In a time when Technology Supported Learning Systems are being widely used, there is a lack of tools that allows their development in an automatic or semi-automatic way. Technology Supported Learning Systems require an appropriate Domain Module, ie. the pedagogical representation of the domain to be mastered, in order to be effective. However, content authoring is a time and effort consuming task, therefore, efforts in automatising the Domain Module acquisition are necessary.Traditionally, textbooks have been used as the main mechanism to maintain and transmit the knowledge of a certain subject or domain. Textbooks have been authored by domain experts who have organised the contents in a means that facilitate understanding and learning, considering pedagogical issues.Given that textbooks are appropriate sources of information, they can be used to facilitate the development of the Domain Module allowing the identification of the topics to be mastered and the pedagogical relationships among them, as well as the extraction of Learning Objects, ie. meaningful fragments of the textbook with educational purpose.Consequently, in this work DOM-Sortze, a framework for the semi-automatic construction of Domain Modules from electronic textbooks, has been developed. DOM-Sortze uses NLP techniques, heuristic reasoning and ontologies to fulfill its work. DOM-Sortze has been designed and developed with the aim of automatising the development of the Domain Module, regardless of the subject, promoting the knowledge reuse and facilitating the collaboration of the users during the process.

Coral reefs exist in warm, clear, and relatively shallow marine waters worldwide. These complex assemblages of marine organisms are unique, in that they support highly diverse, luxuriant, and essentially self-sustaining ecosystems in otherwise nutrient-poor and unproductive waters. Coral reefs are highly valued for their great beauty and for their contribution to marine productivity. Coral reefs are favorite destinations for recreational diving and snorkeling, as well as commercial and recreational fishing activities. The Florida Keys reef tract draws an estimated 2 million tourists each year, contributing nearly $800 million to the economy. However, these reef systems represent a very delicate ecological balance, and can be easily damaged and degraded by direct or indirect human contact. Indirect impacts from human activity occurs in a number of different forms, including runoff of sediments, nutrients, and other pollutants associated with forest harvesting, agricultural practices, urbanization, coastal construction, and industrial activities. Direct impacts occur through overfishing and other destructive fishing practices, mining of corals, and overuse of many reef areas, including damage from souvenir collection, boat anchoring, and diver contact. In order to protect and manage coral reefs within U.S. territorial waters, the National Oceanic and Atmospheric Administration (NOAA) of the U.S. Department of Commerce has been directed to establish and maintain a system of national marine sanctuaries and reserves, and to monitor the condition of corals and other marine organisms within these areas. To help carry out this mandate the NOAA Coastal Services Center convened a workshop in September, 1996, to identify current and emerging sensor technologies, including satellite, airborne, and underwater systems with potential application for detecting and monitoring corals. For reef systems occurring within depths of 10 meters or less (Figure 1), mapping location and monitoring the condition of corals can be accomplished through use of aerial photography combined with diver surveys. However, corals can exist in depths greater than 90 meters (Figure 2), well below the limits of traditional optical imaging systems such as aerial or surface photography or videography. Although specialized scuba systems can allow diving to these depths, the thousands of square kilometers included within these management areas make diver surveys for deeper coral monitoring impractical. For these reasons, NOAA is investigating satellite and airborne sensor systems, as well as technologies which can facilitate the location, mapping, and monitoring of corals in deeper waters. The following systems were discussed as having potential application for detecting, mapping, and assessing the condition of corals. However, no single system is capable of accomplishing all three of these objectives under all depths and conditions within which corals exist. Systems were evaluated for their capabilities, including advantages and disadvantages, relative to their ability to detect and discriminate corals under a variety of conditions. The study is focused on structural aspects of interaction between silencing suppressor p19 and CUG-repeating small RNAs. The work involves crystal structure determination of a protein-unbound RNA form and RNA fragments of various lengths (19, 20, 21 nucleotides) complexed with p19-suppressor. Results prove the ability of silencing suppressor p19 to bind CUG-repeating small RNAs, as well as reveal features of U•U mismatches flanked by Watson-Crick C•G base pairs in p19-bound and p19-unbound states. In addition, structural data reveal a p19 specific site for anchoring extra nucleotides in small RNAs. In general, the study extends our knowledge about the mechanism of small RNA recognition by silencing suppressor p19.

Since 2001, a research group in the Institute of Mechanics, Chinese Academy of Sciences, has been devoted to the research of essential mechanics issues for submerged floating tunnel (SFT). In addition to the structural design of the SFT prototype in Qiandao Lake, the relevant researches cover a number of topics. This paper briefly describes the research procedure and results, including dynamic response of SFT due to surface wave, vortex-induced vibration of anchoring system, structural analysis of curved SFT, temperature effects of curved SFT, structural dynamic response due to accidental load, and effects of structural parameters (buoyancy-weight ratio, tunnel length,tether stiffness,etc.) on dynamic response.

[EN] One universal feature of human languages is the division between grammatical functors and content words. From a learnability point of view, functors might provide entry points or anchors into the syntactic structure of utterances due to their high frequency. Despite its potentially universal scope, this hypothesis has not yet been tested on typologically different languages and on populations of different ages. Here we report a corpus study and an artificial grammar learning experiment testing the anchoring hypothesis in Basque, Japanese, French, and Italian adults. We show that adults are sensitive to the distribution of functors in their native language and use them when learning new linguistic material. However, compared to infants’ performance on a similar task, adults exhibit a slightly different behavior, matching the frequency distributions of their native language more closely than infants do. This finding bears on the issue of the continuity of language learning mechanism.

The work presented in this thesis revolves around erasure correction coding, as applied to distributed data storage and real-time streaming communications.

First, we examine the problem of allocating a given storage budget over a set of nodes for maximum reliability. The objective is to find an allocation of the budget that maximizes the probability of successful recovery by a data collector accessing a random subset of the nodes. This optimization problem is challenging in general because of its combinatorial nature, despite its simple formulation. We study several variations of the problem, assuming different allocation models and access models, and determine the optimal allocation and the optimal symmetric allocation (in which all nonempty nodes store the same amount of data) for a variety of cases. Although the optimal allocation can have nonintuitive structure and can be difficult to find in general, our results suggest that, as a simple heuristic, reliable storage can be achieved by spreading the budget maximally over all nodes when the budget is large, and spreading it minimally over a few nodes when it is small. Coding would therefore be beneficial in the former case, while uncoded replication would suffice in the latter case.

Second, we study how distributed storage allocations affect the recovery delay in a mobile setting. Specifically, two recovery delay optimization problems are considered for a network of mobile storage nodes: the maximization of the probability of successful recovery by a given deadline, and the minimization of the expected recovery delay. We show that the first problem is closely related to the earlier allocation problem, and solve the second problem completely for the case of symmetric allocations. It turns out that the optimal allocations for the two problems can be quite different. In a simulation study, we evaluated the performance of a simple data dissemination and storage protocol for mobile delay-tolerant networks, and observed that the choice of allocation can have a significant impact on the recovery delay under a variety of scenarios.

Third, we consider a real-time streaming system where messages created at regular time intervals at a source are encoded for transmission to a receiver over a packet erasure link; the receiver must subsequently decode each message within a given delay from its creation time. For erasure models containing a limited number of erasures per coding window, per sliding window, and containing erasure bursts whose maximum length is sufficiently short or long, we show that a time-invariant intrasession code asymptotically achieves the maximum message size among all codes that allow decoding under all admissible erasure patterns. For the bursty erasure model, we also show that diagonally interleaved codes derived from specific systematic block codes are asymptotically optimal over all codes in certain cases. We also study an i.i.d. erasure model in which each transmitted packet is erased independently with the same probability; the objective is to maximize the decoding probability for a given message size. We derive an upper bound on the decoding probability for any time-invariant code, and show that the gap between this bound and the performance of a family of time-invariant intrasession codes is small when the message size and packet erasure probability are small. In a simulation study, these codes performed well against a family of random time-invariant convolutional codes under a number of scenarios.

Finally, we consider the joint problems of routing and caching for named data networking. We propose a backpressure-based policy that employs virtual interest packets to make routing and caching decisions. In a packet-level simulation, the proposed policy outperformed a basic protocol that combines shortest-path routing with least-recently-used (LRU) cache replacement.

We have used the technique of non-redundant masking at the Palomar 200-inch telescope and radio VLBI imaging software to make optical aperture synthesis maps of two binary stars, β Corona Borealis and σ Herculis. The dynamic range of the map of β CrB, a binary star with a separation of 230 milliarcseconds is 50:1. For σ Her, we find a separation of 70 milliarcseconds and the dynamic range of our image is 30:1. These demonstrate the potential of the non-redundant masking technique for diffraction-limited imaging of astronomical objects with high dynamic range.

We find that the optimal integration time for measuring the closure phase is longer than that for measuring the fringe amplitude. There is not a close relationship between amplitude errors and phase errors, as is found in radio interferometry. Amplitude self calibration is less effective at optical wavelengths than at radio wavelengths. Primary beam sensitivity correction made in radio aperture synthesis is not necessary in optical aperture synthesis.

The effects of atmospheric disturbances on optical aperture synthesis have been studied by Monte Carlo simulations based on the Kolmogorov theory of refractive-index fluctuations. For the non-redundant masking with τ_c-sized apertures, the simulated fringe amplitude gives an upper bound of the observed fringe amplitude. A smooth transition is seen from the non-redundant masking regime to the speckle regime with increasing aperture size. The fractional reduction of the fringe amplitude according to the bandwidth is nearly independent of the aperture size. The limiting magnitude of optical aperture synthesis with τ_c-sized apertures and that with apertures larger than τ_c are derived.

Monte Carlo simulations are also made to study the sensitivity and resolution of the bispectral analysis of speckle interferometry. We present the bispectral modulation transfer function and its signal-to-noise ratio at high light levels. The results confirm the validity of the heuristic interferometric view of image-forming process in the mid-spatial-frequency range. The signal-to- noise ratio of the bispectrum at arbitrary light levels is derived in the mid-spatial-frequency range.

The non-redundant masking technique is suitable for imaging bright objects with high resolution and high dynamic range, while the faintest limit will be better pursued by speckle imaging.

As a necessary condition for the validity of the present value model, the price-dividend ratio must be stationary. However, significant market episodes seem to provide evidence of prices significantly drifting apart from dividends while other episodes show prices anchoring back to dividends. This paper investigates the stationarity of this ratio in the context of a Markov- switching model à la Hamilton (1989) where an asymmetric speed of adjustment towards a unique attractor is introduced. A three-regime model displays the best regime identification and reveals that the first part of the 90’s boom (1985-1995) and the post-war period are characterized by a stationary state featuring a slow reverting process to a relatively high attractor. Interestingly, the latter part of the 90’s boom (1996-2000), characterized by a growing price-dividend ratio, is entirely attributed to a stationary regime featuring a highly reverting process to the attractor. Finally, the post-Lehman Brothers episode of the subprime crisis can be classified into a temporary nonstationary regime.

This thesis consists of three parts. Chapter 2 deals with the dynamic buckling behavior of steel braces under cyclic axial end displacement. Braces under such a loading condition belong to a class of "acceleration magnifying" structural components, in which a small motion at the loading points can cause large internal acceleration and inertia. This member-level inertia is frequently ignored in current studies of braces and braced structures. This chapter shows that, under certain conditions, the inclusion of the member-level inertia can lead to brace behavior fundamentally different from that predicted by the quasi-static method. This result is to have significance in the correct use of the quasi-static, pseudo-dynamic and static condensation methods in the simulation of braces or braced structures under dynamic loading. The strain magnitude and distribution in the braces are also studied in this chapter.

Chapter 3 examines the effect of column uplift on the earthquake response of braced steel frames and explores the feasibility of flexible column-base anchoring. It is found that fully anchored braced-bay columns can induce extremely large internal forces in the braced-bay members and their connections, thus increasing the risk of failures observed in recent earthquakes. Flexible braced-bay column anchoring can significantly reduce the braced bay member force, but at the same time also introduces large story drift and column uplift. The pounding of an uplifting column with its support can result in very high compressive axial force.

Chapter 4 conducts a comparative study on the effectiveness of a proposed non-buckling bracing system and several conventional bracing systems. The non-buckling bracing system eliminates buckling and thus can be composed of small individual braces distributed widely in a structure to reduce bracing force concentration and increase redundancy. The elimination of buckling results in a significantly more effective bracing system compared with the conventional bracing systems. Among the conventional bracing systems, bracing configurations and end conditions for the bracing members affect the effectiveness.

The studies in Chapter 3 and Chapter 4 also indicate that code-designed conventionally braced steel frames can experience unacceptably severe response under the strong ground motions recorded during the recent Northridge and Kobe earthquakes.

Considerando-se que a família vivencia e partilha junto com o doente todos os sentimentos, emoções e angústias que envolve o diagnóstico e o tratamento do câncer, o presente estudo teve como objetivos descrever as dimensões das representações sociais acerca do câncer para familiares do cliente oncológico em tratamento quimioterápico ambulatorial em uma unidade de referência para o seu tratamento; analisar a representação social do câncer elaborada por familiares do cliente oncológico em tratamento quimioterápico ambulatorial; e discutir as contribuições do enfermeiro junto à família do cliente oncológico em tratamento quimioterápico ambulatorial a partir da construção representacional do câncer para os sujeitos do estudo. De caráter qualitativo, o caminho metodológico foi construído com base na Teoria das Representações Sociais. Os sujeitos foram 30 familiares que estavam acompanhando o doente durante o tratamento quimioterápico. Os dados foram coletados a partir da realização de entrevistas semi-estruturadas e analisados através da análise de conteúdo de Bardin (1979), sistematizada por Oliveira (2008), com o auxílio do software QRS Nvivo 2.0. Da análise dos dados emergiram seis categorias que compõem o campo representacional e se expressa através das dimensões representacionais concretizadas nas seguintes categorias: sentimentos compartilhados por familiares de clientes oncológicos em tratamento quimioterápico, que mostra que, ao se depararem com a doença e sua dura realidade, os familiares são acometidos por diversos tipos de sentimentos; imagens, metáforas e conceitos no existir da família que enfrenta a doença, onde os familiares revelaram que o câncer é percebido, entre outras coisas, como um monstro que invade a vida das pessoas e dela passa a tomar conta e a dominá-la; preconceitos e estigmas na vivência do câncer, que revelou que ainda hoje existem representações e estigmas presentes na sociedade e em suas construções culturais acerca do câncer; diferentes práticas desenvolvidas no contexto da doença e do processo de adoecimento pelo câncer, que evidenciou as diferentes práticas presentes no discurso dos sujeitos, quais sejam, a de religiosidade no contexto do câncer, a de enfrentamento da doença, a de comunicação-ocultamento e de atitudes da família ao estar no mundo frente ao câncer; o processo de ancoragem e o conhecimento adquirido após a experiência do câncer, onde surgiram os conhecimentos que os sujeitos adquiriram acerca do câncer e alguns elementos do processo de ancoragem do câncer; as vivências do enfermeiro que trabalha em oncologia e suas contribuições junto à família que alerta os enfermeiros para a necessidade de intervenções efetivas direcionadas à assistência integral do indivíduo, levando em consideração a importância da família. Conclui-se que ao se descobrir acompanhando um familiar que tem câncer, a família passa a viver um outro mundo, no qual a possibilidade de morte se mostra de forma inevitável e iminente. Diante disso, a família passa a valorizar não apenas o cuidado dispensado ao doente, mas também anseia por uma atenção profissional que contemple seu existir e seu modo de viver.

Nesta dissertação, duas resinas reticuladas comerciais (denominadas, AmberliteGT73 e AmberliteIRC748) foram empregadas para suportar partículas de prata. Os grupos tiol da GT73 e ácido iminodiacético da IRC748 presentes nessas resinas foram empregados para a fixação de íons Ag+ a partir de solução aquosa. Posteriormente, os íons Ag+ foram reduzidos pelo emprego de três redutores diferentes em pH alcalino, denominados hidrazina, hidroxilamina e formaldeído (pH  12). A morfologia e a impregnação de prata dos materiais binários assim obtidos foram avaliadas por meio de microscópio eletrônico de varredura equipado com detector de elétrons retro-espalhados (SEM-BSE). O detector de espectrometria de energia dispersiva de raios-X (EDAX) acoplado ao SEM permitiu a observação de partículas de prata. Os espectros de raios-X revelaram a presença do metal nas superfícies interna e externa das microesferas dos compósitos. A quantidade de prata incorporada foi determinada pelo método titulométrico, empregando solução padrão de tiocianato de potássio. As características antibactericidas dos compósitos foram avaliadas em colunas contendo pérolas de resina por onde foram percoladas suspensões da bactéria Escherichia coli auxotrópica AB1157 (tipo selvagem) nas concentrações de 103 a 107 células/mL. A avaliação biocida mostrou que estes materiais foram completamente bactericidas, sendo efetivos na eliminação da bactéria em poucos minutos. Esta ação biocida foi atribuída à combinação da atuação da prata e dos grupos funcionais das resinas

The proper targeting of membrane proteins is essential to the viability of all cells. Tail-anchored (TA) proteins, defined as having a single transmembrane helix at their C-terminus, are post-translationally targeted to the endoplasmic reticulum (ER) membrane by the GET pathway (Guided Entry of TA proteins). In the yeast pathway, the handover of TA substrates is mediated by the heterotetrameric Get4/Get5 (Get4/5) complex, which tethers the co-chaperone Sgt2 to the central targeting factor, the Get3 ATPase. Although binding of Get4/5 to Get3 is critical for efficient TA targeting, the mechanisms by which Get4 regulates Get3 are unknown. To understand the molecular basis of Get4 function, we used a combination of structural biology, biochemistry, and cell biology. Get4/5 binds across the Get3 dimer interface, in an orientation only compatible with a closed Get3, providing insight into the role of nucleotide in complex formation. Additionally, this structure reveals two functionally distinct binding interfaces for anchoring and ATPase regulation, and loss of the regulatory interface leads to strong defects in vitro and in vivo. Additional crystal structures of the Get3-Get4/5 complex give rise to an alternate conformation, which represents an initial binding interaction mediated by electrostatics that facilitates the rate of subsequent inhibited complex formation. This interface is supported by an in-depth kinetic analysis of the Get3-Get4/5 interaction confirming the two-step complex formation. These results allow us to generate a refined model for Get4/5 function in TA targeting.

Neste trabalho, é proposta uma nova família de métodos a ser aplicada à otimização de problemas multimodais. Nestas técnicas, primeiramente são geradas soluções iniciais com o intuito de explorar o espaço de busca. Em seguida, com a finalidade de encontrar mais de um ótimo, estas soluções são agrupadas em subespaços utilizando um algoritmo de clusterização nebulosa. Finalmente, são feitas buscas locais através de métodos determinísticos de otimização dentro de cada subespaço gerado na fase anterior com a finalidade de encontrar-se o ótimo local. A família de métodos é formada por seis variantes, combinando três esquemas de inicialização das soluções na primeira fase e dois algoritmos de busca local na terceira. A fim de que esta nova família de métodos possa ser avaliada, seus constituintes são comparados com outras metodologias utilizando problemas da literatura e os resultados alcançados são promissores.

Nesta dissertação, foram preparados materiais poliméricos com atividade bactericida a partir de copolímeros de estireno (Sty) e divinilbenzeno (DVB) e de celulose bacteriana. Três copolímeros à base de Sty e DVB foram sintetizados através da técnica de polimerização em suspensão aquosa. Os copolímeros foram preparados com diferentes estruturas porosas, por meio da variação da composição do sistema diluente. Um copolímero comercial macrorreticulado, Amberlite XAD4, de elevada área superficial, também foi usado neste estudo com o objetivo de comparar sua estrutura polimérica e seu desempenho com o dos copolímeros sintetizados. Os copolímeros de Sty-DVB foram caracterizados por meio da densidade aparente, área específica, diâmetro médio de poros, volume de poros, microscopias ótica e eletrônica de varredura, grau de inchamento, espectrometria de infravermelho com transformada de Fourier (FTIR), análise termogravimétrica e análise elementar. Os copolímeros de Sty-DVB foram modificados quimicamente através da reação de nitração, seguida da reação de redução do grupo nitro a amino e posterior reação de diazotação e hidrólise do grupo sal de diazônio. Os copolímeros modificados contendo os grupos NH2 e OH foram utilizados como suporte para a ancoragem das nanopartículas de prata. As partículas de prata foram obtidas pela redução dos íons Ag+ empregando-se cloridrato de hidroxilamina como redutor e PVP como protetor de colóide. O teor de prata impregnada foi determinado pelo método volumétrico. A celulose bacteriana (BC) foi empregada como uma matriz para impregnação de nanopartículas de prata. Membranas de BC foram impregnadas com solução aquosa de AgNO3, a seguir foi realizada a redução dos íons Ag+ com três diferentes agentes redutores (hidrazina, hidroxilamina e ácido ascórbico) empregando ou não protetor de colóide (PVP ou gelatina). As membranas de BC/Ag0 obtidas foram caracterizadas por difração de raios-X, análise termogravimétrica e microscopia eletrônica de varredura. A capacidade bactericida dos copolímeros de Sty-DVB, dos copolímeros funcionalizados e impregnados com prata e das membranas de BC/Ag0 foi avaliada através do método da contagem em placa contra suspensões de Escherichia coli, em diferentes concentrações (103 a 107 células/mL). Foi observado que os copolímeros de origem e os copolímeros modificados contendo os grupos NO2 e NH2 não apresentaram atividade bactericida contra suspensões de E. coli em nenhuma concentração. Por outro lado, os copolímeros modificados contendo o grupo OH apresentaram alguma atividade bactericida, embora abaixo do esperado devido à baixa incorporação de hidroxilas fenólicas. Os copolímeros modificados impregnados com Ag apresentaram maior ação bactericida do que os copolímeros modificados, o que comprova que a ação bactericida é devida às partículas de prata ancoradas nos copolímeros. Os copolímeros modificados contendo o grupo OH impregnados com Ag mesmo possuindo menores teores de prata apresentaram maior ação bactericida do que os copolímeros modificados contendo o grupo NH2 impregnados com Ag. A maior ação bactericida pode ser atribuída à combinação da atividade bactericida das partículas de prata com a atividade dos grupos hidroxilas fenólicas mesmo que estes grupos estejam pouco incorporados nos copolímeros. Os compósitos de BC/Ag0 exibiram atividade bactericida contra E. coli que foi atribuída à ação das nanoparticulas de prata obtidas quando foi empregada a hidroxilamina como agente redutor e a gelatina como protetor de colóide
