Alison Margaret Gill y Peter Denham Smithson son, o eran, ambos murieron hace ya algunos años, dos arquitectos. Así dicho, esto suena a obviedad. Sin embargo no lo es, porque siempre han sido compactados en una unidad proyectual, bien bajo las siglas Alison y Peter Smithson, o bien, más puntualmente todavía, como los Smithson. Cuando Leibniz en 1696 le dice a la princesa electora Sofía que en algo tan puntual y homogéneo aparentemente como una gota de agua, vista al microscopio, resulta haber más de un millón de animales vivos, se puede empezar a vislumbrar el resultado de la Tesis. El resultado de este trabajo no se contempla un millón de partículas pero sí dos. La obra de Alison y Peter Smithson esta conformada desde su inicio probablemente por la particularidad de que en ella intervienen tanto Gill como Smithson, aunque cada uno sea en mayor o menor medida la o el responsable de un proyecto. Puesto al microscopio el proyecto del que se ocupa la Tesis, el concurso de 1953 para la ampliación de la Universidad de Sheffield, se estima que está compuesto de dos partes diferentes y que como Los Smithson o como una gota de agua, su unidad es una mera convención, una forma de designarlo y tratarlo, que falsea en algo su apreciación, aunque, como se verá, es lo que provoca el extraño efecto que como imagen ejerce. El Capítulo I descubre la duplicidad. Reyner Banham fijó en 1955, en el artículo The New Brutalism, el modo de encarar el proyecto de Sheffield cuando proyectó sobre él lo más novedoso que en el campo de la pintura ocurría en ese momento, el Art Autre. Michel Tapié ve en el informalismo presente en las obras que él engloba como un <> rasgos que puede presentar a la luz de conceptos que extrae de Riemann, matemático del s. XIX, introductor de una nueva geometría. La Tesis remonta por tanto hasta Riemann, a la búsqueda del concepto del que penden tanto Tapié como Banham. El concepto es la variedad continua; un conjunto, cuyos elementos pueden recorrer la extensión de la variedad mediante una deformación continua, como si dijéramos, diluyéndose unos en otros sin cortes entre sí. Los ejemplos del informalismo serían, por tanto, los puntos de la variedad, formas inestables o detenciones aleatorias en este transitar continuo de un mismo objeto. La visión de cuerpos en cortes aleatorios a la evolución otorga a sus formas cualesquiera la condición de extrañeza o perturbación, produciendo un impacto en la mente que lo graba instintivamente en la materia de la memoria. Así, es la memorabilidad, dada precisamente por el impacto que supone ver cuerpos en estado evolutivo, sin una forma precisa, la propiedad que gobierna a los objetos que caen bajo el Art Autre, y es la memorabilidad el rasgo distintivo de Sheffield para Banham. Este sentido evolutivo que se puede asociar a los elementos de una variedad entra en consonancia con una variedad de estados en evolución en el marco de las especies orgánicas. No sólo como individuo sino también como especie. La coincidencia no sólo es formal, también es temporal entre el descubrimiento de Riemann de la variedad continua y el de Darwin de la evolución de las especies. Ve Darwin que algunos ejemplares presentan en su cuerpo conformaciones normales de especies diferentes, que puede entenderse como una anormalidad o defecto evolutivo, lo que él llama <>. Un ejemplar monstruoso no es capaz de abrir una escisión de la variedad y en este sentido rompe su cadena evolutiva. Es un elemento, por tanto extraño a la variedad. Lo peculiar que expone Darwin, es que la monstruosidad puede ser provocada artificialmente por motivos estéticos, como ocurre en determinados animales o plantas ornamentales. Bien, esto es exactamente lo que descubre la Tesis en el proyecto de Sheffield. La unión en un solo proyecto de dos conformaciones de naturaleza diferente. El Capítulo II indaga en la naturaleza dual de Sheffield. Y la descubre en la aplicación de dos principios que tienen múltiples derivas, pero interesante en una en particular, referida al espacio. Uno es el Principio de Posición. Bajo él se contempla la situación del concurso dada en su globalidad, en una visión totalizadora que permite buscar y destacar posiciones singulares y tener un control preciso del proyecto desde el inicio. Se dice que es la opción normal en arquitectura comprender el espacio en su conjunto y que el trazado de ejes y el reparto del espacio obedecen a este principio. Son así vistos la propuesta ganadora del equipo GMW y fundamentalmente el trazado de ejes en la obra de Le Corbusier. Una parte de Sheffield es un producto típico de este modo de proceder. Hay un segundo principio que es el Principio de Transición Continua. En los términos expuestos para la variedad de Riemann, sería como si en lugar de ver cuerpos en estados concretos, por deformados que pudieran ser, nos introdujéramos dentro de la propia variedad acoplándonos al curso de la evolución misma, en un acto de unión interior con la marcha de la evolución. Para ejercer esta acción se toma como punto inicial el extremo del edificio universitario existente, el Firth Court, y se lleva cabo un estiramiento continuo. En esto radica lo que la Tesis distingue como la ampliación, el cuerpo que se enrosca paulatinamente. Para exponer este concepto, la Tesis se apoya en un ejercicio llevado a cabo por Raymond Unwin en Hampstead, el close, o estiramiento lateral del borde de un viario para hacer surgir el espacio útil residencial. A partir del concepto de close, se deriva en la urbanística americana de principios del s. XX el concepto de cluster, que pasa a ser uno de los argumentos principales de la teoría de Alison y de Peter Smithson en los años 50. El Capítulo III plantea la dificultad de mantener la dualidad de Sheffield a primeros de los 50, sometido el proyecto de arquitectura a la unicidad que impone el criterio hegemónico de Rudolf Wittkower desde “Los Fundamentos de la Arquitectura en la Edad del Humanismo”. Como en el Capítulo I, la obligación que se traza la Tesis es remontar los orígenes de las cosas que se tratan, y en este caso, entrar en las fuentes del principio proporcional. Se descubren así los fundamentos de la proporción aritmética y geométrica en el seno del pitagorismo, sus problemas y su evolución. La postura de los dos arquitectos frente a Wittkower es de admiración, pero también de libertad. Esta libertad suya es la que defiende la Tesis. Cotejando lo que da de sí la arquitectura basada en los principios proporcionales con otra arquitectura estancada en el paso del Renacimiento al Barroco como es la arquitectura perspectiva y proyectiva, la arquitectura oblicua, recuperada a la sazón por medio de la intervención en la Tesis de Panofsky, se dirimen aspectos colaterales pero fundamentales de una renovación de los planteamientos: hay planteamientos que se dicen esencialistas y objetivos y otros funcionales y que derivan en subjetivos y relacionales. Sobre estas dos marcos de categorías el propósito que persigue la Tesis es dar cuenta de los dos principios que rigen lo visto en el Capítulo II, el Principio de Posición y el Principio de Transición Continua, responsables ambos, al 50%, de la monstruosidad detectada en Sheffield, que no es negativa, sino, como decía Darwin, la combinación en un solo cuerpo de conformaciones normales en animales de especies diferentes, incluso con fines estéticos. En Sheffield existe, y esta dibujada, la figura de una cabeza escultural con un doble rostro. Todo el trabajo de la Tesis se encamina, en definitiva, a explicar, con toda la claridad y extensión que me ha sido posible, en qué consiste ese doble rostro. ABSTRACT Peter and Alison Smithson’s work is, is from its start, very influenced by both Alison Margaret Gill and Peter Denham Smithson. The matter of study of this Thesis, the 1953 competition for the expansion of the University of Sheffield, estimated after its exam, which is composed by two different parts, which, together produce a strange effect as an image. Reyner Banham in 1955 described the image in his article “The New Brutalism” as the key argument (most iconic) for the Sheffield’s project. The way his image powerfully influences sensitivity, by perturbation, makes this a singular and memorable image. This feature, only present in Sheffield, over any other building of the time, even from the same architects, allow Banham to associate the project to Art Autre, thought by Michel Tapié as a new artistic movement, to which Sheffield will belong, in the architecture part. Tapié sees in the informalism of works considered Art Autre some aspects that can bring concepts he extracts from Riemann, XIX Century mathematician, father of the new geometry. This Thesis discovers Riemann’s concept of continuous variety, a set whose elements are able to go through variety by a continuous deformation, diluting themselves without touching each other. Examples of informalism would be, points of that variety, unstable forms or random detentions in the continuous transit of the same object. Therefore, the condition of memorability comes precisely from the impact that seeing bodies in state of evolution creates. That evolutive sense that can be associated to elements of a variety, comes together with a variety of states of evolution in the world of organic species. Not only as an individual, but as well as a species. Coincidence between Riemann and Darwin’s discoveries is not only formal, but as well temporary. Darwin observes that some individuals of concrete species present on their bodies some features, typical of other species, which may be interpreted as evolutive failure. But the most peculiar part of what Darwin exposes is that monstrosity can indeed be artificially made for aesthetical purposes, like it happens in certain animals and plants. Well, this is what the Thesis discovers in Sheffield’s project. The union in a single project of two different nature forms, of which none on the parts is a deformation of the other, but they are both irreducible. Once both parts are collated, a new system which adapts well is discovered. It is a system created by Leibniz in the XVII Century, created to distinguish to principles that clear the difference between the equation methods and differential calculus. This principles are the Principle of Position and the Principle of Continuity. This two principles, translated to the spatial analysis field are key for the two parts of the project. On the one hand, the part developing in a lineal axis belongs to the Principle of Position. This means that that there is a global vision of space after which it is decided which operation to follow, which in this case consists of establishing an axis between two singular positions of the university area. The part which is being deformed while it goes is studied as a continuous action of stretching an existing building, the Firth Court, in which there is no previous spatial analysis. The only way to understand and to explain this action is by the Principle of Continuity. So, all in all, the Thesis changes the view of Sheffield from an Art Autre work to a “monstrosity”, without the negative meaning of it, just as a combination of two different nature formations, which, at the same time, justifies its power as iconic image. Finally, I would like to point out that in the Sheffield’s project there is (drawn and also physically) a sculptural head which has the feature of representing both, a man and a woman’s face. All the work of this Thesis leads to explaining the double nature of the project, taking this double expression head as inspiration.

La obra fílmica del director francés Jacques Tati podría considerarse como el perfecto reflejo del paradigma edificatorio de mediados del siglo XX en plena posguerra europea, una época ávida de transformaciones de las que el cine supo hacerse eco. Particularmente, el cine de Tati refleja las preocupaciones del ciudadano europeo de posguerra sobre las consecuencias de las masivas construcciones erigidas en sus devastados núcleos urbanos y la puesta en práctica de la ciudad funcional propuesta por la Carta de Atenas (1931). Pero, además, el análisis del cine de Jacques Tati permite un acercamiento a la modernidad desde diversos puntos de vista como la movilidad, el diseño urbano, las nuevas construcciones, los espacios de trabajo en los nuevos distritos terciarios, la vivienda -tradicional, moderna y experimental- o el diseño mobiliario en la posguerra. A través de su alter ego -Monsieur Hulot- Tati interacciona curioso con las nuevas construcciones geométricas de paños neutros y cuestiona su ruptura con la tradición edificatoria francesa, enfatizando la oposición entre el pasado nostálgico y la modernidad de las décadas de los 50 y 60, salpicadas por el consumismo feroz del recién estrenado estado de bienestar. La confrontación funcional, volumétrica, estética e incluso cromática entre ambos mundos construidos –el tradicional y el moderno- invita al espectador a un ejercicio de reflexión y crítica sobre la arquitectura moderna de este período en Europa. En particular, la mirada cinematográfica de Tati se centra en dos conceptos fundamentales. Por una parte, su atención se dirige a la famosa casa mecanicista Le Corbuseriana materializada en la ultra-moderna casa Arpel (Mon Oncle, 1958) y proyectada en la misma época en la que se desarrollaban importantes prototipos de vivienda experimental como la Casa de Futuro de Alison y Peter Smithson o las viviendas de Jean Prouvé. Debe ponerse de manifiesto que la crítica de Jacques Tati no se centraba en la arquitectura moderna en sí misma sino en el empleo erróneo que los usuarios pudieran hacer de ella. Por otro lado, Tati centra su atención en el prisma miesiano a través de los bloques de oficinas que conforman la ciudad de Tativille en Playtime (1967). Se trataba de una gran ciudad moderna construida explícitamente para el rodaje de la película y basada en los casi idénticos tejidos urbanos residenciales y terciarios ya en funcionamiento en las principales capitales europeas y norteamericanas en aquellos años. Tativille funcionaría como una ciudad autónoma disponiendo de diversas instalaciones y con el objetivo de integrarse y consolidarse en la trama urbana parisina. Lamentablemente, su destino al final del rodaje fue bien distinto. En definitiva, el análisis de la producción fílmica de Jacques Tati permite un acercamiento a la arquitectura y al urbanismo modernos de posguerra y al contexto socio-económico que favoreció su crecimiento y expansión. Por ello, su obra constituye una herramienta visual muy útil que aún hoy es consultada y mostrada por su claridad y humor y que invita a los ciudadanos –telespectadores- a participar en un ejercicio crítico arquitectónico hasta entonces reservado a los arquitectos. ABSTRACT The film work of French director Jacques Tati could be considered as the perfect reflection of the mid-20th century European post-war building paradigm, a period of time plenty of transformations perfectly echoed by cinema. In particular, Tati’s film work reflects the European post-war citizen’s concerns about the consequences of massive constructions built in their desvastated urban centres, as well as the development of functional cities proposed by the Athens Charter (1931). But, on top of that, an analysis of Jacques Tati’s cinematography allows for an approach to modernity from different perspectives, such as mobility, urban design, new buildings, working spaces in the new tertiary districts, housing -traditional, modern, and experimental-, or furniture design during the post-war period. Embodied by his alter-ego –Monsieur Hulot,- Tati curiously interacts with the new geometric constructions of neutral facades and questions the break with the French building tradition, highlighting the opposition between the nostalgic past and modernity of the 50s and 60s, affected by the fierce consumerism of the new welfare state. The functional, volumetric, aesthetic and even chromatic confrontation between both built worlds –traditional vs modern- invites the viewer to an exercise of meditation and criticism on the European modern architecture of that period. Tati’s film look is particularly focused on two basic concepts: on the one hand, his attention addresses Le Corbusier’s famous mechanistic house which is materialized in the ultra-modern Arpel house (Mon Oncle, 1958) and designed, in turn, when the development of other important experimental dwelling prototypes like Alison and Peter Smithson’s House of the Future or Jean Prouvé´s houses was taking place. It must be highlighted that Jacques Tati’s criticism was not addressed to modern architecture itself but to the wrong use that citizens could make of it. On the other hand, Tati focuses on the Miesian prism through the office buildings that shape the city of Tativille in Playtime (1967). It was a big, modern city built specifically for the film shooting, and based on the almost identical residential and tertiary urban fabrics already active in the main European and American capitals those years. Tativille would work as an autonomous city, having several facilities at its disposal and with the goal of getting integrated and consolidated into the Parisian urban weave. However, its final use was, unfortunately, quite different. In conclusion, an analysis of Jacques Tati’s film production allows for an approach to modern post-war architecture and urbanism, as well as to the socio-economic context that favoured its growth and expansion. As a result of this, Jacques Tati’s film production constitutes a suitable visual tool which, even nowadays, is consulted and shown due to its clarity and humour, and at the same time invites citizens –viewers- to participate in an architectural criticism exercise that, so far, had been reserved to architects.

Esta tesis doctoral busca estudiar el espacio desde la premisa de que el espacio es, fundamentalmente, lo intermedio. El entre, lo que hay entre las cosas mismas y que les da, precisamente, su definición como cosas en un constante proceso de delimitación. Este entre, lo que hay entre las cosas, no es sin embargo un resto que queda, sino bien al contrario el principio activo que hace que las cosas, desde lo que hay alrededor, se configuren en su ser sensible y puedan percibirse. El entre, lo intermedio, no es así una línea, un corte puro, sino un intervalo, un espacio en sí mismo, un ámbito en que se desarrolla un proceso. Es por tanto un espacio de formación, en el que las cosas aún no son y ya están siendo, un intervalo ambiguo pleno de virtualidades, unas desplegadas otras que permanecen en espera, siempre en movimiento. Es la hipótesis principal de esta tesis que en el llamado Movimiento Moderno en la arquitectura del siglo veinte esta condición ambigua del límite se pone de manifiesto como la raíz articuladora de los cambios espaciales y formales que definen su ser y su estética. Para desarrollar esta hipótesis, se parte de una doble premisa: por una parte, como un entre que es en sí misma, la arquitectura se encuentra delimitada por las disciplinas artísticas contemporáneas, especialmente pintura y escultura y a la vez las delimita. Por tanto, se explorarán una serie de momentos claves en las vanguardias de comienzos del siglo veinte en paralelo a una serie de arquitecturas contemporáneas para estudiar esta doble dialéctica entre límites. Por otra parte, y entendiendo que lo propio de la arquitectura es el espacio, se estudiarán en profundidad las concepciones espaciales que en la física y la estética del finales del siglo diecinueve y principios del veinte se desarrollan, para comprender cómo estas nuevas concepciones, centradas en las ideas de relatividad y de espacio-tiempo como magnitudes interlimitadas, determinan a su vez las concepciones espaciales que se desarrollan simultáneamente en las artes plásticas y en la arquitectura. La tríada espacio/tiempo/luz regirá este recorrido, en un doble camino: a través de la física y la teoría del arte, y a través de la arquitectura y las artes plásticas. Se trata por tanto de incardinar la arquitectura en su contexto artístico y científico, y comprobar cómo la comprensión del espacio como un entre, como un intervalo del límite y no como un absoluto trascendente, se generaliza en dichos ámbitos en una exploración paralela que condiciona los resultados en todos ellos y que define, por tanto, la arquitectura de la modernidad como una arquitectura del entre. El encuadre enmarca como motivo principal el Movimiento Moderno a través de la figura de Le Corbusier. Se muestra el modo en que los recursos y mecanismos empleados provocan el encuentro del espacio indecible a través de la polifonía de las artes. Desde el espaciamiento del límite, su borradura, su ambigüedad, producidos mediante el entrelazamiento de los recursos artísticos, la continuidad de lo lineal, el encabalgamiento, el maridaje, la ambivalencia, la relatividad del color y la luz, el intersticio. Ello se analiza a través de la obra de le Corbusier, destacando la dialéctica entre Le Cheminée y Nature morte aux nombreux objets; Ma maison y el pabellón del Zurichhorn; Unité y L’espace indicible. Aclaradas las premisas fundamentales y las consecuencias inmediatas para la arquitectura del Movimiento Moderno, la cuarta parte y final de la tesis, mucho más breve que las anteriores, expone una primera aproximación a las consecuencias que todo esto ha tenido en un momento liminal también: el comienzo de la postmodernidad. En realidad se trata de una apertura a lo que ha de venir, pues establecida la operatividad de la hipóteisi inicial, se trata aquí solamente de abrir el camino a una interpretación que en realidad escapa del alcance de esta tesis. Así se deja apuntado cómo, a través de la obra de tres arquitectos, teóricos y artistas trabajando en los años setenta y comienzos de los ochenta del pasado siglo, una comprensión más clara de la revolución moderna precipitada por esta nueva comprensión del espacio como límite-entre queda patente. Chamberworks de Daniel Libeskind, Wexner Center for the Visual Arts de Peter Eisenman y Manhattan Transcripts de Bernard Tschumi servirían de privilegiados ejemplo. Como queda patente que las consecuencias de todo ello aún las estamos sintiendo en nuestra contemporaneidad. ABSTRACT This thesis aims to study the space from the premise that space is mainly the in-between. The in-between, what is in-between the things themselves and gives them precisely its definition as things in a constant process of delimitation. This in-between, what is in between things, is not a remnant that remains, but on the contrary it is the active ingredient that produces things from what is around, that configures them in a sensitive to be perceived. The in-between, the intermediate, is not a line, a pure cut, but an interval, a space in itself, an area in which a process is developed. It is therefore a space in development, where things are still not and already been, delimiting an ambiguous range full of potentialities, some present and others that remain only as virtualities. It is the main hypothesis of this thesis that in the so-called Modern Movement in the architecture of the twentieth century this ambiguous boundary condition is revealed as the result of the articulatation of spatial and formal changes. To develop this hypothesis, I posit this double premise: On the one hand, as an in-between which is in itself, architecture is defined by contemporary artistic disciplines, especially painting and sculpture and simultaneously defines them. Therefore, a number of key moments in the avantgarde of the early twentieth century in parallel with a number of contemporary architecture to study this double dialectic between boundaries will be explored. On the other hand, and considering that the “essence” of the architecture is space, I will study in depth the spatial concepts in physics and aesthetics of late-nineteenth century and early twentieth century to understand how these new concepts centered on the ideas of relativity and space-time as interlimited magnitudes in turn determine the spatial conceptions that take place simultaneously in the visual arts and architecture. The space / time / light triad governs this journey in a double way: through physics and theory of art, and through architecture and the arts. The aim is to anchor architecture in its artistic and scientific context, and to see how the understanding of space as an in-between interval and not as an absolute transcendent determines the results in all of them, defining, therefore, the architecture of modernity as an architecture of the in-between. The frame of the study is centered in the modern movement through the figure of Le Corbusier and how rthe different resources and mechanisms employed provoke an unspeakable space of encounter through the polyphony of the arts. From the spacing of the limit, its erasure, its ambiguity, produced by the intertwining of artistic resources to continuity of the linear, the marriage, the ambivalence, the relativity of color and light, the gap. All this is analyzed through the work of Le Corbusier, highlighting the dialectic between Le Cheminée and Nature morte aux nombreux objets, Ma maison and Zürichhorn pavilion, Unité and L’espace indicible. Once the basic premises and immediate consequences for the architecture of the Modern Movement are set, the fourth and final part of the thesis aims to explore the impact that all this has had in another liminal time: the beginning of postmodernism. Of course, this part is shorter, in fact it is more a prospective survey and an opening of possibilities than a truly articulated body of theory, since the conceptual core of this thesis has been developed in the central part. Hence, through the work of three architects, theorists and artists working in the seventies and early eighties of last century, a clearer understanding of the modern revolution precipitated by this new understanding of space as a boundary- between is proposed. Chamberworks by Daniel Libeskind, Wexner Center for the Visual Arts by Peter Eisenman and Manhattan Transcripts by Bernard Tschumi will exemplify this approach. The consequences of all this are still haunting our contemporaneity.

We are interested in using recombinant adeno-associated viral vectors in the treatment of hemophilia A. Because of the size constraints of recombinant adeno-associated viral vectors, we delivered the heavy and light chains of the human factor 8 (hFVIII) cDNA independently by using two separate vectors. Recombinant AAV vectors were constructed that utilized the human elongation factor 1α promoter, a human growth factor polyadenylation signal, and the cDNA sequences encoding either the heavy or light chain of hFVIII. Portal vein injections of each vector alone, a combination of both vectors, or a hFIX control vector were performed in C57BL/6 mice. An ELISA specific for the light chain of hFVIII demonstrated very high levels (2–10 μg/ml) of protein expression in animals injected with the light chain vector alone or with both vectors. We utilized a chromogenic assay in combination with an antibody specific to hFVIII to determine the amount of biologically active hFVIII in mouse plasma. In animals injected with both the heavy and light chain vectors, greater than physiological levels (200–400 ng/ml) of biologically active hFVIII were produced. This suggests that coexpression of the heavy and light chains of hFVIII may be a feasible approach for treatment of hemophilia A.

Src family tyrosine kinases are involved in modulating various signal transduction pathways leading to the induction of DNA synthesis and cytoskeletal reorganization in response to cell-cell or cell-matrix adhesion. The critical role of these kinases in regulating cellular signaling pathways requires that their activity be tightly controlled. Src family proteins are regulated through reversible phosphorylation and dephosphorylation events that alter the conformation of the kinase. We have found evidence that Src also is regulated by ubiquitination. Activated forms of Src are less stable than either wild-type or kinase-inactive Src mutants and can be stabilized by proteasome inhibitors. In addition, poly-ubiquitinated forms of active Src have been detected in vivo. Taken together, our results establish ubiquitin-mediated proteolysis as a previously unidentified mechanism for irreversibly attenuating the effects of active Src kinase.

Antigen-induced stimulation of the immune system can generate heterogeneity in CD4+ T cell division rates capable of explaining the temporal patterns seen in the decay of HIV-1 plasma RNA levels during highly active antiretroviral therapy. Posttreatment increases in peripheral CD4+ T cell counts are consistent with a mathematical model in which host cell redistribution between lymph nodes and peripheral blood is a function of viral burden. Model fits to patient data suggest that, although therapy reduces HIV replication below replacement levels, substantial residual replication continues. This residual replication has important consequences for long-term therapy and the evolution of drug resistance and represents a challenge for future treatment strategies.

Apoptosis, also called programmed cell death, has attracted great attention in recent years. After its discovery by Carl Vogt in 1842, apoptosis research was dormant for more than a century. Its rediscovery in the second half of this century, and the coining of the term apoptosis in 1972 by Kerr, Wyllie, and Currie, ignited an unparalleled interest in this field of science. The number of publications related to apoptosis has been growing exponentially every year ever since. This is mainly due to three major advances, two of which have been made recently and one that is currently seen. First, studies with the small nematode Caenorhabditis elegans have identified a number of apoptosis regulating genes—the first evidence that cell death is an active process under genetic control. Many of these genes have mammalian homologs that, like their worm counterparts, seem to regulate mammalian apoptosis. Second, elucidation of the signal transduction pathways of apoptosis has lead especially to the identification of specific death signaling molecules such as a new family of cysteine proteases, the caspases. Third, it has now become clear that many diseases are characterized by dysregulation of apoptotic programs. Many of these programs involve a family of receptors and their ligands, the death receptor/ligand family. The hope now is to interfere with apoptosis regulation in these systems and to develop new therapeutic concepts.

Modification of damaged replication forks is emerging as a crucial factor for efficient chromosomal duplication and the avoidance of genetic instability. The RecG helicase of Escherichia coli, which is involved in recombination and DNA repair, has been postulated to act on stalled replication forks to promote replication restart via the formation of a four-stranded (Holliday) junction. Here we show that RecG can actively unwind the leading and lagging strand arms of model replication fork structures in vitro. Unwinding is achieved in each case by simultaneous interaction with and translocation along both the leading and lagging strand templates at a fork. Disruption of either of these interactions dramatically inhibits unwinding of the opposing duplex arm. Thus, RecG translocates simultaneously along two DNA strands, one with 5′-3′ and the other with 3′-5′ polarity. The unwinding of both nascent strands at a damaged fork, and their subsequent annealing to form a Holliday junction, may explain the ability of RecG to promote replication restart. Moreover, the preferential binding of partial forks lacking a leading strand suggests that RecG may have the ability to target stalled replication intermediates in vivo in which lagging strand synthesis has continued beyond the leading strand.

Archaeological excavations in northern Madagascar during the first half of the 20th century have revealed the presence of a former prosperous civilisation known as the Rasikajy civilisation. Little is known about the origin of this civilisation and how and when they first arrived in Madagascar. The most striking evidence for the Rasikajy civilisation comes from excavations at a necropolis in Vohemar located along the northeast coast, where more than 600 tombs containing spectacular objects were unearthed in the 1940s (Vernier & Millot 1971). The findings in the tombs included, amongst others, Chinese ceramics, silver and gold jewellery, iron weapons, glassware, bronze mirrors and chlorite-schist objects (ibid.). The latter objects were produced from chlorite schist mined at quarries in northern and eastern Madagascar and there is evidence that jewellery and iron objects were also produced by the Rasikajy from locally available raw material. Chlorite-schist objects have not only been found in coastal sites in Madagascar, but also in the Comores and eastern Africa suggesting an active engagement of the Rasikajy in western Indian Ocean trade. Our re-evaluation of published literature on archaeological sites in northern Madagascar indicates that the majority of Chinese ceramics found in the tombs at Vohemar dates from the 15th and first half of the 16th century with some dating back to the 14th century or earlier. Our comparative analysis of burial objects at Vohemar shows that locally produced chlorite-schist tripod vessels exhibit remarkable resemblances to ancient Chinese bronze ritual tripod vessels. The objects encountered in the tombs and their positions with respect to the body indicate that the Rasikajy practiced burial rites similar to those practised in the past in China. Our re-evaluation of the literature suggests that communities with Chinese roots were present in northeastern Madagascar prior to the arrival of the first Europeans in 1500 and participated in the Indian Ocean trade network. The demise of the Rasikajy civilisation seems to have occurred in the second half of the 16th century when production of chlorite-schist objects ceased. It is still unclear why this occurred.

Most of the policy debate surrounding the actions needed to mitigate and adapt to anthropogenic climate change has been framed by observations of the past 150 years as well as climate and sea-level projections for the twenty-first century. The focus on this 250-year window, however, obscures some of the most profound problems associated with climate change. Here, we argue that the twentieth and twenty-first centuries, a period during which the overwhelming majority of human-caused carbon emissions are likely to occur, need to be placed into a long-term context that includes the past 20 millennia, when the last Ice Age ended and human civilization developed, and the next ten millennia, over which time the projected impacts of anthropogenic climate change will grow and persist. This long-term perspective illustrates that policy decisions made in the next few years to decades will have profound impacts on global climate, ecosystems and human societies — not just for this century, but for the next ten millennia and beyond.

Surface active substances (SAS) in the water column were measured by voltammetry using the electrochemical probe o-nitrophenol (ONP) during EIFEX, a mesoscale open ocean iron enrichment experiment in the Southern Ocean. SAS levels were low throughout the experiment (<0.005 - 0.03 mg/L Triton X-100 equivalents). Initially SAS was extremely low in the photic zone, but as the phytoplankton bloom developed concentrations markedly increased throughout the upper 100 m (~0.02 mg/L Triton X-100 equivalents). Highest concentrations of SAS (>0.02 mg/L Triton X-100 equivalents) were found at the end of the bloom particularly at density discontinuities where organic material may accumulate. Exudates from diatoms appeared to be the major source of SAS during EIFEX, either from direct extracellular release or in the action of being grazed upon by zooplankton.
