Digital Microfluidics (DMF) is a second generation technique, derived from the conventional microfluidics that instead of using continuous liquid fluxes, it uses only individual droplets driven by external electric signals. In this thesis a new DMF control/sensing system for visualization, droplet control (movement, dispensing, merging and splitting) and real time impedance measurement have been developed. The software for the proposed system was implemented in MATLAB with a graphical user interface. An Arduino was used as control board and dedicated circuits for voltage switching and contacts were designed and implemented in printed circuit boards. A high resolution camera was integrated for visualization. In our new approach, the DMF chips are driven by a dual-tone signal where the sum of two independent ac signals (one for droplet operations and the other for impedance sensing) is applied to the electrodes, and afterwards independently evaluated by a lock-in amplifier. With this new approach we were able to choose the appropriated amplitudes and frequencies for the different proposes (actuation and sensing). The measurements made were used to evaluate the real time droplet impedance enabling the knowledge of its position and velocity. This new approach opens new possibilities for impedance sensing and feedback control in DMF devices.

Neste artigo, apresentamos uma discussão sobre as potencialidades da rádio e dos media digitais para uma educação para os media. Esta contextualização, que inclui o contexto português relativamente ao uso da rádio como meio em contexto educativo, pretende criar o pano de fundo para a exposição da metodologia aplicada pelo investigação-ação, concentrados em formas de empoderamento que efetivamente constituam elementos de crescimento curricular dos jovens envolvidos e que possam ser usados na procura de emprego. Em seguida, estaremos em condições de expor e refletir sobre os dados preliminares de aplicação do projeto, que decorre entre 2013 e 2014 em cinco países europeus, centrando-nos em processos de ligação ao digital e de ligação à comunidade envolvente.

Most of today’s systems, especially when related to the Web or to multi-agent systems, are not standalone or independent, but are part of a greater ecosystem, where they need to interact with other entities, react to complex changes in the environment, and act both over its own knowledge base and on the external environment itself. Moreover, these systems are clearly not static, but are constantly evolving due to the execution of self updates or external actions. Whenever actions and updates are possible, the need to ensure properties regarding the outcome of performing such actions emerges. Originally purposed in the context of databases, transactions solve this problem by guaranteeing atomicity, consistency, isolation and durability of a special set of actions. However, current transaction solutions fail to guarantee such properties in dynamic environments, since they cannot combine transaction execution with reactive features, or with the execution of actions over domains that the system does not completely control (thus making rolling back a non-viable proposition). In this thesis, we investigate what and how transaction properties can be ensured over these dynamic environments. To achieve this goal, we provide logic-based solutions, based on Transaction Logic, to precisely model and execute transactions in such environments, and where knowledge bases can be defined by arbitrary logic theories.

Digital microfluidics (DMF) is a field which has emerged in the last decade as a re-liable and versatile tool for sensing applications based on liquid reactions. DMF allows the discrete displacement of droplets, over an array of electrodes, by the application of voltage, and also the dispensing from a reservoir, mixing, merging and splitting fluidic operations. The main drawback of these devices is due to the need of high driving volt-ages for droplet operations. In this work, alternative dielectric layers combinations were studied aiming the reduction of these driving voltages. DMF chips were designed, pro-duced and optimized according to the theory of electrowetting-on-dielectric, adopting different combinations of parylene-C and tantalum pentoxide (Ta2O5) as dielectric ma-terials, and Teflon as hydrophobic layer. With both devices’ configurations, i.e., Parylene as single dielectric, and multilayer chips combining Parylene and Ta2O5, it was possible to perform all the fluidic opera-tions in the microliter down to hundreds of nanoliters range. Multilayer chips presented significant reduction on driving voltages for droplet op-erations in silicone oil filler medium: from 70 V (parylene only) down to 30 V (parylene/Ta2O5) for dispensing; and from 50 V (parylene only) down to 15 V (parylene/Ta2O5) for movement. Peroxidase colorimetric reactions were successfully performed as proof-of-concept, using multilayer configuration devices.

This thesis is one of the first reports of digital microfluidics on paper and the first in which the chip’s circuit was screen printed unto the paper. The use of the screen printing technique, being a low cost and fast method for electrodes deposition, makes the all chip processing much more aligned with the low cost choice of paper as a substrate. Functioning chips were developed that were capable of working at as low as 50 V, performing all the digital microfluidics operations: movement, dispensing, merging and splitting of the droplets. Silver ink electrodes were screen printed unto paper substrates, covered by Parylene-C (through vapor deposition) as dielectric and Teflon AF 1600 (through spin coating) as hydrophobic layer. The morphology of different paper substrates, silver inks (with different annealing conditions) and Parylene deposition conditions were studied by optical microscopy, AFM, SEM and 3D profilometry. Resolution tests for the printing process and electrical characterization of the silver electrodes were also made. As a showcase of the applications potential of these chips as a biosensing device, a colorimetric peroxidase detection test was successfully done on chip, using 200 nL to 350 nL droplets dispensed from 1 μL drops.

As the complexity of markets and the dynamicity of systems evolve, the need for interoperable systems capable of strengthening enterprise communication effectiveness increases. This is particularly significant when it comes to collaborative enterprise networks, like manufacturing supply chains, where several companies work, communicate, and depend on each other, in order to achieve a specific goal. Once interoperability is achieved, that is once all network parties are able to communicate with and understand each other, organisations are able to exchange information along a stable environment that follows agreed laws. However, as markets adapt to new requirements and demands, an evolutionary behaviour is triggered giving space to interoperability problems, thus disrupting the sustainability of interoperability and raising the need to develop monitoring activities capable of detecting and preventing unexpected behaviour. This work seeks to contribute to the development of monitoring techniques for interoperable SOA-based enterprise networks. It focuses on the automatic detection of harmonisation breaking events during real-time communications, and strives to develop and propose a methodological approach to handle these disruptions with minimal or no human intervention, hence providing existing service-based networks with the ability to detect and promptly react to interoperability issues.

INTRODUCTION: This study analyzed the occurrence and the contamination of triatomines by trypanosomatids in Orbignya speciosa (babassu) specimens in the State of Rondônia, Brazil, in two different environments (pasture and woods). METHODS: Capture of triatomines on babassus and microscopic search for trypanosomatids in their digestive tube were carried out. RESULTS: Four hundred ninety-four (494) specimens were captured (Rhodnius prolixus and R.robustus), of which 35.6% of the triatomines were positive for trypanosomatids. CONCLUSIONS: The high index of natural infection along with the abundance of triatomines points out to the necessity to create an epidemiological surveillance system to monitor vector-borne transmission and deepen the studies on the ecology of such vectors in the Amazon.

INTRODUCTION: Trypanosoma cruzi-infected specimens of Triatoma costalimai have been detected in domiciliary units of Central Brazil, thereby maintaining the potential risk of vectorial transmission of Chagas disease. The aim of this study was to determine the occurrence and natural infection of T. costalimai in different environments (gallery forest, dry forest and peridomicile) and climatic seasons (wet and dry), in the municipality of Mambaí, State of Goiás, Brazil. METHODS: Triatomines were captured in October 2010 and in June 2011, employing two different methods (manual capture and mouse-baited adhesive traps). The insects were later separated by sex and nymphal stage, counted and examined parasitologically by abdominal compression and microscopic analysis of feces. RESULTS: Triatoma costalimai was found in three environments and in the two seasons studied. Overall, capture success of 900 traps and 60 blocks of rocks inspected was 5.8% and 11.7%, respectively. The occurrence of T. costalimai was higher among rocks in the peridomicile, where 97% of the 131 specimens were captured. The proportion of nymphs (98%) was much higher than that of adults, which were only detected in peridomicile. Most (95%) insects were captured during the wet season, with predominance of early-stage nymphs. None of the 43 specimens examined were infected by trypanosomatids. CONCLUSIONS: The results indicate a greater occurrence of T. costalimai in peridomiciliary environments and during the wet season in Mambaí, Goiás, highlighting the synanthropic behavior of this triatomine species in one area of the Brazilian savanna and the importance of entomological surveillance.

Digital Businesses have become a major driver for economic growth and have seen an explosion of new startups. At the same time, it also includes mature enterprises that have become global giants in a relatively short period of time. Digital Businesses have unique characteristics that make the running and management of a Digital Business much different from traditional offline businesses. Digital businesses respond to online users who are highly interconnected and networked. This enables a rapid flow of word of mouth, at a pace far greater than ever envisioned when dealing with traditional products and services. The relatively low cost of incremental user addition has led to a variety of innovation in pricing of digital products, including various forms of free and freemium pricing models. This thesis explores the unique characteristics and complexities of Digital Businesses and its implications on the design of Digital Business Models and Revenue Models. The thesis proposes an Agent Based Modeling Framework that can be used to develop Simulation Models that simulate the complex dynamics of Digital Businesses and the user interactions between users of a digital product. Such Simulation models can be used for a variety of purposes such as simple forecasting, analysing the impact of market disturbances, analysing the impact of changes in pricing models and optimising the pricing for maximum revenue generation or a balance between growth in usage and revenue generation. These models can be developed for a mature enterprise with a large historical record of user growth rate as well as for early stage enterprises without much historical data. Through three case studies, the thesis demonstrates the applicability of the Framework and its potential applications.

O presente relatório é o resultado de uma reflexão que tem como base um estágio de três meses na redacção do jornal Expresso, nomeadamente na secção do Diário e do Online. A partir de uma análise às publicações dedicadas à cultura, pretende-se explorar as potencialidades digitais como contributo para uma narrativa dinâmica, interactiva e imersiva. Procura-se compreender se, de facto, os elementos multimédia ganham mais possibilidades criativas quando aplicados no jornalismo cultural. Para esse efeito, este relatório encontra-se dividido em quatro partes consoante o enquadramento dos temas analisados. Primeiramente estabelece-se um enquadramento teórico que conceptualiza as duas temáticas centrais deste relatório: jornalismo cultural e jornalismo digital. Numa segunda fase é descrita a experiência de um estágio no Expresso, bem como se contextualizam os seus anos de estórias. De seguida, surgem as questões de investigação devidamente aplicadas no âmbito de análise. Através de uma reflexão qualitativa às publicações do Expresso Online e Diário dedicadas a temáticas culturais e publicadas durante o período de um mês, conclui-se que o jornalismo cultural pode possibilitar ao leitor uma experiência completa. As potencialidades digitais servem como contributo para o despertar de sensações, para uma narrativa abrangente e reflexiva. Há falta de activismo jornalístico ou, antes, de exploração das capacidades da área que vão muito para além do carácter informativo

O intuito desta dissertação é investigar em que medida a Teoria Moderna do Portfólio pode ser aplicada à selecção de um portfólio de canais ou de tácticas de Marketing Digital, de modo a obter o melhor compromisso entre o retorno esperado do investimento e o respectivo risco. Anteriores aplicações da teoria do portfólio ao Marketing, enquanto meio de racionalizar a selecção do portfólio, concentraram-se no uso deste modelo em decisões relacionadas com portfólios de produtos, de clientes e segmentos de clientes, de formatos de retalho e de promoções de preço. Não se encontraram evidências de que a abordagem proposta neste texto tenha já sido tentada. A evolução tecnológica nas últimas duas décadas trouxe a adopção generalizada de smartphones e tablets e também a revolução da internet, originando a proliferação do e-mail, dos blogs e dos sites de redes sociais. Devido à infinidade de meios para chegar aos consumidores, os marketers enfrentam actualmente grandes desafios no planeamento e na definição das suas estratégias de Marketing. Em resposta, consultoras detêm ferramentas para a análise de portfólios de canais de Marketing que permitem às empresas e aos marketers, em particular, melhorar o desempenho da sua actividade e maximizar o rendimento da sua despesa de Marketing. Contudo, além de dispendiosas e estritamente analíticas, estas ferramentas são muito sofisticadas, baseando-se em algoritmos e raciocínios complexos que nem sempre são evidentes para os marketers, o que dificulta a sua compreensão e consequentemente a sua adopção. Sem uma ferramenta de análise, é frequente os marketers fundamentarem as suas decisões nos resultados das campanhas online anteriores, registados pelas Web Analytics, e na sua experiência profissional. Assim, crê-se que a Teoria Moderna do Portfólio, baseando-se em conceitos estatísticos simples, possa representar para os marketers uma ferramenta útil no apoio à selecção do conjunto de canais ou de tácticas de Marketing Digital que melhor se adequa aos interesses e aos objectivos estratégicos das organizações. A Teoria Moderna do Portfólio tem como principais vantagens a análise e selecção de portfólios com base na sua combinação de retorno esperado e risco de investimento (retorno ajustado ao risco), por oposição a considerar exclusivamente o retorno esperado, e a simplicidade estatística do modelo, por oposição aos softwares das consultoras. O presente estudo fundamentou-se na literatura existente acerca da Teoria Moderna do Portfólio, concebida em 1952 por Harry Markowitz, bem como no conceito de Sharpe Ratio, por William Sharpe, tendo-se revisto também conceitos genéricos de Marketing e Marketing Digital. Dado que a medição do retorno financeiro do investimento em Marketing – central ao estudo – não é tão linear quanto a medição dos retornos de activos financeiros, revelou-se necessário introduzir algumas alterações à teoria do portfólio original. Estas alterações tomaram em consideração as principais conclusões dos estudos anteriores sobre a aplicação do modelo a outras áreas. Para desenvolver o modelo proposto, utilizou-se o Microsoft Excel. Seguidamente, procedeu-se à sua prova de conceito através de uma abordagem empírica, utilizando dados de uma empresa portuguesa de Marketing Digital. Concluiu-se que é possível aplicar a Teoria Moderna do Portfólio à selecção de canais ou tácticas de Marketing Digital de uma empresa, com as devidas adaptações, para determinar as parcelas do orçamento de comunicação de uma organização a alocar a cada tipo de canal ou táctica.

Esta dissertação pretende analisar as práticas de dois artistas contemporâneos, Ryan Trecartin e Jacolby Satterwhite, nomeadamente a nível do modo como trabalham a intersecção do vídeo digital e da performance como forma de manifestação de corporalizações queer através do corpo masculino. Mais concretamente, interessa-nos focar processos de “queerificação” e de produção de corpos que emergem intrinsecamente pelo médium digital, e que exploram as possibilidades transformativas do médium tanto a nível de representação, criação e mutação de formas, como a nível comunicacional e de possibilidades de subjectivação e corporalizações múltiplas. Para tal, partimos dos dois campos teóricos que se mostraram mais pertinentes relativamente aos processos artísticos em análise, nomeadamente: a teoria queer pelo modo como definiu o carácter performativo das normas de género e as categorizações identitárias sexuais como suporte da heteronormatividade, identificando estratégias para a sua re-significação; e o pensamento sobre a relação do corpo com o médium digital, pelo modo como delineou processos de corporalização através da tecnologia digital. A análise dos processos artísticos de Ryan Trecartin e Jacolby Satterwhite demonstra como a arte contemporânea é um território fértil não apenas para a exposição de matrizes normativas de opressão, mas também para a criação de mundos radicalmente queer em que, fazendo uso do potencial transformativo do médium digital, se manifestam realidades e corpos onde a normatividade é totalmente implodida.
