A comparative study of nine assay methods for dextransucrase and related enzymes has been made. A relatively widespread method for the reaction of dextransucrase with sucrose is the measurement of the reducing value of D-fructose by alkaline 3,5-dinitrosalicylate (DNS) and thereby the amount of D-glucose incorporated into dextran. Another method is the reaction with C-14-sucrose with the addition of an aliquot to Whatman 3MM paper squares that are washed three times with methanol to remove C-14-D-fructose and unreacted C-14-sucrose, followed by counting of C-14-dextran on the paper by liquid scintillation counting (LSC). It is shown that both methods give erroneous results. The DNS reducing value method gives extremely high values due to over-oxidation of both D-fructose and dextran, and the C-14-paper square method gives significantly low values due to the removal of some of the C-14-dextran from the paper by methanol washes. In the present study, we have examined nine methods and find two that give values that are identical and are an accurate measurement of the dextransucrase reaction. They are (1) a C-14-sucrose/dextransucrase digest in which dextran is precipitated three times with three volumes of ethanol, dissolved in water, and added to paper and counted in a toluene cocktail by LSC: and (2) precipitation of dextran three times with three volumes of ethanol from a sucrose/dextransucrase digest, dried, and weighed. Four reducing value methods were examined to measure the amount of D-fructose. Three of the four (two DNS methods, one with both dextran and D-fructose and the other with only D-fructose, and the ferricyanide/arsenomolybdate method with is-fructose) gave extremely high values due to over-oxidation of D-fructose, D-glucose, leucrose, and dextran. Synthesis and characterization of a new Pt(II) complex with the amino acid L-alliin (S-allyl-L-cysteine sulfoxide, C(6)H(11)NO(3)S) are described. Elemental and mass spectrometric analyses of the solid complex are consistent with [PtCl(2)(alliin)], or [PtCl(2)(C(6)H(11)NO(3)S)]. (13)C nuclear magnetic resonance (NMR), [(1)H-(15)N] two dimensional (2D) NMR and infrared spectroscopy indicate coordination of the ligand to Pt(II) through the N and S atoms. The complex is very soluble in dimethyl sulfoxide. Biological analysis for evaluation of a potential cytotoxic effect of the complex was performed using HeLa cells derived from human cervical adenocarcinoma. The complex presented moderate cytotoxic activity, inducing about 40% cell death at a concentration of 400 mol L(-1).

The electromagnetic tensor for inclusive electron scattering off the pion Wμν for momentum transfers such that q+ = 0, (q+ = q0 + q3) is shown to obey a sum-rule for the component W++. From this sum-rule, one can define the quark-antiquark correlation function in the pion, which characterizes the transverse distance distribution between the quark and antiquark in the light-front pion wave-function. Within the realistic models of the relativistic pion wave function (including instanton vacuum inspired wave function) it is shown that the value of the two-quark correlation radius (rqq̄) is near twice the pion electromagnetic radius (rπ), where rπ ≈ 2/3 fm. We also define the correlation length lcorr where the two-particle correlation have an extremum. The estimation of lcorr ≈ 0.3-0,5 fm is very close to estimations from instanton models of QCD vacuum. It is also shown that the above correlation is very sensitive to the pion light-front wave-function models. © 1997 Elsevier Science B.V.

The lower bound masses of the ground-state relativistic three-boson system in 1 + 1, 2 + 1 and 3 + 1 spacetime dimensions are obtained. We have considered a reduction of the ladder Bethe-Salpeter equation to the lightfront in a model with renormalized two-body contact interaction. The lower bounds are deduced with the constraint of reality of the two-boson subsystem mass. It is verified that, in some cases, the lower bound approaches the ground-state binding energy. The corresponding non-relativistic limits are also verified.

Antimicrobial activity of 45 extracts of medicinal plants were tested on Xanthomonas campestris pv. vesicatoria, Ralstonia solanacearum and Clavibacter michiganense subsp. michiganense. Some assays were done to verify the capability of these plants extrats to show an antibiosis. Five extracts (EAFQ, SM1, SM12, SM16, SA1) shown positive activity. The extract EAFQ expressed bactericide activity on C. michiganense. It suggests the possibility of using these actives substances in natura or as a model to synthesize industrialized products, intendind field utilization.

The digestive system of the capybara has been investigated because of its coprofagia habits, important for their absorptive activity. These species present differences in terms of gastrointestinal morphological characters when compared with other rodents. Macroscopiclly, the stomach of the capybara is constituted of the following parts: cardiac, pyloric, body, fundic and gastric diverticulum. It presents two curvatures, one big and another small. Externally, the presence of gastric bands (tenias) is observed. With regards to the volumetric view, the gastric capacity varies from 850 to 2010 ml, with an average of 1498.57 ml. So, the stomach of this animal can be classified as a simple stomach, in the format of a curved sack and similar to an inverted letter 'J'. The gastric mucous membrane presents a surface filled by numerous tortuous gastric folds and longitudinally distributed along all its extension. The mucous tunic also possesses recesses located among the successive gastric folds, which were denoted as gastric parts with numerous openings described as gastric pits. In the cardiac part, a glandular epithelium with cardiac glands is noticed containing a lot of parietal and mucous neck cells. The fundic part, body and gastric diverticulum contain proper gastric glands with main, parietal and mucous neck cells. Finally, the pyloric part has pyloric glands with two cellular types, mucous neck and parietal cells.

Broiler chicks aged 12 h after hatching were allotted according to a block design in a 7 x 2 factorial schedule of 14 treatments and four replications of 50 chicks each one. The main experimental factors were fasting for 0, 6, 12, 18, 24, 30, and 36 h after chick placement and sex. Independent of sex, fasting had a negative linear effect on weight and productivity of broilers at market age (42 d) without affecting feed conversion or mortality index. Groups subjected to 18 and 36 h of fasting after placement, corresponding to 30 and 48 h posthatching fasting, had lower biometrical values for small intestine (length, weight, and size; villus height; and crypt depth) than chicks fed immediately after placement. According to the Pearson test, BW of birds at 21 and 42 d were significantly correlated to BW at 7 d (r = 0.77) and 21 d (r = 0.45), respectively. Males performed better than females but had higher mortality rates. Fasting did not influence serum concentrations of corticosterone or sexual steroid hormones. Nevertheless, early signs of sexual dimorphism arose from the high estradiol (E2) concentration on female serum. Heterophil:lymphocyte ratio was not different among treatments, indicating that early fasting did not seem to be a stress factor 21 or 42 d after fasting. The results suggested a maximum fasting of 24 h after hatching in order to preserve broiler productivity at market age.

The pion electromagnetic form factor is calculated in the space- and time-like regions from -10 (GeV/c)2 up to 10 (GeV/c)2, within a front-form model. The dressed photon vertex where a photon decays in a quark-antiquark pair is depicted generalizing the vector meson dominance ansatz, by means of the vector meson vertex functions. An important feature of our model is the description of the on-mass-shell vertex functions in the valence sector, for the pion and the vector mesons, through the front-form wave functions obtained within a realistic quark model. The theoretical results show an excellent agreement with the data in the space-like region, while in the time-like region the description is quite encouraging. © 2003 Elsevier B.V. An approach for a unified description of the pion electromagnetic form factor in the space- and time-like regions, within a constituent quark model on the light front is briefly illustrated. Three main ingredients enter our approach: i) the on-shell quark-hadron vertex functions in the valence sector, ii) the dressed photon vertex where a photon decays in a quark-antiquark pair, and iii) the emission and absorption amplitudes of a pion by a quark. © 2005 American Institute of Physics.

A general technique to embed non-uniform displacement discontinuities into standard solid finite elements is presented. The technique is based on the decomposition of the kinematic fields into a component related to the deformation of the solid portion of the element and one related to the rigid-body motion due to a displacement discontinuity. This decomposition simplifies the incorporation of discontinuity interfaces and provides a suitable framework to account for non-uniform discontinuity modes. The present publication addresses two families of finite element formulations: displacement-based and stress hybrid finite element. © 2005 Elsevier Ltd. All rights reserved.

Toxoplasmosis is a zoonosis caused by Toxoplasma gondii, a coccidian protozoan of worldwide distribution. The seroprevalence in canine population can be an alternative for measuring T. gondii urban spreading. A total of 780 blood samples from dogs were collected, during the yearly anti-rabies campaign, carried out by the Department of Veterinary Hygiene and Public Health, School of Veterinary Medicine and Animal Husbandry (FMVZ), São Paulo State University, UNESP, together with the county health authorities, in August 1999. Using Indirect Fluorescent Antibody Test (IFAT) for detecting antibodies anti-T. gondii in the sera samples, we observed that 258 dogs (33.1%) were positive. The associations between the serological results and the epidemiological variables were studied. Statistically significant differences were not found regarding sex (32.2% male and 34.3% female reactors). Dogs without a defined breed showed seropositivity statistically higher than the pedigreed group. The occurrence of infection was considered higher with age.

The putative translation initiation factor 5A (eIF5A) is a highly abundant and conserved protein in all eukaryotes and archaebacteria. This factor is essential for cell viability and is the only cellular protein known to contain the unusual amino acid residue hypusine. In Saccharomyces cerevisiae eIF5A is expressed in aerobic conditions by the gene TIF51A. Although eIF5A has been known for almost 30 years, the biological role of this protein is still obscure. This article reviews the research on the function of eIF5A, discussing the evidence for its involvement in various steps of mRNA metabolism, including translation initiation, nucleocytoplasmic transport and mRNA decay. Moreover, it indicates other studies that have associated eIF5A with cell proliferation and cell cycle progression. Finally, this review presents recent results obtained in our laboratory that reemphasize the role of eIF5A in the translation scenario. Further experiments will be necessary to define the role played by eIF5A in the translational machinery.

Twelve elephant grass genotypes were evaluated in a randomized blocks design with three repetitions. The experimental parcel was of 4 lines with 3m of length, spaced of 1 m and fertilized with 100 kg of P2O5, 100 kg of N, 60 kg of K2O and 25 kg of micronutrients/ha. Significant differences were observed among the genotypes for neutral detergent fiber (FND), acid detergent fiber (FAD) and lignin. The genotypes CNPGL 92-70-02, CNPGL 91-25-01 and CNPGL 93-32-02 showed low values for FND, FAD and lignin, simultaneously, outstanding between the others. There were no significant differences in the levels of dry matter and crude protein contents and in in vitro dry matter digestibility between the genotypes.

The main objective of this work is to analyze the ability of FACTS devices like TCSC and UPFC to damp low frequency oscillations and a POD controller is also included. A comparative study of damping effect of those devices IS carried out. The Power Sensitivity Model (PSM) is used to the representation of the electric power system. Sensibility analysis using the residue method shows the best place for the installation of FACTS and the procedure to determine POD parameters. ©2008 IEEE.
