In the Shackleton Range of East Antarctica, garnet-bearing ultramafic rocks occur as lenses in supracrustal high-grade gneisses. In the presence of olivine, garnet is an unmistakable indicator of eclogite facies metamorphic conditions. The eclogite facies assemblages are only present in ultramafic rocks, particularly in pyroxenites, whereas other lithologies - including metabasites - lack such assemblages. We conclude that under high-temperature conditions, pyroxenites preserve high-pressure assemblages better than isofacial metabasites, provided the pressure is high enough to stabilize garnet-olivine assemblages (i.e. >=18-20 kbar). The Shackleton Range ultramafic rocks experienced a clockwise P-T path and peak conditions of 800-850 °C and 23-25 kbar. These conditions correspond to ~70 km depth of burial and a metamorphic gradient of 11-12 °C/km that is typical of a convergent plate-margin setting. The age of metamorphism is defined by two garnet-whole-rock Sm-Nd isochrons that give ages of 525 ± 5 and 520 ± 14 Ma corresponding to the time of the Pan-African orogeny. These results are evidence of a Pan-African suture zone within the northern Shackleton Range. This suture marks the site of a palaeo-subduction zone that likely continues to the Herbert Mountains, where ophiolitic rocks of Neoproterozoic age testify to an ocean basin that was closed during Pan-African collision. The garnet-bearing ultramafic rocks in the Shackleton Range are the first known example of eclogite facies metamorphism in Antarctica that is related to the collision of East and West Gondwana and the first example of Pan-African eclogite facies ultramafic rocks worldwide. Eclogites in the Lanterman Range of the Transantarctic Mountains formed during subduction of the palaeo-Pacific beneath the East Antarctic craton.

The compositions, mineralogies, and textures of gabbros recovered in polymict breccias in Hole 453 indicate that they are the cumulus assemblages of calc-alkalic crystal fractional on that occurred beneath the West Mariana Ridge. They are among a class of gabbros known only from other calc-alkalic associations (e.g., the Lesser Antilles and the Peninsular Ranges batholith of Southern California) and differ from gabbros of stratiform complexes, ophiolites, and the ocean crust. Particularly abundant in the Hole 453 breccias are olivine-bearing gabbros with extremely calcic Plagioclase (An94-97) but with fairly iron-rich olivines (Fo76-77). Other gabbros contain biotite and amphibole and occur in breccias with fairly high-grade greenschist facies (amphibole-chlorite-stilpnomelane) metabasalts. One unusual gabbro has experienced almost complete subsolidus recrystallization to an assemblage of aluminous magnesio-hornblende, anorthite, and green hercynitic spinel. This reaction, the extremely calcic Plagioclase, the occurrence of biotite and amphibole, and the association with greenschist facies metamorphic rocks suggest that crystallization of the gabbros occurred at elevated P(H2O). Comparisons with other calc-alkalic gabbro suites suggest pressures in excess of 4 kbar (about 12 km depth). The gabbros were exposed by the early stages of opening of the Mariana Trough and imply that considerable uplift may have attended rifting. They were also subjected to hydrothermal alteration after breccia formation, resulting in formation of chlorite, epidote, actinolite, and prehnite. Temperatures of at least 200°C - and probably 350°C - were reached, and most likely could not have been attained without extrusion or intrusion of magmas nearby, even though no such rocks were cored.

The composition of 31 samples of Lower Cretaceous (Valanginian to Aptian) sandstone from ODP Sites 638 through 641 was analyzed using the Gazzi-Dickinson point-counting method. The results show that the source of the Valanginian to Hauterivian sand was a continental block, dominated by granitic and/or high-grade-metamorphic rocks. Although these petrologic results do not allow discrimination between various potential continental block provinces, they suggest, in conjunction with seismic profiles and regional considerations, that the source was the Galicia margin or western Iberia. In contrast, the Barremian and Aptian sand is dominated by carbonate grains that were derived from a carbonate platform, probably on Galicia Bank.

We report results from boron, carbon and oxygen stable isotope analyses of faulted and veined rocks recovered by scientific ocean drilling during ODP Leg 180 in the western Woodlark Basin, off Papua New Guinea. In this area of active continental extension, crustal break-up and incipient seafloor spreading, a shallow-dipping, seismically active detachment fault accommodates strain, defining a zone of mylonites and cataclasites, vein formation and fluid infiltration. Syntectonic microstructures and vein-fill mineralogy suggest frictional heating during slip during extension and exhumation of Moresby Seamount. Low carbon and oxygen isotope ratios of calcite veins indicate precipitation from hydrothermal fluids (delta13C PDB down to -17?; delta18O PDB down to -22?) formed by both dehydration and decarbonation. Boron contents are low (<7 ppm), indicating high-grade metamorphic source rock for the fluids. Some of the delta11B signatures (17-35?; parent solutions to calcite vein fills) are low when compared to deep-seated waters in other tectonic environments, likely reflecting preferential loss of 11B during low-grade metamorphism at depth. Pervasive devolatilization and flux of CO2-rich fluids are evident from similar vein cement geochemistry in the detachment fault zone and splays further updip. Multiple rupture-and-healing history of the veins suggests that precipitation may be an important player in fluid pressure evolution and, hence, seismogenic fault movement.

Recent research has discovered high-grade Au ores in NNE-SSW trending shear zones in metamorphic proterozoic and palaeozoic terranes, some 40 km NW of Santiago de Compostela (NW Spain). The orebodies are bound to late-stage Hercynian structures, mainly due to brittle deformation, which are superimposed on earlier ductile shear zones, cutting through various catazonal lithologies, including ortho- and paragneisses, amphibolites, eclogites, and granites. Ore mineralogy, alteration, and ore textures define a frame whose main features are common to all prospects in the area. Main minerals are arsenopyrite and pyrite - accompanied by quartz, adularia, sericite, + (tourmaline, chlorite, carbonates, graphite), as main gangue minerals - with subordinate amounts of boulangerite, bismuthinite, kobellite, jamesonite, chalcopyrite, marcasite, galena, sphalerite, rutile, titanite, scheelite, beryl, fluorite, and minor native gold, electrum, native bismuth, fahlore, pyrrhotite, mackinawite, etc., defining a meso-catathermal paragenesis. Detailed microscopic study allows the author to propose a general descriptive scheme of textural classification for this type of ore. Most of the ores fill open spaces or veins, seal cracks or cement breccias; disseminated ores with replacement features related to alteration (mainly silicification, sericitization, and adularization) are also observed. Intensive and repeated cataclasis is a common feature of many ores, suggesting successive events of brittle deformation, hydrothermal flow, and ore precipitation. Gold may be transported and accumulated in any of these events, but tends to be concentrated in later ones. The origin of the gold ores is explained in terms of hydrotherreal discharge, associated with mainly brittle deformation and possibly related to granitic magmas, in the global tectonic frame of crustal evolution of West Galicia. The mineralogical and textural study suggests some criteria which will be of practical value for exploration and for ore processing. Ore grades can be improved by flotation of arsenopyrite. Non-conventional methods, such as pressure or bacterial leaching, may subsequently obtain a residue enriched in gold.

La heterogeneidad del medio geológico introduce en el proyecto de obra subterránea un alto grado de incertidumbre que debe ser debidamente gestionado a fin de reducir los riesgos asociados, que son fundamentalmente de tipo geotécnico. Entre los principales problemas a los que se enfrenta la Mecánica de Rocas moderna en el ámbito de la construcción subterránea, se encuentran la fluencia de roca en túneles (squeezing) y la rotura de pilares de carbón. Es ampliamente conocido que su aparición causa importantes perjuicios en el coste y la seguridad de los proyectos por lo que su estudio, ha estado tradicionalmente vinculado a la predicción de su ocurrencia. Entre las soluciones existentes para la determinación de estos problemas se encuentran las que se basan en métodos analíticos y numéricos. Estas metodologías son capaces de proporcionar un alto nivel de representatividad respecto del comportamiento geotécnico real, sin embargo, su utilización solo es posible cuando se dispone de una suficiente caracterización geotécnica y por tanto de una detallada definición de los parámetros que alimentan los complejos modelos constitutivos y criterios de rotura que los fenómenos estudiados requieren. Como es lógico, este nivel de definición solo es posible cuando se alcanzan etapas avanzadas de proyecto, incluso durante la propia construcción, a fin de calibrar adecuadamente los parámetros introducidos en los modelos, lo que supone una limitación de uso en etapas iniciales, cuando su predicción tiene verdadero sentido. Por su parte, los métodos empíricos permiten proporcionar soluciones a estos complejos problemas de un modo sencillo, con una baja parametrización y, dado su eminente enfoque observacional, de gran fiabilidad cuando se implementan sobre condiciones de contorno similares a las originales. La sencillez y escasez de los parámetros utilizados permiten a estas metodologías ser utilizadas desde las fases preliminares del proyecto, ya que estos constituyen en general, información habitual de fácil y económica adquisición. Este aspecto permite por tanto incorporar la predicción desde el principio del proceso de diseño, anticipando el riesgo en origen. En esta tesis doctoral, se presenta una nueva metodología empírica que sirve para proporcionar predicciones para la ocurrencia de squeezing y el fallo de pilares de carbón basada en una extensa recopilación de información de casos reales de túneles y minas en las que ambos fenómenos fueron evaluados. Esta información, recogida de referencias bibliográficas de prestigio, ha permitido recopilar una de las más extensas bases de datos existentes hasta la fecha relativa a estos fenómenos, lo que supone en sí mismo una importante contribución sobre el estado del arte. Con toda esta información, y con la ayuda de la teoría de clasificadores estadísticos, se ha implementado sobre las bases de datos un clasificador lineal de tipo regresión logística que permite hacer predicciones sobre la ocurrencia de ambos fenómenos en términos de probabilidad, y por tanto ponderar la incertidumbre asociada a la heterogeneidad incorporada por el medio geológico. Este aspecto del desarrollo es el verdadero valor añadido proporcionado por la tesis y la principal ventaja de la solución propuesta respecto de otras metodologías empíricas. Esta capacidad de ponderación probabilística permite al clasificador constituir una solución muy interesante como metodología para la evaluación de riesgo geotécnico y la toma de decisiones. De hecho, y como ejercicio de validación práctica, se ha implementado la solución desarrollada en un modelo coste-beneficio asociado a la optimización del diseño de pilares involucrados en una de mina “virtual” explotada por tajos largos. La capacidad del clasificador para cuantificar la probabilidad de fallo del diseño, junto con una adecuada cuantificación de las consecuencias de ese fallo, ha permitido definir una ley de riesgo que se ha incorporado al balance de costes y beneficios, que es capaz, a partir del redimensionamiento iterativo del sistema de pilares y de la propia configuración de la mina, maximizar el resultado económico del proyecto minero bajo unas condiciones de seguridad aceptables, fijadas de antemano. Geological media variability introduces to the subterranean project a high grade of uncertainty that should be properly managed with the aim to reduce the associated risks, which are mainly geotechnical. Among the major problems facing the modern Rock Mechanics in the field of underground construction are both, the rock squeezing while tunneling and the failure of coal pillars. Given their harmfulness to the cost and safety of the projects, their study has been traditionally linked to the determination of its occurrence. Among the existing solutions for the determination of these problems are those that are based on analytical and numerical methods. Those methodologies allow providing a high level of reliability of the geotechnical behavior, and therefore a detailed definition of the parameters that feed the complex constitutive models and failure criteria that require the studied phenomena. Obviously, this level of definition is only possible when advanced stages of the project are achieved and even during construction in order to properly calibrate the parameters entered in the models, which suppose a limited use in early stages, when the prediction has true sense. Meanwhile, empirical methods provide solutions to these complex problems in a simple way, with low parameterization and, given his observational scope, with highly reliability when implemented on similar conditions to the original context. The simplicity and scarcity of the parameters used allow these methodologies be applied in the early stages of the project, since that information should be commonly easy and cheaply to get. This aspect can therefore incorporate the prediction from the beginning of the design process, anticipating the risk beforehand. This thesis, based on the extensive data collection of case histories of tunnels and underground mines, presents a novel empirical approach used to provide predictions for the occurrence of both, squeezing and coal pillars failures. The information has been collected from prestigious references, providing one of the largest databases to date concerning phenomena, a fact which provides an important contribution to the state of the art. With all this information, and with the aid of the theory of statistical classifiers, it has been implemented on both databases, a type linear logistic regression classifier that allows predictions about the occurrence of these phenomena in terms of probability, and therefore weighting the uncertainty associated with geological variability. This aspect of the development is the real added value provided by the thesis and the main advantage of the proposed solution over other empirical methodologies. This probabilistic weighting capacity, allows being the classifier a very interesting methodology for the evaluation of geotechnical risk and decision making. In fact, in order to provide a practical validation, we have implemented the developed solution within a cost-benefit analysis associated with the optimization of the design of coal pillar systems involved in a "virtual" longwall mine. The ability of the classifier to quantify the probability of failure of the design along with proper quantification of the consequences of that failure, has allowed defining a risk law which is introduced into the cost-benefits model, which is able, from iterative resizing of the pillar system and the configuration of the mine, maximize the economic performance of the mining project under acceptable safety conditions established beforehand.

Agonist ligands for the nuclear receptor peroxisome proliferator-activated receptor-γ have been shown to induce terminal differentiation of normal preadipocytes and human liposarcoma cells in vitro. Because the differentiation status of liposarcoma is predictive of clinical outcomes, modulation of the differentiation status of a tumor may favorably impact clinical behavior. We have conducted a clinical trial for treatment of patients with advanced liposarcoma by using the peroxisome proliferator-activated receptor-γ ligand troglitazone, in which extensive correlative laboratory studies of tumor differentiation were performed. We report here the results of three patients with intermediate to high-grade liposarcomas in whom troglitazone administration induced histologic and biochemical differentiation in vivo. Biopsies of tumors from each of these patients while on troglitazone demonstrated histologic evidence of extensive lipid accumulation by tumor cells and substantial increases in NMR-detectable tumor triglycerides compared with pretreatment biopsies. In addition, expression of several mRNA transcripts characteristic of differentiation in the adipocyte lineage was induced. There was also a marked reduction in immunohistochemical expression of Ki-67, a marker of cell proliferation. Together, these data indicate that terminal adipocytic differentiation was induced in these malignant tumors by troglitazone. These results indicate that lineage-appropriate differentiation can be induced pharmacologically in a human solid tumor.

Carcinoma of the cervix is one of the most common malignancies. Papanicolaou (Pap) smear tests have reduced mortality by up to 70%. Nevertheless their interpretation is notoriously difficult with high false-negative rates and frequently fatal consequences. We have addressed this problem by using affinity-purified antibodies against human proteins that regulate DNA replication, namely Cdc6 and Mcm5. These antibodies were applied to sections and smears of normal and diseased uterine cervix by using immunoperoxidase or immunofluorescence to detect abnormal precursor malignant cells. Antibodies against Cdc6 and Mcm5 stain abnormal cells in cervical smears and sections with remarkably high specificity and sensitivity. Proliferation markers Ki-67 and proliferating cell nuclear antigen are much less effective. The majority of abnormal precursor malignant cells are stained in both low-grade and high-grade squamous intraepithelial lesions. Immunostaining of cervical smears can be combined with the conventional Pap stain so that all the morphological information from the conventional method is conserved. Thus antibodies against proteins that regulate DNA replication can reduce the high false-negative rate of the Pap smear test and may facilitate mass automated screening.

X-linked lymphoproliferative syndrome (XLP) is an inherited immunodeficiency characterized by increased susceptibility to Epstein–Barr virus (EBV). In affected males, primary EBV infection leads to the uncontrolled proliferation of virus-containing B cells and reactive cytotoxic T cells, often culminating in the development of high-grade lymphoma. The XLP gene has been mapped to chromosome band Xq25 through linkage analysis and the discovery of patients harboring large constitutional genomic deletions. We describe here the presence of small deletions and intragenic mutations that specifically disrupt a gene named DSHP in 6 of 10 unrelated patients with XLP. This gene encodes a predicted protein of 128 amino acids composing a single SH2 domain with extensive homology to the SH2 domain of SHIP, an inositol polyphosphate 5-phosphatase that functions as a negative regulator of lymphocyte activation. DSHP is expressed in transformed T cell lines and is induced following in vitro activation of peripheral blood T lymphocytes. Expression of DSHP is restricted in vivo to lymphoid tissues, and RNA in situ hybridization demonstrates DSHP expression in activated T and B cell regions of reactive lymph nodes and in both T and B cell neoplasms. These observations confirm the identity of DSHP as the gene responsible for XLP, and suggest a role in the regulation of lymphocyte activation and proliferation. Induction of DSHP may sustain the immune response by interfering with SHIP-mediated inhibition of lymphocyte activation, while its inactivation in XLP patients results in a selective immunodeficiency to EBV.
