LRRK2 is one of the most important genetic contributors to Parkinson’s disease (PD). Point mutations in this gene cause an autosomal dominant form of PD, but to date no cellular phenotype has been consis- tently linked with mutations in each of the functional domains (ROC, COR and Kinase) of the protein product of this gene. In this study, primary fibroblasts from individuals carrying pathogenic mutations in the three central domains of LRRK2 were assessed for alterations in the autophagy/lysosomal pathway using a combination of biochemical and cellular approaches. Mutations in all three domains resulted in alterations in markers for autophagy/lysosomal function compared to wild type cells. These data high- light the autophagy and lysosomal pathways as read outs for pathogenic LRRK2 function and as a marker for disease, and provide insight into the mechanisms linking LRRK2 function and mutations.

We study a series of transient entries into the low-latitude boundary layer (LLBL) of all four Cluster spacecraft during an outbound pass through the mid-afternoon magnetopause ([X(GSM), Y(GSM), Z(GSM)] approximate to [2, 7, 9] R(E)). The events take place during an interval of northward IMF, as seen in the data from the ACE satellite and lagged by a propagation delay of 75 min that is well-defined by two separate studies: (1) the magnetospheric variations prior to the northward turning (Lockwood et al., 2001, this issue) and (2) the field clock angle seen by Cluster after it had emerged into the magnetosheath (Opgenoorth et al., 2001, this issue). With an additional lag of 16.5 min, the transient LLBL events cor-relate well with swings of the IMF clock angle (in GSM) to near 90degrees. Most of this additional lag is explained by ground-based observations, which reveal signatures of transient reconnection in the pre-noon sector that then take 10-15 min to propagate eastward to 15 MLT, where they are observed by Cluster. The eastward phase speed of these signatures agrees very well with the motion deduced by the cross-correlation of the signatures seen on the four Cluster spacecraft. The evidence that these events are reconnection pulses includes: transient erosion of the noon 630 nm (cusp/cleft) aurora to lower latitudes; transient and travelling enhancements of the flow into the polar cap, imaged by the AMIE technique; and poleward-moving events moving into the polar cap, seen by the EISCAT Svalbard Radar (ESR). A pass of the DMSP-F15 satellite reveals that the open field lines near noon have been opened for some time: the more recently opened field lines were found closer to dusk where the flow transient and the poleward-moving event intersected the satellite pass. The events at Cluster have ion and electron characteristics predicted and observed by Lockwood and Hapgood (1998) for a Flux Transfer Event (FTE), with allowance for magnetospheric ion reflection at Alfvenic disturbances in the magnetopause reconnection layer. Like FTEs, the events are about 1 R(E) in their direction of motion and show a rise in the magnetic field strength, but unlike FTEs, in general, they show no pressure excess in their core and hence, no characteristic bipolar signature in the boundary-normal component. However, most of the events were observed when the magnetic field was southward, i.e. on the edge of the interior magnetic cusp, or when the field was parallel to the magnetic equatorial plane. Only when the satellite begins to emerge from the exterior boundary (when the field was northward), do the events start to show a pressure excess in their core and the consequent bipolar signature. We identify the events as the first observations of FTEs at middle altitudes.

Most studies concerned with the representations of local people in tourism discourse point to the prevalence of stereotypic images asserting that contemporary tourism perpetuates colonial legacy and gendered discursive practices. This claim has been, to some extent, contested in research that explores representations of hosts in local tourism materials claiming that tourism can also discursively resist the dominant Western imagery. While the evidence for the existence of hegemonic and diverging discourses about the local ‘Other’ seems compelling, the empirical basis of this research is rather small and often limited to one geographic context. The present study addresses these shortcomings by examining representations of hosts in a larger corpus of promotional tourism materials including texts produced by Western and local tourism industries. The data is investigated using the methodology of Corpus-Assisted Discourse Studies (CADS). By comparing external with internal (self) representations, this study verifies and refines some of the claims on the subject and offers a much more nuanced picture of representations that defies the black and white scenarios proposed in previous research

The Team Formation problem (TFP) has become a well-known problem in the OR literature over the last few years. In this problem, the allocation of multiple individuals that match a required set of skills as a group must be chosen to maximise one or several social positive attributes. Speci�cally, the aim of the current research is two-fold. First, two new dimensions of the TFP are added by considering multiple projects and fractions of people's dedication. This new problem is named the Multiple Team Formation Problem (MTFP). Second, an optimization model consisting in a quadratic objective function, linear constraints and integer variables is proposed for the problem. The optimization model is solved by three algorithms: a Constraint Programming approach provided by a commercial solver, a Local Search heuristic and a Variable Neighbourhood Search metaheuristic. These three algorithms constitute the first attempt to solve the MTFP, being a variable neighbourhood local search metaheuristic the most effi�cient in almost all cases. Applications of this problem commonly appear in real-life situations, particularly with the current and ongoing development of social network analysis. Therefore, this work opens multiple paths for future research.

A large amount of biological data has been produced in the last years. Important knowledge can be extracted from these data by the use of data analysis techniques. Clustering plays an important role in data analysis, by organizing similar objects from a dataset into meaningful groups. Several clustering algorithms have been proposed in the literature. However, each algorithm has its bias, being more adequate for particular datasets. This paper presents a mathematical formulation to support the creation of consistent clusters for biological data. Moreover. it shows a clustering algorithm to solve this formulation that uses GRASP (Greedy Randomized Adaptive Search Procedure). We compared the proposed algorithm with three known other algorithms. The proposed algorithm presented the best clustering results confirmed statistically. (C) 2009 Elsevier Ltd. In this paper we present a genetic algorithm with new components to tackle capacitated lot sizing and scheduling problems with sequence dependent setups that appear in a wide range of industries, from soft drink bottling to food manufacturing. Finding a feasible solution to highly constrained problems is often a very difficult task. Various strategies have been applied to deal with infeasible solutions throughout the search. We propose a new scheme of classifying individuals based on nested domains to determine the solutions according to the level of infeasibility, which in our case represents bands of additional production hours (overtime). Within each band, individuals are just differentiated by their fitness function. As iterations are conducted, the widths of the bands are dynamically adjusted to improve the convergence of the individuals into the feasible domain. The numerical experiments on highly capacitated instances show the effectiveness of this computational tractable approach to guide the search toward the feasible domain. Our approach outperforms other state-of-the-art approaches and commercial solvers. (C) 2009 Elsevier Ltd. All rights reserved.

This paper deals with the classical one-dimensional integer cutting stock problem, which consists of cutting a set of available stock lengths in order to produce smaller ordered items. This process is carried out in order to optimize a given objective function (e.g., minimizing waste). Our study deals with a case in which there are several stock lengths available in limited quantities. Moreover, we have focused on problems of low demand. Some heuristic methods are proposed in order to obtain an integer solution and compared with others. The heuristic methods are empirically analyzed by solving a set of randomly generated instances and a set of instances from the literature. Concerning the latter. most of the optimal solutions of these instances are known, therefore it was possible to compare the solutions. A lot sizing and scheduling problem prevalent in small market-driven foundries is studied. There are two related decision levels: (I the furnace scheduling of metal alloy production, and (2) moulding machine planning which specifies the type and size of production lots. A mixed integer programming (MIP) formulation of the problem is proposed, but is impractical to solve in reasonable computing time for non-small instances. As a result, a faster relax-and-fix (RF) approach is developed that can also be used on a rolling horizon basis where only immediate-term schedules are implemented. As well as a MIP method to solve the basic RF approach, three variants of a local search method are also developed and tested using instances based on the literature. Finally, foundry-based tests with a real-order book resulted in a very substantial reduction of delivery delays and finished inventory, better use of capacity, and much faster schedule definition compared to the foundry`s own practice. The focus of study in this paper is the class of packing problems. More specifically, it deals with the placement of a set of N circular items of unitary radius inside an object with the aim of minimizing its dimensions. Differently shaped containers are considered, namely circles, squares, rectangles, strips and triangles. By means of the resolution of non-linear equations systems through the Newton-Raphson method, the herein presented algorithm succeeds in improving the accuracy of previous results attained by continuous optimization approaches up to numerical machine precision. We present approximation algorithms for the three-dimensional strip packing problem, and the three-dimensional bin packing problem. We consider orthogonal packings where 90 degrees rotations are allowed. The algorithms we show for these problems have asymptotic performance bounds 2.64, and 4.89, respectively. These algorithms are for the more general case in which the bounded dimensions of the bin given in the input are not necessarily equal (that is, we consider bins for which the length. the width and the height are not necessarily equal). Moreover, we show that these problems-in the general version-are as hard to approximate as the corresponding oriented version. (C) 2009 Elsevier Ltd. All rights reserved.

Given a fixed set of identical or different-sized circular items, the problem we deal with consists on finding the smallest object within which the items can be packed. Circular, triangular, squared, rectangular and also strip objects are considered. Moreover, 2D and 3D problems are treated. Twice-differentiable models for all these problems are presented. A strategy to reduce the complexity of evaluating the models is employed and, as a consequence, instances with a large number of items can be considered. Diferentes espécies vegetais são denominadas popularmente como arnica, devido ao uso em ferimentos, traumatismos e contusões, semelhantes ao da espécie oficial Arnica montana L. Dentre elas, destacam-se as seguintes: Porophyllum ruderale (Jacq.) Cass., nativa do Brasil, com maior frequência na região Sudeste, constituída por óleo essencial, flavonoides, taninos, alcaloides e saponinas; Solidago sp., planta originária da América do Sul e encontrada em vários terrenos brasileiros, constituída por flavonoides, óleo essencial e saponinas. Este trabalho teve por objetivo realizar a avaliação farmacognóstica das espécies Porophyllum ruderale e Solidago sp. A parte aérea seca das espécies foi pulverizada e tamisada, e analisaram-se as características organolépticas dos pós: aspecto, cor, odor e sabor; a triagem fitoquímica foi realizada a partir de métodos tradicionais. Verificou-se a presença de taninos condensados e flavonoides em ambas as espécies, e não foi confirmada a presença de saponinas e alcaloides em nenhuma das amostras.

O café é um produto de forte apelo nacional e tem estado presente nos hábitos do consumidor deste produto, denotando uma busca por cafés diferenciados e de qualidade. As avaliações do produto se baseiam nas posições descritivas, dedutivas e de informações do mesmo. As características percebidas podem ser intrínsecas como o sabor, o aroma, a cor, a aparência e a consistência e extrínsecas, que referem-se a parte externa e que envolve o produto, tais como o ambiente, a embalagem, a marca e também o país de origem. Este estudo teve por objetivo investigar se as características intrínsecas (sabor, aroma, cor, aparência e consistência) da bebida café e as características extrínsecas associadas ao produto estimulam o consumidor em frequentar cafeterias (ambiente especializado em café). Para tal foi realizada uma pesquisa de caráter exploratório, com levantamento de campo e bibliográfico cujos resultados foram tratados por meio de análise de conteúdo. Utilizou-se como instrumento de coleta de dados um roteiro de entrevistas e foram entrevistadas 87 pessoas, frequentadoras de cafeterias. As cafeterias pesquisadas estão localizadas na cidade de São Paulo sendo: duas num Shopping Center do bairro de Higienópolis e uma cafeteria em uma universidade no centro da capital. Os resultados obtidos demonstraram que o sabor, prioritariamente e, depois o aroma do café foram as características intrínsecas consideradas mais relevantes entre os respondentes. Por outro lado, um ambiente agradável, confortável e requintado foram fatores valorizados pelos entrevistados. Ambas características se mostraram complementares e estes fatores citados foram os apontados como aqueles que estimulam o consumidor a frequentar as cafeterias.

O Rio Grande do Sul possui seis regiões vitícolas, sendo a denominada “Serra Gaúcha” a maior região vitícola brasileira. Os índices bioclimáticos, bem como os modelos existentes para estimativas da fenologia e do rendimento qualitativo da videira foram estabelecidos para regiões vitícolas de diversos países, com clima distinto do encontrado nas regiões vitivinícolas do Rio Grande do Sul, sendo necessário verificar a sua eficácia nestas condições. Por esse motivo, estudos no sentido de estabelecer o comportamento da cultura frente às condições do ambiente, em especial o clima, são necessários para o planejamento do seu cultivo. Os objetivos deste estudo foram caracterizar a fenologia das principais cultivares, estabelecer modelos para a estimativa das fases fenológicas e determinar relações entre variáveis meteorológicas e o rendimento qualitativo para a videira da “Serra Gaúcha”. Para isso, foram utilizados dados fenológicos e de qualidade do mosto, nos ciclos vegetativos de 1984 a 1994 e dados meteorológicos e do teor de açúcar do mosto da cv. Cabernet Franc de 1961 a 2000 Os resultados fenológicos permitiram classificar as videiras, quanto à época de brotação, em precoces, médias e tardias e, quanto à maturação, como de primeira, segunda e terceira época. A data da brotação foi estimada com duas a três semanas de antecedência. Foi possível prever as datas da floração, mudança de cor das bagas e colheita, a partir da data da brotação, com desvio padrão médio de 2, 3 e 5 dias, respectivamente. A duração do brilho solar, a deficiência hídrica e o quociente heliopluviométrico foram positivamente correlacionados com a qualidade da uva, enquanto que a precipitação pluvial e o excesso hídrico apresentaram efeito contrário. A melhor estimativa do teor de açúcar da uva Cabernet Franc foi obtida com a duração do brilho solar e a precipitação pluvial ou duração do brilho solar e deficiência hídrica dos meses de dezembro a fevereiro.

Objetivo: Determinar a prevalência de má absorção de lactose e sua associação com a cor da pele e a idade em crianças e adolescentes de escolas públicas do município de Porto Alegre. Material e Métodos: Foi realizado um estudo transversal que incluiu 225 indivíduos de 8 a 18 anos, freqüentadores de escolas públicas do município de Porto Alegre. Os participantes foram classificados segundo a cor e a faixa etária. A má absorção de lactose foi diagnosticada através do teste do hidrogênio expirado após ingestão de 250 ml de leite. O teste teve duração de 3 horas e foi considerado como critério de positividade o aumento ≥ 20 partes por milhão na concentração de hidrogênio em relação ao nível basal. Resultados: Foram estudados 225 alunos, com uma média e desvio padrão de idade de 12,2 ± 2,0 anos. Cento e cinqüenta e quatro alunos eram de cor branca (68,4%) e os restantes, de cor não-branca (preta ou parda). A má absorção de lactose foi evidenciada em 19/225 casos (8,4%). Foram diagnosticados 8/154 casos (5,2%) nos alunos de cor branca e 11/71 casos (15,5%) nos alunos de cor não-branca (p = 0,02). Em relação à faixa etária, ocorreram 15/143 casos (10,5%) nos alunos entre 8 e 12 anos e 4/82 casos (4,9%) entre 13 e 18 anos (p = 0,227). Conclusões: A prevalência de má absorção de lactose encontrada em alunos de escolas pú-blicas de Porto Alegre é alta, especialmente se considerarmos que foram utilizadas doses fi-siológicas (250 ml de leite) para o diagnóstico. As taxas de má absorção foram maiores entre as crianças de cor não-branca em relação às crianças de cor branca, confirmando a influência racial na hipolactasia primária do tipo adulto.
