Manu National Park of southern Peru is one of the most renowned protected areas in the world, yet large-bodied vertebrate surveys conducted to date have been restricted to Cocha Cashu Biological Station, a research station covering <0.06 percent of the 1.7Mha park. Manu Park is occupied by >460 settled Matsigenka Amerindians, 300-400 isolated Matsigenka, and several, little-known groups of isolated hunter-gatherers, yet the impact of these native Amazonians on game vertebrate populations within the park remains poorly understood. On the basis of 1495 km of standardized line-transect censuses, we present density and biomass estimates for 23 mammal, bird, and reptile species for seven lowland and upland forest sites in Manu Park, including Cocha Cashu. We compare these estimates between hunted and nonhunted sites within Manu Park, and with other Neotropical forest sites. Manu Park safeguards some of the most species-rich and highest biomass assemblages of arboreal and terrestrial mammals ever recorded in Neotropical forests, most likely because of its direct Andean influence and high levels of soil fertility. Relative to Barro Colorado Island, seed predators and arboreal folivores in Manu are rare, and generalist frugivores specializing on mature fruit pulp are abundant. The impact of such a qualitative shift in the vertebrate community on the dynamics of plant regeneration, and therefore, on our understanding of tropical plant ecology, must be profound. Despite a number of external threats, Manu Park continues to serve as a baseline against which other Neotropical forests can be gauged.

Crocodilians are quite vocal relative to other reptile groups, and the alligators are among the most vocal of the crocodilians. The Chinese alligator, Alligator sinensis, is usually solitary but engages in bellowing choruses in certain waters during the mating season. This paper reports the organization of Chinese alligator's bellowing choruses based upon field observations and playback experiments. Alligators of both genders engaged in the choruses, remaining immobile throughout and inclining toward bellowing synchronously (i.e., starting and finishing at about the same time). The choruses lasted about 10 min with abrupt onset and offset. Moreover, playback experiments revealed that both male and female alligators responded equally to bellowing stimuli from the same and opposite sexes and that none of the tested alligators approached the loudspeaker in spite of playback of male or female stimuli. These suggest that Chinese alligators. may not bellow to compete for or attract mates during the choruses. Instead, when their ecological behaviors, namely, dispersed inhabitation, multi-copulation, restricted mating season, etc., are considered, we hypothesize that they may synchronize bellows to enhance group detectability for assembling individuals into certain waters for subsequent copulations. Based on geophysical and geological data in Jiyang depression, the paper has identified main unconformity surfaces (main movement surfaces) and tectonic sequences and established tectonic and strata framework for correlation between different sags. Based on different sorts of structural styles and characteristics of typical structures, the paper summarized characteristics and distribution of deep structures, discussed evolution sequence of structure, analyzed the relation between tectonic evolution and generation of petroleum. The major developments are as following: Six tectonic sequences could be divided from bottom to top in the deep zone of Jiyang depression. These tectonic sequences are Cambrian to Ordovician, Carboniferous to Permian, lower to middle Jurassic, upper Jurassic to lower Cretaceous, upper Cretaceous and Kongdian formation to the fourth member of Shahejie formation. The center of sedimentation and subsidence of tectonic sequences distinguished from each other in seismic profiles is controlled by tectonic movements. Six tectonic evolution stages could be summarized in the deep zone in Jiyang depression. Among these stages, Paleozoic stage is croton sedimentation basin; Indosinian stage, open folds of EW direction are controlled by compression of nearly SN direction in early Indosinian (early to middle Triassic) while fold thrust fault of EW – NWW direction and arch protruding to NNE direction are controlled by strong compression in late Indosinian (latter Triassic); early Yanshanian stage (early to middle Jurassic), in relatively weak movement after Indosinian compressional orogeny, fluviolacustrine is deposited in intermontane basins in the beginning of early Yanshanian and then extensively denudated in the main orogenic phase; middle Yanshanian (late Jurassic to early Cretaceous), strike-slipping basins are wide distribution with extension (negative reversion) of NW – SE direction; latter Yanshanian (late Cretaceous), fold and thrust of NE – NNE direction and positive reversion structure of late Jurassic to early Cretaceous strike-slipping basin are formed by strong compression of NW–SE direction; sedimentation stage of Kongdian formation to the fourth member of Shahejie formation of Cenozoic, half graben basins are formed by extension of SN direction early while uplift is resulted from compression of nearly EW direction latterly. Compression system, extension system and strike-slip system are formed in deep zone of Jiyang depression. According to identifying flower structure of seismic profiles and analysis of leveling layer slice of 3D seismic data and tectonic map of deep tectonic interface, strike-slip structures of deep zone in Jiyang depression are distinguished. In the middle of the Jiyang depression, strike-slip structures extend as SN direction, NNW direction in Huimin sag, but NNE in Zhandong area. Based on map of relict strata thickness, main faults activity and regional tectonic setting, dynamic mechanisms of deep structure are preliminary determination. The main reason is the difference of direction and character of the plate’s movement. Development and rework of multi-stage tectonic effects are benefit for favorable reservoir and structural trap. Based on tectonic development, accumulation conditions of deep sub-sags and exploration achievements in recent years, potential zones of oil-gas reservoir are put forward, such as Dongying sag and Bonan sag.

Estudou-se a ação acaricida dos extratos brutos etanólicos de 14 plantas do Pantanal sobre fêmeas ingurgitadas do carrapato-do-boi. Os bioensaios foram realizados em triplicatas pelo método de imersão das teleóginas e estimadas a produção de ovos, a eclodibilidade e a eficácia dos extratos. Extratos do lenho da raiz e da casca da raiz de Annona dioica, da raiz e da casca do caule de Simarouba versicolor, da raiz de Annona cornifolia e de Duguetia furfuracea tiveram atividade acaricida entre 50% e 100% e resposta dose-dependente. Extratos de Dimorphandra mollis, Magonia pubescens, Protium heptaphyllum, Hyptis crenata, Sebastiana hispida, Aspidosperma australe, Senna occidentalis e de Elyonurus muticus mostraram atividade acaricida baixa ou ausente (0% a 10%). Extratos da parte aérea de A. cornifolia, da planta inteira de Croton glandulosus e da casca do caule de Stryphnodendron obovatum mostraram atividade acaricida intermediária (10% a 20%). A alta atividade acaricida observada nos extratos de A. dioica, A. cornifolia e D. furfuracea indica ação de acetogeninas, moléculas naturais com várias atividades farmacológicas já descritas e presentes, principalmente, nas plantas da família Annonaceae.

Tryptophyllins are a heterogenous group of amphibian skin peptides originally identified in skin extracts of Neotropical leaf frogs, Phyllomedusa sp., by chemical means. Until now, biosynthetic precursor structure and biological activity remain unreported. Here we describe the isolation of a novel, post-translationally modified tryptophyllin, Lys-Pro-Hyp-Ala-Trp-Val-Pro.amide (PdT-1), from the skin secretion of the Mexican leaf frog, Pachymedusa dacnicolor. Using a 3'- and 5'-RACE strategy and an in vitro skin cDNA library, the PdT-1-encoding precursor was cloned and found to consist of an open-reading frame of 62 amino acids with a single copy of PdT-1 located towards the C-terminus. A synthetic replicate of PdT-1 was found to be a potent myoactive agent, relaxing mammalian arterial smooth muscle and contracting small intestinal smooth muscle at nanomolar concentrations. PdT-1 is thus the first amphibian skin tryptophyllin to be pharmacologically characterized and the first whose precursor cDNA has been cloned.

The defensive strategy of amphibians against predator attack relies heavily on the secretion of noxious/toxic chemical cocktails from specialized skin granular glands. Bioactive peptides constitute a major component of secretions in many species and the most complex are produced by neotropical leaf frogs of the sub-family Phyllomedusinae. We recently reported that these skin secretions contain elements of both the granular gland peptidome and transcriptome and that polyadenylated mRNAs constituting the latter are protected from degradation by interactions with endogenous amphipathic peptides. This thus permits parallel amino acid sequencing of peptides and nucleic acid sequencing of cloned precursor transcripts from single lyophilized samples of secretion. Here we report that the protection afforded is sufficiently robust to permit transcriptome studies by cloning of full-length polyadenylated peptide precursor encoding mRNAs from libraries constructed using ambient temperature air-dried skin from recently deceased specimens as source material. The technique was sufficiently sensitive to permit the identification of cDNAs encoding antimicrobial peptides constituted by six different isoforms of phylloseptin and two dermaseptins. Also, for the first time, establishment of the nucleic acid and amino acid sequence of the precursor encoding the phyllomedusine frog skin bradykinin-related peptide, phyllokinin, from cloned cDNA, was achieved. These data unequivocally demonstrate that the granular gland transcriptome persists in air-dried amphibian skin—a finding that may have fundamental implications in the study of archived materials but also in the wider field of molecular biology.

The Neotropical Euglossini (Hymenoptera: Apidae) are important pollinators of many flowering plants, particularly orchids. Lack of highly polymorphic genetic markers for euglossine species has limited the study of their social organization and inbreeding. We therefore developed microsatellite markers for two species, Eulaema nigrita (11 loci) and Euglossa cordata (nine loci), most of which were highly polymorphic in the source species and in a range of related euglossine bees.

Neotropical orchid bees (Euglossini) are conspicuously different from other corbiculate bees (Apinae) in their lack of advanced sociality and in male use of acquired odors (fragrances) as pheromone-analogues. In both contexts, orchid bee mating systems, in particular the number of males a female mates with, are of great interest but are currently unknown. To assess female mating frequency in the genus Euglossa, we obtained nests from three species in Mexico and Panama and genotyped mothers and their brood at microsatellite DNA loci. In 26 out of 29 nests, genotypes of female brood were fully consistent with being descended from a singly mated mother. In nests with more than one adult female present, those adult females were frequently related, with genotypes being consistent with full sister-sister (r = 0.75) or mother-daughter (r = 0.5) relationships. Thus, our genetic data support the notions of female philopatry and nest-reuse in the genus Euglossa. Theoretically, single mating should promote the evolution of eusociality by maximizing the relatedness among individuals in a nest. However, in Euglossini this genetic incentive has not led to the formation of eusocial colonies as in other corbiculate bees, presumably due to differing ecological or physiological selective regimes. Finally, monandry in orchid bees is in agreement with the theory that females select a single best mate based on the male fragrance phenotype, which may contain information on male age, cognitive ability, and competitive strength.

The skin secretions of Neotropical phyllomedusine leaf frogs have proven to be a rich source of biologically-active peptides, including antimicrobials. The major families of antimicrobial peptides (AMPs) reported are the dermaseptins and phylloseptins and the minor families, the dermatoxins, phylloxins, plasticins, distinctins and the medusins. Here, we report a novel AMP of 10 amino acid residues (LRPAILVRIKamide), named balteatide, from the skin secretion of wild Peruvian purple-sided leaf frogs, Phyllomedusa baltea. Balteatide was found to exhibit a 90% sequence identity with sauvatide, a potent myotropic peptide from the skin secretion of Phyllomedusa sauvagei. However, despite both peptides exhibiting only a single amino acid difference (I/T at position 9), sauvatide is devoid of antimicrobial activity and balteatide is devoid of myotropic activity. Balteatide was found to have differential activity against the Gram-positive bacterium, Staphylococcus aureus, the Gram-negative bacterium, Escherichia coli and the yeast, Candida albicans, and unusually for phyllomedusine frog skin AMPs, was most potent (MIC 32 mg/L) against the yeast. Balteatide was also devoid of haemolytic activity up to concentrations of 512 mg/L. Phyllomedusine frog skin secretions thus continue to provide novel AMPs, some of which may provide templates for the rational design of new classes of anti-infective therapeutics.

Apesar da fauna de mamíferos Neotropicais ser uma das mais ricas do mundo, o nosso conhecimento sobre os limites de espécies, distribuições geográficas e relações filogenéticas está ainda agora no seu início. As áreas de transição entre os dois maiores biomas da América do Sul, o Cerrado e a Amazónia, são ainda menos conhecidas. Até ao momento, escassos estudos focaram os pequenos mamíferos destas áreas. Destes estudos, apenas dois apresentam dados taxonómicos e de distribuição geográfica de uma lista de espécies reduzida e, nenhum é focado nos processos evolutivos que conduziram à diversidade destas áreas. O presente trabalho tem como objectivo aumentar o conhecimento básico sobre a diversidade do médio Rio Araguaia, na região central do Brasil, através da amostragem e análise de espécies de pequenos mamíferos, integrando um intenso trabalho de campo, de laboratório e de museu. Desta forma, um total de 22 espécies é registado para o médio Araguaia. De entre estas espécies, descreve-se uma espécie nova de Rhipidomys, regista-se uma espécie não descrita de Thrichomys e uma potencial nova forma de Oligoryzomys, e também se apresenta uma diagnose emendada do obscuro Oecomys cleberi. Para cada espécie, são também descritas as suas características morfológicas e resumem-se os seus aspectos de distribuição geográfica e história natural. Para os quatro géneros acima referidos, são apresentadas as análises filogenéticas que permitem a identificação das espécies. Adicionalmente, os princípios da filogeografia são aplicados para estudar os padrões da distribuição geográfica da diversidade genética de três roedores sigmodontíneos e seis marsupiais didelphídeos. Os resultados obtidos demonstram que o Rio Araguaia forma uma barreira geográfica para espécies especialistas em florestas não-alagáveis; por outro lado, espécies generalistas apresentam partilha de haplótipos em ambas as margens do rio. Argumentamos também que os refúgios florestais e os gradientes poderão ter tido um papel importante para moldar a estrutura genética de populações de pequenos mamíferos no Brasil central. Em suma, os resultados apresentados corroboram a proposição de que a diversidade Neotropical não poderá ser explicada através de um único modelo de especiação e que estes não são mutuamente exclusivos. O entendimento integral dos processos ecológicos e históricos que deram origem à fauna Neotropical, assim como a continuidade de estudos sistemáticos, depende da realização de novas amostragens e consequente enriquecimento dos museus com colecções apropriadas.

A conservação da biodiversidade nunca foi uma assunto tão popular como nas últimas décadas, mas esta popularidade crescente é devida à pior das razões: o passo acelerado da extinção de espécies e habitats. Os ecossistemas tropicais são, ao mesmo tempo, os mais diversos e os mais ameaçados, em parte porque muitos países destas regiões emergem ainda de situações de instabilidade social, económica e política. O Brasil é o maior país Neotropical, onde se encontram alguns dos biomas com maior diversidade e mais ameaçados do planeta. Actualmente, é também um país líder ao nível da planificação e implementação de medidas de conservação da biodiversidade. Vários dos biomas tropicais mais diversos e ameaçados encontram-se em território brasileiro. Dois destes biomas, a Amazónia e o Cerrado, convergem numa região ecotonal sujeita a uma elevada pressão humana, conhecida como o arco do desmatamento. O Araguaia, um dos maiores rios do Brasil, corre ao longo desta paisagem e os efeitos do desmatamento são já evidentes em toda a sua bacia. Por causa do acelerado ritmo de degradação deste ecossistema, torna-se urgente obter uma imagem clara da biodiversidade regional e compreender como e se a estratégia de conservação para esta região é capaz de lidar com as correntes ameaças e alcançar o seu objectivo a longo prazo: conservar a biota regional. Tendo a herpetofauna como grupo-alvo, os nossos objectivos principais foram: aumentar o conhecimento das comunidades de anfíbios e répteis squamata da região do curso médio do Rio Araguaia; compreender a importância deste rio nos padrões intraespecíficos de estrutura e diversidade genética para diferentes espécies com diferentes características ecológicas; avaliar o potencial de diferentes metodologias para o estudo e monitorização da herpetofauna regional. Os nossos resultados revelam que a amostragem continuada e o uso de diferentes técnicas são essenciais para a obtenção de uma imagem precisa da diversidade da herpetofauna local. As comunidades locais de anfíbios e lagartos apresentaram maior riqueza específica na Área de Protecção Ambiental Bananal/Cantão (APABC), uma área tampão, do no Parque Estadual do Cantão (PEC), uma área de conservação estrita. A APABC é caracterizada por uma maior heterogeneidade de habitats e os nosso resultados corroboram a teoria da heterogeneidade espacial e resultados recentes que revelam uma maior diversidade de lagartos nas zonas interfluviais do Cerrado, do que nas matas de galeria. Os resultados aqui apresentados não corroboram a hipótese de que os ecótonos apresentam maior diversidade do que os biomas em redor. Os nossos resultados revelaram ainda que o Rio Araguaia afecta de forma diferente a estrutura genética de várias espécies de anfíbios e lagartos. Estas diferenças poderão estar relacionadas com a ecologia das espécies, nomeadamente com o uso de diferentes habitats, a vagilidade, ou a estratégia alimentar. Sugerimos que a gestão integração de diferentes unidades de conservação, com diferentes estatutos, podem ajudar a preservar melhor a biota regional.

Instead of developing easily degradable, and low-priced insecticides, we are going after highly sophisticated chemicals. Here, an attempt is being made to develop safer formulations of insecticides of botanical origin. Different parts of the plants were chosen based on their use in countryside and villages The dried plant materials were extracted with petroleum ether, and were applied on Tribolium castaneum. The results were statistically analysed. The active principles from Croton tigilium and Leea sambucina, the most potential plants, were isolated using Column Chromatography, TLC, and Hydrolysis. The isolated principles were analysed spectroscopically ( UV-Vis., IR, NMR, and MS ) to identify their chemical nature. The active principles from Leea and Croton were identified as a cholisterate derivative and a phorbol derivative respectively. In order to ascertain the environmental combatibility of the principles, degradation by soil bacteria was studied. The isolated principles were made into three type of formulations using stabilizers .The formulations were applied on Snake gourd semilooper, Pulse beetle, and mosquito larvae. Also the biocidal activity of the formulations was studied. Both Leea derivative and Croton derivative could be formulated effectively and were effective against a variety of pests. They are eco-friendly, as there is no artificial chemicals involved.
