For those who have read even one of my musings, it will come as no surprise that I find Facebook, Twitter, social networking sites (SNS), and the rest of Webology less than inspiring. If you had read nothing other than the screed I blathered about Google a few columns back, you’d know that I find all this talk about the Web replacing libraries more than a little silly; I find it downright idiotic. Still, one must keep an open mind.

Durante o longo período de experiências de magistério no ensino tecnológico de segundo e terceiro graus, o autor desta dissertação de mestrado vem aplicando uma metodologia que consiste em cativar os estudantes através de projetos inusitados, elaborados de conformidade com as habilitações específicas dos cursos, empregando-os como elemento motivador para realizar empreendimentos, paralelamente ao ensino curricular. Esse motivo, escolhido e planejado pedagogicamente, serve de estímulo para concentrar as atenções dos alunos na solução de problemas sócio-econômicos do país e da humanidade em geral, com o propósito de integrá-los no desenvolvimento científico e tecnológico, despertando o interesse pelo estudo e pela pesquisa, nas atividades dos projetos realizados por grupos de colegas de vocações afins. o projeto, caracterizado por um complexo cibernético ou biônico, tem a propriedade de efetivar a conjugação das diversas disciplinas, cujos conhecimentos são aplicados nessas oportunidades, ajudando os alunos a compreenderem melhor o sentido pragmático do conjunto curricular do seu curso. Atuando, de início, autodidatamente, mas orientado pelos princípios da Psicologia Educacional e mais tarde apoiado na Pedagogia de Dewey, o autor aprimorou sua metodologia nos trabalhos de Decroly, Kilpatrick, Skinner e Mc Clelland. A aplicação desse método de ensino suscitou a criaçao de um Centro de Pesquisas, integrado na organização escolar, através de uma de suas assessorias, sendo dirigido por um professor responsável e estruturado na forma de um setor onde se reunem as equipes de Planejamento Educacional e Pesquisa, Planejamento Técnico, Desenvolvimento de Atividades e Apoio, todas formadas por professores convidados, especialistas nos assuntos envolvidos pelos projetos. Este setor-ambiente, bem arquitetado,é instalado com recursos auxiliares didáticos e motivacionais, pequena biblioteca, museu, arquivo, uma pequena oficina de precisão, um laboratório para pesquisas tecnológicas e outro para tratamento fotográfico. Evidencia-se, assim, que o Centro de Pesquisas, como agente de motivação realizadora, atua na escola produzindo mudanças significativas no processo de ensino e consequentemente na formação dos alunos, pela sua metodologia e dinâmica das atividades a que estes se dedicam,manifestand sua capacidade de produzir, aplicando a criatividade aliada à sua aprendizagem, ao mesmo tempo valiosas para sua educação e para a sociedade. Fundamentado nos bons resultados obtidos com a aplicação dessa metodologia de ensino, o autor sugere a conveniência de serem implantados nas demais Escolas Técnicas e nos Centros Federais de Educação Tecnológica, centros ou núcleos de pesquisas semelhantes ao descrito nesta dissertação de mestrado.

Much research has explored the relationship between economics and elections, and scholars have begun to explore how institutions mediate that link. However, the relationship between presidential institutions and electoral accountability remains largely unexplored in comparative politics. Because voters in presidential systems can cast votes for executive and legislative elections separately, we have good reasons to suspect that the institutions of presidentialism might generate different forms or degrees of accountability than parliamentarism. Powell and Whitten (1993) suggest that the partisan or institutional “clarity of responsibility” might mediate the relationship between economics and elections: when responsibility for outcomes is clear, the relationship should be strong, and vice-versa. I develop this notion for use in presidential systems, and explore executive and legislative elections in 24 countries. The results indicate that economics always influences the incumbent vote in executive elections, regardless of the partisan or institutional clarity of responsibility. Economics also affects vote swings in legislative elections, but the institutional clarity of responsibility does mediate this relationship: legislative accountability for national economic outcomes is lowest when clarity of responsibility is highest, a situation that arises when the president is relatively more powerful and the bases for electing legislators and the president differ. By providing an empirical basis for a discussion of accountability under presidentialism, these findings contribute to important debates in comparative politics.

A city is made not only to streets, sidewalks, buildings, bridges and viaducts. The city is also built by layers of sediment from the past that blend with layers of sediments present. The city's it's art, it's sociability is written. The aim of this study is to analyze how Mossoró historiographically the narrative is built by Luís da Câmara Cascudo. With this objective, we divide the work into three chapters. At first, we investigate the investment that the prefecture of the Mossoró city at the beginning of the forties, under the administration of Dix-sept Rosado, held for the construction of what would be the city's culture. A culture that was linked to the creation of a library, a museum, a university, the completion of several lectures on the themes of the city, and writing the history of Mossoró. The second chapter discusses the historical conditions of possibility that made Luís da Câmara Cascudo of the town historian. In the last chapter, we show how Mossoró was built by Cascudo from the specific analysis of the book Notas e Documentos para a história de Mossoró (1955). We discuss the conditions for its emergence, examining the interplay of interests that enabled its production

Neste trabalho, apresenta-se a descrição histórica da Biblioteca Pública do Maranhão. Aborda-se a situação econômica, política e social vivenciada no estado, no século XIX, que contribuiu para a sua criação e expõem-se, ainda, os fatores que condicionaram a sua falência e o seu ressurgimento no início da Primeira República, marcada pela identidade sociocultural progressista. Além disso, pontuam-se os principais aspectos educativos existentes na biblioteca, na gestão de Antônio Lobo. O método empregado se caracteriza como histórico-descritivo, baseando-se, principalmente, na análise e na descrição dos Relatórios Presidenciais do Maranhão, referentes ao século XIX e início do XX. Desta forma, o estudo constatou que a história da biblioteca do Maranhão foi marcada pelas dificuldades relacionadas, especialmente, à falta de interesse dos governantes em proporcionar condições necessárias para a promoção da cultura.

An 11-year-old girl with short stature, mental retardation, and mild dysmorphic features was found to have an inverted duplication of most of the short arm of the X chromosome [dic inv dup(X)(qter --> p22.3 = p22.3 --> cen:)]. Her mother, who is also short and retarded, carries the same duplication. Fluorescence in situ hybridization with an X chromosome library, and with X centromere-specific alpha satellite and telomere probes, was useful in characterizing the duplication. In most females with structurally abnormal X chromosomes, the abnormal chromosome is inactivated. Although the duplicated X was consistently late replicating in the mother, X chromosome inactivation studies in the proband indicated that in 11 % of her lymphocytes the duplicated X was active.

The evolutionarily conserved factor eIF5A is the only protein known to undergo hypusination, a unique posttranslational modification triggered by deoxyhypusine synthase (Dys1). Although eIF5A is essential for cell viability, the function of this putative translation initiation factor is still obscure. To identify eIF5A-binding proteins that could clarify its function, we screened a two-hybrid library and identified two eIF-5A partners in S. cerevisiae: Dys1 and the protein encoded by the gene YJR070C, named Lia1 (Ligand of eIF5A). The interactions were confirmed by GST pulldown. Mapping binding sites for these proteins revealed that both eIF5A domains can bind to Dys1, whereas the C-terminal domain is sufficient to bind Lia1. We demonstrate for the first time in vivo that the N-terminal α-helix of Dys1 can modulate enzyme activity by inhibiting eIF5A interaction. We suggest that this inhibition be abrogated in the cell when hypusinated and functional eIF5A is required. © 2003 Published by Elsevier B.V. on behalf of the Federation of European Biochemical Societies.

Background: Yellow fever virus (YFV) belongs to the Flavivirus genus and causes an important disease. An alarming resurgence of viral circulation and the expansion of YFV-endemic zones have been detected in Africa and South America in recent years. NS5 is a viral protein that contains methyltransferase and RNA-dependent RNA polymerase (RdRp) domains, which are essential for viral replication, and the interactions between NS5 and cellular proteins have been studied to better understand viral replication. The aim of this study was to characterize the interaction of the NS5 protein with eukaryotic translation initiation factor 3 subunit L (eIF3L) and to evaluate the role of eIF3L in yellow fever replication. Methods. To identify interactions of YFV NS5 with cellular proteins, we performed a two-hybrid screen using the YFV NS5 RdRp domain as bait with a human cDNA library, and RNApol deletion mutants were generated and analyzed using the two-hybrid system for mapping the interactions. The RNApol region involved was segmented into three fragments and analyzed using an eIF3L-expressing yeast strain. To map the NS5 residues that are critical for the interactions, we performed site-direct mutagenesis in segment 3 of the interaction domain (ID) and confirmed the interaction using in vitro assays and in vivo coimmunoprecipitation. The significance of eIF3L for YFV replication was investigated using eIF3L overexpression and RNA interference. Results: In this work, we describe and characterize the interaction of NS5 with the translation factor eIF3L. The interaction between NS5 and eIF3L was confirmed using in vitro binding and in vivo coimmunoprecipitation assays. This interaction occurs at a region (the interaction domain of the RNApol domain) that is conserved in several flaviviruses and that is, therefore, likely to be relevant to the genus. eIF3L overexpression and plaque reduction assays showed a slight effect on YFV replication, indicating that the interaction of eIF3L with YFV NS5 may play a role in YFV replication. Conclusions: Although the precise function of eIF3L on interactions with viral proteins is not entirely understood, these results indicate an interaction of eIF3L with YF NS5 and that eIF3L overexpression facilitates translation, which has potential implications for virus replication. © 2013 Morais et al.; licensee BioMed Central Ltd.

The traditional characteristics and challenges for organizing and searching information on the World Wide Web are outlined and reviewed. The classification features of two of these methods, such as Google, in the case of automated search engines, and Yahoo! Directory, in the case of subject directories are analyzed. Recent advances in the Semantic Web, particularly the growing application of ontologies and Linked Data are also reviewed. Finally, some problems and prospects related to the use of classification and indexing on the World Wide Web are discussed, emphasizing the need of rethinking the role of classification in the organization of these resources and outlining the possibilities of applying Ranganathan's facet theories of classification.

SynopsisBackgroundCellulite refers to skin relief alterations in womens thighs and buttocks, causing dissatisfaction and search for treatment. Its physiopathology is complex and not completely understood. Many therapeutic options have been reported with no scientific evidence about benefits. The majority of the studies are not controlled nor randomized; most efficacy endpoints are subjective, like not well-standardized photographs and investigator opinion. Objective measures could improve severity assessment. Our purpose was to correlate non-invasive instrumental measures and standardized clinical evaluation.MethodsTwenty six women presenting cellulite on buttocks, aged from 25 to 41, were evaluated by: body mass index; standardized photography analysis (10-point severity and 5-point photonumeric scales) by five dermatologists; cutometry and high-frequency ultrasonography (dermal density and dermis/hypodermis interface length). Quality of life impact was assessed. Correlations between clinical and instrumental parameters were performed.ResultsGood agreement among dermatologists and main investigator perceptions was detected. Positive correlations: body mass index and clinical scores; ultrasonographic measures. Negative correlation: cutometry and clinical scores. Quality of life score was correlated to dermal collagen density.ConclusionCellulite caused impact in quality of life. Poor correlation between objective measures and clinical evaluation was detected. Cellulite severity assessment is a challenge, and objective parameters should be optimized for clinical trials.
