T-cell development depends upon interactions between thymocytes and thymic epithelial cells (TECs). The engagement of delta-like 4 (DL4) on TECs by Notch1 expressed by blood-borne BM-derived precursors is essential for T-cell commitment in the adult thymus. In contrast to the adult, the earliest T-cell progenitors in the embryo originate in the fetal liver and migrate to the nonvascularized fetal thymus via chemokine signals. Within the fetal thymus, some T-cell precursors undergo programmed TCRγ and TCRδ rearrangement and selection, giving rise to unique γδ T cells. Despite these fundamental differences between fetal and adult T-cell lymphopoiesis, we show here that DL4-mediated Notch signaling is essential for the development of both αβ and γδ T-cell lineages in the embryo. Deletion of the DL4 gene in fetal TECs results in an early block in αβ T-cell development and a dramatic reduction of all γδ T-cell subsets in the fetal thymus. In contrast to the adult, no dramatic deviation of T-cell precursors to alternative fates was observed in the fetal thymus in the absence of Notch signaling. Taken together, our data reveal a common requirement for DL4-mediated Notch signaling in fetal and adult thymopoiesis.

We present a segmentation method for fetal brain tissuesof T2w MR images, based on the well known ExpectationMaximization Markov Random Field (EM- MRF) scheme. Ourmain contribution is an intensity model composed of 7Gaussian distribution designed to deal with the largeintensity variability of fetal brain tissues. The secondmain contribution is a 3-steps MRF model that introducesboth local spatial and anatomical priors given by acortical distance map. Preliminary results on 4 subjectsare presented and evaluated in comparison to manualsegmentations showing that our methodology cansuccessfully be applied to such data, dealing with largeintensity variability within brain tissues and partialvolume (PV).

One of the world's largest wollastonite deposits was formed at the contact of the northern Hunter Mountain Batholith (California, USA) in Paleozoic sediments. Wollastonite occurs as zones of variable thickness surrounding layers or nodules of quartzite in limestones. A minimum formation temperature of 650 degrees C is estimated from isolated periclase-bearing lenses in that area. Contact metamorphism of siliceous carbonates has produced mineral assemblages that are consistent with heterogeneous, and partly limited infiltration of water-rich fluids, compatible with O-18/O-16 and C-13/C-12 isotopic patterns recorded in carbonates. Oxygen isotope compositions of wollastonites in the study area may also not require infiltration of large quantities of externally-derived fluids that were out of equilibrium with the rocks. 8180 values of wollastonite are high (14.8 parts per thousand to 25.0 parts per thousand; median: 19.7 parts per thousand) and close to those of the host limestone (19.7 parts per thousand to 28 parts per thousand; median: 24.9 parts per thousand) and quartz (18.0 parts per thousand. to 29.1 parts per thousand; median: 22.6 parts per thousand). Isotopic disequilibrium exists at quartz/wollastonite and wollastonite/calcite boundaries. Therefore, classical batch/Rayleigh fractionation models based on reactant and product equilibrium are not applicable to the wollastonite rims. An approach that relies on local instantaneous mass balance for the reactants, based on the wollastonite-forming reaction is suggested as an alternative way to model wollastonite reaction rims. This model reproduces many of the measured delta O-18 values of wollastonite reaction rims of the current study to within +/- 1 parts per thousand, even though the wollastonite compositions vary by almost 10 parts per thousand. Previous studies showed a fetal sheep liver extract (FSLE), in association with monophosphoryl lipid A, MPLA (a bioactive component of lipid A of LPS), could interact to induce the development of dendritic cells (DCs) which regulated production of Foxp3+ Treg. This interaction was associated with an altered gene expression both of distinct subsets of TLRs and of CD200Rs. Prior studies had suggested that major interacting components within FSLE were gamma-chain of fetal hemoglobin (Hgbgamma) and glutathione (GSH). We investigated whether differentiation/maturation of DCs in vitro in the presence of either GM-CSF or Flt3L to produce preferentially either immunogenic or tolerogenic DCs was itself controlled by an interaction between MPLA, GSH and Hgbgamma. At low (approximately 10 microg/ml) Hgbgamma concentrations, DCs developing in culture with GSH and MPLA produced optimal stimulation of allogeneic CTL cell responses in vitro (and enhanced skin graft rejection in vivo). At higher concentrations (>40 microg/ml Hgbgamma) and equivalent concentrations of MPLA and GSH, the DCs induce populations of Treg which can suppress the induction of allogeneic CTL and graft rejection in vivo. These different populations of DCs express different patterns of mRNAs for the CD200R family. Addition of anti-TLR or anti-MD-1 mAbs to DCs developing in this mixture (Hgbgamma+GSH+MPLA), suggests that one effect of (GSH+Hgbgamma) on MPLA stimulation may involve altered signaling through TLR4.

The aim of this study was to evaluate splenic eosinophil and mast cell accumulation using pagoda red stain in a series of anaphylaxis-related deaths that underwent medico-legal investigations. Our goal was to assess whether fatal reactions to insect stings, intramuscularly administered antibiotics and intravenously injected contrast media are responsible for specific patterns of eosinophil and mast cell accumulation. Two study groups were prospectively formed, an anaphylaxis-related death group and a control group. Autopsy, histology (haematoxylin-eosin stain, pagoda red stain and immunohistochemistry using anti-tryptase antibodies), toxicology and postmortem biochemistry (beta-tryptase, total IgE and specific IgE) were performed in all cases. All tested parameters (spleen weight, beta-tryptase and total IgE levels as well as eosinophil, mast cell and degranulated mast cell numbers in the spleen) were significantly higher in the anaphylaxis-related death group. No statistically significant differences were observed among the various groups (intramuscular antibiotic injection, intravenous contrast medium administration and stinging insects) in any combination, suggesting that mast cell and eosinophil accumulation in the spleen during anaphylaxis does not have any specific pattern related to the triggering allergen. Despite a lower sensitivity than immunohistochemical staining in discriminating eosinophil and mast cells, pagoda red stain allowed these cells to be identified and could therefore be proposed as a low-cost, first-line diagnostic procedure in those situations where immunohistochemistry is not systematically performed or cannot be carried out.

Death receptors (DRs) of the TNFR superfamily contribute to antiviral immunity by promoting apoptosis and regulating immune homeostasis during infection, and viral inhibition of DR signaling can alter immune defenses. Here we identify the human cytomegalovirus (HCMV) UL141 glycoprotein as necessary and sufficient to restrict TRAIL DR function. Despite showing no primary sequence homology to TNF family cytokines, UL141 binds the ectodomains of both human TRAIL DRs with affinities comparable to the natural ligand TRAIL. UL141 binding promotes intracellular retention of the DRs, thus protecting virus infected cells from TRAIL and TRAIL-dependent NK cell-mediated killing. The identification of UL141 as a herpesvirus modulator of the TRAIL DRs strongly implicates this pathway as a regulator of host defense to HCMV and highlights UL141 as a pleiotropic inhibitor of NK cell effector function.

The otter shrews of mainland Africa are the closest relatives of the Madagascar tenrecs. We sought for similarities in placentation between the two groups and, in a wider context, with other mammals of the Afrotheria clade. Specimens of the Nimba otter shrew (Micropotamogale lamottei) were obtained from the Ivory Coast and examples of the giant otter shrew (Potamogale velox) from the Hill Collection. The Nimba otter shrew has a central haemophagous organ similar to that in tenrecs. The labyrinth of the Nimba otter shrew, however, is endotheliochorial with syncytial trophoblast enclosing the maternal vessels. On the other hand tenrecs have cellular haemomonochorial placentae and an associated spongy zone, which is not present in the Nimba otter shrew. The placenta of the giant otter shrew is also endotheliochorial. The central region of its placenta is particularly interesting, since the juxtafetal portion is clearly a haemophagous region whereas the labyrinth feeding this region is endotheliochorial. Thus there is considerable variation in placental morphology within Tenrecidae. Importantly, however, both otter shrews have a large allantoic sac divided into four intercommunicating lobes by two pairs of septal folds. A similar arrangement has been described for representatives of each of the remaining five orders within Afrotheria. This is significant because previous anatomical studies have failed to establish a single synapomorphy in support of Afrotheria.

A previously described extract of sheep fetal liver was reported to reverse many of the cytokine changes associated with aging in mice, including an augmented spleen cell ConA-stimulated production of IL-4 and decreased production of IL-2. Similar effects were not seen with adult liver preparations. These changes were observed in various strains of mice, including BALB/c, DBA/2 and C57BL/6, using mice with ages ranging from 8 to 110 weeks. Preliminary characterization of this crude extract showed evidence for the presence of Hb gamma chain, as well as of lipid A of LPS. We show below that purified preparations of sheep fetal Hb, but not adult Hb, in concert with suboptimally stimulating doses of LPS (lipid A), cooperate in the regulation of production of a number of cytokines, including TNFalpha and IL-6, in vitro. Furthermore, isolated fresh spleen or peritoneal cells from animals treated in vivo with the same combination of Hb and LPS, showed an augmented capacity to produce these cytokines on further culture in vitro. Evidence was also obtained for a further interaction between CLP, LPS and fetal Hb itself in this augmented cytokine production. These data suggest that some of the functional activities in the fetal liver extract reported earlier can be explained in terms of a novel immunomodulatory role of a mixture of LPS (lipid A) and fetal Hb.

Hereditary non-structural diseases such as catecholaminergic polymorphic ventricular tachycardia (CPVT), long QT, and the Brugada syndrome as well as structural disease such as hypertrophic cardiomyopathy (HCM) and arrhythmogenic right ventricular cardiomyopathy (ARVC) cause a significant percentage of sudden cardiac deaths in the young. In these cases, genetic testing can be useful and does not require proxy consent if it is carried out at the request of judicial authorities as part of a forensic death investigation. Mutations in several genes are implicated in arrhythmic syndromes, including SCN5A, KCNQ1, KCNH2, RyR2, and genes causing HCM. If the victim's test is positive, this information is important for relatives who might be themselves at risk of carrying the disease-causing mutation. There is no consensus about how professionals should proceed in this context. This article discusses the ethical and legal arguments in favour of and against three options: genetic testing of the deceased victim only; counselling of relatives before testing the victim; counselling restricted to relatives of victims who tested positive for mutations of serious and preventable diseases. Legal cases are mentioned that pertain to the duty of geneticists and other physicians to warn relatives. Although the claim for a legal duty is tenuous, recent publications and guidelines suggest that geneticists and others involved in the multidisciplinary approach of sudden death (SD) cases may, nevertheless, have an ethical duty to inform relatives of SD victims. Several practical problems remain pertaining to the costs of testing, the counselling and to the need to obtain permission of judicial authorities.

Abstract: The human protozoan parasite Leishmania major has been shown to exhibit several morphological and biochemical features characteristic of a programmed cell death (PCD) when differentiating into infectious stages and under a variety of stress conditions. In mammalian cells, the principal effector molecules of PCD or apoptosis are caspases. Although some caspase-like peptidase activity has been reported in dying parasites, no caspase gene is present in the L. major genome. However, a single metacaspase gene is present in L. major whose encoded protein harbors the predicted secondary structure and the catalytic dyad histidine/cysteine described for caspases and other metacaspases identified in plants and yeast. Metacaspases are also present in other protozoan parasites such as Trypanosoma and Plasmodium species and are not present in mammalian cells, which make them a possible drug target for the treatment of the parasitic diseases they cause. The Saccharomyces cerevisiae metacaspase YCA1 has been implicated in the death of aging cells, cells defective in some biological functions, and cells exposed to different environmental stresses. In this study, we evaluated the functional heterologous complementation of a S. cerevisiae ycal null mutant with the L. major metacaspase (LmjMCA} in cell death induced by oxidative stress. We show that LmjMCA is involved in yeast cell death, similar to YCA1, and that this function depends on its catalytic activity. LmjMCA was found to be auto-processed as occurs for caspases, however, LmjMCA did not exhibit any activity with caspase substrates. In contrast, LmjMCA was active towards substrates with arginine in the P1 position, with the activity being abolished following H147A and C202A catalytic site mutations and addition of the arginal inhibitor leupeptin. In order to identify the L. major proteins that may function as substrates, inhibitors, or may bind and recruit LmjMCA, a yeast two-hybrid screening with cDNA libraries from different life cycle stages of the parasite was conducted. Proteins putatively involved in PCD were identified as interacting with LmjMCA, however, the interaction of LmjMCA with proteins involved in other physiological processes such as vesicle transport, suggests that LmjMCA could have additional roles in the different life cycle stages of the parasite. Résumé: Plusieurs caractéristiques morphologiques et biochimiques rappelant la mort cellulaire programmée ont été identifiées dans les stades infectieux et sous des conditions de stress, chez le parasite protozoaire humain, Leishmania major. Dans les cellules de mammifères, les caspases sont les molécules effectrices principales impliquées dans la mort cellulaire programmée et l'apoptose. Bien qu'une activité caspase ait été retrouvée dans des parasites en mon` cellulaire, le génome de Leishmania ne contient aucun gène qui pourrait coder pour une caspase. À la place, on retrouve un gène unique codant pour une métacaspase. Une prédiction de la structure secondaire de la métacaspase montre que cette métacaspase a un domaine catalytique contenant la dyade histidine/cystéine présente dans les caspases et les autres métacaspases décrites chez les plantes et la levure. Les métacaspases sont aussi présentes dans d'autres parasites protozoaires tels que Trypanosome et Plasmodium, mais ne sont pas présentes dans les cellules de mammifères, ce qui en fait des cibles intéressantes pour le développement de drogue. Dans la levure, Saccharomyces cerevisiae, la métacaspase YCA1 est impliquée dans la mort des cellules âgées, la mort des cellules défectueuses dans certaines fonctions biologiques et dans les cellules exposées à différents stress environnementaux. Dans cette étude, une complémentation hétérologue fonctionnelle d'un mutant de la levure déficient en YCA1 par le gène LmjMCA de L. major lors de l'induction de ta mort par stress oxydatif a été évaluée. Nos résultats montrent que LmjMCA peut remplacer YGA1 dans le programme de mort cellulaire chez la levure et que celte fonction dépend de son activité catalytique. De plus, LmjMCA subit une auto clivage comme les caspases mais n'exhibe aucune spécificité pour les substrats des caspases. Plusieurs protéines potentiellement impliquées dans un programme de mort cellulaire ont été identifiées comme interagissant avec LmjMCA. Cependant, l'identification de protéines impliquées dans le transport vésiculaire suggère aussi que LmjMCA pourrait avoir un rôle additionnel dans les différents stades du cycle parasitaire. Résumé destiné à un large public: De nos jours, la leishmaniose est la deuxième plus importante maladie parasitaire après la malaria. Malgré les avancées accomplies dans les stratégies de contrôle, près de deux millions de nouveaux cas apparaissent chaque année. Actuellement, la principale stratégie pour faire face à ce problème épidémiologique consiste en un traitement pharmacologique des personnes infectées. Pourtant, seule une dizaine de médicaments, dont la plupart sont toxiques, est disponible pour traiter la leishmaniose et des cas de résistance émergent dans certains pays endémiques. Il devient donc urgent de mettre au point de nouveaux traitements anti-leishmaniens capables d'éliminer le parasite sans effets indésirables sur le patient. Récemment, des caractéristiques morphologiques et biochimiques de la mort cellulaire programmée (MCP) semblables au processus de l'apoptose chez les mammifères ont été décrites dans Leishmania. Cependant, des gènes codant pour des protéines similaires à celles qui sont impliquées dans l'apoptose, comme les caspases, ne se retrouvent pas dans le génome de Leishmanía major. Néanmoins, les espèces de Leishmanía, aussi bien que d'autres parasites protozoaires responsables des trypanosomiases et de la malaria, possèdent des métacaspases qui sont des protéines homologues aux caspases mais qui ne sont pas présentes chez les mammifères. C'est pourquoi, la caractérisation de la métacaspase de Leishmania (LmjMCA) ainsi que ses mécanismes d'activation pourrait être une piste d'investigation intéressante dans l'identification de nouvelles cibles thérapeutiques dans les voies de signalisation de la MCP des parasites protozoaires. Aproximadamente 2 millones de nuevos casos ocurren cada año a pesar de los avances logrados en el desarrollo de nuevos métodos de control. El tratamiento farmacológico de las personas infectadas es actualmente la principal estrategia de control, sin embargo, menos de una decena de medicamentos se encuentran disponibles en el mercado, la mayoría de ellos son tóxicos, y ya empiezan a encontrarse parásitos resistentes en algunos países endémicos para la leishmaniasis. El desarrollo de nuevos medicamentos capaces de eliminar los parásitos sin producir efectos indeseables en los humanos, es una necesidad inminente. Recientemente, algunas de las características morfológicas y bioquímicas de la muerte celular programada (MCP) similares al proceso de la apoptosis en mamíferos, han sido descritas en parasitos de Leishmania. Sin embargo, genes que codifiquen proteínas similares a aquellas involucradas en la apoptosis, como las caspasas, no se encuentran en el genoma de Leishmania major. AI contrario, las especies de Leishmania, así como de los otros parásitos responsables de la tripanosomiasis y de la malaria, poseen metacaspases, proteínas homologas a las caspases pero que no están presentes en las células de mamíferos. La caracterización de la metacaspasa de L. major y de sus mecanismos de activación constituye, por lo tanto, un área de investigación interesante para la identificación de nuevos blancos terapéuticos en el proceso de MCP de los parásitos protozoarios. En la levadura Saccharomyces cerevisiae, la metacaspasa YCA1 ha sido descrita como implicada en la muerte de células envejecidas, células defectuosas en algunas funciones biológicas, y en células expuestas a diferentes tipos de estrés ambiental. En el presente estudio se evaluó la complementación heteróloga funcional de una levadura mutante deficiente en YCA1 con el gen de metacaspase de L. major (LmjMCA) en la MCP inducida por estrés oxidativo. Nuestros resultados muestran que la LmjMCA puede reemplazarla YCA1 en la MCP de la levadura dependiente de su actividad catalítica y que la LmjMCA se auto-procesa similar a las caspasas. Sin embargo, LmjMCA no reconoce los substratos de caspasas sino substratos con una arginina en ta posición P1. Dicha actividad enzimática fue abolida con la inducción de las mutaciones puntuales H147A y C202A en la díada catalítica de LmjMCA y con la adición de leupeptina, un inhibidor con arginina. Con el fin de identificar proteínas que pudieran funcionar como substratos, inhibidores o moléculas modificadoras de LmjMCA, se aplicó el método de doble-híbrido en levadura con bibliotecas de ADNc provenientes de diferentes estadios del ciclo de vida del parásito. Algunas proteínas potencialmente implicadas. en la MCP del parásito fueron identiñcadas interactuando con LmjMCA. La identificación de otras proteínas involucradas en transporte vesicular sugiere que la LmjMCA podría tener una función biológica adicional en los diferentes estadios del ciclo de vida dei parásito.

Activation of proteolytic cell death pathways may circumvent drug resistance in deadly protozoan parasites such as Plasmodium falciparum and Leishmania. To this end, it is important to define the cell death pathway(s) in parasites and thus characterize proteases such as metacaspases (MCA), which have been reported to induce cell death in plants and Leishmania parasites. We, therefore, investigated whether the cell death function of MCA is conserved in different protozoan parasite species such as Plasmodium falciparum and Leishmania major, focusing on the substrate specificity and functional role in cell survival as compared to Saccharomyces cerevisae. Our results show that, similarly to Leishmania, Plasmodium MCA exhibits a calcium-dependent, arginine-specific protease activity and its expression in yeast induced growth inhibition as well as an 82% increase in cell death under oxidative stress, a situation encountered by parasites during the host or when exposed to drugs such as artemisins. Furthermore, we show that MCA cell death pathways in both Plasmodium and Leishmania, involve a z-VAD-fmk inhibitable protease. Our data provide evidence that MCA from both Leishmania and Plasmodium falciparum is able to induce cell death in stress conditions, where it specifically activates a downstream enzyme as part of a cell death pathway. This enzymatic activity is also induced by the antimalarial drug chloroquine in erythrocytic stages of Plasmodium falciparum. Interestingly, we found that blocking parasite cell death influences their drug sensitivity, a result which could be used to create therapeutic strategies that by-pass drug resistance mechanisms by acting directly on the innate pathways of protozoan cell death.

OBJECTIVES: Ultrasound scan in the mid-trimester of pregnancy is now a routine part of prenatal care in most European countries. The objective of this study was to evaluate the prenatal diagnosis of dysmorphic syndromes by fetal ultrasound examination. METHODS: Data from 20 registries of congenital malformations in 12 European countries were included in the study. RESULTS: There were 2454 cases with congenital heart diseases, 479 of which were recognized syndromes, including 375 chromosomal anomalies and 104 syndromes without chromosomal anomalies. Fifty-one of the 104 were detected prenatally (49.0%). One hundred and ninety-two of 1130 cases with renal anomalies were recognized syndromes, including 128 chromosomal anomalies and 64 syndromes without chromosomal anomalies; 162 of them (84.4%) were diagnosed prenatally, including 109 chromosomal anomalies and 53 non-chromosomal syndromes. Fifty-four of the 250 cases with limb defects were recognized syndromes, including 16 chromosomal syndromes and 38 syndromes without chromosomal anomalies; 21 of these 54 syndromes were diagnosed prenatally (38.9%), including 9 chromosomal syndromes. There were 243 cases of abdominal wall defects including 57 recognizable syndromes, 48 with omphalocele and 9 with gastroschisis; 48 were diagnosed prenatally (84.2%). Twenty-six of the 187 cases with diaphragmatic hernia had recognized syndromes, including 20 chromosomal aberrations and 6 syndromes without chromosomal anomalies. Twenty-two of them (84.6%) were detected prenatally. Sixty-four of 349 cases with intestinal anomalies were recognized syndromes; 24 were diagnosed prenatally (37.5%). There were 553 cases of cleft lip and palate (CL(P)) and 198 of cleft palate (CP) including 74 chromosomal anomalies and 73 recognized non-chromosomal syndromes. Prenatal diagnosis was made in 51 cases of CL(P) (53.7%) and 7 of CP (13.7%). Twenty-two of 188 anencephalic cases were syndromic and all were diagnosed prenatally. Of 290 cases with spina bifida, 18 were recognized syndromes, and of them 17 were diagnosed prenatally. All 11 syndromic encephaloceles were diagnosed prenatally. CONCLUSIONS: Around 50% of the recognized syndromes which are associated with major congenital anomalies (cardiac, renal, intestinal, limb defects, abdominal wall defects and oral clefts) can be detected prenatally by the anomaly scan. However the detection rate varies with the type of syndrome and with the different countries' policies of prenatal screening.

Fas, a death domain-containing member of the tumor necrosis factor receptor family and its ligand FasL have been predominantly studied with respect to their capability to induce cell death. However, a few studies indicate a proliferation-inducing signaling activity of these molecules too. We describe here a novel signaling pathway of FasL and the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) that triggers transcriptional activation of the proto-oncogene c-fos, a typical target gene of mitogenic pathways. FasL- and TRAIL-mediated up-regulation of c-Fos was completely dependent on the presence of Fas-associated death domain protein (FADD) and caspase-8, but caspase activity seemed to be dispensable as a pan inhibitor of caspases had no inhibitory effect. Upon overexpression of the long splice form of cellular FADD-like interleukin-1-converting enzyme (FLICE) inhibitory protein (cFLIP) in Jurkat cells, FasL- and TRAIL-induced up-regulation of c-Fos was almost completely blocked. The short splice form of FLIP, however, showed a rather stimulatory effect on c-Fos induction. Together these data demonstrate the existence of a death receptor-induced, FADD- and caspase-8-dependent pathway leading to c-Fos induction that is inhibited by the long splice form FLIP-L.
