The van der Waals and Platteuw (vdVVP) theory has been successfully used to model the thermodynamics of gas hydrates. However, earlier studies have shown that this could be due to the presence of a large number of adjustable parameters whose values are obtained through regression with experimental data. To test this assertion, we carry out a systematic and rigorous study of the performance of various models of vdWP theory that have been proposed over the years. The hydrate phase equilibrium data used for this study is obtained from Monte Carlo molecular simulations of methane hydrates. The parameters of the vdWP theory are regressed from this equilibrium data and compared with their true values obtained directly from simulations. This comparison reveals that (i) methane-water interactions beyond the first cage and methane-methane interactions make a significant contribution to the partition function and thus cannot be neglected, (ii) the rigorous Monte Carlo integration should be used to evaluate the Langmuir constant instead of the spherical smoothed cell approximation, (iii) the parameter values describing the methane-water interactions cannot be correctly regressed from the equilibrium data using the vdVVP theory in its present form, (iv) the regressed empty hydrate property values closely match their true values irrespective of the level of rigor in the theory, and (v) the flexibility of the water lattice forming the hydrate phase needs to be incorporated in the vdWP theory. Since methane is among the simplest of hydrate forming molecules, the conclusions from this study should also hold true for more complicated hydrate guest molecules.

The envelope protein (E1-E2) of Hepatitis C virus (HCV) is a major component of the viral structure. The glycosylated envelope protein is considered to be important for initiation of infection by binding to cellular receptor(s) and also known as one of the major antigenic targets to host immune response. The present study was aimed at identifying mouse monoclonal antibodies which inhibit binding of virus like particles of HCV to target cells. The first step in this direction was to generate recombinant HCV-like particles (HCV-LPs) specific for genotypes 3a of HCV (prevalent in India) using the genes encoding core, E1 and E2 envelop proteins in a baculovirus expression system. The purified HCV-LPs were characterized by ELISA and electron microscopy and were used to generate monoclonal antibodies (mAbs) in mice. Two monoclonal antibodies (E8G9 and H1H10) specific for the E2 region of envelope protein of HCV genotype 3a, were found to reduce the virus binding to Huh7 cells. However, the mAbs generated against HCV genotype 1b (D2H3, G2C7, E1B11) were not so effective. More importantly, mAb E8G9 showed significant inhibition of the virus entry in HCV JFH1 cell culture system. Finally, the epitopic regions on E2 protein which bind to the mAbs have also been identified. Results suggest a new therapeutic strategy and provide the proof of concept that mAb against HCV-LP could be effective in preventing virus entry into liver cells to block HCV replication.

The rapid recent increase in microarray-based gene expression studies in the corpus luteum (CL) utilizing macaque models gathered increasing volume of data in publically accessible microarray expression databases. Examining gene pathways in different functional states of CL may help to understand the factors that control luteal function and hence human fertility. Co-regulation of genes in microarray experiments may imply common transcriptional regulation by sequence-specific DNA-binding transcriptional factors. We have computationally analyzed the transcription factor binding sites (TFBS) in a previously reported macaque luteal microarray gene set (n = 15) that are common targets of luteotropin (luteinizing hormone (LH) and human chorionic gonadotropin (hCG)) and luteolysin (prostaglandin (PG) F-2 alpha). This in silico approach can reveal transcriptional networks that control these important genes which are representative of the interplay between luteotropic and luteolytic factors in the control of luteal function. Our computational analyses revealed 6 matrix families whose binding sites are significantly over-represented in promoters of these genes. The roles of these factors are discussed, which might help to understand the transcriptional regulatory network in the control of luteal function. These factors might be promising experimental targets for investigation of human luteal insufficiency. (C) 2012 Elsevier B.V. All rights reserved.

A molecular dynamics (MD) investigation of LiCl in water, methanol, and ethylene glycol (EG) at 298 K is reported. Several; structural and dynamical properties of the ions as well as the solvent such as self-diffusivity, radial distribution functions, void and neck distributions, velocity autocorrelation functions, and mean residence times of solvent in the first solvation shell have been computed. The results show that the reciprocal relationship between the self-diffusivity of the ions and the viscosity is valid in almost all solvents with the exception of water. From an analysis of radial distribution functions and coordination numbers the nature of hydrogen bonding within the solvent and its influence on the void and neck distribution becomes evident. It is seen that the solvent solvent interaction is important in EG while solute solvent interactions dominate in water and methanol. From Voronoi tessellation, it is seen that the voids and necks within methanol are larger as compared to those within water or EG. On the basis of the void and neck distributions obtained from MD simulations and literature experimental data of limiting ion conductivity for various ions of different sizes we show that there is a relation between the void and neck radius on e one hand and dependence of conductivity on the ionic radius on the other. It is shown that the presence of large diameter voids and necks in methanol is responsible for maximum in limiting ion conductivity (lambda(0)) of TMA(+), while in water in EG, the maximum is seen for Rb+. In the case of monovalent anions, maximum in lambda(0) as a function ionic radius is seen for Br- in water EG but for the larger ClO4- ion in methanol. The relation between the void and neck distribution and the variation in lambda(0) with ionic radius arises via the Levitation effect which is discussed. These studies show the importance of the solvent structure and the associated void structure.

The electrocaloric effect (ECE) of 0.85PbMg(1/3)Nb(2/3)O(3-)0.15PbTiO(3) (0.85PMN-0.15PT) thin films deposited on (111) Pt/TiO2/SiO2/Si substrate by pulsed laser deposition (PLD) has been calculated. The reversible adiabatic temperature was calculated indirectly using the Maxwell's relation Delta T = -T/C rho integral(E2)(E1) (partial derivative P/partial derivative T)(sigma,E)dE. Permittivity and P-E measurements show an anomaly at 11 degrees C on heating only. This anomaly previously reported are claimed to arise due to the PNR depolarization upon heating. The absence of this anomaly during cooling suggests that no structural phase transition takes place. A negative electrocaloric effect is observed which is explained by the increase in the entropy term.

Aims: Administration of estradiol or compounds with estrogenic activity to newborn female rats results in irreversible masculinization as well as defeminization in the brain and the animals exhibit altered reproductive behavior as adults. The cellular and molecular mechanism involved in inducing the irreversible changes is largely unknown. In the present study, we have monitored the changes in the expression of selected synaptogenesis related genes in the sexually dimorphic brain regions such as POA, hypothalamus and pituitary following 17 beta-estradiol administration to neonatal female rats. Main methods: Female Wistar rats which were administered 17 beta-estradiol on day 2 and 3 after birth were sacrificed 120 days later and the expression levels of genes implicated in synaptogenesis were monitored by semi-quantitative reverse transcription PCR. Since estradiol induced up-regulation of COX-2 in POA is a marker for estradiol induced masculinization as well as defeminization, in the present study only animals in which the increase in expression of COX-2 gene was observed in POA were included in the study. Key findings: Down-regulation of genes such as NMDA-2B, NETRIN-1, BDNF, MT-5 MMP and TNF-alpha was observed in the pre-optic area of neonatally E2 treated female rat brain but not in hypothalamus and pituitary compared to the vehicle- treated controls as assessed by RT-PCR and Western blot analysis. Significance: Our results suggest a possibility that down-regulation of genes associated with synaptogenesis in POA, may be resulting in disruption of the cyclical regulation of hormone secretion by pituitary the consequence of which could be infertility and altered reproductive behavior. El presente trabajo se realizó en el periodo comprendido entre Noviembre y Diciembre de 1989, con el objetivo de investigar la infestación de parásitos gastrointestinales y pulmonares de mayor importancia económica en los bovinos del complejo 1 y 2 de la empresa genética Agenor Gómez. Boaco, Nicaragua. Se muestrearon 242 animales, entre bovinos de la raza Aberdeen Angus y Brahman, distribuidos en las categorías siguientes: C1 (0-1 año), C2 (1-3 años) y C3 (>3 años). Las muestras fueron extraídas directamente del recto animal, luego refrigeradas a 5°C y fueron analizadas en laboratorio de parasitología del MIDINRA, localizado en municipio de Camoapa V región. La cantidad de huevos y larvas se registró para cada género empleando las técnicas de sedimentación en agua, flotación con solución salina saturada y el método de larvoscopia e identificación con fotografías. Al realizar el ANDEVA por género de parásitos reportados en el análisis coprológico para la variable N° de huevos y larvas/gr de heces, se encontró que existen diferencias significativas con P>0.05 entre los promedios observados de los diferentes hatos. Los niveles de infestación promedio en los distintos hatos oscilaron entre 0u y 2lu; donde u= número de huevos y larvas/gramos de heces. De estos 48 promedios (8 hatos por 6 géneros de parásitos), 6 alcanzaron valores >9; 1 hato C1 de la upe Las Brisas con media de 2lu del género Haemonchus, 1 hato e de la upe San Miguel con media de 20.87u del género Strongyloides, 1 hato C2 de la upe San Jorge con media de 17.69u del género Strongyloides, 2 hatos C1 de las upe las Brisas y San Miguel con medias de 10.67u y medias de 10.5u respectivamente, ambos del género Coccidia y 1 hato e2 de la upe San Jorge con media de 9.6lu del género Coccidia. En los promedios restantes se obtuvieron valores <6u perteneciendo a la calificación leve. Posteriormente se realizó un análisis de correlación para determinar el grado de asociación existente entre cada uno de los géneros de parásitos económicamente importantes, encontrándose que el género Coccidia presentó el mayor grado de correlación con respecto a los demás géneros encontrados, se encontró que las correlaciones >= 0.25 corresponden a los géneros Coccidia, Moniezia, Neoascaris, Strongyloides y Haemonchus con r de 0.25, 0.25, 0.48, 0.30 respectivamente, similar para Haemonchus y Moniezia con r de 0.289.

Through LSC funding, Thurrock Adult Community College (TACC) purchased a single-deck bus, equipped with all the essential equipment that has allowed them to deliver fundamental IT skills within local communities. The bus travels to various local towns and villages which in turn has brought learning to the learners. This has proved to be very useful for those who perhaps have difficulty travelling to the main campuses or do not have the confidence to enrol on a course.

South Essex College has developed a college intranet called C_Space. This has grown from a handful of HTML pages to an intranet hosting 70 in-house developed applications and two million staff and student-generated pages. In the past three years, the College has developed a range of social networking tools within the site to engage students. This has provided a 360-degree appraisal system for the College, which has informed the content and design of the intranet, as well as influenced the overall development of the College.

Paul Kendrick, Vocational Trainer for Action Training, has created a bespoke virtual learning environment (VLE) utilising Google Apps for Education. Learners and trainers are now more fulfilled as they have greater ownership of their learning environment and can pick which applications best suit their learning needs. They can also work more collaboratively as they are able to share documents with other learners and trainers between different locations.

e-Learning Developer, Charlie Williams, has improved efficiencies at Oaklands College with the introduction of LAMS, the Learning Activity Management System. Charlie has worked alongside teachers at the College to create activities for a range of individual tasks, small group work and whole classes, based on online content and collaboration. Since 2010, this blended learning environment has improved retention as students can work at hours that suit them, and has given teachers more time to work on future lessons and other tasks.

As part of an LSIS Regional Response Fund project, Essex Adult Community Learning (ACL) has created a toolkit. The toolkit provides training for foreign language tutors in producing digital resources which combine audio, video, text and communication activities. The toolkit which is now an integral part of a blended learning language course, has also developed tutors' skills in using technology for teaching and learning. The main aim has also been to provide an alternative and flexible method of delivery, especially where funding cuts have impacted on the cost of running taught courses.

They have saved time and money on paper and travel, diversified evidence capture and improved their overall engagement with learners.

Broadland Council Training Services have reined in their reliance on traditional learning methods by introducing Xerte/Maxos to their equine-based students. Learners who were once deluged by stacks of paper and unable to utilise an internet connection in a horse yard are now able to access interactive learning exercises using Maxos: Xerte on a memory stick. Students are now more engaged and focused on their studies, teaching methods are much more diverse, and success rates have improved.
