisation dynamique d’une surface générée par plusieurs actionneurs d’un miroir ferrofluidique intégré dans un montage d’optique adaptative. Elle conclut un projet de doctorat au sein du groupe Borra s’étant échelonné sur 4 années. Un bref portrait général de l’optique adaptative est d’abord présenté, suivi d’une section sur la théorie du contrôle. Les types de contrôleurs y sont abordés, soit PID et surcharge. L’effet de la viscosité sur la réponse dynamique du système, ainsi que les analyseurs de front d’onde utilisés sont ensuite expliqués. La section résultats est subdivisée en plusieurs sous-sections ordonnées de façon chronologique. Dans un premier temps, il est question des résultats obtenus dans le cadre d’une caractérisation d’un nouveau miroir de 91 actionneurs fabriqué au sein du groupe. Il est ensuite question des résultats obtenus avec diverses techniques telles le PSD et l’imagerie déclenchée. Il y aura toute une section sur les résultats en vitesse, en fonction de la viscosité du liquide, suivie d’une section sur les simulations réalisées avec Simulink afin de bien cibler les limites du système. Les résultats portant sur la technique de surcharge des actionneurs seront ensuite présentés avec des projections futures. La dernière partie de cette thèse portera sur une innovation apportée par un autre membre du groupe. Nous parlerons de la déposition d’une membrane élastomère réfléchissante et de ses effets sur la dynamique du système.
Développement des bétons autoplaçants à faible teneur en poudre, Éco-BAP: formulation et performance

Abstract : Although concrete is a relatively green material, the astronomical volume of concrete produced worldwide annually places the concrete construction sector among the noticeable contributors to the global warming. The most polluting constituent of concrete is cement due to its production process which releases, on average, 0.83 kg CO[subscript 2] per kg of cement. Self-consolidating concrete (SCC), a type of concrete that can fill in the formwork without external vibration, is a technology that can offer a solution to the sustainability issues of concrete industry. However, all of the workability requirements of SCC originate from a higher powder content (compared to conventional concrete) which can increase both the cost of construction and the environmental impact of SCC for some applications. Ecological SCC, Eco-SCC, is a recent development combing the advantages of SCC and a significantly lower powder content. The maximum powder content of this concrete, intended for building and commercial construction, is limited to 315 kg/m[superscript 3]. Nevertheless, designing Eco-SCC can be challenging since a delicate balance between different ingredients of this concrete is required to secure a satisfactory mixture. In this Ph.D. program, the principal objective is to develop a systematic design method to produce Eco-SCC. Since the particle lattice effect (PLE) is a key parameter to design stable Eco-SCC mixtures and is not well understood, in the first phase of this research, this phenomenon is studied. The focus in this phase is on the effect of particle-size distribution (PSD) on the PLE and stability of model mixtures as well as SCC. In the second phase, the design protocol is developed, and the properties of obtained Eco-SCC mixtures in both fresh and hardened states are evaluated. Since the assessment of robustness is crucial for successful production of concrete on large-scale, in the final phase of this work, the robustness of one the best-performing mixtures of Phase II is examined. It was found that increasing the volume fraction of a stable size-class results in an increase in the stability of that class, which in turn contributes to a higher PLE of the granular skeleton and better stability of the system. It was shown that a continuous PSD in which the volume fraction of each size class is larger than the consecutive coarser class can increase the PLE. Using such PSD was shown to allow for a substantial increase in the fluidity of SCC mixture without compromising the segregation resistance. An index to predict the segregation potential of a suspension of particles in a yield stress fluid was proposed. In the second phase of the dissertation, a five-step design method for Eco-SCC was established. The design protocol started with the determination of powder and water contents followed by the optimization of sand and coarse aggregate volume fractions according to an ideal PSD model (Funk and Dinger). The powder composition was optimized in the third step to minimize the water demand while securing adequate performance in the hardened state. The superplasticizer (SP) content of the mixtures was determined in next step. The last step dealt with the assessment of the global warming potential of the formulated Eco-SCC mixtures. The optimized Eco-SCC mixtures met all the requirements of self-consolidation in the fresh state. The 28-day compressive strength of such mixtures complied with the target range of 25 to 35 MPa. In addition, the mixtures showed sufficient performance in terms of drying shrinkage, electrical resistivity, and frost durability for the intended applications. The eco-performance of the developed mixtures was satisfactory as well. It was demonstrated in the last phase that the robustness of Eco-SCC is generally good with regards to water content variations and coarse aggregate characteristics alterations. Special attention must be paid to the dosage of SP during batching.

Los resultados morfológicos del Programa de Vivienda de Interés Social Ciudadela Desepaz se caracterizan por la desarticulación y baja funcionalidad del espacio urbano. Esta condición urbanística no deriva de una falta de planificación integral, ya que el Municipio de Santiago de Cali evidenció la intención de desarrollar una porción de ciudad bien articulada, sino más bien de un conjunto de factores de los procesos de gestión urbana y urbanística llevados a cabo en este Programa. A partir de una investigación juiciosa de estos procesos, y del análisis morfológico de cada de uno de los proyectos que conformaron la Ciudadela, se definieron 6 factores principales de la gestión que incidieron en que la intención inicial del Municipio no se pudiera materializar.
