Contraction of cardiac muscle is regulated through the Ca2+ dependent protein-protein interactions of the troponin complex (Tn). The critical role cardiac troponin C (cTnC) plays as the Ca2+ receptor in this complex makes it an attractive target for positive inotropic compounds. In this study, the ten Met methyl groups in cTnC, [98% 13C ϵ]-Met cTnC, are used as structural markers to monitor conformational changes in cTnC and identify sites of interaction between cTnC and cardiac troponin I (cTnI) responsible for the Ca2+ dependent interactions. In addition the structural consequences that a number of Ca2+-sensitizing compounds have on free cTnC and the cTnC·cTnI complex were characterized. Using heteronuclear NMR experiments and monitoring chemical shift changes in the ten Met methyl 1H-13C correlations in 3Ca2+ cTnC when bound to cTnI revealed an anti-parallel arrangement for the two proteins such that the N-domain of cTnI interacts with the C-domain of cTnC. The large chemical shifts in Mets-81, -120, and -157 identified points of contact between the proteins that include the C-domain hydrophobic surface in cTnC and the A, B, and D helical interface located in the regulatory N-domain of cTnC. TnI association [cTnI(33–80), cTnI(86–211), or cTnI(33–211)] was found also to dramatically reduce flexibility in the D/E central linker of cTnC as monitored by line broadening in the Met 1H- 13C correlations of cTnC induced by a nitroxide spin label, MTSSL, covalently attached to cTnC at Cys 84. TnI association resulted in an extended cTnC that is unlike the compact structure observed for free cTnC. The Met 1H-13C correlations also allowed the binding characteristics of bepridil, TFP, levosimendan, and EMD 57033 to the apo, 2Ca2+, and Ca2+ saturated forms of cTnC to be determined. In addition, the location of drug binding on the 3Ca2+cTnC·cTnI complex was identified for bepridil and TFP. Use of a novel spin-labeled phenothiazine, and detection of isotope filtered NOEs, allowed identification of drug binding sites in the shallow hydrophobic cup in the C-terminal domain, and on two hydrophobic surfaces on N-regulatory domain in free 3Ca2+ cTnC. In contrast, only one N-domain drug binding site exists in 3Ca2+ cTnC·cTnI complex. The methyl groups of Met 45, 60 and 80, which are grouped in a hydrophobic patch near site II in cTnC, showed the greatest change upon titration with bepridil or TFP, suggesting that this is a critical site of drug binding in both free cTnC and when associated with cTnI. The strongest NOEs were seen for Met-60 and -80, which are located on helices C and D, respectively, of Ca2+ binding site II. These results support the conclusion that the small hydrophobic patch which includes Met-45, -60, and -80 constitutes a drug binding site, and that binding drugs to this site will lead to an increase in Ca2+ binding affinity of site II while preserving maximal cTnC activity. Thus, the subregion in cTnC makes a likely target against which to design new and selective Ca2+-sensitizing compounds. ^

Sediment samples from ODP Site 1085 were investigated in order to obtain more information on the initiation and development of the Benguela upwelling system during the middle and upper Miocene. In particular, our intent was to establish the causes of the upwelling as well as the response of the upwelling regime to the development of the Antarctic Circumpolar Current. Based on changes in the calcareous dinoflagellate cyst association, we found an initial increase of the dinoflagellate cyst productivity, probably related to the initiation of upwelling about 11.8 Ma ago. Two distinct increases in cyst productivity in conjunction with temperature decreases of the upper water masses reflect upwelling pulses off Namibia and occur at the end of the Miocene cooling events Mi5 (about 11.5 Ma) and Mi6 (about 10.5 Ma). Both cooling events are associated with an ice volume increase in Antarctica and are thought to have led to an increase in southeasterly winds, possibly causing these two upwelling pulses. We demonstrate a decrease in dinoflagellate cyst productivity and enhanced terrigenous input via the Orange River after the Mi5 event. At about 11.1 Ma, the dinoflagellate cyst productivity increases again. The polar cyst species Caracomia arctica occurs here for the first time. This implies an influence of subantarctic mode water and therefore a change in the quality of the upwelling water which allowed the Benguela upwelling to develop into modern conditions. From about 10.4 Ma, C. arctica forms a permanent part of the association, pointing to an establishment of the upwelling regime.

The logic of territorial ordainment in Recife has been developed mainly through the seclusion of the unwanted and the removal of stilt houses and slums in order to make room and prepare the space for new private enterprises. As an example of ordainment we took the 'Via Mangue' project, which is part of Recife's mobility plan and is one of the main projects aimed at the city's preparation for the 2014 FIFA World Cup Brazil. We believe that the project makes use of the 'great social benefits' discourse in order to cover its actual and practical consequences which, taken as whole, lead to the favoring of private sector over the public interest. That being so, the main goal of the present work is to make an analysis of territorial ordainment in Recife through the execution of the Via Mangue project; observing at the same time the urban and social impacts caused by the relocation of communities to the Via Mangue III housing complex, and verifying whether this policy actually promoted substantial improvement of habitability or only a precarious social inclusion of these populations. Our research was conducted and operated at three levels. First, the conceptual reconstitution of territorial ordainment; second, documental and cartographic research on the Via Mangue project; and last, fieldwork with observation of the constructed space and personal interviews with members of the families relocated to the Via Mangue III housing complex. We hope the present work could be a valuable contribution to the comprehension of the complexities involved in the relocation of families to housing complexes built by the government

A preliminary palynological survey of 118 samples from the Mississippi Fan (Sites 615, 616, and 620) and from 2 intraslope basins (Sites 619 and 618) shows pollen in all samples. Reworked pollen is generally abundant, forming over 50% of the sum of pollen and reworked pollen. Concentration of nonreworked pollen is usually low (on the order of tens to hundreds of pollen grains per cubic centimeter wet sediment). Conifers, primarily Pinus, Picea, and Tsuga, dominate Pleistocene marine pollen spectra; significant percentages of Quercus are present in Holocene sediments and in sediments deposited during oxygen-isotope Stage 5.

About one third of the anthropogenic carbon dioxide (CO2) released into the atmosphere in the past two centuries has been taken up by the ocean. As CO2 invades the surface ocean, carbonate ion concentrations and pH are lowered. Laboratory studies indicate that this reduces the calcification rates of marine calcifying organisms, including planktic foraminifera. Such a reduction in calcification resulting from anthropogenic CO2 emissions has not been observed, or quantified in the field yet. Here we present the findings of a study in the Western Arabian Sea that uses shells of the surface water dwelling planktic foraminifer Globigerinoides ruber in order to test the hypothesis that anthropogenically induced acidification has reduced shell calcification of this species. We found that light, thin-walled shells from the surface sediment are younger (based on 14C and d13C measurements) than the heavier, thicker-walled shells. Shells in the upper, bioturbated, sediment layer were significantly lighter compared to shells found below this layer. These observations are consistent with a scenario where anthropogenically induced ocean acidification reduced the rate at which foraminifera calcify, resulting in lighter shells. On the other hand, we show that seasonal upwelling in the area also influences their calcification and the stable isotope (d13C and d18O) signatures recorded by the foraminifera shells. Plankton tow and sediment trap data show that lighter shells were produced during upwelling and heavier ones during non-upwelling periods. Seasonality alone, however, cannot explain the 14C results, or the increase in shell weight below the bioturbated sediment layer. We therefore must conclude that probably both the processes of acidification and seasonal upwelling are responsible for the presence of light shells in the top of the sediment and the age difference between thick and thin specimens.

The Cariaco Basin is a 1400-m-deep depression approximately 160 km long by 70 km wide located off the central Venezuelan coast . It is connected to the Atlantic Ocean by a sill ~100-m-deep, and two slightly deeper channels that breech it; Canal Centinela (146-m-deep) and Canal de la Tortuge (135-m-deep). High surface production rates and restricted circulation result in anoxic waters below ca. 275 m. The depth of the oxycline varies between 250 and 320 m and is independent of density. Rather, fluctuations in oxycline depth appear to be due to lateral intrusions of Caribbean Sea water that are linked to eddies along the continental shelf. A mooring with five sediment traps (Z, A-D) is located in the eastern Cariaco Basin. Traps A-D have been in place since November 1995. Trap A is located in oxic waters at 226 ± 6 m. Trap B is located at 407 ± 3 m and Trap D is located at 1205 ± 3 m. Trap C was located at a depth of 880 ± 2 m from Jan. 1996 to Nov. 2000, and was moved to 807 ± 2 m in Nov. 2000. A fifth trap, Z, was added in November 2003 at 110 m for the first 6 months, and at 150 m thereafter. All five sediment traps are coneshaped with a 0.5 m**2 opening that is covered with a baffle top to reduce turbulence. The mooring is deployed for six-month intervals and each sample collection cup is filled with a buffered 3.2% formalin solution as a preservative for the accumulating organic matter. The cups are numbered 1-13, with cup 1 collecting for the two-week interval immediately following deployment, and cup 13 collecting for the 2 weeks immediately before recovery.

Ocean acidification, the assimilation of atmospheric CO2 by the oceans that decreases the pH and CaCO3 saturation state (Omega) of seawater, is projected to have severe adverse consequences for calcifying organisms. While strong evidence suggests calcification by tropical reef-building corals containing algal symbionts (zooxanthellae) will decline over the next century, likely responses of azooxanthellate corals to ocean acidification are less well understood. Because azooxanthellate corals do not obtain photosynthetic energy from symbionts, they provide a system for studying the direct effects of acidification on energy available for calcification. The solitary azooxanthellate orange cup coral Balanophyllia elegans often lives in low-pH, upwelled waters along the California coast. In an 8-month factorial experiment, we measured the effects of three pCO2 treatments (410, 770, and 1220 µatm) and two feeding frequencies (3-day and 21-day intervals) on "planulation" (larval release) by adult B. elegans, and on the survival, skeletal growth, and calcification of newly settled juveniles. Planulation rates were affected by food level but not pCO2. Juvenile mortality was highest under high pCO2 (1220 µatm) and low food (21-day intervals). Feeding rate had a greater impact on calcification of B. elegans than pCO2. While net calcification was positive even at 1220 µatm (~3 times current atmospheric pCO2), overall calcification declined by ~25-45%, and skeletal density declined by ~35-45% as pCO2 increased from 410 to 1220 µatm. Aragonite crystal morphology changed at high pCO2, becoming significantly shorter but not wider at 1220 µatm. We conclude that food abundance is critical for azooxanthellate coral calcification, and that B. elegans may be partially protected from adverse consequences of ocean acidification in habitats with abundant heterotrophic food.

Using shells collected from a sediment trap series in the Madeira Basin, we investigate the effects of seasonal variation of temperature, productivity, and optimum growth conditions on calcification in three species of planktonic Foraminifera. The series covers an entire seasonal cycle and reflects conditions at the edge of the distribution of the studied species, manifesting more suitable growth conditions during different parts of the year. The seasonal variation in seawater carbonate saturation at the studied site is negligible compared to other oceanic regions, allowing us to assess the effect of parameters other than carbonate saturation. Shell calcification is quantified using weight and size of individual shells. The size-weight scaling within each species is robust against changes in environmental parameters, but differs among species. An analysis of the variation in calcification intensity (size-normalized weight) reveals species-specific response patterns. In Globigerinoides ruber (white) and Globigerinoides elongatus, calcification intensity is correlated with temperature (positive) and productivity (negative), whilst in Globigerina bulloides no environmental forcing is observed. The size-weight scaling, calcification intensity, and response of calcification intensity to environmental change differed between G. ruber (white) and G. elongatus, implying that patterns extracted from pooled analyses of these species may reflect their changing proportions in the samples. Using shell flux as a measure of optimum growth conditions, we observe significant positive correlation with calcification intensity in G. elongatus, but negative correlation in G. bulloides. The lack of a consistent response of calcification intensity to optimum growth conditions is mirrored by the results of shell size analyses. We conclude that calcification intensity in planktonic Foraminifera is affected by factors other than carbonate saturation. These factors include temperature, productivity, and optimum growth conditions, but the strength and sign of the relationships differ among species, potentially complicating interpretations of calcification data from the fossil record.

A medical cyclotron accelerating H- ions to 18 MeV is in operation at the Bern University Hospital (Inselspital). It is the commercial IBA 18/18 cyclotron equipped with a specifically conceived 6 m long external beam line ending in a separate bunker. This feature is unique for a hospital-based facility and makes it possible to conduct routine radioisotope production for PET diagnostics in parallel with multidisciplinary research activities, among which are novel particle detectors, radiation biophysics, radioprotection, radiochemistry and radiopharmacy developments. Several of these activities, such as radiobiology experiments for example, require low current beams down to the pA range, while medical cyclotrons are designed for high current operation above 10 μA. In this paper, we present the first results on the low current performance of a PET medical cyclotron obtained by ion source, radio-frequency and main coil tuning. With this method, stable beam currents down to (1.5+/- 0.5 ) pA were obtained and measured with a high-sensitivity Faraday cup located at the end of the beam transport line.

Steel is, together with concrete, the most widely used material in civil engineering works. Not only its high strength, but also its ductility is of special interest from the point of view of the structural safety, since it enables stress distribution with adjacent elements and, therefore, more energy can be stored before reaching the structural failure. However, despite of being extensively used, there are still some aspects related to its fracture behaviour that need to be clarified and that will allow for a better use of its properties. When a steel item is tested under tension and reaches the maximum load point, necking process begins, which makes difficult to define the material behaviour from that moment onward. The ISO standard 6892-1, which defines the tensile testing method for metallic materials, describes the procedures to obtain some parameters related to this last section of the F − E curve. Nevertheless, these parameters have proved to be controversial, since they have low reproducibility and repeatibility rates that are difficult to explain. This Thesis tries to deepen the knowledge of the last section of the F − E curve for construction steels. An extensive experimental campaign has been carried out with two representative steels used in civil engineering works: a steel rod used for manufacturing prestressing steel wires, before the cold-drawing process is applied, and steel bars used in reinforced concrete structures. Both materials have different fracture surfaces: while the first of them shows a flat fracture surface, perpendicular to the loading direction with a dark region in the centre of it, the second one shows the classical cup-cone fracture surface. The cup-cone fracture surface has been deeply studied in the past and different numerical models have been able to reproduce it with success, with a special mention to the Gurson-Tvergaard-Needleman model (GTN). Regarding the failure surface shown by the first material, in principle it can be numerically reproduced by a GTN model, but the differences observed between both materials in the experimental campaign suggest thinking of a different failure criterium. In the present Thesis, an extensive experimental campaign has been carried out using cylindrical specimens made of two representative construction steels with different fracture behaviours. On one hand, the initial eutectoid steel rod used for manufacturing prestressing steel wires is analysed, which presents a flat fracture surface, perpendicular to the loading direction, and with a dark region in the centre of it. On the other hand, B 500 SD steel bars, typically used in reinforced concrete structures and with the typical cup-cone fracture surface, are studied. These experimental works have allowed distinguishing two clearly different fracture behaviours between both materials and, in the case of the first one, a fragile-like behaviour has been identified. For the first material, which shows a flat fracture surface perpendicular to the loading direction, the following hypothesis is proposed in this study: a quasi-brittle fracture is developed as a consequence of a decohesion process, with the dark region acting as a circular crack perpendicular to the loading direction. To reproduce numerically the fracture behaviour shown by the first material, a failure criterium based on a cohesive model is proposed in this Thesis. As an innovative contribution, this failure criterium depends on the stress triaxiality state of the material, which is a key parameter when studying fracture in this kind of materials. This type of models have some advantages when compared to the widely used GTN models. While GTN models need a high number of parameters to be defined, cohesive models need basically two parameters to define the softening curve: the decohesion stress ft and the fracture energy GF . In addition to this, GTN models parameters cannot be measured experimentally, while GF is indeed. Regarding ft, although no experimental procedure is defined for its obtention, it has an easier interpretation than the parameters used by the GTN models like, for instance, the void volume needed for the coalescence process to start or the void volume that leads to a total loss of the bearing capacity. In order to implement this failure criterium, a triaxiality-dependent cohesive interface element has been developed. The experimental results obtained in the experimental campaign have been successfully reproduced by using this interface element. Furthermore, in these models the failure mechanism is developed in the same way as observed experimentally: with a circular decohesive process taking place around the longitudinal axis of the specimen. In summary, the works developed in this Thesis, both experimental and numerical, contribute to clarify the behaviour of construction steels in the last section of the F − E curve and the mechanisms responsible for the eventual material failure, an aspect that can lead to a better use of the properties of these steels in the future and a safety improvement in the structures built with them.

The aim of this research was to determine the running technique profile during a triathlon competition from a biomechanical perspective. Also, to analyze the triathlete gender’s and level of performance’s influence on this profile in competition. An accurate (internal validity) and reliable methodology with a high external validity (ecological) had to be developed to get those aims in competition. Sixty-four triathletes were analyzed. 32 (16 males, 16 females) took part in the Madrid 2008 Triathlon World Cup and 32 (16 males and 16 females) took part in the Spanish Triathlon National Championships. The biomechanical analyses were carried out by a photogrammetric system that allow to calculate the landmarks coordinates (x,y) with a 1.66% error in x axis, and a 2.10% error in y axis. The frames were obtained with a camera situated perpendicular to the triathletes’ trajectory, filming the saggittal plane. DLT based algorithms (Abdel-Aziz & Karara, 1971) were used to calculate the real coordinates from the digitalized ones and the final variables afterwards. The biomechanical analisys itself was performed in four different moments during the competition, according to each 2.5 km lap the triathletes had to do. Running speed was highly related to performance level. Also, 3 of the 4 analyzed groups showed speed values under the 3 minutes and 30 seconds per kilometer. It demonstrated the very high performance level of the analized triathletes. Furthermore, it seems that men get higher speeds because their longer stride length, while women shows higher stride frequency values. The highest stride frequency values were found in the first lap. Women and the international level triathletes showed the highest values. Stride length showed different tendencies according to the gender and level of performance. Men and international level triathletes showed the highest level in the first lap and a decreasing tendency after that. The accumulated fatigue was probably the reason of this tendency. On the other hand, higher values than in first lap were found in the second one in women and national level triathletes. It demonstrated the previous cycling can affect to those groups in terms of biomechanics. Flight times remained constant during the running part, while the contact times showed an increasing tendency that caused a variation in flight times percents. The lowest contact times were found in the first lap and in men and international triathletes’ values. Also, these two groups were more consistent during the whole running. On the other hand, increasing tendencies were found in women and national level triathletes, who were not able to maintain the same values probably due to their lower level of performance. Higher level triathletes showed more consistent hip vertical oscillation values than lower level triathletes, who presented increasing tendencies. The highest values were found in women and national level triathletes. The horizontal distance hip-toe cap remained constant among the laps in all the groups. Men and international level triathletes showed the highest values. The support knee angle at toe-off did not show a clear tendency. The lowest values were found in men and international level triathletes. A high correlation was found between the non-support knee angle and the running speed. Furthermore, men and international level triathletes showed the smallest values, due to the higher velocities reached by these two groups. Ankles angles did not show any tendency during the running part. Similar values were found in the four analyzed groups, so this variable does not seem to represent an important one within the triathlete’s performance. The results obtained in the present research support the use of the bidimensional photogrammetric video-system to analyze the running technique during a triathlon competition. Its application in international triathlon meetings has allowed determining the triathletes’ technique profile during the running part. Also, it has been demonstrated the laboratory-based studies does not reproduce a top-level competition.
Resumo:
The aim of this study was to examine the effect of positioning on the correctness of decision making of top-class referees and assistant referees during international games. Match analyses were carried out during the Fe´de´ration Internationale de Football Association (FIFA) Confederations Cup 2009 and 380 foul play incidents and 165 offside situations were examined. The error percentage for the referees when indicating the incidents averaged 14%. The lowest error percentage occurred in the central area of the field, where the collaboration of the assistant referee is limited, and was achieved when indicating the incidents from a distance of 11–15 m, whereas this percentage peaked (23%) in the last 15-min match period. The error rate for the assistant referees was 13%. Distance of the assistant referee to the offside line did not have an impact on the quality of the offside decision. The risk of making incorrect decisions was reduced when the assistant referees viewed the offside situations from an angle between 46 and 608. Incorrect offside decisions occurred twice as often in the second as in the first half of the games. Perceptual-cognitive training sessions specific to the requirements of the game should be implemented in the weekly schedule of football officials to reduce the overall error rate.
