Agaro-oligosaccharides were hydrolytically obtained from agar using hydrochloric acid, citric acid, and cationic exchange resin (solid acid). The FT-IR and NMR data showed that the hydrolysate has the structure of agaro-oligomers. Orthogonal matrix method was applied to optimize the preparation conditions based on alpha-naphthylamine end-labeled HPLC analysis method. The optimal way for oligosaccharides with different degree of polymerization (DP) was achieved by using solid acid degradation, which could give high yield and avoid solution neutralization process. Agaro-oligosaccharides with high purity were consequently obtained by activated carbon column isolation. Furthermore, the antioxidant and alpha-glucosidase inhibitory activity of three fractions were also investigated. The result indicated that 8% ethanol-eluted fraction showed highest activity against alpha-glucosidase with IC50 of 8.84 mg/mL, while 25% ethanol-eluted fraction possessed excellent antioxidant ability.

Large yellow croaker, Pseudosciaena crocea, exhibit sexually dimorphic growth, with females growing faster and reaching larger adult sizes than males. Thus, development of techniques for preferentially producing females is necessary to optimize production of these species. We have established a protocol to produce all-female croaker P. crocea through induction of meiotic gynogenesis with homologous sperm. The first set of experiments investigated the ultra-violet (UV) irradiation on sperm motility and duration of sperm activity to determine the optimal UV dosage for genetic inactivation of sperm, yet retaining adequate motility for activation of eggs. Milt from several males was diluted 1: 100 with Ringer's solution and UV irradiated with doses ranging from 0-150 J cm (-2). The results indicated that motility and duration of activity generally decreased with increased UV doses. At UV doses greater than 105 J cm(-2), after fertilization, motility was < 10% and fertilization rates were significantly lower. Highest hatching rate was obtained at 75 J cm -2. A second set of experiments was carried out to determine appropriate conditions of cold shock for retention of the 2nd polar body in P. crocea eggs after fertilization with UV-inactivated sperm by altering the timing, temperature and duration of shock. At 208 degrees C, shock applied at 3 min after fertilization resulted in higher survival rate of larvae at 6 h after hatching. Results of different combinations of three shock temperatures ( 28 degrees C, 38 degrees C or 48 degrees C) and five shock durations ( 4 min, 8 min, 12 min, 16 min or 20 min) at 3 min after fertilization demonstrated that shocks of 12 min gave highest production of diploid gynogens. Statistical analysis revealed that maximum production of diploid gynogens (44.55 +/- 2.99%) were obtained at 38 degrees C. The results of this study indicate that the use of UV-irradiated homologous sperm for activation of P. crocea eggs and cold shock for polar body retention is an effective method for producing gynogenetic offspring.

Turbot Scophthalmus maximus exhibits sexually dimorphic growth, with females growing faster and reaching larger adult sizes than males. Thus, development of techniques for preferentially producing females is necessary to optimize production of these species. In this paper, gynogenetic diploids of turbot were induced by activating egg development with ultraviolet (UV)-irradiated left-eyed flounder Paralichthys olivaceus sperm combined with cold shock to prevent extrusion of the second polar body. The results of UV irradiation experiments showed that survival, motility, and duration of activity of P. olivaceus sperm generally decreased with increase in UV dose. The typical Hertwig's effect was observed after fertilized turbot eggs with UV-irradiated P. olivaceus sperm and the optimal UV dose for gynogenetic haploid production was 36,000 erg mm(-2). At 15 degrees C, appropriate timing of cold shock for retention of the second polar body in turbot eggs was at 6 min after fertilization. Results of different combinations of two shock temperatures (1 or 3 degrees C) and four shock durations (15, 25, 35 or 45 min) at 6 min after fertilization demonstrated that shock of 25 min at 1 degrees C gave the highest production of diploid gynogens (39.58% relative to its diploid control). The results of this study reveal that the use of UV-irradiated P. olivaceus sperm for activation of turbot eggs and cold shock for polar body retention is an effective method to produce gynogenetic offspring.

Batch cultivation for transgenic kelp gametophyte cells was investigated in an online controlled 5 L stirred-tank photo-bioreactor to rapidly optimize the process conditions by monitoring the rate of increase of pH. The transgenic kelp gametophytes with heterologous gene encoding hepatitis B surface antigen (HBsAg) could rapidly grow in the bioreactor. Optimal temperature and agitation rate for bioreactor cultivation of gametophytes were 15 degrees C and 200 rpm. Optimal incident light intensities depended on the initial cell densities. 数值模式是潮波研究的一种有利手段,但在研究中会面临各种具体问题,包括开边界条件的确定、底摩擦系数和耗散系数的选取等。数据同化是解决这些问题的一种途径,即利用有限数量的潮汐观测资料对潮波进行最优估计,其根本目的是迫使模型预报值逼近观测值,使模式不要偏离实际情况太远。本文采用了一种优化开边界方法,沿着数值模型的开边界优化潮汐水位信息,目的是设法使数值解在动力约束的意义下接近观测值,获得研究区域的潮汐结果。边界值由指定优化问题的解来定,以提高模拟区域的潮汐精度,最优问题的解是基于通过开边界的能量通量的变化,处理开边界处的观测值与计算值之差的最小化。这里提供了辐射型边界条件,由Reid 和Bodine(本文简称为RB)推导,我们将采用的优化后的RB方法(称为ORB)是优化开边界的特殊情况。 本文对理想矩形海域( E- E, N- N, 分辨率 )进行了潮波模拟,有东部开边界,模式采用ECOM3D模式。对数据结果的误差分析采用,振幅平均偏差,平均绝对偏差,平均相对误差和均方根偏差四个值来衡量模拟结果的好坏程度。 需要优化入开边界的解析潮汐值本文采用的解析解由方国洪《海湾的潮汐与潮流》(1966年)方法提供,为验证本文所做的解析解和方文的一致,本文做了其第一个例子的关键值a,b,z,结果与其结果吻合的相当好。但略有差别,分析的可能原因是两法在具体迭代方案和计算机保留小数上有区别造成微小误差。另外,我们取m=20,得到更精确的数值,我们发现对前十项的各项参数值,取m=10,m=20各项参数略有改进。当然我们可以获得m更大的各项参数值。 同时为了检验解析解的正确性讨论m和l变化对边界值的影响,结果指出,增大m,m=20时,u的模最大在本身u1或u2的模的6%;m=100时,u的模最大在本身u1或u2的模的4%;m再增大,m=1000时,u的模最大在本身u1或u2的模的4%,改变不大。当l<1时, =0处u的模最大为2。当l=1时, =0处u的模最大为0.1,当l>1时,l越大,u的模越小,当l=10时,u的模最大为0.001,可以认为为0。 为检验该优化方法的应用情况,我们对理想矩形区域进行模拟,首先将本文所采用的优化开边界方法应用于30m的情况,在开边界优化入开边界得出模式解,所得模拟结果与解析解吻合得相当好,该模式解和解析解在整个区域上,振幅平均绝对偏差为9.9cm,相位平均绝对偏差只有4.0 ,均方根偏差只有13.3cm,说明该优化方法在潮波模型中有效。 为验证该优化方法在各种条件下的模拟结果情况,在下面我们做了三类敏感性试验: 第一类试验:为证明在开边界上使用优化方法相比于没有采用优化方法的模拟解更接近于解析解,我们来比较ORB条件与RB条件的优劣,我们模拟用了两个不同的摩擦系数,k分别为:0,0.00006。 结果显示,针对不同摩擦系数,显示在开边界上使用ORB条件的解比使用RB条件的解无论是振幅还是相位都有显著改善,两个试验均方根偏差优化程度分别为84.3%,83.7%。说明在开边界上使用优化方法相比于没有采用优化方法的模拟解更接近于解析解,大大提高了模拟水平。上述的两个试验得出, k=0.00006优化结果比k=0的好。 第二类试验,使用ORB条件确定优化开边界情况下,在东西边界加入出入流的情况,流考虑线性和非线性情况,结果显示,加入流的情况,潮汐模拟的效果降低不少,流为1Sv的情况要比5Sv的情况均方根偏差相差20cm,而不加流的情况只有0.2cm。线性流和非线性流情况两者模式解相差不大,振幅,相位各项指数都相近, 说明流的线性与否对结果影响不大。 第三类试验,不仅在开边界使用ORB条件,在模式内部也使用ORB条件,比较了内部优化和不优化情况与解析解的偏差。结果显示,选用不同的k,振幅都能得到很好的模拟,而相位相对较差。另外,在内部优化的情况下,考虑不同的k的模式解, 我们选用了与解析解相近的6个模式解的k,结果显示,不同的k,振幅都能得到很好的模拟,而相位较差。 总之,在开边界使用ORB条件比使用RB条件好,振幅相位都有大幅度改进,在加入出入流情况下,流的大小对模拟结果有影响,但线形流和非线性流差别不大。内部优化的结果显示,模式采用不同的k都能很好模拟解析解的振幅。

Studies were carried out to optimize the conditions for the recovery of protein. The results showed that pH of 6.00 for wastewater, the dosage of 1% chitosan solution in 1% acetic acid aqueous solution of 2.0 ml for 50 ml wastewater and 1% FeCl3 aqueous solution of 2 ml for 50 ml wastewater, the flocculation time of 4.0 h were the optimal conditions for the recovery of protein. The obtained protein sediment contained abundant amino acids, especially isoleucine, methione and lysine that are absent in other protein resource. (c) 2007 Elsevier Ltd. All rights reserved.

The aim of this study was to optimize the cryopreservation protocols for the sperm of red seabream, Pagrus major. The 2-mL cryovials and programmable freezer were employed for cryopreservation. Six extenders, six cryoprotectants in various concentrations ranging from 6 to 20% (v/v), four cooling rates, and three thawing temperatures were evaluated by postthaw sperm motility and fertility. The ratio of sperm to egg for postthaw sperm fertilization trials was experimentally standardized and was optimal at 500:1. The best motility of postthaw sperm (79.4 +/- 4.7% to 88.6 +/- 8.0%), fertilization rates (89.6 +/- 2.9 to 95.6 +/- 1.9%), and hatching rates (85.3 +/- 5.1% to 91.4 +/- 4.3%) were achieved when Cortland extender, dimethyl sulfoxide (15, 18, and 20%) or ethylene glycol (9, 12%) as cryoprotectants, 20 C/min as the cooling rate, and 40 C as the thawing temperature were employed. Moreover, the results on embryonic development were not significantly different between cryopreserved sperm and fresh sperm during incubation process. In conclusion, these methods of cryopreservation of red seabream sperm are suitable for routine aquaculture application and preservation of genetic resources.

Studies on reproduction, hatchery management, and culture of Manila clams Ruditapes philippinarum were carried out in an attempt to optimize their culture conditions and techniques. Results from these studies led to the development of a three-phase culture method for Manila clam farming in northern China. The key components of the new method were: 1) early spawning and over-wintering indoors (greenhouse); 2) optimized larval culture conditions and techniques; 3) juvenile rearing in shallow, fertilized nursery ponds; 4) optimized stocking size and density and substrate for mudflat grow out. Broodstock were maturated indoors for a month from early April to early May. Primarily because of higher water temperatures in the greenhouse the clams spawned more than one month earlier than in the natural environment. From May to July, juveniles were reared for 1-2 months indoors to a size of 2.0-3.0 mm in shell length before being moved to outdoor, pre-disinfected, nursery ponds. Juveniles were then reared in the nursery ponds for one month to about 1.0 cm before being transferred to the mudflat for grow out. Juvenile clams in nursery ponds grew considerably faster than in the natural environment probably because of higher temperatures and more abundant natural food. During grow out, the clams were reared for 4-7 months until they reached a market size (3.0-3.3 cm). Juveniles produced after August were over-wintered in the greenhouse in which the water temperature was about 3 degrees C higher than that of the outdoor environment. Juveniles grew at an average rate of > 20 mu m day(-1), while in the natural environment no growth was observed during winter because of low temperatures. Juveniles in the greenhouse grew to 2-3 mm by the following March before being moved into outdoor nursery ponds. The three-phase culture method not only shortened the production period from spawn to market size from 24-36 months to about 10-14 months, but also prolonged the spawning season from 2 to 7 months, resulting in increased production of seed and market-size clams. Compared with the traditional method, the new method could increase the yield of market-size clams by 10-11 times, and increase the profit per ha mudflat by as much as 124 times and the profit per kg market-size clams produced by 13 times. 针对室内动态非结构化环境下的移动机器人路径规划问题,提出了一种能够将全局路径规划方法和局部路径规划方法相结合、将基于反应的行为规划和基于慎思的行为规划相结合的路径规划方法.全局路径规划器采用A*算法生成到达目标点的子目标节点序列;局部路径规划器采用改进的人工势场方法对子目标节点序列中相邻两节点进行路径平滑和优化处理.在考虑了移动机器人运动学约束的前提下,该方法不但能够充分利用已知环境信息生成全局最优路径,而且还能及时处理所遇到的随机障碍信息.仿真研究与在室内复杂环境下的实际运行结果验证了该方法的有效性.

为解决供应链生产计划协调问题,通过市场价格和中间库存因素使供应链上下游企业结合成一个整体,建立一种供应链上下游一体化计划模型,从整体考虑供应链合作计划问题.为获取问题的可行解,采用拉格朗日松弛技术进行优化,为供应链上下游企业在信息共享条件下实现"多赢"目标,提供了理论依据.仿真结果验证了模型和算法的有效性。

可变形机器人AMOEBA-I具有多种构形和多种转向方式.为实现机器人转向性能的优化,提出了可变形机器人的协同转向方法,并建立了相应的数学模型,对不同构形下的协同转向方式进行了理论分析.设定了机器人三个模块在协同转向过程中的运动关系,在此基础上给出了可变形机器人协同转向性能的评价指标.通过理论和实验比较了不同构形下的协同转向方式,实验验证了协同转向方法的有效性.

本文介绍了星球探测机器人视觉系统的标定方法,首先介绍了一种新的从视觉坐标系到车体坐标系的变换方法,随后给出了像机的模型,在像机参数优化过程中将三维重投影误差作为评价函数,利用遗传算法完成寻优过程,以保证估计出的像机参数全局最优。真实环境实验结果表明:该方法具有较高的空间定位精度。

提出了一种新的最优模糊PID控制器,它由两部分组成,即在线模糊推理机构和带有不完全微分的常规PID控制器,在模糊推理机构中,引入了三个可调节因子xp,xi和xd,其作用是进一步修改和优化模糊推理的结果,以使控制器对一个给定对象具有最优的控制效果,可调节因子的最优值采用ITAE准则及Nelder和Mead提出的柔性多面体最优搜索算法加以确定,这种PID控制器被用来控制由作者设计的智能人工腿中的一个直流电机,仿真结果表明该控制器的设计是非常有效的,它可被用于控制各种不同的对象和过程。

提出了一种既能够在陆地上爬行,又能够在一定深度的水下浮游和在海底爬行的新概念轮桨腿一体化两栖机器人;多运动模式和复合移动机构是该机器人的突出特点.分析了轮桨腿复合式驱动机构的运动机理,并采用多目标优化设计理论和算法,对驱动机构的爬行性能和浮游特性进行了综合优化,得到了两栖机器人驱动机构的结构优化参数.虚拟样机的仿真结果证明了该轮桨腿一体化两栖机器人驱动机构的综合运动性能良好,对非结构环境具有一定的适应能力。
