BACKGROUND: Faculties face the permanent challenge to design training programs with well-balanced educational outcomes, and to offer various organised and individual learning opportunities. AIM: To apply our original model to a postgraduate training program in rheumatology in general, and to various learning experiences in particular, in order to analyse the balance between different educational objectives. METHODS: Learning times of various educational activities were reported by the junior staff as targeted learners. The suitability of different learning experiences to achieve cognitive, affective and psychomotor learning objectives was estimated. Learning points with respect to efficacy were calculated by multiplication of the estimated learning times by the perceived appropriateness of the educational strategies. RESULTS: Out of 780 hours of professional learning per year (17.7 hours/week), 37.7% of the time was spent under individual supervision of senior staff, 24.4% in organised structured learning, 22.6% in self-studies, and 15.3% in organised patient-oriented learning. The balance between the different types of learning objectives was appropriate for the overall program, but not for each particular learning experience. Acquisition of factual knowledge and problem solving was readily aimed for during organised teaching sessions of different formats, and by personal targeted reading. Attitudes, skills and competencies, as well as behavioural and performance changes were mostly learned during caring for patients under interactive supervision by experts. CONCLUSION: We encourage other faculties to apply this approach to any other curriculum of undergraduate education, postgraduate training or continuous professional development in order to foster the development of well-balanced learning experiences.

The hypothesis of ecological divergence giving rise to premating isolation in the face of gene flow is controversial. However, this may be an important mechanism to explain the rapid multiplication of species during adaptive radiation following the colonization of a new environment when geographical barriers to gene flow are largely absent but underutilized niche space is abundant. Using cichlid fish, we tested the prediction of ecological speciation that the strength of premating isolation among species is predicted by phenotypic rather than genetic distance. We conducted mate choice experiments between three closely related, sympatric species of a recent radiation in Lake Mweru (Zambia/DRC) that differ in habitat use and phenotype, and a distantly related population from Lake Bangweulu that resembles one of the species in Lake Mweru. We found significant assortative mating among all closely related, sympatric species that differed phenotypically, but none between the distantly related allopatric populations of more similar phenotype. Phenotypic distance between species was a good predictor of the strength of premating isolation, suggesting that assortative mating can evolve rapidly in association with ecological divergence during adaptive radiation. Our data also reveals that distantly related allopatric populations that have not diverged phenotypically, may hybridize when coming into secondary contact, e.g. upon river capture because of diversion of drainage systems.

Bilingual education programs implicitly assume that the acquired knowledge is represented in a language-independent way. This assumption, however, stands in strong contrast to research findings showing that information may be represented in a way closely tied to the specific language of instruction and learning. The present study aims to examine whether and to which extent cognitive costs appear during arithmetic learning when language of instruction and language of retrieving differ. Thirty-nine high school students participating in a bilingual education program underwent a four-day training on multiplication and subtraction problems in one language (German or French), followed by a test session in which they had to solve trained as well as untrained problems in both languages. We found that cognitive costs related to language switching appeared for both arithmetic operations. Implications of our findings are discussed with respect to bilingual education as well as to cognitive mechanisms underlying different arithmetic operations.

Maximal amounts of prodigiosin were synthesized in either minimal or complete medium after incubation of cultures at 27 C for 7 days. Biosynthesis of prodigiosin began earlier and the range of temperature for formation was greater in complete medium. No prodigiosin was formed in either medium when cultures were incubated at 38 C; however, after a shift to 27 C, pigmentation ensued, provided the period of incubation at 38 C was not longer than 36 hr for minimal medium or 48 hr for complete medium. Washed, nonpigmented cells grown in either medium at 38 C for 72 hr could synthesize prodigiosin when suspended in saline at 27 C when casein hydrolysate was added. These suspensions produced less prodigiosin at a slower rate than did cultures growing in casein hydrolysate at 27 C without prior incubation at 38 C. Optimal concentration of casein hydrolysate for pigment formation by suspensions was 0.4%; optimal temperature was 27 C. Anaerobic incubation, shift back to 38 C, killing cells by heating, or chloramphenicol (25 mug/ml) inhibited pigmentation. Suspensions of washed cells forming pigment reached pH 8.0 to 8.3 rapidly and maintained this pH throughout incubation for 7 days. Measurements of viable count and of protein, plus other data, indicated that cellular multiplication did not occur in suspensions of washed cells during pigment formation. By this procedure utilizing a shift down in temperature, biosynthesis of prodigiosin by washed cells could be separated from multiplication of bacteria.

Bacterial meningitis is fatal in 5% to 40% of patients and causes neurologic sequelae in up to 30% of survivors. Much has been learned recently about the mechanisms that lead to brain injury during meningitis. Once bacteria have gained access to the central nervous system, their multiplication triggers a complex host response consisting of humoral and cellular immune mediators, reactive oxygen intermediates, matrix-metalloproteinases, and other host-derived factors. Alterations of the cerebral vasculature, with disruption of the blood brain barrier and global and focal ischemia, ultimately lead to functional and structural brain damage. This article reviews current concepts of the pathophysiology of bacterial meningitis and emphasizes possible therapeutic strategies to prevent its harmful consequences.

The twenty-first century has seen a further dramatic increase in the use of quantitative knowledge for governing social life after its explosion in the 1980s. Indicators and rankings play an increasing role in the way governmental and non-governmental organizations distribute attention, make decisions, and allocate scarce resources. Quantitative knowledge promises to be more objective and straightforward as well as more transparent and open for public debate than qualitative knowledge, thus producing more democratic decision-making. However, we know little about the social processes through which this knowledge is constituted nor its effects. Understanding how such numeric knowledge is produced and used is increasingly important as proliferating technologies of quantification alter modes of knowing in subtle and often unrecognized ways. This book explores the implications of the global multiplication of indicators as a specific technology of numeric knowledge production used in governance. Combination of insights from anthropology of law, history of science, science and technology studies, sociology of quantification, economics and geography will appeal to those who are uncomfortable with the separation between 'theoretical' and 'empirical' approaches and with the current weakness of critique that address the main trends shaping the relations between capitalism, markets, law and democracy Theoretical discussion of the nature and historical formation of quantification will appeal to those who ask questions such as, 'What is new or different about our contemporary reliance on quantitative knowledge?' Groundbreaking empirical case studies uncover the social work and politics that often go into the making of indicators and explore the far-reaching effects and impacts of these numerical representations in specific settings

Buruli ulcer, caused by infection with Mycobacterium ulcerans, is a necrotizing disease of the skin and subcutaneous tissue, which is most prevalent in rural regions of West African countries. The majority of clinical presentations seen in patients are ulcers on limbs that can be treated by eight weeks of antibiotic therapy. Nevertheless, scarring and permanent disabilities occur frequently and Buruli ulcer still causes high morbidity. A vaccine against the disease is so far not available but would be of great benefit if used for prophylaxis as well as therapy. In the present study, vesicular stomatitis virus-based RNA replicon particles encoding the M. ulcerans proteins MUL2232 and MUL3720 were generated and the expression of the recombinant antigens characterized in vitro. Immunisation of mice with the recombinant replicon particles elicited antibodies that reacted with the endogenous antigens of M. ulcerans cells. A prime-boost immunization regimen with MUL2232-recombinant replicon particles and recombinant MUL2232 protein induced a strong immune response but only slightly reduced bacterial multiplication in a mouse model of M. ulcerans infection. We conclude that a monovalent vaccine based on the MUL2232 antigen will probably not sufficiently control M. ulcerans infection in humans.

En este trabajo se presentan estudios de germinación y propagación in vitro de L. heterophylla, nativa con potencial ornamental. Se evaluó el efecto del momento y lugar de recolección sobre la germinación. Se recolectaron semillas en plena floración y al final de la misma, en dos localidades de la provincia de Mendoza: Gualtallary y Chacras de Coria. Las pruebas de germinación se realizaron en estufa a 20 °C. Las semillas provenientes de Gualtallary germinaron en mayor proporción que las de Chacras de Coria: en ambas localidades la máxima germinación se obtuvo en la recolección de plena floración. Para establecer un protocolo de micropropagación se realizaron dos ensayos de introducción y uno de multiplicación. En la introducción se evaluó el medio de cultivo MS entero o ½ de macronutrientes, en combinación con BA y AIB. En la multiplicación se evaluaron los medios MS ½ o ¼ de macronutrientes con 20 o 40 g.L-1 de sacarosa. No se encontraron diferencias en la sobrevivencia, el BA incrementó significativamente la proliferación de brotes. El mejor medio de multiplicación fue MS ¼ adicionado de 20 g.L-1 de sacarosa.

La azucena del campo (Az-del campo) (Habranthus gracilifolius) y la azucenita colorada (Az-colorada) (Rhodophiala bifida) son bulbosas nativas de Argentina de valor ornamental potencial, de las que se tiene escasa información sobre la germinación y el crecimiento. Para establecer la aptitud germinativa, el régimen de incubación más adecuado, y el crecimiento de las hojas y los bulbos, se realizó un ensayo de germinación y las plántulas se cultivaron en macetas durante un año. Se emplearon cuatro regímenes de incubación: temperatura constante de 20°C y alternadas de 20°/30°C y de 10°/20°C, todos con 8 horas de luz, y de 10°/20°C en oscuridad. Las semillas resultaron fotoblásticas neutras. A temperaturas de 20°C o de 10°/20°C, en ambas especies, la energía y el poder germinativo superaron el 75 y 92%, respectivamente, pero temperaturas de 20°/30°C demoraron la germinación. De otoño a fin de primavera las plantas presentaron hasta 4 hojas en Az-del campo y 2,5 hojas/ planta en Az-colorada, pero en verano no exhibieron hojas. Los bulbos presentaron crecimiento lineal, siendo mayores las tasas en Az-del campo (ancho = 0,022 y longitud = 0,049 mm.día-1) que en Az-colorada (ancho = 0,011 y longitud = 0,014 mm.día-1). La multiplicación por semillas es efectiva, pero en ambas especies se necesita más de un año para alcanzar el estadio reproductivo.

Se micropropagó Cissus tiliacea, recurso fitogenético con potencial agronómico y farmacológico, en los medios de cultivo Murashige-Skoog (MS) y Lloyd y McCown (WPM). En ambos medios se generaron resultados similares para número de brotes, nudos, hojas y raíces adventicias, sólo existió diferencia significativa (p ≤ 0,05) en la formación de callo. Para la multiplicación in vitro se utilizó WPM adicionado con 0; 0,5; 1,0; 1,5 ó 2,0 mg L-1 de benciladenina (BA) y se emplearon tres tipos de segmentos nodales (basal, medio y apical). Las concentraciones de 0 y 0,5 mg L-1 de BA resultaron en un mayor tamaño y desarrollo del explante, además permitieron la formación de 1,2 a 1,6 raíces por explante. Las concentraciones de 1,5 y 2,0 mg L-1 de BA indujeron la formación de callo. No existió diferencia significativa en las variables evaluadas por efecto del tipo de segmento nodal establecido in vitro. En el enraizamiento, en el medio MS, se evaluaron tres tipos de auxinas: ácido naftalen-1-acético (ANA), ácido indol-3-butírico (AIB) y ácido indol- 3-acético (AIA) a 0,5 mg L-1; el mayor número de raíces secundarias y diámetro de la raíz principal fue inducido por ANA, sin embargo AIB indujo una mayor elongación de la raíz principal. Los resultados del presente trabajo sugieren que el cultivo in vitro de C. tiliacea es una alternativa para su conservación y multiplicación.

La especie nativa Mutisia subspinosa es una enredadera perenne, resistente a heladas, con hermosas flores anaranjadas que presenta gran interés como ornamental. En trabajos previos de propagación a través de semillas o estacas se obtuvieron escasos resultados, lo que justificó el uso de la micropropagación. Para el establecimiento in vitro se utilizó el medio de Murashige Skoog (MS) diluido a la mitad. Se evaluó la adición de 6-bencilaminopurina (BAP) sola o en combinación con thidiazurón (TDZ) y un testigo sin hormonas. En la etapa de multiplicación se utilizó el mismo medio basal y se probaron dos auxinas: los ácidos indol butírico (AIB) y naftalén acético (ANA), además del uso de carbón activado. La composición del medio de cultivo más adecuada para el establecimiento fue la del testigo, mientras que para la multiplicación los mejores resultados se obtuvieron con la adición de 4 μmoles.L-1 de ANA. El agregado de 1 g.L-1 de carbón activado al medio de cultivo con 2,7 μmoles.L-1 mejoró la tasa de multiplicación y el enraizamiento.

The flower market is characterized by being both eager for novelties and highly competitive. The exploration of native species with ornamental potential represents a remarkable area of research, since it entails the introduction and development of novel promising ornamental crops. The genus Glandularia, widely distributed in Argentina, holds an enormous ornamental potential, due to the variety of colors of its inflorescences (red, violet, white, rose and lily), and extended flowering period. There is little information on tissue culture of Glandularia, thus highlighting the relevance of this research. In this work, the conditions for in vitro multiplication of G. peruviana were optimized. It was concluded that WPM supplemented with TDZ, in concentrations ranging from 1.1 to 9.0 μM, was the most adequate treatment, rendering a multiplication rate of approximately 10 de novo shoots per explant. This paper presents a protocol for the in vitro propagation of this species and introduces interesting prospects in the application of biotechnological tools to breed Glandularia.

Con el objeto de micropropagar portainjertos de vid de interés para la provincia de Misiones (Paulsen 1103 y Vr 04343) se cultivaron segmentos nodales y ápices meristemáticos. Para el establecimiento de segmentos nodales se evaluaron diferentes concentraciones del medio propuesto por Murashige y Skoog (MS) -¼, ½, 1- adicionando diferentes concentraciones de benciladenina (0; 1; 3 y 5 mg.L-1) y ácido naftalenacético (0 y 0,01 mg.L-1). Se evaluaron estado fisiológico y topófisis de yemas. En fase de multiplicación se evaluaron ápices, segmentos uni y binodales con o sin hoja desplegada. Para el establecimiento de ápices se evaluó el medio MS ½ suplementado con bencilaminopurina (0; 0,5; 1; 2 mg.L-1). Los mejores resultados para el establecimiento de segmentos nodales se obtuvieron con yema despierta en medio MS ¼ sin adición de reguladores. La multiplicación de los mismos puede realizarse partiendo de un explanto uni o binodal con o sin hoja. Al utilizar ápices como explanto, se debe adicionar al medio MS ½ bencilaminopurina en una concentración de 0,5 mg.L-1. En la aclimatación de las plantas, se obtuvieron valores de supervivencia de 80 a 90%.

En este trabajo se estableció un protocolo de propagación in vitro de tres especies nativas del género Glandularia: G. peruviana, G. sp. y G. laciniata. Para el establecimiento in vitro se evaluó el medio de Murashige Skoog (MS) con macro y micronutrientes diluidos a la mitad adicionado con 0,05 μM de bencilaminopurina (BAP) sola o en combinación con 0,1 μM thiadiazuron (TDZ) y un testigo sin reguladores del crecimiento. En la etapa de multiplicación se evaluó el medio de cultivo MS diluido a la ½ ó ¼ y adicionado de 20 ó 40 gr.L-1 de sacarosa. Es posible establecer y micropropagar estas especies en medios de cultivo sencillos. El medio más eficiente para el establecimiento fue aquel sin reguladores, mientras que el más adecuado para la multiplicación fue MS ½ adicionado de 20 gr.L-1 de sacarosa, en el cual la tasa de multiplicación cada 30 días fue de 6 en G. sp. y G. peruviana y 4 para G. laciniata.

Multiplication rate and production of zooflagellates were determined in the Central Indian Ocean. In the surface zone of the areas studied zooflagellate generation time ranged from 24 to 48 hours and production from 0.5 to 10.6 mg/m**3. The lowest multiplication rate was found at depth of 100 m (92 hours); no multiplication was found in water samples taken from depth of 120 m or below.
