The activity of the Na(+)/H(+) exchanger NHE3 is regulated by a number of factors including parathyroid hormone (PTH). In the current study, we used a renal epithelial cell line, the opossum kidney (OKP) cell, to elucidate the mechanisms underlying the long-term effects of PTH on NHE3 transport activity and expression. We observed that NHE3 activity was reduced 6 h after addition of PTH, and this reduction persisted almost unaltered after 24 h. The decrease in activity was associated with diminished NHE3 cell surface expression at 6, 16, and 24 h after PTH addition, total cellular NHE3 protein at 16 and 24 h, and NHE3 mRNA abundance at 24 h. The lower levels of NHE3 mRNA were associated to a small, but significant, decrease in mRNA stability. Additionally, by analyzing the rat NHE3 gene promoter activity in OKP cells, we verified that the regulatory region spanning the segment -152 to +55 was mildly reduced under the influence of PTH. This effect was completely abolished by the presence of the PKA inhibitor KT 5720. In conclusion, long-term exposure to PTH results in reduction of NHE3 mRNA levels due to a PKA-dependent inhibitory effect on the NHE3 promoter and a small reduction of mRNA half-life, and decrease in the total amount of protein which is preceded by endocytosis of the apical surface NHE3. The decreased NHE3 expression is likely to be responsible for the reduction of sodium, bicarbonate, and fluid reabsorption in the proximal tubule consistently perceived in experimental models of PTH disorders.

We examined the effect of storage time on culture viability and some rheological properties (yield stress, storage modulus, loss modulus, linear viscoelastic region, structural recuperation and firmness) of fermented milk made with Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus acidophilus (LA) and Bifidobacterium animalis ssp. lactis in coculture with Streptococcus thermophilus (ST). Acidification profiles and factors that affect viability (postfermentation acidification, acidity and dissolved oxygen) were also studied during 35 days at 4C. Fermented milk prepared with a coculture of ST and Bifidobacterium lactis gave the most constant rheological behavior and the best cell viability during cold storage; it was superior to ST plus LA for probiotic fermented milk production.

This study aimed to optimize the rheological properties of probiotic yoghurts supplemented with skimmed milk powder (SMP) whey protein concentrate (WPC) and sodium caseinate (Na-Cn) by using an experimental design type simplex-centroid for mixture modeling It Included seven batches/trials three were supplemented with each type of the dairy protein used three corresponding to the binary mixtures and one to the ternary one in order to increase protein concentration in 1 g 100 g(-1) of final product A control experiment was prepared without supplementing the milk base Processed milk bases were fermented at 42 C until pH 4 5 by using a starter culture blend that consisted of Streptococcus thermophilus Lactobacillus delbrueckii subsp bulgaricus and Bifidobacterium (Humans subsp lactis The kinetics of acidification was followed during the fermentation period as well the physico-chemical analyses enumeration of viable bacteria and theological characteristics of the yoghurts Models were adjusted to the results (kinetic responses counts of viable bacteria and theological parameters) through three regression models (linear quadratic and cubic special) applied to mixtures The results showed that the addition of milk proteins affected slightly acidification profile and counts of S thermophilus and B animal`s subsp lactis but it was significant for L delbrueckii subsp bulgaricus Partially-replacing SMP (45 g/100 g) with WPC or Na-Cn simultaneously enhanced the theological properties of probiotic yoghurts taking into account the kinetics of acidification and enumeration of viable bacteria (C) 2010 Elsevier Ltd All rights reserved

A mineração de carvão em lavra a céu aberto provoca a mistura dos materiais sobrejacentes ao carvão e que são, posteriormente, utilizados no preenchimento das cavas mineradas. O processo de construção dos solos e a mistura podem ocasionar problemas ambientais pelas alterações quimicas e fisicas provocadas nos solos construidos. A fimde comparar solos construidos de diferentes idades, caracterizou-se a coluna geológica. o ~Io natural originale solos construidos de 2 anos e de 24 anos da mina de carvão Boa Vista, no Municípiode Minas do Leão, RS. A descrição morfológicado perfil do solo e as análises quimicas e fisicas demonstram que os solos construidos apresentam sérias limitações físicas, impedindo o desenvolvimento normal da vegetação. Ademais. diferenças quimicas entre os perfis de diferentes idades originam-se principalmente da utilização de materiais geológicos e processos de construção distintos, do que pelos processos pedogenéticos. Quanto à evolução temporal dos solos construidos, observou-se que está fortemente caracterizada pela intemperização acido-sulfatada, intensificando-se na sub-área mais antiga. O potencial de acidificação e o potencial de neutralização determínados na coluna geológica indicam a existência de camadas com alto potencial gerador de Drenagem Ácida de Minas, recomendando-se o cuidado na deposição final destas camadas. Os resultados deste estudo indicam que as propriedades químicas e fisicas determinadas nos solos construidos servem como indicadores para o monitoramentode áreas construídas,visando a prevenção de danos ambientais.

Milk from different animals can be used for dairy production. Yoghurt is a popular fermented milk product and considered to be one of the greatest importance in terms of consumer acceptance and consumption. The present research deals with the production of strawberry set-type yoghurt by mixing goat and buffalo s milk and it has the objective of taking advantage of the intrinsic characteristics of each milk to produce a final product with desirable attributes. It was conducted by analyzing five experimental groups with different proportions of goat and buffalo s milk: C 100% goat s milk; 7C3B - 70% goat s milk and 30% buffalo s milk, 5C5B - 50% goat s milk and 50% buffalo s milk, 3C7B 30% goat s milk and 70% buffalo s milk; B - 100% buffalo s milk. Each group was evaluated for total solids content and the acidification profile was monitored every 30 minutes by pH analysis. The yoghurt samples were analyzed for physical-chemical (pH, acidity, protein, fat, total and reducing sugars, ash and total solids), rheological (syneresis and viscosity) and sensory characteristics (appearance, odor, consistency and flavour). Samples with higher percentual of bubaline milk reached Vm faster, but the time necessary for pH 4.6 (Te) were similar between groups. Statistical differences (p<0.05) were observed for fat and total solids content of yoghurt, with superior values for groups higher proportions of buffalo s milk. The parameters of behavior reached by the model of Ostwald of Waale pointed yoghurt samples as non-Newtonian and pseudoplastic fluids. Yoghurt made only with goat s milk (C) had higher values (p<0.05) for syneresis, which can be explained by its fragile coagulum. Additionally, this group also had the lowest sensory scores for the attributes consistence and taste, while bubaline yoghurt (B) obtained the best acceptance indexes for all of the appraised parameters

Among the main challenges in the beer industrial production is the market supply at the lowest cost and high quality, in order to ensure the expectations of customers and. consumers The beer fermentation stage represents approximately 70% of the whole time necessary to its production, having a obligatoriness of strict process controls to avoid becoming bottleneck in beer production. This stage is responsible for the formation of a series of subproducts, which are responsible for the composition of aroma/bouquet existing in beer and some of these subproducts, if produced in larger quantities, they will confer unpleasant taste and odor to the final product. Among the subproducts formed during the fermentation stage, total vicinal diketones is the main component, since it is limiting for product transfusion to the subsequent steps, besides having a low perception threshold by the consumer and giving undesirable taste and odor. Due to the instability of main raw materials quality and also process controls during fermentation, the development of alternative forms of beer production without impacting on total fermentation time and final product quality is a great challenge to breweries. In this work, a prior acidification of the pasty yeast was carried out, utilizing for that phosphoric acid, food grade, reducing yeast pH of about 5.30 to 2.20 and altering its characteristic from flocculent to pulverulent during beer fermentation. An increase of six times was observed in amount of yeast cells in suspension in the second fermentation stage regarding to fermentations by yeast with no prior acidification. With alteration on two input variables, temperature curve and cell multiplication, which goal was to minimize the maximum values for diketones detected in the fermenter tank, a reduction was obtained from peak of formed diacetyl and consequently contributed to reduction in fermentation time and total process time. Several experiments were performed with those process changes in order to verify the influence on the total fermentation time and total vicinal diketones concentration at the end of fermentation. This experiment reached as the best production result a total fermentation time of 151 hours and total vicinal diketone concentration of 0.08 ppm. The mass of yeast in suspension in the second phase of fermentation increased from 2.45 x 106 to 16.38 x 106 cells/mL of yeast, which fact is key to a greater efficiency in reducing total vicinal diketones existing in the medium, confirming that the prior yeast acidification, as well as the control of temperature and yeast cell multiplication in fermentative process enhances the performance of diketones reduction and consequently reduce the total fermentation time with diketones concentration below the expected value (Max: 0.10 ppm)

O objetivo deste trabalho foi avaliar a dinâmica de nutrientes na solução do solo após a aplicação, via fertirrigação, de nitrogênio, fósforo e potássio a laranjeiras. O experimento foi realizado entre setembro de 2007 e outubro de 2009, em pomares de laranjeiras 'Valência' e 'Hamlin', enxertadas sobre citrumeleiro 'Swingle'. Foram avaliadas cinco doses de N, P2O5 e K2O (0, 25, 50, 100 e 200% da dose recomendada). A solução do solo foi extraída a 30 e 60 cm de profundidade, com o auxílio de extratores com cápsulas porosas. Foram realizadas 11 avaliações durante o período experimental, com as extrações iniciadas após 12 horas das fertirrigações. O aumento das doses reduziu o pH (pH~3,5, na maior dose), e aumentou a condutividade elétrica (CE~1,5 dS m-1, na maior dose) e os teores de NH4, NO3, P, K, Mn e Zn na solução do solo, nas duas profundidades amostradas. Nos meses com maior precipitação pluvial, houve perda potencial de nutrientes por lixiviação, pois maiores concentrações de NO3, K e B foram observadas à profundidade de 60 cm. A análise da solução do solo, obtida por extratores com cápsula de cerâmica porosa, pode ser considerada ferramenta auxiliar para monitorar e avaliar a disponibilidade de nutrientes às plantas.

A viabilização e aumento da produção de novas cultivares podem ser obtidos com a prática da adubação nitrogenada. O objetivo desse trabalho foi verificar o efeito de fontes e doses de adubação nitrogenada, em profundidades, na colonização micorrízica e na fertilidade do solo com Brachiaria brizantha cv. Xaraés. O delineamento experimental em blocos casualizados e os tratamentos em esquema fatorial 3 x 4, envolvendo três fontes de nitrogênio (ureia, sulfato de amônio e ajifer-L40), quatro doses (0, 100, 200 e 400 kg ha-1) estudadas em duas profundidades (0-0,10 e 0,10-0,20 m), com três repetições. Por parcela, as amostras compostas, por cinco amostras simples de solo, foram coletadas, peneiradas, homogeneizadas e destinadas às análises. O uso do ajifer e da ureia proporcionou maior produtividade de massa seca da parte aérea de B. brizantha cv. Xaraés. em contrapartida, esta sofre redução à medida que as doses se elevaram. A melhor dose de adubação foi a aplicação de 100 kg ha-1 de N. As doses mais elevadas de N, na camada 0 a 0,10 m de profundidade, determinam acidificação do solo e reduzem os valores de MO, K+, Ca2+, Mg2+, SB e V%. A esporulação, mas não colonização micorrízica, é sensivelmente estimulada em elevadas doses.

As pastagens geralmente perdem seu potencial produtivo e vigor de rebrota nos primeiros anos, frequentemente associada à falta de adubação nitrogenada. O objetivo deste foi verificar o efeito de fontes e doses de adubação nitrogenada na atividade microbiana (carbono da biomassa microbiana e carbono do CO2 liberado) e na fertilidade do solo cultivado com Brachiaria brizantha cv. Xaraés. O delineamento experimental foi em blocos casualizados, em arranjo fatorial 3 x 4, envolvendo três fontes de nitrogênio (uréia, sulfato de amônio e Ajifer-L40) e quatro doses de nitrogênio (0, 100, 200 e 400 kg ha-1), nas profundidades 0 - 0,10 e 0,10 - 0,20 m, com três repetições. O aumento nas doses de nitrogênio, na camada 0,0 a 0,10 m de profundidade, reduziu os valores de pH, MO, K+, Ca2+, Mg2+, SB e V%. O Ajifer e a uréia elevaram os valores de Ca2+ na camada 0,10 - 0,20 m, e as doses de 100 e 200 kg ha-1 aumentaram os valores de Mg2+ e SB. em doses elevadas o sulfato de amônio acidificou o solo. A perda de carbono pela elevada atividade microbiana pode estar relacionado ao estresse metabólico devido à acidificação do solo. A aplicação de 100 kg ha-1 de nitrogênio acarretou as menores perdas de carbono pela atividade microbiana.

Racional — A resposta do esfíncter esofágico superior ao refluxo gastroesofágico é controvertida. A perfusão esofágica com ácido clorídrico é modelo de estudo da ação do ácido o componente mais agressivo do refluxo sobre o esfíncter. Objetivos - Estudar o efeito da acidificação esofágica sobre o esfíncter esofágico superior através da eletromanometria esofágica. - Material e Métodos - em 30 cães adultos de ambos os sexos foram registrados estudos eletromanométricos do esôfago. A técnica utilizada foi a de puxada intermitente da sonda e perfusão contínua dos cateteres com água destilada. Estes exames permitiram as medidas da amplitude da pressão (mm Hg) e do comprimento (cm) do esfíncter superior do esôfago em condições basais (momento 1). Após esta fase, os animais foram submetidos a perfusão esofágica e divididos em 3 grupos de 10, na dependência da solução utilizada na perfusão e do momento do estudo: Grupo 1: perfusão esofágica com água destilada e estudos eletromanométricos realizados aos 15 minutos (momento 2) e 30 minutos (momento 3) do término da perfusão; Grupo 2: perfusão esofágica com HCl 0,1 N e estudos eletromanométricos realizados 15 minutos após o término da perfusão (momento 2); Grupo 3: perfusão esofágica com HCl 0,1 N e estudos eletromanométricos realizados 30 minutos após o término da perfusão (momento 3). Resultados/Conclusão - Os resultados observados permitiram concluir que a acidificação do esôfago não provocou alterações significativas sobre a amplitude de pressão e comprimento do esfíncter superior do esôfago.

INTRODUCTION: Metabolic investigation in patients with urinary lithiasis is very important for preventing recurrence of disease. The objective of this work was to diagnose and to determine the prevalence of metabolic disorders, to assess the quality of the water consumed and volume of diuresis as potential risk factors for this pathology. PATIENTS and METHODS: We studied 182 patients older than 12 years. We included patients with history and/or imaging tests confirming at least 2 stones, with creatinine clearance > 60 mL/min and negative urine culture. The protocol consisted in the collection of 2, 24-hour urine samples, for dosing Ca, P, uric acid, Na, K, Mg, Ox and Ci, glycemia and serum levels of Ca, P, Uric acid, Na, K, Cl, Mg, U and Cr, urinary pH and urinary acidification test. RESULTS: 158 patients fulfilled the inclusion criteria. Among these, 151 (95.5%) presented metabolic changes, with 94 (62.2%) presenting isolated metabolic change and 57 (37.8%) had mixed changes. The main disorders detected were hypercalciuria (74%), hypocitraturia (37.3%), hyperoxaluria (24.1%), hypomagnesuria (21%), hyperuricosuria (20.2%), primary hyperparathyroidism (1.8%) secondary hyperparathyroidism (0.6%) and renal tubular acidosis (0.6). CONCLUSION: Metabolic change was diagnosed in 95.5% of patients. These results warrant the metabolic study and follow-up in patients with recurrent lithiasis in order to decrease the recurrence rate through specific treatments, modification in alimentary and behavioral habits.

Nanostructured calcium phosphate (CaP) has been histologically and biomechanically proven to enhance osseointegration of implants; however, conventional techniques were not sufficiently sensitive to capture its biological effects fully. Here, we compared the conventional removal torque (RTQ) evaluation and gene expression in tissues around nanostructured CaP-coated implants, using real-time RT-PCR, with those of uncoated implants, in a rabbit model. At 2 wks, RTQ values were significantly higher, alkaline phosphatase (ALP) expression was significantly higher, and runt-related transcription factor 2 and tumor necrosis factor-alpha expressions were significantly lower in the coated than in the uncoated implants. This indicates that inflammatory responses were suppressed and osteoprogenitor activity increased around the CaP-coated surface. At 4 wks, although RTQ values did not significantly differ between the 2 groups, ALP and osteocalcin (OCN) were significantly up-regulated in the coated group, indicating progressive mineralization of the bone around the implant. Moreover, an osteoclast marker, adenosine triphosphatase, which indicates acidification of the resorption lacunae, was significantly higher for the coated implants, suggesting gradual resorption of the CaP coating. This study reveals detailed genetic responses to nanostructured CaP-coated implants and provides evidence that the effect of nanotopography is significant during the osseointegration cascade.
