Mucetin (Trimeresurus mucrosquamatus venom activator, TMVA) is a potent platelet activator purified from Chinese habu (Trimeresurus mucrosquamatus) venom. It belongs to the snake venom heterodimeric C-type lectin family and exists in several multimeric forms. We now show that binding to platelet glycoprotein (GP) lb is involved in mucetin-induced platelet aggregation. Antibodies against GPIb as well as the GPIb-blocking C-type lectin echicetin inhibited mucetin-induced platelet aggregation. Binding of GPIb was confirmed by affinity chromatography and Western blotting. Antibodies against GPVI inhibited convulxin- but not mucetin-induced aggregation. Signalling by mucetin involved rapid tyrosine phosphorylation of a number of proteins including Syk, Src, LAT and PLCgamma2. Mucetininduced phosphorylation of the Fcgamma chain of platelet was greatly promoted by inhibition of alpha(llb)beta(3) by the peptidomimetic EMD 132338, suggesting that phosphatases downstream Of alpha(llb)beta(3) activation are involved in dephosphorylation of Fcgamma. Unlike other multimeric snake C-type lectins that act via GPIb and only agglutinate platelets, mucetin activates alpha(llb)beta(3). Inhibition Of alpha(llb)beta(3) strongly reduced the aggregation response to mucetin, indicating that activation Of alpha(llb)beta(3) and binding of fibrinogen are involved in mucetin-induced platelet aggregation. Apyrase and aspirin also inhibit platelet aggregation induced by mucetin, suggesting that ADP and thromboxaneA(2) are involved in autocrine feedback. Sequence and structural comparison with closely related members of this protein family point to features that may be responsible for the functional differences.

The giant panda skeletal muscle cells, uterus epithelial cells and mammary gland cells from an adult individual were cultured and used as nucleus donor for the construction of interspecies embryos by transferring them into enucleated rabbit eggs. All the three kinds of somatic cells were able to reprogram in rabbit ooplasm and support early embryo development, of which mammary gland cells were proven to be the Lest, followed by uterus epithelial cells and skeletal muscle cells. The experiments showed that direct injection of mammary gland cell into enucleated rabbit ooplasm, combined with in vivo development in ligated rabbit oviduct, achieved higher blastocyst development than in vitro culture after the somatic cell was injected into the perivitelline space and fused with the enucleated egg by electrical stimulation. The chromosome analysis demonstrated that the genetic materials in reconstructed blastocyst cells were the same as that in panda somatic cells. In addition, giant panda mitochondrial DNA (mtDNA) was shown to exist in the interspecies reconstructed blastocyst. The data suggest that (i) the ability of ooplasm to dedifferentiate somatic cells is not species-specific; (ii) there is compatibility between interspecies somatic nucleus and ooplasm during early development of the reconstructed egg.

A method for DNA isolation from early development of blastocyst and further analysis of nuclear and mitochondrial DNA was developed in present study. Total DNA was prepared from interspecies reconstructed blastocyst and a giant panda specific microsatellite locus g(010) was successfully amplified. DNA sequencing of the PCR product showed that two sequences of reconstructed blastocysts are the same as that of positive control giant panda. Our results prove that the nucleus of interspecies reconstructed blastocyst comes from somatic nucleus of donor giant panda.

The extent of idle capacity in fish processing (freezing) plants estimated by stratified random sampling is reported. The estimates for 1978 and 1979 for the processing plants on the west coast of India were 76.9 % and 73.2 % respectively at the rate of 250 working days per annum and two shifts per day. The percentage error of estimates worked out to 6.04 for 1978 and 6.98 for 1979. Substantial under utilization of processing plants noticed in all the states accounts mainly to the non-availability of raw material (prawn), high cost of production and shortage of power.

The extent of idle capacity in the fish processing (freezing) plants in the east coast of India estimated by stratified random sampling and the factors responsible for the same are reported. The estimates of idle capacity of fish processing plants in the east coast for the years 1978 and 1979 were respectively 75.9% and 72.5% on the basis of 250 working days per annum and double shift per day. The percentage errors of estimates worked out to 6.9 for 1978 and 4.7 for 1979. The corresponding figures were worked out on the basis of 200 working days also. Substantial under-utilisation of plants in all the maritime states in the east coast accounted mainly to non-availability of raw material, high cost of production, shortage of power, scarcity of ice and potable water during peak season and frequent labour troubles.

Shrimp culture in Bangladesh has emerged as an important aquaculture industry over the last three decades although its culture in greater parts of the farming area is done in traditional ways. In the meantime, the government of Bangladesh has taken necessary measures along with the private sectors to increase production, upgrade processing industries and to promote export performance. Long supply chain in raw material collection, inadequate infrastructure facilities, poor level of cool chain and lack of adequate HACCP-based training on hygiene and sanitation of different groups of people involved in the field level are the main problems of quality loss of raw materials. Shortage of raw materials results in poor capacity utilization of the processing plants. The growth of bagda (P. monodon) hatchery has expanded rapidly over the last few years, remaining mostly concentrated in Cox's Bazar region is enough to meet the target production. However, there is a shortage of pelleted shrimp feed in Bangladesh. A large number of export processors are now producing increasing amounts of value-added products such as individually quick-frozen, peeled and divined, butterfly cut shrimp, as well as cooked products. The export earnings from value added products is about half of the total export value. About 95% of total fish products are exported to European countries, USA and Japan and the remaining to the Southeast Asia and the Middle East. Most of the EU approved shrimp processing industries have been upgraded with laboratory facilities and provided HACCP training to their workers. As of now, HACCP is applied on the processing plants, but to ensure the quality of raw materials and to reduce risks, shrimp farms are also required to adopt HACCP plan. There is increased pressure time to time from importing countries for fish processors to establish effective quality assurance system in processing plants. Fish Inspection and Quality Control (FIQC) of the Department of Fisheries while having moderately equipped laboratories with chemical, bio-chemical and microbiological testing facilities and qualified technical personnel, the creation of facilities for testing of antibiotics is underway. FIQC mainly supervises quality aspects of the processing plants and has little or no control over raw material supply chains from farm to processing plants. Bangladesh export consignments sometimes face rejection due to reported poor quality of the products. Three types of barriers are reported for export of shrimp to EU countries. These are:(1) government participation in trade and restrictive practices (state aid, countervailing duties, state trading enterprises, government monopoly practices), customs and administrative entry procedures (anti-dumping duties, customs valuation, classification, formalities, rules of origin); (2) technical barriers to trade or TBT (technical regulations, standards, testing, certification arrangement); (3) specific limitations (quantitative restrictions, import licensing, embargoes, exchange control, discriminatory sourcing, export restraints, measures to regulate domestic prices, requirements concerning marking, labeling and packaging).

 目的 观察丙泊酚对大鼠海马CA1 区锥体神经元产生的长时程抑制(L TD) 的影响,并 分析其可能机制。方法 断头法分离wistar 大鼠(13~19 d) 海马半脑,用切片机切出400μm 厚度的 海马脑片。实验分三组:脂肪乳剂组( I 组) ,丙泊酚组(P 组) ,SR95531 + 丙泊酚组( GP 组) 。I 组和P 组以90μL 脂肪乳剂或丙泊酚(相当于100μmol/ L) 预孵脑片60 min ,然后给予低频刺激(L FS) ,记录 L TD 的表达情况; GP 组先在循环液中加入10μmol/ L SR95531 预孵脑片30 min ,再加入100μmol/ L 丙泊酚继续孵育60 min ,继而给予L FS ,记录L TD 的表达情况。结果 I 组给予L FS 后,产生L TD , L FS 后10~40 min 的兴奋性突触后电流( EPSC) 值为基础值的57185 %;P 组给予L FS 后10~40 min 的EPSC 值为基础值的40182 % ,明显低于I 组( P < 0105) ; GP 组给予L FS 后10~40 min 的EPSC 值为基础值的56151 % ,与I 组比较差异无显著意义( P > 0105) ,与P 组比较差异有显著意义( P < 0105) 。结论 100μmol/ L 丙泊酚使大鼠海马CA1 区锥体神经元L TD 表达增强,这种作用与其增强 GABAA 受体功能有关;当阻断GABAA 受体后,这种易化作用消失。

A brief outline is given of the Philippine seaweed industry, which involves mainly the red seaweed Kappaphycus alvarezii, produced as a source of the semi-refined or Philippine natural grade (PNG) carrageenan. Other species cultured and marketed include: Gracilaria changii, G. firma, G. heteroclada, G. manilaensis and G. tenuistipitata. The Philippines has the largest carrageenan refinery in Asia. However, production of seaweeds is rather erratic, which is attributed to the following factors: 1) indiscriminate harvesting, seasonal abundance of different species in different areas and lack of proper management; 2) shortage and deteriorating quality of seedlings, especially for Kappaphycus; and 3) natural calamities.

Purpose - The purpose of this paper is to develop a framework of total acquisition cost of overseas outsourcing/sourcing in manufacturing industry. This framework contains categorized cost items that may occur during the overseas outsourcing/sourcing process. The framework was tested by a case study to establish both its feasibility and usability. Design/methodology/approach - First, interviews were carried out with practitioners who have the experience of overseas outsourcing/sourcing in order to obtain inputs from industry. The framework was then built up based on combined inputs from literature and from practitioners. Finally, the framework was tested by a case study in a multinational high-tech manufacturer to establish both its feasibility and usability. Findings - A practical barrier for implementing this framework is shortage of information. The predictability of the cost items in the framework varies. How to deal with the trade off between accuracy and applicability is a problem needed to be solved in the future research. Originality/value - There are always limitations to the generalizations that can be made from just one case. However, despite these limitations, this case study is believed to have shown the general requirement of modeling the uncertainty and dealing with the dilemma between accuracy and applicability in practice. © Emerald Group Publishing Limited.

in the last 10 years many designs and trial implementations of holonic manufacturing systems have been reported in the literature. Few of these have resulted in any industrial take up of the approach and part of this lack of adoption might be attributed to a shortage of evaluations of the resulting designs and implementations and their comparison with more conventional approaches. This paper proposes a simple approach for evaluating the effectiveness of a holonic system design, with particular focus on the ability of the system to support reconfiguration (in the face of change). A case study relating to a laboratory assembly system is provided to demonstrate the evaluation approach. Copyright © 2005 IFAC.
