BACKGROUND: This study was designed to investigate, for the first time, the short-term molecular evolution of the HIV-2 C2, V3 and C3 envelope regions and its association with the immune response. Clonal sequences of the env C2V3C3 region were obtained from a cohort of eighteen HIV-2 chronically infected patients followed prospectively during 2-4 years. Genetic diversity, divergence, positive selection and glycosylation in the C2V3C3 region were analysed as a function of the number of CD4+ T cells and the anti-C2V3C3 IgG and IgA antibody reactivity RESULTS: The mean intra-host nucleotide diversity was 2.1% (SD, 1.1%), increasing along the course of infection in most patients. Diversity at the amino acid level was significantly lower for the V3 region and higher for the C2 region. The average divergence rate was 0.014 substitutions/site/year, which is similar to that reported in chronic HIV-1 infection. The number and position of positively selected sites was highly variable, except for codons 267 and 270 in C2 that were under strong and persistent positive selection in most patients. N-glycosylation sites located in C2 and V3 were conserved in all patients along the course of infection. Intra-host variation of C2V3C3-specific IgG response over time was inversely associated with the variation in nucleotide and amino acid diversity of the C2V3C3 region. Variation of the C2V3C3-specific IgA response was inversely associated with variation in the number of N-glycosylation sites. CONCLUSION: The evolutionary dynamics of HIV-2 envelope during chronic aviremic infection is similar to HIV-1 implying that the virus should be actively replicating in cellular compartments. Convergent evolution of N-glycosylation in C2 and V3, and the limited diversification of V3, indicates that there are important functional constraints to the potential diversity of the HIV-2 envelope. C2V3C3-specific IgG antibodies are effective at reducing viral population size limiting the number of virus escape mutants. The C3 region seems to be a target for IgA antibodies and increasing N-linked glycosylation may prevent HIV-2 envelope recognition by these antibodies. Our results provide new insights into the biology of HIV-2 and its relation with the human host and may have important implications for vaccine design.

The aim of this work was to study the self-assembly process of C3-symmetric molecules. To accomplish this objective 1,3,5 – benzentricarboxamides (BTA) derivatives were obtained. Five C3-symmetric molecules were synthesized in moderate to good yields (39-72%) using azo-benzene, aniline, benzylamine, tryptophan and tyrosine. The aggregation behavior of the BTA derivatives was probed with 1H-NMR spectroscopy, 1H-1H 2D Nuclear Overhauser Effect Spectroscopy (NOESY) and Diffusion Ordered Spectroscopy (DOSY). These experiments allowed to study the influence of H-bonding groups, aromatic rings, unsaturated bonds and the overall geometry in the molecular self-assembly associated with the different structural patterns present on these molecules. The stacking and large molecule behavior where observed in BTA 1, aniline derivative, BTA 4, tyrosine derivative or BTA 5, tryptophan derivative, with several of those discussed functional groups such as unsaturated bonds and H-bonding groups. BTA 5 was used in a few preliminary interaction studies with glucose and ammonium chloride showing interaction with the ammonium ion.

Las Gramíneas se caracterizan por la presencia de diferentes mecanismos fotosínteticos. Casi la mitad de sus miembros -unos 5000 - posee la vía fotosintética C4, siendo los restantes C3. Además, dentro de las especies que siguen la vía fotosíntetica C4, pueden reconocerse 3 variantes bioquímicas diferentes (NADP-ME y PCK). En los trópicos y subtrópicos la vía C4 confiere a las Gramíneas ventajes adaptativas que son atribuibles al mecanismo bioquímico de concentración de CO2 que se produce en las hojas. La taxonomía y biología de los pastos indican que la diversificación que experimentaron los mecanismos fotosintéticos fue crucial en la historia evolutiva y radiación adaptativa de la familia; asimismo, puede inferirse que tal diversificación se desencadenó en respuesta a cambios naturales en el pasado, en las mismas variables climáticas que están siendo antropogénicamente alteradas en la actualidad: CO, temperatura y precipitaciones. En el marco aludido, surge la pregunta de cuáles serán las especies capaces de adaptarse -y prosperar- cuando la concentración de CO2 se duplique en la atmósfera y, a la par, cuáles serán aquellas que no sobrevivirán. Mientras que el incremento en los niveles de CO2 puede favorecer el desarrollo de los pastos C3 en un ambiente con "efecto invernadero", el concomitante aumento en la temperatura favorecerá sin duda la implementación de los que exhiban el mecanismo C4; además, los cambios en la cantidad, intensidad y estacionalidad de las precipitaciones también ejercerán su influencia en la distribución relativa de las especies C3 y C4. El análisis empírico de las interrelaciones entre las variables climáticas y la distribución actual de los pastos C3 y C4 -incluidas las 3 variantes conocidas de estos últimos-, junto a modelos de cambio global, permiten predecir la distribución que, en el futuro, pueden llegar a tener los pastos en un escenario que exhiba un cambio climático determinado. Es por ello que, en este proyecto, se propone analizar, en el centro y norte de Argentina, la distribución actual de Gramíneas C3 y C4 y su relación con variables climáticas, con la intención de predecir la posible respuesta de los pastos ante determinados cambios climáticos utilizando modelos globales tales como el GIS o similares.

The levels of complement C3 and C4 components were determined in non-indigenous (creoles) and indigenous (Warao) populations, the latter with an extremely high tuberculosis (TB) rate. Serum samples from 209 adults were studied and classified in 4 groups taking into account tuberculin skin tests (TST): (1) the group of Warao patients (58 positive for the TST, WP TST+ and 9 negative for the TST, WP TST-), (2) the group of creole patients (34 positive for the TST, CP TST+ and 9 negative for the TST, CP TST-), (3) the group of healthy Warao controls (38 positive and 14 negative for TST, WC TST+ and WC TST-, respectively), (4) the creole controls (26 positive and 21 negative for the TST, CC TST+ and CC TST-, respectively). With respect to the results concerning the measurement of both complement C3 and C4 components with the exception of the WC TST and the CC groups, the WP TST+ and WP TST- as well as WC TST+ groups showed a significant frequency of individuals with decreased levels of complement C3 component (20.6, 33.3, and 26.3%, respectively) and also C4 component (12.0, 11.1, and 13.3%, respectively) in comparison to both creole patients (CP TST+, 8.82% and CP TST-, 0% and CP TST+, 5.88% and CP TST-, 0%) for C3 and C4, respectively. The study of these parameters carried out in 15 Warao subjects with active infection, before and after anti-TB chemotherapy,statisticallyconfirmedthat the effective chemotherapy did not restore normal levels of the complement C3 and C4 components among Warao patients. Aditional tests for hepatitis B or hepatitis C infection, and the profile of the hepatic proteins were not associated to the deficiency in production of the complement components.In conclusion, the results show that within the Warao population, a high percentage of subjects exhibit decreased levels of both complement C3 and C4 components independent of latent or active infection and the status of TST.

Human cytosolic thymidine kinase (hTK1) has proven to be a suitable target for the noninvasive imaging of cancer cell proliferation using radiolabeled thymidine analogues such as [(18)F]3'-fluoro-3'-deoxythymidine ([(18)F]FLT). A thymidine analogue for single photon emission computed tomography (SPECT), which incorporates the readily available and inexpensive nuclide technetium-99m, would be of considerable practical interest. hTK1 is known to accommodate modification of the structure of the natural substrate thymidine at the positions N3 and C3' and, to a lesser extent, C5. In this work, we used the copper-catalyzed azide-alkyne cycloaddition to synthesize two series of derivatives in which thymidine is functionalized at either the C3' or N3 position with chelating systems suitable for the M(CO)(3) core (M = (99m)Tc, Re). The click chemistry approach enabled complexes with different structures and overall charges to be synthesized from a common precursor. Using this strategy, the first organometallic hTK1 substrates in which thymidine is modified at the C3' position were identified. Phosphorylation of the organometallic derivatives was measured relative to thymidine. We have shown that the influence of the overall charge of the derivatives is dependent on the position of functionalization. In the case of the C3'-functionalized derivatives, neutral and anionic substrates were most readily phosphorylated (20-28% of the value for the parent ligand thymidine), whereas for the N3-functionalized derivatives, cationic and neutral complexes were apparently better substrates for the enzyme (14-18%) than anionic derivatives (9%).

Interaction between CD40, a member of the tumor necrosis factor receptor (TNFR) superfamily, and its ligand CD40L, a 39-kDa glycoprotein, is essential for the development of humoral and cellular immune responses. Selective blockade or activation of this pathway provides the ground for the development of new treatments against immunologically based diseases and malignancies. Like other members of the TNF superfamily, CD40L monomers self-assemble around a threefold symmetry axis to form noncovalent homotrimers that can each bind three receptor molecules. Here, we report on the structure-based design of small synthetic molecules with C3 symmetry that can mimic CD40L homotrimers. These molecules interact with CD40, compete with the binding of CD40L to CD40, and reproduce, to a certain extent, the functional properties of the much larger homotrimeric soluble CD40L. Architectures based on rigid C3-symmetric cores may thus represent a general approach to mimicking homotrimers of the TNF superfamily.

Crop species with the C4 photosynthetic pathway are more efficient in assimilating N than C3 plants, which results in different N amounts prone to be washed from its straw by rain water. Such differences may affect N recycling in agricultural systems where these species are grown as cover crops. In this experiment, phytomass production and N leaching from the straw of grasses with different photosynthetic pathways were studied in response to N application. Pearl millet (Pennisetum glaucum) and congo grass (Brachiaria ruziziensis) with the C4 photosynthetic pathway, and black oat (Avena Strigosa) and triticale (X Triticosecale), with the C3 photosynthetic pathway, were grown for 47 days. After determining dry matter yields and N and C contents, a 30 mm rainfall was simulated over 8 t ha-1 of dry matter of each plant residue and the leached amounts of ammonium and nitrate were determined. C4 grasses responded to higher fertilizer rates, whereas N contents in plant tissue were lower. The amount of N leached from C4 grass residues was lower, probably because the C/N ratio is higher and N is more tightly bound to organic compounds. When planning a crop rotation system it is important to take into account the difference in N release of different plant residues which may affect N nutrition of the subsequent crop.

Este estudo comparou a fenologia de três gramíneas nativas do Cerrado em dois níveis estruturais: variação do número de folhas por perfilho (colmo) e do número de perfilhos por planta. As espécies C4 Axonopus marginatus (Trin.) Chase e Trachypogon marginatus (L.f.) Kuntze e a C3 Echinolaena inflexa (Poir.) Chase foram estudadas na Reserva Ecológica do IBGE em Brasília, Distrito Federal, entre novembro de 1996 e junho de 1998. A fenologia de perfilhos foi comparada em três comunidades vegetais: cerrado denso protegido de fogo por 26 anos, campo sujo protegido de fogo por 22 anos e campo sujo queimado bienalmente. A fenologia de folhas foi estudada apenas no cerrado denso. O número de folhas da espécie C3 foi sempre maior que das C4. A área foliar foi constante durante todo o período na espécie C3, mas decresceu nas espécies C4 durante a estação seca. Para as espécies A. marginatus e E. inflexa, o número de perfilhos foi maior no campo sujo protegido, seguido de cerrado denso protegido e do campo sujo queimado. O número de perfilhos de T. spicatus no campo sujo protegido foi o dobro do encontrado no campo sujo queimado; o campo sujo queimado, não diferiu do cerrado denso. As diferenças fenológicas foram mais claras no nível foliar que no de perfilhos. O fogo promoveu a simplificação da arquitetura da planta nas três espécies.

Genetic studies have suggested that polymorphisms of genes coding for apolipoproteins are significant determinants of serum lipoprotein and lipid levels in adults. However, only a few studies have investigated the association of these polymorphisms in children. Therefore, in the present investigation we studied the distribution of APOA1 -75 G>A, +83 C>T, APOC3 -482 C>T, -455 T>C and 3238 C>G, and APOA4 Q360H and T347S polymorphisms and their influence on plasma lipoprotein levels in children from a Brazilian northeastern admixed population. The seven polymorphic sites were genotyped in 414 children aged 5 to 15 years (mean 8.9 ± 2.9). The genotypes of the seven polymorphic sites were assessed by PCR-RFLP methods. The frequencies of the less common alleles were, in general, intermediate among parental populations, as expected. Strong linkage disequilibrium was detected between polymorphisms at the APOA1, APOC3 and APOA4 loci in this admixed population sample. Overall the genotype effects seen in adults were weaker or absent in children. The APOC3/-455 and APOA4 T347S variants showed significant effects on HDL cholesterol in girls (P = 0.033 and P = 0.016, respectively). Significantly higher plasma total (P = 0.003) and LDL cholesterol (P = 0.004) levels were observed in boys who were carriers of the 3238G allele at the APOC3/3238 C>G site. These results disclosed an overall absence of associations between these polymorphisms and lipids in children. This finding is not unexpected because expression of the effect of these polymorphisms might depend on the interaction with environmental variables both internal and external to the individual.

Se presenta el proyecto INCLUES que tiene como objetivo la creación de una red europea de profesionales, colegios e instituciones de formación de profesorado para promover la educación inclusiva y cognitiva. Se trata, por tanto de integrar los niños con dificultades de aprendizaje por diversas causas, entre ellas, la discapacidad, modificando las pautas habituales de enseñanza. Se entiende por educación inclusiva aquella que defiende el derecho a una educación de calidad personalizada y adaptada a todos los niños, sin exclusión alguna; mientras que la educación cognitiva se refiere a las habilidades sociales, motoras, artísticas y emocionales del niño. El proyecto se justifica en la existencia de un movimiento mundial, con normas en algunos países europeos y en las demandas sociales para el acceso a la información, tecnología, vida económica y relaciones sociales. Se expone la falta de formación del profesorado en la inclusión, enseñanzas y evaluación; y la búsqueda de nuevos enfoques con una educación cognitiva-mediadora, utilizando la creatividad, la música y el arte. Las innovaciones de INCLUES están en la combinación de la inclusión con la activación cognitiva; el aprovechamiento y uso de la experiencia y práctica de los colegios específicos de educación especial; la difusión de una selección de métodos y enfoques eficaces; y la evaluación dinámica. Los antecedentes del proyecto se basan en otros: Helios II; Leonardo; Comenius; Equal; y en especial en el Proyecto Comenius de Desarrollo, y el Proyecto INSIDE que permite crear materiales y métodos. Se señala la variedad de destinatarios directos: entre otros los profesores; profesionales de rehabilitación y psicólogos; y destinatarios indirectos: alumnos en riesgo de fracaso escolar y niños con dificultades de aprendizaje general o específica. Se señalan algunas actividades que hay que poner en marcha como los foros de discusión en la web; revistas digitales cuatrimestrales; recopilaciones y publicaciones e intercambio de informaciones.
