Radical cyclization continues to be a central methodology for the preparation of natural products containing heterocyclic rings. Hence, some electrochemical results obtained by cyclic voltammetry and controlled-potential electrolysis in the study of electroreductive intramolecular cyclization of ethyl (2S, 3R)-2-bromo-3-propargyloxy-3-(2’,3’,4’,6’-tetra-O-acetyl-beta-D-glucopyranosyloxy) propanoate (1a), 2-bromo-3-allyloxy-3-(2’,3’,4’,6’-tetra-O-acetyl-beta-D-glucopyranosyloxy)propanoate (1b), 2-bromo-[1-(prop-2-yn-1-yloxy)propyl]benzene (1c) and [1-bromo-2-methoxy-2-(prop-2’-yn-1-yloxy)ethyl]benzene (1d) promoted by (1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetradecane)nickel(I), [Ni(tmc)]+, electrogenerated at glassy carbon cathodes in ethanol and ethanol:water mixtures containing tetraalkylammonium salts, are presented. During controlled-potential electrolyses of solutions containing [Ni(tmc)]2+ and bromoalkoxylated compounds (1) catalytic reduction of the latter proceeds via one-electron cleavage of the carbon–bromine bond to form a radical intermediate that undergoes cyclization to afford the substituted tetrahydrofurans.

We investigated the reductive intramolecular cyclization of bromopropargyl ethers derivatives, catalyzed by electrogenerated (1,4,8,11-tetramethyl-1,4,8,11-tetraaza-cyclotetradecane)nickel(I), [Ni(tmc)]+ as the catalysts in N,N,N-trimethyl-N-(2- hydroxyethyl)ammonium bis(trifluoromethylsulfonyl)imide,[N1 1 1 2(OH)][NTf2] and 1-ethyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide, [C2mim][NTf2] by cyclic voltammetry and controlled-potential electrolysis. The results show that the reaction leads to the formation of the expected cyclic compounds, which are important intermediates in the synthesis of natural products with possible biological activities.

Phenolic acids are present in our diet in different foods. In particular, mushrooms are a good source of these molecules. Due to their bioactive properties, phenolic acids are extensively studied and there is evidence of their role in disease prevention. Nevertheless, in vivo, these compounds are metabolized and circulate in the organism as glucuronated, sulfated and methylated metabolites, displaying higher or lower bioactivity. To clarify the importance of the metabolism of phenolic acids, the knowledge about the bioactivity of the metabolites is extremely important. In this review, chemical features, biosynthesis and bioavailability of phenolic acids are discussed as well as the chemical and enzymatic synthesis of their metabolites. Finally, the metabolites bioactive properties are compared with that of the corresponding parental compounds.

The Euterpe oleracea Mart. (açaí) is a plant from the Amazon region, classified as "super fruit" because of its various functional properties. However, limited investigation has been performed on açaí by-products, such as seeds. Therefore, the aim of this work was to characterized the phenolic compounds of the aqueous extract of açaí seeds and further evaluate its bioactivity (antioxidant and cytotoxic activities. Only proanthocyanidins were detected, being a B-type (epi)catechin tetramer the most abundant; however, procyanidin trimmers were the most predominant form. Açaí seeds extract revealed a high antioxidant (EC50 ranging from 3.6 to 19.4 μg/mL) and cytotoxic activity, being more effective in the cervical carcinoma cell line (HeLa; GI50 = 18 μg/mL); it did not show toxicity for non-tumor cells. Açaí seeds are considered a waste and could have an added economic benefit, through the extraction of natural antioxidants, particularly proanthocyanidins, that could find applications in food and pharmaceutical industries.

Mushrooms contain a multitude of biomolecules with nutritional and/or biological activity. Among the bioactive molecules, phenolic compounds and tocopherols are the most responsible for their antioxidant activity. In the present work, Boletus edulis, Lentinus edodes and Xerocomus badius, three edible mushroom species originated from Poland, were analyzed for their chemical composition and antioxidant activity. Carbohydrates were the most abundant macronutrients, followed by proteins and ash. Fructose, mannitol and trehalose were the prevalent sugars, but glucose was only found in B. edulis. Polyunsaturated fatty acids predominated over mono and saturated fatty acids. Palmitic, oleic and linoleic acids were abundant in the three samples. α- and β- Tocopherols were quantified in all the samples, but γ-tocopherol was only identified in X. badius. Oxalic and fumaric acids were quantified in the three samples; quinic acid was only present in L. edodes, and malic and citric acids were only found in X. badius. p-Hydroxybenzoic, protocatechuic and cinnamic acids were quantified in all the species, while p-coumaric acid was only found in B. edulis. This species and X. badius revealed the highest antioxidant properties, being B. edulis more effective in radicals scavenging activity and reducing power, and X. badius in lipid peroxidation inhibition, which is related with the highest amounts in phenolic compounds and tocopherols, respectively.

This work describes the synthesis and characterisation of Ni(II) complexes of the following neutral bidentate nitrogen ligands containing pyrazole (pz), pyrimidine (pm) and pyridine (py) aromatic rings: 2-pyrazol-1-yl-pyrimidine (pzpm), 2-(4-methyl-pyrazol-1-yl)-pyrimidine (4-Mepzpm), 2-(4-bromo-pyrazol-1-yl)-pyrimidine (4-Brpzpm), 2-(3,5-dimethyl-pyrazol-1-yl)-pyrimidine (pz*pm), 2-pyrazol-1-yl-pyridine (pzpy) and bis(3,5-dimethylpyrazol-1-yl)phenylmethane (bpz*mph). The complexes [NiBr2(pzpm)] (1), [NiBr2(4-Mepzpm)] (2), [NiBr2(4-Brpzpm)] (3), [NiBr2(pz*pm)] (4), [NiBr2(pzpy)] (5) and [NiBr2(bpz*mph)] (6) were tested as catalysts for ethylene polymerisation, in the presence of the cocatalysts methylaluminoxane (MAO) or diethylaluminium chloride (AlEt2Cl), the catalyst systems 1-3/MAO showing moderate to high activities up to the temperature of 20 °C only in the presence of MAO, whereas 4-6/MAO revealed to be inactive. Other related Pd(II) complexes, already reported in previous works, such as [PdClMe(pzpm)], [PdClMe(pz*pm)], [PdClMe(pzpy)] and [PdClMe(bpz*mph)], also showed to be inactive in the polymerisation of ethylene, when activated by MAO or AlEt2Cl. Selected samples of polyethylene products were characterised by GPC/SEC, 1H and 13C NMR and DSC, showing to be low molecular weight polymers with Mn values ranging from ca. 550 to 1500 g mol−1 and unusually low dispersities of 1.2–1.7, with total branching degrees generally varying between 2 and 12%, melting temperatures from 40 to 120 °C and crystallinities from 40 to 70%.

Partition behavior of eight small organic compounds and six proteins was examined in poly(ethylene glycol)-8000sodium sulfate aqueous two-phase systems containing 0.215 M NaCl and 0.5 M osmolyte (sorbitol, sucrose, TMAO) and poly(ethylene glycol)-10000sodium sulfate0.215 M NaCl system, all in 0.01 M sodium phosphate buffer, pH 6.8. The differences between the solvent properties of the coexisting phases (solvent dipolarity/polarizability, hydrogen bond donor acidity, and hydrogen bond acceptor basicity) were characterized with solvatochromic dyes using the solvatochromic comparison method. Differences between the electrostatic properties of the phases were determined by analysis of partitioning of sodium salts of dinitrophenylated (DNP-) amino acids with aliphatic alkyl side-chain. The partition coefficients of all compounds examined (including proteins) were described in terms of solutesolvent interactions. The results obtained in the study show that solutesolvent interactions of nonionic organic compounds and proteins in polyethylene glycolsodium sulfate aqueous two-phase system change in the presence of NaCl additive.

Partition behavior of adenosine and guanine mononucleotides was examined in aqueous dextran-polyethylene glycol (PEG) and PEG-sodium sulfate two-phase systems. The partition coefficients for each series of mononucleotides were analyzed as a functions of the number of phosphate groups and found to be dependent on the nature of nucleic base and on the type of \ATPS\ utilized. It was concluded that an average contribution of a phosphate group into logarithm of partition coefficient of a mononucleotide cannot be used to estimate the difference between the electrostatic properties of the coexisting phases of ATPS. The data obtained in this study were considered together with those for other organic compounds and proteins reported previously, and the linear interrelationship between logarithms of partition coefficients in dextran-PEG, PEG-Na2SO4 and PEG-Na2SO4-0.215 M NaCl (all in 0.01 M Na- or K/Na-phosphate buffer, pH 7.4 or 6.8) was established. Similar relationship was found for the previously reported data for proteins in Dex-PEG, PEG-600-Na2SO4, and PEG-8000-Na2SO4 ATPS. It is suggested that the linear relationships of the kind established in \ATPS\ may be observed for biological properties of compounds as well.

Fat, oils, and grease present in complex wastewater can be readily converted to methane, but the energy potential of these compounds is not always recyclable, due to incomplete degradation of long chain fatty acids (LCFA) released during lipids hydrolysis. Oleate (C18:1) is generally the dominant LCFA in lipid-containing wastewater, and its conversion in anaerobic bioreactors results in palmitate (C16:0) accumulation. The reason why oleate is continuously converted to palmitate without further degradation via β-oxidation is still unknown. In this work, the influence of methanogenic activity in the initial conversion steps of unsaturated LCFA was studied in 10 bioreactors continuously operated with saturated or unsaturated C16- and C18-LCFA, in the presence or absence of the methanogenic inhibitor bromoethanesulfonate (BrES). Saturated Cn-2-LCFA accumulated both in the presence and absence of BrES during the degradation of unsaturated Cn-LCFA, and represented more than 50\% of total LCFA. In the presence of BrES further conversion of saturated intermediates did not proceed, not even when prolonged batch incubation was applied. As the initial steps of unsaturated LCFA degradation proceed uncoupled from methanogenesis, accumulation of saturated LCFA can be expected. Analysis of the active microbial communities suggests a role for facultative anaerobic bacteria in the initial steps of unsaturated LCFA biodegradation. Understanding this role is now imperative to optimize methane production from LCFA.

El objetivo de este trabajo es caracterizar la respuesta de P. putida frente a condiciones ambientales adversas dadas por la presencia del detergente catiónico tetradeciltrimetilamonio (TDTMA). El objetivo final que se persigue es el de utilizar este microorganismo como vehículo en procesos de biorremediación. El proyecto comprende aspectos relacionados con la degradación y con la respuesta adaptativa que le permiten a P. putida tolerar altas concentraciones del biocida. La degradación de TDTMA por P. putida involucra una actividad monooxigenasa, que produce trimetilamina (TMA) y tetradecilalcanal. Parte de la TMA producida es demetilada, por una TMAdehidrogenasa (TMADH), e utilizada por la bacteria como fuente de nitrógeno y parte es acumulada intracelularmente, inhibiendo el crecimiento bacteriano. Considerando la importancia de las oxigenasas y dehidrogenasas en la transformación química de compuestos recalcitrantes, se identificarán los genes responsables de la actividad monooxigenasa y de la TMADH, se caracterizarán las enzimas, lo que permitirá conocer, además, datos evolutivos de las mismas. Teniendo en cuenta que la acumulación intracelular TMA conduce a la degradación parcial del detergente, efecto contrarrestado por la adición de aluminio (Al), se investigarán si otros factores nutricionales participan en el control de la degradación de TDMA por P. putida. Se investigará si el regulador global NtrC, que se activa en respuesta a limitación de nitrógeno, participa en el metabolismo de TDTMA. Se prevé construir mutantes en los genes que codifican para monoxigenasa y TMADH y analizar la respuesta de estas cepas frente al estrés ocasionado por TDTMA y Al. En este proyecto se postula además que los cambios a nivel de fosfolípidos (PL) de membrana son una estrategia de P. putida para sobrevivir en presencia del TDTMA. Para concluir si fosfatidilglicerol es el principal responsable de la adaptación de P. putida frente al estrés ocasionado por TDTMA, se pretenden obtener mutantes afectadas en la biosíntesis de novo de PL, particularmente en cardiolipina sintasa. Paralelamente se estudiará si fosfolipasa D participa en la respuesta, lo que permitirá asignar un rol a esta enzima en procesos de señalización análogos a los que ocurren en organismos eucariotas. En presencia de TDTMA y Al, P. putida responde aumentando el contenido de fosfatidilcolina y posiblemente este PL actúe como un reservorio temporario del ión. Identificar en P. putida los genes que codifican para las enzimas responsables de su biosíntesis, particularmente fosfatidilcolina sintasa y/o fosfolípido N-metiltranferasa, conducirá a conocer el mecanismo por el cual fosfatidilcolina estaría involucrada en la respuesta a Al.

La inhibición en la actividad de ciertas enzimas esenciales puede generar disturbios en la fisiología de algunos organismos como insectos, plantas y microorganismos y en muchos casos puede conducir a su muerte. Por otro lado, la inhibición de estas proteínas logra modificar factores implicados en la manifestación de determinadas enfermedades. Entre las enzimas que muestran estas características podemos mencionar a tirosinasa, p-hidroxifenilpiruvato dioxigenasa (HPPD) y acetilcolinesterasa (AChE). Debido a la necesidad de nuevas drogas con acción inhibidora de las mencionadas enzimas,los investigadores están explorando el mundo vegetal con el fin de obtenerlas,ya que se ha comprobado que las plantas son capaces de sintetizar esta clase de moléculas. Plantas nativas de nuestra región presentan esta propiedad. Continuando con la búsqueda de compuestos bioactivos obtenidos de plantas, se propone en este proyecto obtener nuevos agentes naturales altamente efectivos en inhibir las nombradas enzimas a partir de 100 plantas nativas de la región central de Argentina. Los compuestos aislados pueden ser utilizados directamente o servir como modelo para la síntesis de análogos. En primer lugar se determinará la efectividad de los extractos obtenidos a partir de las plantas seleccionadas como inhibidores de HPPD (utilizando el método del enol-borato) con el fin de seleccionar el más potente. A partir de este extracto y de aquellos seleccionados como más potentes en inhibir tirosinasa y AChE se aislarán, mediante aislamiento bioguiado, e identificarán el/los compuesto/s responsables. Este proceso será llevado a cabo por técnicas cromatográficas y espectroscópicas y el seguimiento de actividad se realizará mediante el método de enol-borato, dopacromo y Ellman para HPPD, tirosinasa y AChE,respectivamente. Posterior a determinar el nivel de actividad (IC50) de cada compuesto aislado se estudiará el posible efecto sinergista que pudieran ejercer al combinarlos entre ellos (si más de un compuesto es aislado de una planta) y con compuestos comerciales. Si los resultados muestran que los extractos ensayados y los metabolitos activos presentes en ellos exhiben alta efectividad en inhibir las enzimas, ellos pueden surgir como agentes terapéuticos eficaces para el tratamiento de ciertas enfermedades que las involucran y de esta manera mejorar la calidad de vida de los pacientes afectados. Estos productos pueden dar lugar a las compañías farmaceúticas a producir drogas no convencionales como nuevas alternativas medicinales. Por otro lado estas sustancias pueden derivar en novedosos herbicidas, antimicrobianos o insecticidas.Dado que todas las plantas propuestas crecen fácilmente en Argentina, la producción de estos medicamentos significaría nuevas fuentes laborales para nuestro país. El hecho de poder obtener y posteriormente utilizar estos productos aumenta en forma sustancial el aprovechamiento que podemos darle a nuestra rica flora nativa.
