A argila bentonítica é amplamente utilizada em transporte de sólidos produzidos durante perfuração de poços. Teve por objetivo estudar o escoamento de misturas bentonita-água e determinar suas propriedades reológicas e parâmetros hidráulicos úteis nos projetos de instalações de recalque de misturas sólido-líquido. Foi montado um circuito fechado de tubulações para estudar dados de perda de carga e perfis de velocidade. Realizaram-se ensaios com misturas bentonita-água sob varias concentrações, algumas transportando areia. Observaram-se que a reologia da mistura bentonita-água é melhor descrita pela formulação de Herschell-Bulkley para fluidos não-Newtonianos. O coeficiente de atrito para descrever a perda de carga da mistura bentonita-água observada em tubulações no laboratório coloca-se entre as previsões de Tomita (1959) e Szilas et al (1981). A variação da velocidade da mistura na seção transversal do tubo é melhor aproximada pela equação de Bogue-Metzner (1963).

As doenças tropicais negligenciadas (DTNs) causam um imenso sofrimento para a pessoa acometida e em muitos casos podem levar o indivíduo a morte. Elas representam um obstáculo devastador para a saúde e continuam a ser um sério impedimento para a redução da pobreza e desenvolvimento socioeconômico. Das 17 doenças desse grupo, a leishmaniose, incluindo a leishmaniose cutânea, tem grande destaque devido sua alta incidência, os gastos para o tratamento e as complicações geradas em processos de coinfecção. Ainda mais agravante, os investimentos direcionados ao controle, combate e principalmente a inovação em novos produtos é ainda muito limitado. Atualmente, a academia tem um importante papel na luta contra essas doenças através da busca de novos alvos terapêuticos e também de novas moléculas com potencial terapêutico. É nesse contexto que esse projeto teve como meta a implantação de uma plataforma para a identificação de moléculas com atividade leishmanicida. Como alvo terapêutico, optamos pela utilização da enzima diidroorotato desidrogenase de Leishmania Viannia braziliensis (LbDHODH), enzima de extrema importância na síntese de novo de nucleotídeos de pirimidina, cuja principal função é converter o diidroorotato em orotato. Esta enzima foi clonada, expressa e purificada com sucesso em nosso laboratório. Os estudos permitiram que a enzima fosse caracterizada cineticamente e estruturalmente via cristalografia de raios- X. Os primeiros ensaios inibitórios foram realizados com o orotato, produto da catálise e inibidor natural da enzima. O potencial inibitório do orotato foi mensurado através da estimativa do IC50 e a interação proteína-ligante foi caracterizada através de estudos cristalográficos. Estratégias in silico e in vitro foram utilizadas na busca de ligantes, através das quais foram identificados inibidores para a enzima LbDHODH. Ensaios de validação cruzada, utilizando a enzima homóloga humana, permitiram identificar os ligantes com maior índice de seletividade que tiveram seu potencial leishmanicida avaliado in vitro contra as formas promastigota e amastigota de Leishmania braziliensis. A realização do presente projeto permitiu a identificação de uma classe de ligantes que apresentam atividade seletiva contra LbDHODH e que será utilizada no planejamento de futuras gerações de moléculas com atividade terapêutica para o tratamento da leishmaniose. Além disso, a plataforma de ensaios otimizada permitirá a avaliação de novos grupos de moléculas como uma importante estratégia na busca por novos tratamentos contra a leishmaniose

The purpose of this study was to explore the leadership capacities and practices of assistant principals. The research also sought to determine what relationships existed between capacity and practice and to see if there was a difference based on experience, context and personal characteristics. Since the majority of principals first serve as assistant principals, their work and experiences as assistant principals will have significant consequences (Kwan, 2009). The literature has long held and continues to challenge the notion that the role of assistant principal is adequate preparation for the principalship (Chan, Webb, & Bowen, 2003; Harris, Muijs, & Crawford, 2003; Kwan, 2009; Mertz, 2000; Webb & Vulliamy, 1995). Based on empirical findings, this study has affirmed the need to further research and refine the role of the assistant principal. The results indicate that in addition to strengths, there are explicit gaps and missed opportunities in the leadership practices of assistant principals that impact the potential for building a leadership pipeline within schools. The work of the assistant principal is characterized by a proliferation of duties rather than a strategic set of practices that support distributed leadership and sustainability.

O trabalho aborda a aplicação da técnica de reconciliação de dados para o balanço da movimentação de gás natural em uma malha de escoamento de gás não processado, elaborando também um método de cálculo rápido de inventário de um duto. Foram aplicadas, separadamente, a reconciliação volumétrica à condição padrão de medição e a reconciliação mássica, bem como realizadas comparações dos resultados em relação ao balanço original e verificação do balanço resultante de energia em termos de poder calorífico superior. Dois conjuntos de pesos foram aplicados, um arbitrado de acordo com o conhecimento prévio da qualidade do sistema de medição de cada um dos pontos, outro baseado no inverso da variância dos volumes diários apurados no período. Ambos apresentaram bons resultados e o segundo foi considerado o mais apropriado. Por meio de uma abordagem termodinâmica, foi avaliado o potencial impacto, ao balanço, da condensação de parte da fase gás ao longo do escoamento e a injeção de um condensado de gás natural não estabilizado por uma das fontes. Ambos tendem a impactar o balanço, sendo o resultado esperado um menor volume, massa e energia de fase gás na saída. Outros fatores de considerável impacto na qualidade dos dados e no resultado final da reconciliação são a qualidade da medição de saída do sistema e a representatividade da composição do gás neste ponto. O inventário é calculado a partir de uma regressão que se baseia em um regime permanente de escoamento, o que pode apresentar maior desvio quando fortes transientes estão ocorrendo no último dia do mês, porém a variação de inventário ao longo do mês possui baixo impacto no balanço. Concluiu-se que a reconciliação volumétrica é a mais apropriada para este sistema, pois os dados reconciliados levam os balanços mássicos e de energia em termos de poder calorífico, ambos na fase gás, para dentro do perfil esperado de comportamento. Embora um balanço volumétrico nulo apenas da fase gás não seja por si só o comportamento esperado quando se considera os efeitos descritos, para desenvolver um balanço mais robusto é necessário considerar as frações líquidas presentes no sistema, agregando maior dificuldade na aquisição e qualidade dos dados.

Gasoline Distribution Generally Available Control Technology (GD GACT) is a Federal environmental regulation that is specifically written and enforced to reduce HAP emissions from gasoline distribution (GD) facilities. The regulation targets four specific types of GD facilities: bulk gasoline terminals, bulk gasoline plants, pipeline breakout stations, and pipeline pumping stations. A GD GACT compliance plan was developed for a particular, representative example of each type of GD facility affected by the regulation. Each facility in the study is owned and operated by a single company. The compliance plans were developed to meet the regulatory requirements contained within GD GACT. The compliance plans will be implemented at each facility prior to the January 10, 2011 compliance date.

Feature vectors can be anything from simple surface normals to more complex feature descriptors. Feature extraction is important to solve various computer vision problems: e.g. registration, object recognition and scene understanding. Most of these techniques cannot be computed online due to their complexity and the context where they are applied. Therefore, computing these features in real-time for many points in the scene is impossible. In this work, a hardware-based implementation of 3D feature extraction and 3D object recognition is proposed to accelerate these methods and therefore the entire pipeline of RGBD based computer vision systems where such features are typically used. The use of a GPU as a general purpose processor can achieve considerable speed-ups compared with a CPU implementation. In this work, advantageous results are obtained using the GPU to accelerate the computation of a 3D descriptor based on the calculation of 3D semi-local surface patches of partial views. This allows descriptor computation at several points of a scene in real-time. Benefits of the accelerated descriptor have been demonstrated in object recognition tasks. Source code will be made publicly available as contribution to the Open Source Point Cloud Library.

Context. Young massive clusters are key to map the Milky Way’s structure, and near-infrared large area sky surveys have contributed strongly to the discovery of new obscured massive stellar clusters. Aims. We present the third article in a series of papers focused on young and massive clusters discovered in the VVV survey. This article is dedicated to the physical characterization of VVV CL086, using part of its OB-stellar population. Methods. We physically characterized the cluster using JHKS near-infrared photometry from ESO public survey VVV images, using the VVV-SkZ pipeline, and near-infrared K-band spectroscopy, following the methodology presented in the first article of the series. Results. Individual distances for two observed stars indicate that the cluster is located at the far edge of the Galactic bar. These stars, which are probable cluster members from the statistically field-star decontaminated CMD, have spectral types between O9 and B0 V. According to our analysis, this young cluster (1.0 Myr < age < 5.0 Myr) is located at a distance of 11+5-6 kpc, and we estimate a lower limit for the cluster total mass of (2.8+1.6-1.4) · 103 M⊙. It is likely that the cluster contains even earlier and more massive stars.

The Gaia-ESO Survey is a large public spectroscopic survey that aims to derive radial velocities and fundamental parameters of about 105 Milky Way stars in the field and in clusters. Observations are carried out with the multi-object optical spectrograph FLAMES, using simultaneously the medium-resolution (R ~ 20 000) GIRAFFE spectrograph and the high-resolution (R ~ 47 000) UVES spectrograph. In this paper we describe the methods and the software used for the data reduction, the derivation of the radial velocities, and the quality control of the FLAMES-UVES spectra. Data reduction has been performed using a workflow specifically developed for this project. This workflow runs the ESO public pipeline optimizing the data reduction for the Gaia-ESO Survey, automatically performs sky subtraction, barycentric correction and normalisation, and calculates radial velocities and a first guess of the rotational velocities. The quality control is performed using the output parameters from the ESO pipeline, by a visual inspection of the spectra and by the analysis of the signal-to-noise ratio of the spectra. Using the observations of the first 18 months, specifically targets observed multiple times at different epochs, stars observed with both GIRAFFE and UVES, and observations of radial velocity standards, we estimated the precision and the accuracy of the radial velocities. The statistical error on the radial velocities is σ ~ 0.4 km s-1 and is mainly due to uncertainties in the zero point of the wavelength calibration. However, we found a systematic bias with respect to the GIRAFFE spectra (~0.9 km s-1) and to the radial velocities of the standard stars (~0.5 km s-1) retrieved from the literature. This bias will be corrected in the future data releases, when a common zero point for all the set-ups and instruments used for the survey is be established.

In many regions, seawater desalination is a growing industry that has its impact on benthic communities. This study analyses the effect on benthic communities of a mitigation measure applied to a brine discharge, using polychaete assemblages as indicator. An eight-year study was conducted at San Pedro del Pinatar (SE Spain) establishing a grid of 12 sites at a depth range of 29–38 m during autumn. Brine discharge started in 2006 and produced a significant decrease in abundance, richness and diversity of polychaete families at the location closest to the discharge, where salinity reached 49. In 2010, a diffuser was deployed at the end of the pipeline in order to increase the mixing, to reduce the impact on benthic communities. After implementation of this mitigation measure, the salinity measured close to discharge was less than 38.5 and a significant recovery in polychaete richness and diversity was detected, to levels similar to those before the discharge. A less evident recovery in abundance was also observed, probably due to different recovery rates of polychaete families. Some families like Paraonidae and Magelonidae were more tolerant to this impact. Others like Syllidae and Capitellidae recovered quickly, although still affected by the discharge, while some families such as Sabellidae and Cirratulidae appeared to recover more slowly.

Sensing techniques are important for solving problems of uncertainty inherent to intelligent grasping tasks. The main goal here is to present a visual sensing system based on range imaging technology for robot manipulation of non-rigid objects. Our proposal provides a suitable visual perception system of complex grasping tasks to support a robot controller when other sensor systems, such as tactile and force, are not able to obtain useful data relevant to the grasping manipulation task. In particular, a new visual approach based on RGBD data was implemented to help a robot controller carry out intelligent manipulation tasks with flexible objects. The proposed method supervises the interaction between the grasped object and the robot hand in order to avoid poor contact between the fingertips and an object when there is neither force nor pressure data. This new approach is also used to measure changes to the shape of an object’s surfaces and so allows us to find deformations caused by inappropriate pressure being applied by the hand’s fingers. Test was carried out for grasping tasks involving several flexible household objects with a multi-fingered robot hand working in real time. Our approach generates pulses from the deformation detection method and sends an event message to the robot controller when surface deformation is detected. In comparison with other methods, the obtained results reveal that our visual pipeline does not use deformations models of objects and materials, as well as the approach works well both planar and 3D household objects in real time. In addition, our method does not depend on the pose of the robot hand because the location of the reference system is computed from a recognition process of a pattern located place at the robot forearm. The presented experiments demonstrate that the proposed method accomplishes a good monitoring of grasping task with several objects and different grasping configurations in indoor environments.

The EU relies to a considerable degree on imports to meet its demand for natural gas. Whereas Norwegian export pipelines are directly connected to the EU gas system, a major share of Russian gas flows through the Ukrainian territory before reaching consumers located other consumers located down in the supply chain (e.g. Slovakia, Hungary or Italy). But is the Ukrainian gas transit route still a risk? Will the construction of the South Stream pipeline further reduce the importance of Ukraine as a transit country? Or is there more at stake here than meets the eye?

An act restricting Gazprom’s monopoly in Russian gas exports came into effect on 1 December 2013. Previously Gazprom had had a legal guarantee to its monopoly position. The changes are an effect of consultations between various ministries that had been conducted for many months and were affected by lobbying from Novatek and Rosneft (Gazprom’s competitors on the domestic gas market); they need not, though, be seen as system changes. The ‘liberalisation’ they appear to bring in is feigned. Proof of this are found for example in both the limited material scope of the new law (it concerns only exports of liquefied natural gas, LNG) and the small number of the beneficiaries of the new regulations (the new solutions will be beneficial for Novatek and Rosneft). Contrary to initial announcements, the right to export LNG has not been restricted to South-Eastern Asian markets, which means that Russian liquefied natural gas is also likely to be sold to Europe in the coming years. Although these changes have been motivated above all by the individual interests of Gazprom’s competitors, they are also to a certain extent a response to the processes taking place on regional gas markets. They may, therefore, turn out to be beneficial for the state (increasing Russia’s share on the global LNG market and attracting foreign investors to gas extraction projects being implemented in Russia). The new regulations are probably the first step down the long road to breaking Gazprom’s monopoly in gas exports via the pipeline system.

The energy security of countries importing energy resources depends largely on the shape and quality of operational transport connections. This is particularly important in the case of natural gas supplies. Natural gas is transported mostly by gas pipelines which permanently connect gas producers and consumers. Thus Europe as a consumer is "tied" to certain gas suppliers for anywhere between a dozen and several tens of years. As their own resources are becoming depleted, the EU Member States get increasingly dependent on import of natural gas. The present paper discusses the existing and projected gas transport routes from Russia to the EU. The first part deals with the importance of gas exports to the economy of the Russian Federation, and the second delves into the EU Member States' dependence on gas imports. Then this paper examines the differences in perceiving the energy security issue between the old and the new Member States, those differences stemming from the different degrees of their dependence on Russian supplies. In the third part, two new transport route projects for Russian gas supplies to the EU are compared and it is argued that from the point of view of the Community's interests, the Yamal gas pipeline is a better solution than the North European (Trans-Baltic) gas pipeline.

Nord Stream increases Gazprom’s flexibility as far its export routes are concerned; it enables them to be changed with regard to the market or political situation. Nevertheless, this expensive pipeline may contribute to a further drop in the price competitiveness of Russian gas. Accordingly, increasing the attractiveness of Russian fuel and ensuring profitable sales is steadily becoming the main challenge for Gazprom in the EU against a backdrop of increasing competitiveness on the market.
