Casearin X, Its Degradation Product and Other Clerodane Diterpenes from Leaves of Casearia sylvestris: Evaluation of Cytotoxicity against Normal and Tumor Human Cells

An EtOH extract of the leaves of Casearia sylvestris afforded new clerodane diterpene, casearin X, together with the known compounds casearins B, D, L, and O, and caseargrewiin F Casearin X degraded to the corresponding dialdehyde when stored in CDCl(3). The diterpenes isolated were cytotoxic to human cancer cell lines, with caseargrewiin F being the most active and the new clerodane, casearin X, the second active compound with IC(50) values comparable to the positive control doxorubicin. All isolated diterpenes showed lower activities against normal human cells than against cancer cell lines, which might indicate a possible selective action on cancer cells. Casearin X dialdehyde was not cytotoxic to cancer cells indicating that the occurrence of these CO groups at C(18) and C(19) is incompatible with the cytotoxic activity.

Utilizando resultados de trabalho estatístico com base nos dados da Relação Anual de Informações Sociais (RAIS), que permitem caracterizar a indústria de calçados de Nova Serrana (MG) como um sistema local de produção, e nos resultados de pesquisa de campo efetuada em abril de 2002, quando foram realizadas entrevistas e visitas em empresas e instituições locais, o artigo estuda o sistema local de produção de Nova Serrana tanto em termos do sistema como um todo quanto das empresas e instituições que o compõem. O objetivo é produzir, nesses dois níveis de análise, evidências que possam orientar ações públicas e privadas, visando ao aprimoramento do sistema e das empresas locais em duas esferas: (1) produtiva e tecnológica, em seus vários desdobramentos, e (2) institucional e organizacional. Para isso, caracteriza e dimensiona primeiramente o sistema local em termos de sua localização e região de influência, sua logística, população e empregos gerados, história e evolução, estrutura de produção, instituições de apoio, contexto social, cultural e político. em seguida, caracteriza as empresas em termos de tamanho e quanto a: desenvolvimento de produto, sistemas de comercialização, relações cooperativas, interações com fornecedores, fontes de informação para desenvolvimento de produtos e sobre mercados e processos de fabricação, práticas e políticas quanto à qualidade, ao financiamento e a outros aspectos pertinentes ao caso em estudo. Por fim, ressalta os problemas mais relevantes que podem ser objeto de políticas públicas e/ou de ações conjuntas das empresas e instituições locais.

Determination of solubility product of mercurous acetate at various ionic strengths in aqueous medium

The conditionals (K-ps) and thermodynamics (K-ps(o)) solubility products of mercurous acetate have been determined, in aqueous solution, at 25 degrees C and ionic strength (I) comprised between 0.300 and 3.000 mol/L (NaClO4). The investigation was carried out potentiometrically by using a second class electrodes which responds to acetate ions. The values obtained for [pK(ps) (I)] were: 9.49+/-0.08 (0.000); 9.51+/-0.08 (0.300); 9.53+/-0.08 (0.500); 9.54+/-0.08 (0.700); 9.55+/-0.09 (0.900), 9.57+/-0.09 (1.200); 9.59+/-0.10(1.500); 9.61+/-0.10 (1.800); 9.63+/-0.10 (2.100); 9.65+/-0.11 (2.400); 9.67+/-0.11 (2.700) e 9.69+/-0.12 (3.000).

NATURAL PRODUCT SUBSTITUTION PATTERN AND SKELETAL RECOGNITION METHOD

This work describes a methodology for identification of skeletal types of diterpenes based on data base with 1500 compounds isolated from Asteraceae. One program named BOTOCSYS was built with the codification of the compounds and their botanical sources. An example of identification of a new substance is given.

Indirect polarographic and cathodic stripping voltammetric determination of cefaclor as an alkaline degradation product

Cefaclor is not reducible at a mercury electrode, but it can be determined polarographically and by cathodic stripping voltammetry as its initial alkaline degradation product which is obtained in high yield by hydrolysis of cefaclor in Britton-Robinson (B-R) buffer pH 10 at 50 degrees C for 30 min (reduction peak at pH 10, -0.70 V). Differential pulse polarographic calibration graphs are linear up to at least 1 x 10(-4) mol l(-1). Recoveries of 93% of the cefaclor (n = 3) were obtained from urine spiked with 38.6 mu g ml(-1) using this polarographic method with 1 ml urine made up to 10 ml with pH 10 buffer. Using cathodic stripping voltammetry and accumulating at a hanging mercury drop electrode at -0.2 V for 30 s, linear calibration graphs were obtained from 0.35 to 40 mu g ml(-1) cefaclor in B-R buffer pH 10. A relative standard deviation of 4.2% (eta = 5) was obtained, and the limit of detection was calculated to be 2.9 ng ml(-1). Direct determination of cefaclor in human urine (1 ml of urine was made up to 10 ml with pH 10 buffer) spiked to 0.39 mu g ml(-1) was made (recovery 98.6%). (C) 1999 Elsevier B.V. B.V. Automatic identification by C-13 NMR of substituent groups bonded in natural product skeletons

The aim of this paper is to present a procedure that utilizes C-13 NMR for identification of substituent groups which are bonded to carbon skeletons of natural products. For so much was developed a new version of the program (MACRONO), that presents a database with 161 substituent types found in the most varied terpenoids. This new version was widely tested in the identification of the substituents of 60 compounds that, after removal of the signals that did not belong to the carbon skeleton, served to test the prediction of skeletons by using other programs of the expert system (SISTEMAT). Product development process: Using real options for assessments and to support the decision-making at decision gates

Enterprises need continuous product development activities to remain competitive in the marketplace. Their product development process (PDP) must manage stakeholders' needs - technical, financial, legal, and environmental aspects, customer requirements, Corporate strategy, etc. -, being a multidisciplinary and strategic issue. An approach to use real option to support the decision-making process at PDP phases in taken. The real option valuation method is often presented as an alternative to the conventional net present value (NPV) approach. It is based on the same principals of financial options: the right to buy or sell financial values (mostly stocks) at a predetermined price, with no obligation to do so. In PDP, a multi-period approach that takes into account the flexibility of, for instance, being able to postpone prototyping and design decisions, waiting for more information about technologies, customer acceptance, funding, etc. In the present article, the state of the art of real options theory is prospected and a model to use the real options in PDP is proposed, so that financial aspects can be properly considered at each project phase of the product development. Conclusion is that such model can provide more robustness to the decisions processes within PDP.

Effectiveness and safety of a new vaginal misoprostol product specifically labeled for cervical ripening and labor induction

Objective. The purpose of this study was to evaluate the effectiveness and safety of misoprostol in two different formulations: vaginal tablets of 25 mu g and one-eighth of a 200-mu g oral tablet, also administered intravaginally, for cervical ripening and labor induction of term pregnancies with an indication for that. Methods. A single-blind, randomized, controlled clinical trial was carried out in 120 pregnant women who randomly received one of the two formulations. The main dependent variables were mode of delivery, need for additional oxytocin, time between beginning of induction and delivery, perinatal results, complications, and maternal side effects. Student's t, Mann-Whitney, chi(2), Fisher's Exact, Wilcoxon and Kolmogorov-Smirnoff tests, as well as survival analysis, were used in the data analysis. Results. There were no significant differences between the groups in terms of general characteristics, uterine contractility, and fetal well-being during labor, cesarean section rates, perinatal outcomes, or maternal adverse events. The mean time between the beginning of cervical ripening and delivery was 31.3 h in the vaginal tablet group and 30.1 h in the oral tablet group, a difference that was not statistically significant. Conclusion. The results showed that the 25-mu g vaginal tablets of misoprostol were as effective and safe for cervical ripening and labor induction as the dose-equivalent fraction of 200-mu g oral tablets.

Determination of 2,4-dichlorophenoxyacetic acid and its major transformation product in soil samples by liquid chromatographic analysis

The 2,4-dichlorophenoxyacetic acid (2,4-D) is one of the most applied herbicides around the world to control broad leave herbs in many crops: In this study, a method was developed for simultaneous extraction and determination of 2,4D and its major transformation product, i.e., the 2,4-dichlorophenol (2,4-DCP), in soil samples. The herbicide and its degradation product were extracted twice from soil samples, after acidification, by dichloromethane on ultrasound system for 1 h. Both extracts were combined and filtrated in qualitative filter paper and Celitee. The total extract was concentrated in rotatory evaporator, dried under N-2 and finally dissolved in 1 ml of methanol. High Performance Liquid Chromatography with UV detection at 230 nm was used for analysis. Recoveries were obtained from soil samples fortified at 0.1, 1.0, 2.0, 3.0 and 4.0 mg kg(-1) levels and the results varied from 85 to 111% (for 2,4-D) and from 95 to 98% (for 2,4-DCP). For both compounds, the limits of quantification were 0.1 mg kg(-1), which were the loss level at which the accuracy and the precision were studied. Nevertheless, the limits of detection, calculated by considering the blank standard deviation and the minimum concentration level, were 0.03 and 0.02 mg kg for 2,4-D and 2,4-DCP, respectively. This proposed method was applied to soil samples of eucalyptus crops, which was previously treated with the herbicide. Despite that, neither 2,4-D nor its degradation product were detected 30 days after the herbicide application. (C) 2003 Elsevier B.V. B.V. All rights reserved.
