La citogenética dispone de métodos que permiten evaluar, directa o indirectamente, anomalías cromosómicas causantes de importantes mejoras o pérdidas ganaderas en varios países que las han estudiado. Por ello, diferentes países de zootecnia avanzada dan importancia creciente a los estudios citogenéticos, planteándose en algunos la necesidad o exigencia de realizar estudios del cariotipo en reproductores que se usan en centros de inseminación artificial, como de sus ascendientes o descendientes en test de progenie o bancos de embriones. Existen numerosas anomalías cromosómicas causantes de trastornos del desarrollo, neoplasias e infertilidad en el bovino, ovino, caprino, porcino, equino y otros animales domésticos. Las pérdidas estudiadas son cuantiosas y algunas de estas anomalías se refieren a menor tasa de fertilidad y cantidad de terneros. Debido a que la evaluación citogenética involucra etapas que consumen mucho tiempo, se desarrollan procedimientos semiautomáticos que reducen los tiempos significativamente, sin afectar la seguridad y consistencia. Por tal motivo, debido a que se dispone de un analizador digital de imágenes con el software base y software para el análisis del cariotipo, se capacitarán recursos humanos, que permitan luego agilizar la obtención de resultados. En Argentina, los estudios citogenéticos en el bovino son recientes, por lo que es necesario continuarlos y extenderlos a otros animales de importancia económica, afectiva o en vías de extinción, con el objeto de protegerlos de alteraciones hereditarias que resulten negativas. En este sentido, se han dispuesto algunas acciones que propenden a reglamentar la necesidad del cariotipado en reproductores dadores de semen y los que se emplean en la transferencia de embriones. No conocemos de estudios citogenéticos a nivel nacional en camélidos andinos, ovinos, caprinos, equinos, caninos y/o aves (...) 1. Objetivos Generales: - Desarrollar recursos humanos y técnicos para realizar estudios cariotípicos (cultivo y técnicas de bandeo) en animales de importancia económica. - Capacitación en análisis digital de imágenes para realizar el análisis cromosómico. - Realizar estudios cariotípicos en reproductores con problemas de fertilidad. - Realizar estudios cariotípicos en bovinos con diagnóstico serológico de leucosis. - Realizar estudios de prevalencia de anomalías cromosómicas en toros usados en inseminación artificial. - Ofrecer servicios de registros cariotípicos para reproductores empleados en inseminación artificial, transplante de embriones, animales con tumores o trastornos del desarrollo.

El proyecto tiene como objetivo estudiar la implicancia funcional de los glucocorticoides liberados durante diferentes situaciones estresantes en las consecuencias comportamentales inducidas por las mismas. Asimismo, se evaluará la influencia de los esteroides en los cambios neuroquímicos (neurotransmisión gabaérgica) y neuroendócrinos (actividad del sistema hipotalámico-hipofisario-adrenal, HHA) que subyacen a las alteraciones comportamentales. El estudio contribuye, además, a la caracterización de los mecanismos neurales implicados en los déficits conductuales inducidos por estrés. Durante este período se abordarán los siguientes objetivos específicos: a) Caracterización de los sistemas de receptores centrales (MR o GR) involucrados en la respuesta ansiogénica inducida por una sesión de inmovilización y de las áreas centrales comprometidas. (...) De este modo se extenderán los hallazgos conductuales a la actividad del complejo supramolecular GABAa. b) Evaluación de los efectos inducidos por la inyección sistémica aguda de dexametasona sobre la funcionalidad complejo-receptor GABAa en corteza prefrontal por medio de la determinación de la captación de cloruro estimulada por GABA. (...) De este modo se extenderán los hallazgos conductuales a la actividad del complejo supramolecular GABAa. c) En el paradigma residente-intruso se evaluará la generalización de la acción de Corticoesterona (CS) sobre el diferentes medidas conductuales de reactividad a un evento aversivo novel. (...) d) Se evaluarán los cambios en la actividad del sistema HHA en relación a las diferentes expresiones conductuales (sumisión, "boxing" o "freezing") asociadas a la confrontación social, para lo cual se determinará el curso temporal de secreción de CS y ACTH, antes y después del encuentro agonístico. (...) e) Se investigarán los efectos de una experiencia previa de derrota sobre la respuesta neuroendócrina inducida por exposición a un evento aversivo novel.

The A. went last August to the State of Minas Gerais to continue his studies on transmission of leprosy by insects. He selected ten lepers (all L3 cases) for his experiments. It happened in the middle of August, a few day after freezing temperature. Practically there were no day mosquitoes, even near a river where in March there were very many. Bringing the patients to a wood, near the Peixe River (Fish River), at the dawn they were attacked by very many Anopheles, some flebotomus, a few Simulium and very rare Culex scapularis. All these insects became infected, in different degrees, by biting such patients. CONCLUSIONS. 1. Two species of Anopheles (A. albitarsis and A. tarsimaculatus) became strongly infected by Hansen bacilli. 2. By dissection done by Dr. Oliveira Castro were found lepra bacilli in various points of the proboscis of two Flebotomus (F. intermedius) and in their stomachs. 3. By smearing were found lepra bacilli in two specimens of Simulium sp. (probably pertinax). 4. It was confirmed also the verification done last March, at the same Leper Colony, that Phthirius pubis can be also a carrier of lepra bacillus. 5. There was confirmed also natural infection of nymphs of Amblyomma cajennense in lepers. Dr. Oliveira Castro is dissecting the Anopheles to locate the bacilli in their organisms and he started, with the cooperation of the Director of Colonia Santa Fé, Dr. José Mariano, attenpts to re-infect a group of negative-nerve cases of leprosy with infected mosquitoes.

A technics for prefreezing of blood plasma and serum is described in this paper. The method indicated by Strumia et al. (2), uses a rapid local freezing to obtain the shell-freezing, with refigerated alcohol bath, at temperatures around minus 35ºC. On our work, it has been found that normal horse blood plasma fulfils the instructions given by Strumia, although normal human blood plasma, very often, fails to give the expected results. This is very disadvantageous at the routine work. With the use of small amounts of solid carbon dioxide, spread over the flasks, in the refrigerated bath, it has been possible to start the chrystallization. The technics prescribes a rapid cooling, like the one used by Strumia, to bring the temperature down, to about plus 10ºC. and, with rotating device stopped, the solid carbon dioxide is applied for one minute simultaneously on each flask. Starting rotation again, it begins to form a very uniform shell around the walls of the flasks.

Reproductive performance including mating, semen transfer, egg production and hatching rate were investigated in pairs of Panstrongylus megistus in wich the male was treated with either precocene II or ethoxyprecocene II, topically applied shortly after imaginal molt. Eighty percent of the males were not able to mate for a period as long as 40 days after treatment. Those males wich did mate (20%) produced small spermatophores; a few (10%) were unable to transfer spermatozoa to female, while the others (10%) exhibited reproductive performance similar to that of non-treated males. The cumulative effects of the precocene on the different stages of reproduction resulted in a drastic reduction of offspring production. The anti-allatal compound precocene seems to inhibit successful reproduction through a corpus allatum inactivation and subsequent lack of secretory activity of the male accessory glands.

The effects of radiation on the reproductive capacity and the longevity of Panstrongylus magistus were studied. An indirect correlation between longevity and radiation doses was observed. Males were more affected than females. Longevity of females submitted to 10 Gy was not different when compared to controls. Some of the irradiated males copulated and in these cases semen was transferred to females, but only few eggs were laid. Fertility was seriously affected in all irradiated groups, decreasing with increasing intensity of dose. The dose of 80 Gy induced sterility in males and females. Gelatinous spermatophores that were expelled by females irradiated with 20 and 40 Gy, may be a consequence of irradiation that induced modification in the bursa copulatrix pH.

Using cryo-electron microscopy we reconstructed the three-dimensional trajectories adopted in cryovitrified solutions by double-stranded DNA molecules in which the backbone of one strand lacked a phosphate at regular intervals of 20 nucleotides. The shape of such nicked DNA molecules was compared with that of DNA molecules with exactly the same sequence but without any single-stranded scissions. Upon changing the salt concentration we observed opposite effects of charge neutralization on nicked and non-nicked DNA. In low salt solutions (10 mM Tris-HCl, 10 mM NaCl) the applied dense nicking caused ca 3.5-fold reduction of the DNA persistence length as compared with non-nicked DNA. Upon increasing the salt concentration (to 150 mM NaCl and 10 mM MgCl2) the persistence length of non-nicked DNA appreciably decreased while that of nicked DNA molecules increased by a factor of 2.

The aim of this study was to compare the efficacy of conservation by freezing the strains of Haemophilus influenzae at -20ºC and -70ºC. Skim milk supplemented with glucose, yeast extract and glycerol allowed highest viability of H. influenzae both at -20ºC and -70ºC from the media analyzed. Trypticase soy broth and brain heart infusion broth supplemented with glycerol, allowed excellent recovery. Use of cotton swaps as supporting material, with or without addition of cryoprotective agents, did not modify H. influenzae viability after six months of storage. Concentration of the initial inoculum positively affected viability when stored at -20ºC. Initial concentration did not influence survival after storage at -70ºC. Thawing at room temperature should not exceed 3 h as to get highest survival percentage.

The hypothalamic neuropeptide oxytocin (OT), which controls childbirth and lactation, receives increasing attention for its effects on social behaviors, but how it reaches central brain regions is still unclear. Here we gained by recombinant viruses selective genetic access to hypothalamic OT neurons to study their connectivity and control their activity by optogenetic means. We found axons of hypothalamic OT neurons in the majority of forebrain regions, including the central amygdala (CeA), a structure critically involved in OT-mediated fear suppression. In vitro, exposure to blue light of channelrhodopsin-2-expressing OT axons activated a local GABAergic circuit that inhibited neurons in the output region of the CeA. Remarkably, in vivo, local blue-light-induced endogenous OT release robustly decreased freezing responses in fear-conditioned rats. Our results thus show widespread central projections of hypothalamic OT neurons and demonstrate that OT release from local axonal endings can specifically control region-associated behaviors.

The present study was conducted to investigate a possible correlation between plasma (PVL) and seminal viral load (SVL) on treatment-naïve HIV-1-infected patients in Vitória, ES, Brazil. We also evaluated whether the progressive immunosuppression associated with HIV disease (as evidenced by declining CD4 T cell counts) has any impact on the correlation between PVL and SVL HIV-1. Viral load on paired blood and semen samples from 56 consecutive treatment-naïve patients were evaluated and compared to CD4 cell counts. Viral load and T cell counts (cells/µl) were determined by NASBA and by flow cytometry, respectively. Overall, a strong positive correlation between PVL and SVL (rho = 0.438, p = 0.001) was observed. However, when patients were grouped according to their CD4 counts, this correlation was only significant among patients with CD4 counts > 200 cells/µl. Results presented here demonstrate the existence of a strong correlation between PVL and SVL on patients with CD4 cell counts > 200 cells/µl, suggesting that this association may correlate with disease progression.

Background: Urinary human chorionic gonadotropin (hCG) concentration is routinely measured in all anti-doping laboratories to exclude the misuse of recombinant or urinary hCG preparations. In this study, extended validation of two commercial immunoassays for hCG measurements in urine was performed. Both tests were initially designed for hCG determination in human serum/plasma. Methods: Access (R) and Elecsys (R) 1010 are two automated immunoanalysers for central laboratories. The limits of detection and quantification, as well as intra-laboratory and inter-technique correlation, precision, and accuracy, were determined. Stability studies of hCG in urine following freezing and thawing cycles (n = 3) as well as storage conditions at room temperature, 4 degrees C and 20 degrees C, were performed. Results: Statistical evaluation of hCG concentrations in male urine samples (n = 2429) measured with the Elecsys (R) 1010 system enabled us to draw a skewed frequency histogram and establish a far outside value equal to 2.3 IU/L. This decision limit corresponds to the concentration at which a sportsman will be considered positive for hCG. Intra-assay precision for the Access (R) analyser was less than 4.0 A, whereas the inter-assay precision was closer to 4.5 % (concentrations of the official external controls contained between 5.5 and 195.0 IU/L). Intra and inter-assay precision for the Elecsys (R) 1010 analyser was slightly better. A good inter-technique correlation was obtained when measuring various urine samples (male and female). No urinary hCG loss was observed after two freeze/thaw cycles. On the other hand, time and inappropriate storage conditions, such as temperatures above 10 degrees C for more than 5 days, can deteriorate urinary hCG. Conclusions: Both analysers showed acceptable performances and are suitable for screening urine for anti-doping analyses. Each laboratory should validate and establish its own reference values because hCG concentrations measured in urine can be different from one immunoassay to another. The time delay between urine collection and analysis should be reduced as much as possible, and urine samples should be transported in optimal conditions to avoid a loss of hCG immunoreactivity.

Axial deflection of DNA molecules in solution results from thermal motion and intrinsic curvature related to the DNA sequence. In order to measure directly the contribution of thermal motion we constructed intrinsically straight DNA molecules and measured their persistence length by cryo-electron microscopy. The persistence length of such intrinsically straight DNA molecules suspended in thin layers of cryo-vitrified solutions is about 80 nm. In order to test our experimental approach, we measured the apparent persistence length of DNA molecules with natural "random" sequences. The result of about 45 nm is consistent with the generally accepted value of the apparent persistence length of natural DNA sequences. By comparing the apparent persistence length to intrinsically straight DNA with that of natural DNA, it is possible to determine both the dynamic and the static contributions to the apparent persistence length.

A decline in human sperm quality and quantity has been reported in numerous Western countries. This observation was also accompanied by an increase in urogenital malformations. The need for epidemiological studies dealing with unbiased populations in order to understand the causes of these observations is obvious. In Switzerland, the large majority of young men are asked to attend a military camp to be drafted into the army. A few weeks before this camp, conscripts were contacted and invited to participate in a large national study on semen quality. The participation was totally voluntary and anonymous. From September 2005 to June 2007, 770 volunteers filled out a questionnaire, underwent a clinical examination and provided sperm, blood and urine samples. Using self-rated health assessments, the observed cohort could be considered as healthy and no testicular cancer was found. Moreover, the testicular volumes, measured using Prader's orchidometry and ultrasonography, were comparable to those already published for young male populations. The median sperm concentration was 47 x 10(6)/ml, which is close to the concentration reported in Denmark, known to have the highest incidence of testicular cancer in Europe. Statistically significant differences were observed between regions with a lower sperm concentration for men residing in the Alps (43 x 10(6)/ml) and in the Zurich area (36 x 10(6)/ml) compared to men from West Plateau (54 x 10(6)/ml) and from the Jura (54 x 10(6)/ml). Such a regional discrepancy could be related to environmental factors, including endocrine disruptors. In order to confirm such regional differences more volunteers from the already studied regions should be studied and other parts of the country should be investigated. The rather low sperm concentration of Swiss young volunteers should be considered as a national health issue and investigated further.

Improvement in cancer treatments resulted in an increased number of men surviving cancer. Quality of life has become an important issue in these patients. Anti cancer treatments might have transient or definitive harmful effects on male fertility. Sperm cryoconservation is currently the only proven method to preserve fertility in patients undergoing oncologic treatment. It should be proposed to every patient at reproductive age before chemotherapy, radiotherapy or any surgery involving reproductive tract. Despite low use rate, this simple method could allow patients presenting infertility after treatment to father a child.

Objectives This article presents a psychological approach to substance use in sport using a dynamic and situated activity framework. The aim was to analyze the various relationships between activity and the consumption of substances during the sporting life course of athletes who recognized doping violation. Design Data were collected from secondary sources and biographical and self-confrontational interviews to build traces of the past activity. Method Twelve doping athletes or those admitting to having used banned substances volunteered to participate. The data were coded and compared to identify typical activities and their intrinsic dynamics. Results Six activities were identified: "Agree to use," "Drop out of a non-viable state," Return to a former state," "Prevent a potential deficiency," "Maintain an acquired state," and "Balance the sporting life with substance use," comprising 11 patterns. Conclusions The athletes' activity embedded substance use in reciprocal relationships that consisted of freezing, exploring and exploiting fields of possible actions created and offered by the situation dynamics. Recommendations for situated and dynamic prevention are provided.
