Famous for being the first foreign feature film that obtained permission to shoot in the Forbidden City, The Last Emperor (1987) is also one of the most ambitious and expensive independent productions of its time, awarded four Golden Globes and nine Academy Awards, including Best Picture. In addition, The Last Emperor can be considered as one of the first attempts of cinematic collaboration between West and East, in a period of cultural and economic transformations witnessed by China. This article aims to offer an overview of the production history of The Last Emperor, focusing on the co-production collaborations and the outcomes of a western auteur’s gaze on Chinese history. Questions of Orientalism, travel narrative and critical reception are taken into account in order to engage with the transnational implications of Bertolucci’s film and the western fascination with China.

Today there is a growing interest in the integration of health monitoring applications in portable devices necessitating the development of methods that improve the energy efficiency of such systems. In this paper, we present a systematic approach that enables energy-quality trade-offs in spectral analysis systems for bio-signals, which are useful in monitoring various health conditions as those associated with the heart-rate. To enable such trade-offs, the processed signals are expressed initially in a basis in which significant components that carry most of the relevant information can be easily distinguished from the parts that influence the output to a lesser extent. Such a classification allows the pruning of operations associated with the less significant signal components leading to power savings with minor quality loss since only less useful parts are pruned under the given requirements. To exploit the attributes of the modified spectral analysis system, thresholding rules are determined and adopted at design- and run-time, allowing the static or dynamic pruning of less-useful operations based on the accuracy and energy requirements. The proposed algorithm is implemented on a typical sensor node simulator and results show up-to 82% energy savings when static pruning is combined with voltage and frequency scaling, compared to the conventional algorithm in which such trade-offs were not available. In addition, experiments with numerous cardiac samples of various patients show that such energy savings come with a 4.9% average accuracy loss, which does not affect the system detection capability of sinus-arrhythmia which was used as a test case.

Arsenic (As) uptake and distribution in the roots, shoots, and grain of wheat (Triticum durum) grown in 2 As polluted soils (192 and 304 mg kg -1 respectively), and an uncontaminated soil (14 mg kg-1 ), collected from Scarlino plain (Tuscany, Italy), was investigated with respect with phosphorus fertilization. Three different level of phosphorus (P) fertilization: PO [0 kg ha-1], Pl [75 kg ha-1], and P2 [150 kg ha-1], as KH2PO4 of P, were applied. The presence of high concentrations of As in soils reduced plants growth, decreased grain yield and increased root, shoot and grain As concentrations, especially in the absence of P fertilization. The P fertilization decreased the As concentration in all the tissues as well as the translocation of As to the shoot and grain. This observation may be useful in certain areas of the world with high levels of As in soils, to reduce the potential risk posed to human health by As entering the food-chain. © by PSP.

In this study we have investigated the uptake and distribution of arsenic (As) and phosphate (Pi) in roots, shoots, and grain of wheat grown in an uncontaminated soil irrigated with solutions containing As at three different concentrations (0.5, 1 and 2 mg l-1) and in the presence or in the absence of P fertilization. Arsenic in irrigation water reduced plants growth and decreased grain yield. When Pi was not added (P-), plants were more greatly impacted compared to the plus Pi (P+) treatments. The differences in mean biomass between P- and P+ treatments at the higher As concentrations demonstrated the role of Pi in preventing As toxicity and growth inhibition. Arsenic concentrations in root, shoot and grain increased with increasing As concentration in irrigation water. It appears that P fertilization minimizes the translocation of As to the shoots and grain whilst enhancing P status of plant. The observation that P fertilization minimises the translocation of arsenic to the shoots and grain is interesting and may be useful for certain regions of the world that has high levels of As in groundwater or soils. © 2008 Springer Science+Business Media B.V.

Elevated soil arsenic levels resulting from long-term use of arsenic contaminated ground for irrigation in Bangladesh may inhibit seed germination and seedling establishment of rice, the country's main food crop. A germination study on rice seeds and a short-term toxicity experiment with different concentrations of arsenite and arsenate on rice seedlings were conducted. Percent germination over control decreased significantly with increasing concentrations of arsenite and arsenate. Arsenite was found to be more toxic than arsenate for rice seed germination. There were varietal differences among the test varieties in response to arsenite and arsenate exposure. The performance of the dry season variety Purbachi was the best among the varieties. Germination of Purbachi was not inhibited at all up to 4 mg l-1 arsenite and 8 mg l-1 arsenate treatment. Root tolerance index (RTI) and relative shoot height (RSH) for rice seedlings decreased with increasing concentrations of arsenite and arsenate. Reduction of RTI caused by arsenate was higher than that of arsenite. In general, dry season varieties have more tolerance to arsenite or arsenate than the wet season varieties.

Pinus sylvestris seedlings infected with either the ectomycorrhizal (ECM) fungus Paxillus involutus or Suillus variegatus were exposed to a range of Cd or Zn concentrations. This was done to investigate the relationship between the sensitivity of ECM fungi and their host plants over a wide range of concentrations. P involutus ameliorated the toxicity of Cd and Zn to P. sylvestris with respect to root length, despite significant inhibition of ECM infection levels by Cd (Cd EC50 [effective concentration which inhibits ECM infection by 50%] values were: P. involutus 3.7 μg g-1 Cd; S. variegatus 2.3 μg g-1 Cd). ECM infection by P. involutus also decreased Cd and Zn transport to the plant shoots at potentially toxic concentrations and also influenced the proportion of Zn transported to the roots and shoots, with a higher proportion retained in the roots of the seedlings. ECM infection did increase host biomass production, but this was not affected by the presence of Cd or Zn. Root and shoot biomass production by P. sylvestris, in both the presence and absence of ECM fungi, was unaffected by Cd and Zn at all concentrations tested.

Scots pine seedlings colonized by ectomycorrhizal (ECM) fungi from natural soil inoculum were exposed to a range of Cd or Zn concentrations to investigate the effects of metals on ECM fungi-Scots pine associations in a realistic soil environment. Experiments focused on the relationship between the sensitivity of ECM fungi and their host plants, the influence of metals on ECM community dynamics on Scots pine roots, and the effects of metal exposure on ECM colonization from soil-borne propagules. Ectomycorrhizal colonization was inhibited by Cd and Zn, with a decrease in the proportion of ECM-colonized root tips. Shoot and root biomass, total root length, and total root-tip density, however, were unaffected by Cd or Zn. A decrease in the diversity of ECM morphotypes also occurred, which could have a negative effect on tree vigor. Overall, colonization by ECM fungi was more sensitive than seedling growth to Cd and Zn, and this could have serious implications for successful tree establishment on metal-contaminated soils.

Biomass and phosphorus allocation were determined in arsenate tolerant and non-tolerant clones of the grass Holcus lanatus L. in both solution culture and in soil. Arsenate is a phosphate analogue and is taken up by the phosphate uptake system. Tolerance to arsenate in this grass is achieved by suppression of arsenate (and phosphate) influx. When clones differing in their arsenate tolerance were grown in solution culture with a range of phosphate levels, a tolerant clone did not fare as well as a non-tolerant at low levels of phosphate nutrition in that it had reduced shoot biomass production, increased biomass allocation to the roots and lower shoot phosphorus concentration. At a higher level of phosphate nutrition there was little or no difference in these parameters, suggesting that differences at lower levels of phosphate nutrition were due solely to differences in the rates of phosphate accumulation. In experiments in sterile soil (potting compost) the situation was more complicated with tolerant plants having lower growth rates but higher phosphorus concentrations. The gene for arsenate tolerance is polymorphic in arsenate uncontaminated populations. When phosphorus concentration of tolerant phenotypes was determined in one such population, again tolerants had a higher phosphorus status than non-tolerants. Tolerants also had higher rates of vesicular-arbuscular mycorrhizal (VAM) infection. The ecological implications of these results are that it appears that suppression of the high affinity uptake system, is at least in part, compensated by increased mycorrhizal infection. © 1994 Kluwer Academic Publishers.

Polygenic risk scores have shown great promise in predicting complex disease risk and will become more accurate as training sample sizes increase. The standard approach for calculating risk scores involves linkage disequilibrium (LD)-based marker pruning and applying a p value threshold to association statistics, but this discards information and can reduce predictive accuracy. We introduce LDpred, a method that infers the posterior mean effect size of each marker by using a prior on effect sizes and LD information from an external reference panel. Theory and simulations show that LDpred outperforms the approach of pruning followed by thresholding, particularly at large sample sizes. Accordingly, predicted R(2) increased from 20.1% to 25.3% in a large schizophrenia dataset and from 9.8% to 12.0% in a large multiple sclerosis dataset. A similar relative improvement in accuracy was observed for three additional large disease datasets and for non-European schizophrenia samples. The advantage of LDpred over existing methods will grow as sample sizes increase.

Quantifying the similarity between two trajectories is a fundamental operation in analysis of spatio-temporal databases. While a number of distance functions exist, the recent shift in the dynamics of the trajectory generation procedure violates one of their core assumptions; a consistent and uniform sampling rate. In this paper, we formulate a robust distance function called Edit Distance with Projections (EDwP) to match trajectories under inconsistent and variable sampling rates through dynamic interpolation. This is achieved by deploying the idea of projections that goes beyond matching only the sampled points while aligning trajectories. To enable efficient trajectory retrievals using EDwP, we design an index structure called TrajTree. TrajTree derives its pruning power by employing the unique combination of bounding boxes with Lipschitz embedding. Extensive experiments on real trajectory databases demonstrate EDwP to be up to 5 times more accurate than the state-of-the-art distance functions. Additionally, TrajTree increases the efficiency of trajectory retrievals by up to an order of magnitude over existing techniques.

PURPOSE: To provide a tool to enable gamma analysis software algorithms to be included in a quality assurance (QA) program.

METHODS: Four image sets were created comprising two geometric images to independently test the distance to agreement (DTA) and dose difference (DD) elements of the gamma algorithm, a clinical step and shoot IMRT field and a clinical VMAT arc. The images were analysed using global and local gamma analysis with 2 in-house and 8 commercially available software encompassing 15 software versions. The effect of image resolution on gamma pass rates was also investigated.

RESULTS: All but one software accurately calculated the gamma passing rate for the geometric images. Variation in global gamma passing rates of 1% at 3%/3mm and over 2% at 1%/1mm was measured between software and software versions with analysis of appropriately sampled images.

CONCLUSION: This study provides a suite of test images and the gamma pass rates achieved for a selection of commercially available software. This image suite will enable validation of gamma analysis software within a QA program and provide a frame of reference by which to compare results reported in the literature from various manufacturers and software versions.

No presente trabalho desenvolveram-se estudos visando a valorização do coberto vegetal da Ilha de Porto Santo, através de duas metodologias de investigação complementares: a) preservação e reintrodução na Ilha de uma espécie endémica e em risco do Arquipélago da Madeira (Olea maderensis) e de uma espécie naturalizada (Olea europaea ssp. europaea var. sylvestris), recorrendo para o efeito a técnicas de biotecnologia (micropropagação); b) análise da percepção da comunidade local e visitante sobre o fenómeno da desertificação e a valorização do coberto vegetal bem como a sua aceitação relativamente à aplicação de técnicas de biotecnologia (para micropropagar e reintroduzir espécies de oliveira na Ilha) para minimização do processo de desertificação. A dissertação estrutura-se em quatro partes principais. A Parte I caracteriza a Ilha de Porto Santo em termos geográficos, geológicos, climáticos, sócio-economicos e do uso do solo. Enquadra, ainda, o problema da desertificação, através da caracterização/evolução do coberto vegetal ao longo dos anos. Finalmente apresentam-se os objectivos gerais deste estudo. A Parte II centra-se no desenvolvimento de metodologias no âmbito da biotecnologia vegetal para propagação de espécies de O. maderensis e O. europaea ssp. europaea var. sylvestris. em larga escala. Esta parte está dividida em seis capítulos. O Capítulo II.1 aborda a distribuição geográfica das espécies de oliveira e faz uma revisão bibliográfica dos aspectos mais importantes da micropropagação de oliveira (O. europaea L.), principalmente através da micropropagação por estimulação de gomos axilares. No final deste capítulo apresentam-se os objectivos específicos desta investigação. No Capítulo II.2 faz-se a caracterização genética de genótipos de O. maderensis do Arquipélago da Madeira através da análise da ploidia e do conteúdo em DNA por citometria de fluxo (FCM) e através da detecção de polimorfismos por análise de microssatélites (SSR). Nesta análise usaram-se ainda outros genótipos, nomeadamente: O. europaea ssp. europaea var. sylvestris, O. cerasiformes e O. europaea ssp. europaea var. europaea. Este estudo contribuiu para uma melhor caracterização desta espécie e permitiu a detecção de um nível de ploidia novo no género Olea (tetraploidia). O Capítulo II.3 descreve a optimização das condições de cultura in vitro (e.g. desinfecção, meio de cultura e enraizamento) para propagar e preservar a O. maderensis. Avalia-se ainda a “performance” dos rebentos in vitro (taxas de crescimento, avaliação da aparência das folhas e estudos fisiológicos), de modo a confirmar a optimização das condições de propagação. Neste capítulo define-se um meio novo (OMG) para propagação desta espécie endémica. O Capítulo II.4 descreve dois protocolos de micropropagação e aclimatização de ambas as espécies (O. maderensis e O. europaea ssp. europaea var. sylvestris) e a qualidade das plantas (“true-to-type”) é avaliada através da possível ocorrência de variabilidade genética através de FCM (ploidia) e SSRs. O Capítulo II.5 descreve um protocolo eficiente de aclimatização ao campo de O. maderensis e avalia a “performance” das plantas micropropagadas no campo através da análise de parâmetros fisiológicos durante o processo. O Capítulo II.6 apresenta os estudos em curso relativamente às plantas de O. maderensis em aclimatização no campo, bem como a introdução de plantas micropropagadas num outro local da Ilha com um maior grau de degradação dos solos. Estas estratégias estão a ser aplicadas juntamente com a DRFRAM, no âmbito de programas de florestação em curso. Finalmente é realçada a necessidade de estudos semelhantes com outras espécies nativas. Na Parte III, são apresentados os estudos sobre a percepção da comunidade local relativamente à valorização do coberto vegetal para a minimização dos processos de degradação dos solos/desertificação. A introdução faz o enquadramento teórico sobre o fenómeno da desertificação, particularmente na Ilha de Porto Santo e sobre a percepção social da desertificação. São ainda apresentados os objectivos específicos desta investigação. A metodologia adoptada recorreu à aplicação de inquéritos por questionário à população residente e aos visitantes da Ilha de Porto Santo e ainda a realização de inquéritos por entrevista a algumas entidades e especialistas. Estes estudos permitiram verificar que existe uma nítida consciência da situação de risco da ilha, das medidas tomadas e a tomar e da premência da resolução do problema. Face ao recurso de estratégias alternativas envolvendo biotecnologia, e apesar de existir algum desconhecimento, concluiu-se ainda que a população manifesta aceitação, desde que essas estratégias valorizem o coberto vegetal e, assim, ajudem a combater a degradação biofísica dos solos. Finalmente são apresentadas as conclusões e algumas recomendações. Na Parte IV apresentam-se as conclusões gerais e perspectivas futuras, onde o potencial ambiental destas oliveiras bravas micropropagadas é destacado, bem como é considerado o alargamento da aplicação destas estratégias a outras espécies indígenas em risco, nesta Ilha (e noutros locais). São ainda resumidas as principais visões da população e das entidades e dos especialistas que poderão contribuir para apoiar a elaboração de medidas de mitigação e prevenção no combate ao processo de degradação dos solos/desertificação em curso.

O cádmio (Cd) é um metal não essencial e é considerado um poluente prioritário pela comunidade europeia. Este metal atinge o ambiente no decurso de várias actividades antropogénicas e tende a concentrar-se nos solos e sedimentos, onde está potencialmente disponível para as plantas, sendo posteriormente transferido através da cadeia trófica. Neste contexto, o principal objectivo da presente dissertação foi o estudo dos efeitos da assimilação e da acumulação de Cd em plantas e as suas consequências para animais consumidores. Numa primeira fase, foram estudados os principais efeitos fisiológicos e genotóxicos do Cd em plantas. As plantas de alface (Lactuca sativa L.) expostas a Cd apresentaram um decréscimo na eficiência fotossintética, aumento de peroxidação lipídica e alterações significativas na actividade de enzimas de stress oxidativo. Estas alterações culminaram num decréscimo do crescimento da parte aérea no final da exposição. As respostas obtidas pelos parâmetros bioquímicos sugerem que estes poderão ser utilizados como eventuais biomarcadores em testes ecotoxicológicos com Cd em abordagens integrantes em conjunto com parâmetros clássicos. Os efeitos mutagénicos de Cd foram avaliados através da determinação da instabilidade de microsatélites (IM). Não foi observada IM, nem nas folhas nem nas raízes de plantas de alface com 5 semanas de idade expostas a 100 μM Cd durante 14 dias, no entanto observou-se IM em raízes de alface exposta a 10 μM Cd durante 28 dias desde a germinação. A idade da planta e a maior acumulação de Cd nas raízes poderão explicar os resultados obtidos. A clastogenicidade de Cd foi analisada em três espécies vegetais com diferentes capacidades de destoxificação e acumulação de metais através de citometria de fluxo. Foram detectadas alterações significativas nos parâmetros analisados em raízes alface, mas não nas espécies Thlaspi caerulescens J & C Presl e Thlaspi arvense L. Estes resultados sugerem que o stress provocado pelo Cd originou clastogenicidade como consequência da perda de porções de cromossomas, uma vez que o conteúdo de ADN nuclear diminuiu. A transferência trófica através da cadeia alimentar permanece muito pouco estudada em termos ecotoxicológicos. A distribuição subcellular de metais num organismo pode ser utilizada para compreender a transferência trófica de um metal na cadeia alimentar. Como tal, numa última parte é estudado de que modo a distribuição subcellular do Cd em plantas com perfis de acumulação de Cd distintos afecta a biodisponibilidade e transferência trófica de Cd para isópodes. A distribuição de Cd entre as 4 fracções subcelulares obtidas através de centrifugação diferencial revelou a existência de diferenças significativas entre as espécies de plantas. Estes resultados em conjunto com a avaliação directa da eficiência de assimilação (EA) de Cd individual de cada uma das quatro fracções subcelulares das plantas em estudo, resultou em informação de grande relevância para a explicação das diferenças observadas na EA de Cd por parte de isópodes alimentados com folhas de diferentes espécies de plantas. Com base nos resultados obtidos, o Cd ligado a proteínas estáveis à temperatura (e.g. metaloteoninas e fitoquelatinas) é o menos biodisponível, sendo assim o que menos contribuiu para a transferência trófica, enquanto que o Cd ligado a proteínas desnaturadas pela temperatura foi a fracção mais disponível para transferência trófica de Cd ao isópode. Estes resultados realçam a relevância ecológica da distribuição subcelular de Cd em plantas que tem influência directa na tranferência trófica deste metal para os consumidores e ainda o facto de que alterações na distribuição subcelular de Cd em plantas devido a diferentes mecanismos de destoxificação poderá ter um impacto directo na transferência trófica de Cd para o animal consumidor.

We explored the relationships between perturbation-driven population decline and genetic/genotypic structure in the clonal seagrass Posidonia oceanica, subject to intensive meadow regression around four Mediterranean fish-farms, using seven specific microsatellites. Two meadows were randomly sampled (40 shoots) within 1,600 m2 at each site: the “impacted” station, 5–200 m from fish cages, and the “control” station, around 1,000 m downstream further away (considered a proxy of the pre-impact genetic structure at the site). Clonal richness (R), Simpson genotypic diversity (D*) and clonal sub-range (CR) were highly variable among sites. Nevertheless, the maximum distance at which clonal dispersal was detected, indicated by CR, was higher at impacted stations than at the respective control station (paired t-test: P < 0.05, N = 4). The mean number of alleles (Â) and the presence of rare alleles ( r) decreased at impacted stations (paired t-test: P < 0.05, and P < 0.02, respectively, N = 4). At a given perturbation level (quantified by the organic and nutrient loads), shoot mortality at the impacted stations significantly decreased with CR at control stations (R 2 = 0.86, P < 0.05). Seagrass mortality also increased with  (R 2 = 0.81, P < 0.10), R (R 2 = 0.96, P < 0.05) and D* (R 2 = 0.99, P < 0.01) at the control stations, probably because of the negative correlation between those parameters and CR. Therefore, the effects of clonal size structure on meadow resistance could play an important role on meadow survival. Large genotypes of P. oceanica meadows thus seem to resist better to fish farm-derived impacts than little ones. Clonal integration, foraging advantage or other size-related fitness traits could account for this effect.
