The aim of this study was to evaluate the ability of an Escherichia coli with the multiple antibiotic resistance (MAR) phenotype to withstand the stresses of slaughter compared to an isogenic progenitor strain. A wild type E. coli isolate (345-2RifC) of porcine origin was used to derive 3 isogenic MAR mutants. Escherichia coli 345-2RifC and its MAR derivatives were inoculated into separate groups of pigs. Once colonisation was established, the pigs were slaughtered and persistence of the E. coli strains in the abattoir environment and on the pig carcasses was monitored and compared. No significant difference (P>0.05) was detected between the shedding of the different E. coli strains from the live pigs. Both the parent strain and its MAR derivatives persisted in the abattoir environment, however the parent strain was recovered from 6 of the 13 locations sampled while the MAR derivatives were recovered from 11 of 13 and the number of MAR E. coil recovered was 10-fold higher than the parent strain at half of the locations. The parent strain was not recovered from any of the 6 chilled carcasses whereas the MAR derivatives were recovered from 3 out of 5 (P<0.001). This study demonstrates that the expression of MAR in 345-2RifC increased its ability to survive the stresses of the slaughter and chilling processes. Therefore in E. coli, MAR can give a selective advantage, compared to non-MAR strains, for persistence on chilled carcasses thereby facilitating transit of these strains through the food chain. Objectives: The aims of this study were to determine whether strains of Salmonella enterica serovar Typhimurium which had acquired low-level multiple antibiotic resistance (MAR) through repeated exposure to farm disinfectants were able to colonize and transmit between chicks as easily as the parent strain and, if such strains were less susceptible to fluoroquinolones, would high-level resistance be selected after fluoroquinolone treatment. Methods: Two mutants were compared with the isogenic parent. In the first experiment, day-old chicks were co-infected with both the parent and a mutant to determine their relative fitness. In the second experiment, parent and mutant strains (in separate groups of chicks) were assessed for their ability to transmit from infected (contact) to non-infected (naive) birds and with respect to their susceptibility to fluoroquinolone treatment. Birds were regularly monitored for the presence of Salmonella in caecal contents. Replica plating was used to monitor for the selection of antibiotic-resistant strains. Results: The parent strain was shown to be significantly fitter than the two mutants and was more rapidly disseminated to naive birds. Antibiotic treatment did not preferentially select for the two mutants or for resistant strains. Conclusions: The disinfectant-exposed strains, although MAR, were less fit, less able to disseminate than the parent strain and were not preferentially selected by therapeutic antibiotic treatment. As such, these strains are unlikely to present a greater problem than other salmonellae in chickens.

The virulence of a Salmonella enterica serovar Typhimurium DT014 strain in which marA was insertionally inactivated was compared to its isogenic parent in vitro and in vivo. In vitro, the numbers of the marA mutant phagocytosed by porcine lung macrophages were significantly increased, while survival at 24 h inside macrophages and adherence to human gut cells were significantly reduced in comparison with the parent strain. In vivo, the marA inactivated strain, in competition with its parent strain, persisted for a shorter period in chickens, was present in the caeca at significantly lower levels and invaded the deeper organs to a significantly lesser extent. Therapeutic antibiotic treatment of one group of chickens with oxytetracycline favoured the persistence of both the parent strain and, to a lesser extent, the marA inactivated strain; but interestingly, increased tetracycline resistance of Salmonella isolates after treatment of birds with antibiotic was seen only for the parent strain. Further work is needed to elucidate how mar is involved in virulence and if its inactivation can minimise the ability of bacteria to become antibiotic-resistant in vivo.

In order to understand the role of the mar locus in Salmonella with regard to multiple antibiotic resistance, cyclohexane resistance, and outer membrane protein F (OmpF) regulation, a marA::gfp reporter mutant was constructed in an antibiotic-sensitive salmonella enterica serovar Typhimurium DT104 background. Salicylate induced marA, whereas a number of antibiotics, disinfect ants, and various growth conditions did not. Increased antibiotic resistance was observed upon salicylate induction, although this was shown to be by both mar-dependent and mar-independent pathways. Cyclohexane resistance, however, was induced by salicylate by a mar-dependent pathway. Complementation studies with a plasmid that constitutively expressed marA confirmed the involvement of map in Salmonella with low-level antibiotic resistance and cyclohexane resistance, although the involvement of mar in down regulation of OmpF was unclear. However, marA overexpression did increase the expression of a ca. 50-kDa protein, but its identity remains to be elucidated Passage of the marA::gfp reporter mutant with increasing levels of tetracycline, a method reported to select for mar mutants in Escherichia coli, led to both multiple-antibiotic and cyclohexane resistance. Collectively, these data indicate that low-level antibiotic resistance, cyclohexane resistance, and modulation of OMPs in Salmonella, as in E. coli, can occur in both a mar-dependent and mar-independent manner.

Chromosomally encoded systems involved in low level resistance of bacteria to different classes of antibiotics (mainly beta-lactams, chloramphenicol, quinolones and tetracycline), disinfectants and in resistance to organic solvents have been the focus of considerable interest in recent years. The multiple antibiotic resistance (mar) locus of Escherichia coli and Salmonella is perhaps the best described system involved in this type of resistance which is induced by MarA, the activator protein encoded by the marRAB locus. The mar-locus is reported to mediate resistance primarily by up-regulating efflux of some antibiotics, disinfectants and organic solvents via the AcrAB-TolC efflux pump and down regulating influx through Outer Membrane Protein F (OmpF). Whilst the level of antibiotic resistance conferred by marRAB is only low level, there are increasing data to suggest that marRAB and related systems are important in clinical antibiotic resistance, possibly as a 'stepping stone' to higher levels of resistance. Other related systems include up-regulation of RobA, SoxS and AcrAB which give rise to a similar resistance phenotype to that conferred by up-regulation of MarA. The aim of this paper is to review the function and significance of the mar-locus and related systems with a particular focus on its implications in veterinary medicine. The Mar Menor is a coastal lagoon increasingly threatened by urban and agricultural pressures. The main watercourse draining into the lagoon is the Rambla del Albujón. A fortnightly campaign carried out over one annual cycle enabled us to characterize the treated urban sewage effluents and agricultural sources which contribute to the nutrient fluxes in the watercourse. Multivariate analysis provided information for establishing chemical signatures and for assessing the relative influence of the various sources on the water quality at the outlet. Mass balances were used to examine net gains and losses, and cross-correlations with rainfall to analyze climatic influence and control factors in the trends of the nutrient flux. The rainfall pattern was significantly cross-correlated with nitrate and phosphorus fluxes from agricultural sources, while fluctuations in the resident population explained the phosphorus flux trend in urban sources. 50% of dissolved inorganic nitrogen was from agricultural sources, while 70% of total phosphate and 91% of total organic carbon were from urban point sources. The net amounts of all the nutrients fell as a result of plant uptake and/or denitrification in the channel. The control of urban point sources (phosphorus-enriched) is suggested as a promptly action for improving the health of the coastal lagoon.

The Bocaina Plateau, which is situated on the eastern flank of the continental rift of southeastern Brazil, is the highest part of the Serra do Mar. Topographic relief in this area is suggested to be closely related to its complex tectono-magmatic evolution since the breakup of Western Gondwana and opening of the South Atlantic Ocean. Apatite fission track ages and track length distributions from 27 basement outcrops were determined to assess these hypotheses and reconstruct the denudation history of the Bocaina Plateau. The ages range between 303 +/- 32 and 46 +/- 5 Ma, and are significantly younger than the stratigraphic ages. Mean track lengths vary from 13.44 +/- 1.51 to 11.1 +/- 1.48 mu m, with standard deviations between 1.16 and 1.83 mu m. Contrasting ages within a single plateau and similar ages at different altitudes indicate a complex regional tectonothermal evolution. The thermal histories inferred from these data imply three periods of accelerated cooling related to the Early Cretaceous continental breakup, Early Cretaceous alkaline magmatism, and the Paleogene evolution of the continental rift of southeastern Brazil. The oldest fission track ages (>200 Ma) were obtained in the Serra do Mar region, suggesting that these areas were a long-lived source of sediments for the Parana, Bauru, and Santos basins. This essay is an analysis of the character Mara Josefa in the play 'The House of Bernarda Alba' (1936) by Federico García Lorca. It is hypothesized that the character of Mara Josefa can be considered a distorting mirror of the femininity presented in the play, and that through Mara Josefa this femininity is both revealed and problematized. The analysis adopts theories from the field of the (female) grotesque, using terminology both from Mikhail Bakhtin and Mary Russo. Throughout the analysis it is demonstrated how the character of Mara Josefa challenges the boundaries and norms which are dictated by Bernarda to control the women of the house. These conventions are challenged by Mara Josefa with the use of her loud speech, her dressed up appearance, and in her physical resistance. In this manner Mara Josefa is creating and embodying an alternate feminine view that is uncovered through her consistent provocation and by making herself into "a spectacle", a transboundary behaviour that is well interpreted within the theories of carnival, the grotesque body, and the spectacle of the female grotesque.

In this paper we study the problem of routing in opportunistic wireless network, and propose a novel routing mechanism, message-aware routing (MAR). Using MAR, the messages can be prioritized at mobile nodes and the resources will be allocated accordingly. The MAR uses the message-aware socializing model to classify mobile nodes into different social groups. In MAR, nodes only maintain up-to-date routing information for the nodes in the same social group and the messages for the nodes in the same social group will have higher priority to be delivered. The MAR improves the routing efficiency in terms of reduced traffic and a higher delivery success rate. Further, MAR is constructed in decentralized way and does not require any centralized infrastructure. Experiments using NS2 simulator show that the MAR achieves higher delivery rate than the Epidemic and Prophet routing.

Phosphorylated sperm proteins are crucial for sperm maturation and capacitation as a priori to their fertilization with eggs. In the freshwater prawn, Macrobrachium rosenbergii, a male reproduction-related protein (Mar-Mrr) was known to be expressed only in the spermatic ducts as a protein with putative phosphorylation and may be involved in sperm capacitation in this species. We investigated further the temporal and spatial expression of the Mar-Mrr gene using RT-PCR and in situ hybridization and the characteristics and fate of the protein using immunblotting and immunocytochemistry. The Mar-Mrr gene was first expressed in 4-week-old post larvae and the protein was produced in epithelial cells lining the spermatic ducts, at the highest level in the proximal region and decreased in the middle and distal parts. The native protein had a MW of 17 kDa and a high degree of serine/threonine phosphorylation. It was transferred from the epithelial cells to become a major protein at the anterior region of the sperm. We suggest that it is involved in sperm capacitation and fertilization in this open thelycal species and this is being investigated.

o presente trabalho teve por objetivo estudar o comportamento de um aço de alta resistência e baixa liga (com amostras de composição aproximada de 0,4% C, 0,6% Cr e 0,4% Mo), da classe API scr PIlO, utilizado na perfuração de petróleo offshore, frente a processos de fragilização causados pelo meio. Água do mar sintética foi utilizada como meio, com intuito de padronizar, em laboratório, as condições a que o material fica submetido na prática. Buscou-se avaliar e comparar o comportamento mecânico do material pela modificação dos parâmetros: temperatura, potencial aplicado ao material, e o efeito da presença ou ausência de H2S na solução. Para isso, foram realizados ensaios de tração pelo método de baixa taxa de deformação (da ordem de 10-6S-I), obtendo-se as curvas tensão x deformação nas diferentes situações ensaiadas, comparadas com as obtidas em óleo mineral. Análises fratográficas também foram utilizadas como forma de caracterizar os processos de fragilização. Além disso, estudou-se o comportamento eletroquímico do material nas diferentes condições através de ensaios de polarização potenciostáticos. Com isso, pode-se determinar quais as condições mais danosas e de maior risco para a utilização do material e em quais delas o seu uso é seguro. O aço estudado apresentou-se susceptível a processos de fragilização e todos os parâmetros analisados mostraram-se importantes no estudo desses processos.

Estudo sobre o Curso Pré-vestibular Comunitário da Mar (CPV Mar), a partir de sua inserção no movimento de pré-vestibulares comunitários existente hoje em todo o Brasil. Os dados e fontes coletados permitiram entender como se deu a trajetória do curso, bem como seus resultados e limites. Através do crescimento do curso, é possível entender o processo de crescimento e institucionalização do Centro de Estudos e Ações Solidárias da Mar (CEASM), organização não governamental criada por moradores e ex-moradores da Mar em 1997, que teve crescente e expressivo crescimento desde a sua formação. Em sete anos de existência - 1998 a 2005 -, os estudantes que freqüentaram o CPV Mar obtiveram, ao todo, 539 aprovações para universidades públicas e privadas, sobretudo para a PUC-RJ, totalizando 408 ingressos de alunos no ensino superior. Em consonância com a proposta do curso, e diante da condição econômica de seus alunos, serão aqui consideradas apenas as aprovações para as universidades públicas e para a PUC RJ, tendo em vista a parceria estabelecida entre essa universidade e o CEASM, no tocante à concessão de bolsas de estudo. Assim, são 355 alunos que ingressaram nas universidades públicas e na PUC-Rio. È com esse quantitativo que iremos trabalhar neste estudo.

Estudo de caso sobre a construção de identidades sociais no bairro da Mar. Descreve o processo de formação das favelas cariocas e, em particular, da Favela da Mar, desde o início de sua ocupação até a criação do bairro, instituído pelo poder público municipal, na XXXª Região Administrativa, que abrange 15 localidades, comumente reconhecidas na cidade do Rio de Janeiro como o Conjunto de favelas da Mar ou Complexo da Mar. Discute as reações dos moradores locais ao fato do bairro ter sido criado a partir da favela, espaço cujas representações estão hegemonicamente associadas a tudo o que se opõe à existência da cidade. Analisa as ações empreendidas pela organização não governamental Centro de Estudos e Ações Solidárias da Mar (CEASM), no sentido de “inventar” o bairro, tendo como princípios norteadores de sua atuação, a valorização do lugar e de sua história; as memórias dos moradores; e o protagonismo dos próprios agentes sociais locais. O estudo apóia-se na análise de fontes orais, e na pesquisa documental e bibliográfica.
