L’influence des contaminants environnementaux (CE) a été largement étudié en contexte d’exposition aigüe et d’intoxication. Qu’en est-il d’une exposition chronique à plus faibles doses sur le développement cognitif et sensoriel des enfants? Les études longitudinales de la présente thèse ont été réalisées au Nunavik. Bien que géographiquement éloignée des centres industriels, la communauté inuite est exposée aux CE via leur transport par les courants atmosphériques et océaniques ainsi que par leur bioaccumulation dans le poisson et les mammifères marins. Puisque l’alimentation autochtone traditionnelle, notamment la chair de béluga, est encore fréquemment consommée par la communauté, une proportion substantielle d’enfants inuits est exposée in utero aux CE. Ceux-ci sont également continuellement exposés à ces agents neurotoxiques durant leur développement postnatal. Or, la variation considérable dans l’adoption de l’alimentation traditionnelle au sein de la communauté représente une opportunité pour étudier la relation entre les niveaux d’exposition aux CE et le développement cognitif et sensoriel des enfants. Bien que certains déficits aient déjà été mis en lien avec l’exposition chronique aux CE dans la littérature, la présente thèse s’intéressa à cette relation plus spécifiquement chez les enfants inuits vivant dans le Grand Nord et plus exposés aux CE en raison de leur alimentation. Par ailleurs, les protocoles qui ont été développés pour cette thèse permettront d’évaluer des aspects qui ont été peu étudiés en lien avec les CE, soit l’attention visuospatiale et le traitement visuel cérébral précoce. Dans le premier volet de cette thèse, la relation entre trois CE et l’attention visuospatiale a été étudiée à l’aide d’une version adaptée de la tâche de Posner (M.I. Posner et al., 1980). Cette tâche psychophysique a été administrée à des enfants inuits (âge moyen = 11.2 ans) dont les niveaux d’exposition au mercure (Hg), au plomb (Pb) et aux biphényles polychlorés (BPCs) ont été documentés durant les périodes pré et postnatale. Les expositions in utero au Pb et aux BPCs ont été significativement associées à de l’impulsivité et de l’inattention, respectivement, alors que l’exposition postnatale au Pb a été associée à des temps de réaction plus longs. Bien qu’aucune relation spécifique avec l’attention visuospatiale n’ait été trouvée, les résultats de cette étude suggèrent que l’exposition aux CE est associée à une diminution des capacités attentionnelles générales chez les enfants résidant au Nunavik. Dans le second volet, le traitement cérébral précoce de l’information visuelle a été examiné à l’aide de potentiels évoqués visuels auprès d’enfants de la même communauté (âge moyen = 10.9 ans). La concentration de Hg dans le sang de cordon ombilical mesurée à la naissance a été associée à une réduction de l’amplitude et à une augmentation de la latence de la composante N75. L’exposition prénatale au Pb a quant à elle été associée à un délai dans la latence de la composante N150. Les résultats obtenus suggèrent ainsi que l’exposition aux métaux lourds, en particulier durant la période gestationnelle, serait associée à des altérations dans le développement visuel. Les résultats présentés dans cette thèse soutiennent l’hypothèse selon laquelle le cerveau en développement est vulnérable lors d’une exposition chronique aux CE, et cela même à des niveaux de concentration inférieurs aux limites recommandées par les organismes de santé publique. Les résultats permettent également d’apporter un éclairage nouveau sur les déficits cognitifs et sensoriels associés aux CE.

Acid and alkaline DNase activities in partially purified preparations from young and old chick brain were measured. The specific activity of acid DNase from old brain was lower by about 50% than that of enzyme from young brain , whereas alkaline DNase exhibited only marginal difference in activity of the two preparations . Study of various properties, viz. heat-stability and effect of exogenous compounds like Mg=', Hgl', Zn=', PHM B , on these enzymes revealed that while acid DNase in old brain is more susceptible to heat and heavy metal ion inhibition , alkaline DNase is devoid of any age-dependent variation in its properties.

Liquid Crystalline DNA is emerging as an active area of research, due to its potential applications in diverse fields, ranging from nanoelectronics to therapeutics. Since, counter ion neutralization is an essential requirement for the expression of LC DNA, and the present level of understanding on the LC phase behavior of high molecular weight DNA is inadequate, a thorough investigation is required to understand the nature and stability of these phases under the influence of various cationic species. The present study is, therefore mainly focused on a comparative investigation of the effect of metal ions of varying charge, size, hydration and binding modes on the LC phase behavior of high molecular weight DNA. The main objectives of the works are investigations on the induction and stabilization of LC phases of high molecular weight DNA by alkali metal ions, investigations on the induction and stabilization of LC phases of high molecular weight DNA by alkaline earth metal ions, effects of multivalent, transition and heavy metal ions on the LC phase behavior of high molecular weight DNA and investigations on spermine induced LC behavior of high molecular weight DNA in the presence of alkali and alkaline earth metal ions. The critical DNA concentration (CD) required for the expression of LC phases, phase transitions and their stability varied considerably when the binding site of the metal ions changed from phosphate groups to the nitrogenous bases of DNA, with Li+ giving the highest stability. Multiple LC phases with different textures, sometimes diffused and unstable or otherwise mainly distinct and clear, were observed on mixing metal ions with DNA solutions, which in turn depended on the charge, size, hydration factor, binding modes, concentration of the metal ions and time. Molecular modeling studies on binding of selected metal ions to DNA supported the experimental findings

The present investigation is to find the hypoxic adaptations and role of carotenoids in the anaerobic catabolism of two intertidal bivalves-Sunetta scripta and Perna viridis. Physiological and cytological responses during hypoxic stress have been studied and compared to that of sublethal heavy metal (copper) exposure using two indices : total carotenoid concentration and accumulation of lipofuscin granules. A close similarity has been observed between hypoxic exposed and copper (sublethal) exposed animals regarding the total carotenoid concentration and lipofuscin accumulation. In the case of S.scripta, the total caroteniod increase at 48h of both hypoxic and heavy metal exposure was found to be nearly 40% greater than that of the control (0h). Whereas in P.viridis, the increment in the total carotenoid concentration at 48h of hypoxic exposure and 48h of heavy metal exposure were found to be nearly 87% and 95% higher than that of the control (0h) respectively.Regarding the lipofuscin accumulation, in both S.scripta and P.viridis , the characteristic features of the granule at 48h of hypoxia is very much similar to that observed at 48h of heavy metal exposure. Thus, the present study suggests that the increase in carotenoid concentration and lipofuscin accumulation expressed by bivalves under heavy metal stress can be due to the indirect effect of hypoxia.

This thesis embodies findings on a taxonomical investigation of a group of lower marine invertebrates belonging to the category coelomata. Bryozoans are well known both in fossil and recent taxonomical history. They comprise of about 5,000 living and 16000 fossil species. Bryozoans are well known for their taxonomic abundance and structural diversity,representing the various ecological niches ranging from the intertidal to the abyssal benthic. At a time when global marine biological diversity has become a concern of not only to the scientists but also to the policy makers,an understanding of species diversity and abundance are cardinal aspects of biological studies. Geological time scales which is known that by Pre-Cambrian, marine invertebrate diversity reach the maximum and this diversity has become more comprehensive as time advanced. Taxonomists a vanishing species of scientists have become more concerned in discerning patterns of species diversity. The basic tool for this is identification fo animals. with this idea in mind a detailed study of taxonomy of bryozoan was undertaken . The major part of this thesis is devoted to describe various species of bryozoans with detailed description and ecotypical variations.The pattern of distribution and abundance which are important aspects of animal groups have also been documented. Possible effects of heavy metal contamination on the tolerance and growth of bryozoans, a few species of which have been eliminated from the chronically polluted areas of Cochin backwaters have also been documented.

The present study is an investigation to address relevant chemical aspects of the three varied aquatic environments, such as mangroves, river and the estuary. The sampling locations include a thick mangrove forest with high tidal activity, a mangrove nursery with minimal disturbances and low tidal inundation, a highly polluted riverine system and an estuarine site, as reference. Nutrients and bioorganic compounds in the water column and surface sediment were estimated in an attempt to understand the regeneration properties of these different aquatic systems.Assessment of the trace metal pollution was also carried out.

Asha M. R This thesis Entitled Toxicological effects of copper and mercury on the fish macerones gulio (hamiloton – buchanan).Chapter 1. In this chapter, a broad outline of heavy metal uptake, requirement of a suitable bio — monitoring organism, criteria for a standard test fish, and suitability of Macrones gulig for the toxicological study are given. Chapter 2. This chapter deals with the lethal toxicity bioassays to find the 96 hr LC 50 of copper and mercury for the fish Macrones gglig. The experimental results indicated that of the two metals tested, copper was more toxic than mercury.Chapter 3. The effect of copper and mercury on the haemoglobin, haematocrit, erythrocyte count, MCV, MCH and MCHC was studied.Chapter 4. The glycogen and protein contents of liver and muscle after exposure to copper and mercury were studied. There was a significant decrease of glycogen in the liver and muscle of metal treated fishes.Chapter 5. The histopathological changes of the tissues like liver, kidney and gill after exposure to copper and mercury were studied.

The present study is an attempt at investigating the intercompartmental exchange of trace metals (copper, cadmium, zinc, lead and nickel) in the Cochin estuary. The nature and extent of distribution in the different compartments with special reference to the transport from environmental compartments to biological compartments have been dealt with in detail. The suitability of the shells of Villorita cyprinoides var cochinensis (Hanely) in pollution monitoring activities has been assessed. A mathematical model (SAAMPLE - Shells in the Assessment of Aquatic Metal Pollution Levels) based on kinetic laws that govern the intercompartmental exchange has been proposed.

The thesis is Studies on the Effect or the Obganophosphorus Pesticide Ekalux(R) EC 25 on the Bacterial Flora or Villorita Cyprinoides Var.Cochinensis (Hanley). For the present investigation, the black clam Villorita gyprinoides var. cochinensis (Hanley), a most common clam genus present in this estuarine system has been selected as test organaism and Ekalux (R) EC 25 as toxicant. The aspects dealt with are 1. Total heterotrophic bacterial population, 2. Generic composition, 3. Hydrolytic enzyme producing bacteria, 4. Antibiotic resistance, 5. Heavy metal resistance, 6. The effect of pesticide concentration on the growth of the bacteria and 7. Effect of temperature, pH and sodium chloride on the growth and phosphate release of selected isolates.The samples for the experiment were collected from the Vembanad Lake, near Kumbalam Island during the period of September 1985 to May '86. The THB of the estuarine water and clams contained 6.5 x I04/ml and 2.975 x l06/g respectively, immediately after collection. Untreated water and clam samples showed enormous increase in THB from 0 hr population. The treated samples (water and clams) contained higher THB than 0 hr. In general, THB was observed to increase tremendously in the samples treated with pesticide when compared to their native flora. With reference to various concentrations of pesticides, THB recorded an increase with increase of concentration in water and clam samples.

As a result of the issues of care and conservation and sustainable utilisation, the proper management of mangrove forests have become more pressing than ever. Much recent ecological and toxicological debate has been centered around the question of validity of making predictions about the future of mangrove ecosystemas a result of the newly evolved environmental policy. Though muchinformation exist on the biodiversity, floristic composition and characteristics, geographical distribution and uses of mangroves, systematic documentation of the various sedimentological and geochemical phenomena in relation to the mangrove flora are scarce. Hazardous, persistent, man-made chemicals and waste produces are entering the mangrove ecosystem at from the adjacent watersheds which strengthened alarming rates the indispensible need for further researches on the environmental behaviours, fate and the effect of such products. Studies on the effect of heavy metals, pesticides and the other toxic signals through bioassay and toxicity tests on mangrove species as well as in sediments definitely will furnish ample clues to establish the actual operative mechanisms of these environments. A thorough review of literature made in this angle reveals that some attempts have already been initiated the world over the record the physico-chemical characteristics of major abiotic components such as sediments and water of many mangrove ecosystem, however, adequate information is lacking in the Indian Environmental Science scenario. The present investigation is an attempt to record the sedimentological, mineralogical and geochemical characteristics of sediments as well as the heavy metal enrichment in the various species ofmangrove flora of three important mangrove ecosystems of Kerala, located at Veli (SouthKerala), Kochi (Central Kerala) and Kannur (North Kerala). The results of the above investigation have been analysed statistically, discussed based on the available literature and presented in this thesis under seven chapters

The group cyanobacteria includes a large number of organisms characterised by a low state of cellular organization. Their cells lack a well defined nucleus. Cell division is by division of the protoplast by an ingrowth of the septum. These organisms are characterised generally by a blue green colouration of the cell, the chief pigments being chlorophyll-a, carotenes, xanthophylls, C phycocyanin and C phycoerythrin. The product of photosynthesis is glycogen. These organisms lack flagellate reproductive bodies and there is a total lack of sexual reproduction. They are also unique because of the presence of murein in the place of cellulose (cell wall) and the absence of chloroplast, mitochondria and endoplasmic reticulum. Just like bacteria some of them possess Plasmids and can fix atmospheric nitrogen. In the present study growth kinetics, heavy metal tolerance, tolerance mechanisms, heavy metal intake, and antibacterial activity of §ynechocystics salina Wislouch - a nanoplanktonic, euryhaline, Cyanobacterium present in Cochin back waters has been carried out for the potential biotechnological application of this organism. _§; salina occur as small spherical cells of 3n diameter (sometimes in pairs) with bluish green colour. The species is characterised by jerky movement of the cells and is structrually similar to other cyanobacteria

Environmental persistence, fate and interactive effects with living organisms - beneficial or toxic - of trace elements are directly related to the physico-chemical forms in which they occur. Knowledge on the association of trace metals with different environmental compartments in an aquatic system are, therefore, essential for monitoring the trace metal pollution as well as transport, fate and bio-geochemical cycles of trace metals. This thesis is a modest attempt in assessing the trace metal levels and their behaviour in the aquatic environment of Kuttanad, an aquatic system that is severely affected by man's intervention on natural processes, by seriously evaluating the levels of trace metals in dissolved and particulate phases and also in the different chemical fractions of the sediments.

Heavy metals are major toxic pollutants with severe health effects on humans. They are released into the environment from a variety of industrial activities. Cadmium, lead, zinc, chromium and copper are the most toxic metals of widespread use in industries such as tanning, electroplating, electronic equipment manufacturing and chemical processing plants. Heavy metals contribute to a variety of adverse health environmental effects due to their acute and chronic exposure through air, water and food chain. Conventional treatment methods of metal removal are often limited by their cost and ineffectiveness at low concentrations. Adsorption, the use of inactivated biomass as adsorbents offers an attractive potential alternative to their conventional methods. Mango peel and Alisma plantago aquatica are naturally occurring and abundant biomass can offer an economical solution for metal removal.The Cd(II), Pb(II), Zn(II), Cr(III) and Cu(II) adsorption by milled adsorbents of mango peel and Alisma plantago aquatica were evaluated in batches.

This thesis is an attempt by the author to assess the suitability of Metapenaeus dobsoni (Miers), an economically important crustacean species as a sentinel organism of trace metal pollution. The results of detailed investigations on seasonal variation, bioassay, accumulation and depuration of three metals viz., mercury, copper and zinc are presented and discussed. The importance of trace metals in the aquatic environment and their present status in the study area - Cochin backwaters, the significance of crustacean fisheries, the species M. dobsoni and the objectives of the present studies are described in Chapter 1. The methodology adopted during the investigation is given in Chapter 2. Chapter 3 delineates the seasonal variation of Hg, Cu and Zn in the edible and non-edible parts of M. dobsoni collected from Cochin backwaters for a period of one year (June 1984-May 1985). The results of bioassay experiments are given in Chapter 4. Kinetics of accumulation ,retention and depuration of trace metals, their biological half-life, the influence of size group and environmental factors are given in Chapter 5. The effect of these metals on the physiological response of M. dobsoni viz. oxygen consumption is included in Chapter 6. A summary and list of references are also appended.
