Transcription elongation by RNA polymerase II is regulated by the general elongation factor TFIIS. This factor stimulates RNA polymerase II to transcribe through regions of DNA that promote the formation of stalled ternary complexes. Limited proteolytic digestion showed that yeast TFIIS is composed of three structural domains, termed I, II, and III. The two C-terminal domains (II and III) are required for transcription activity. The structure of domain III has been solved previously by using NMR spectroscopy. Here, we report the NMR-derived structure of domain II: a three-helix bundle built around a hydrophobic core composed largely of three tyrosines protruding from one face of the C-terminal helix. The arrangement of known inactivating mutations of TFIIS suggests that two surfaces of domain II are critical for transcription activity.

We have investigated the influence of genetic instability [replication error (RER) phenotype] on APC (adenomatous polyposis coli), a gene thought to initiate colorectal tumorigenesis. The prevalence of APC mutations was similar in RER and non-RER tumors, indicating that both tumor types share this step in neoplastic transformation. However, in a total of 101 sequenced mutations, we noted a substantial excess of APC frameshift mutations in the RER cases (70% in RER tumors versus 47% in non-RER tumors, P < 0.04). These frameshifts were characteristic of mutations arising in cells deficient in DNA mismatch repair, with a predilection for mononucleotide repeats in the RER tumors (P < 0.0002), particularly (A)n tracts (P < 0.00007). These findings suggest that the genetic instability that is reflected by the RER phenotype precedes, and is responsible for, APC mutation in RER large bowel tumors and have important implications for understanding the very earliest stages of neoplasia in patients with tumors deficient in mismatch repair.

Tumors result from disruptions in the homeostatic mechanisms that regulate cell birth and cell death. In colon cancer, one of the earliest manifestation of this imbalance is the formation of polyps, caused by somatic and inherited mutations of the adenomatous polyposis coli (APC) tumor suppressor gene in both humans and mice. While the importance of APC in tumorigenesis is well documented, how it functions to prevent tumors remains a mystery. Using a novel inducible expression system, we show that expression of APC in human colorectal cancer cells containing endogenous inactive APC alleles results in a substantial diminution of cell growth. Further evaluation demonstrated that this was due to the induction of cell death through apoptosis. These results suggest that apoptosis plays a role not only in advanced tumors but also at the very earliest stages of neoplasia.

A área da Tecnologia da Informação no Brasil sofre um problema latente com a falta de planejamento e atrasos constantes em projetos, determinando para os profissionais vinculados a ela um ambiente altamente desmotivador para a condução de seus trabalhos. Supõe-se que o que possa corroborar para tal problema seja a formação educacional deficitária dos indivíduos que atuam neste segmento, principalmente aqueles relacionados a cargos executivos e que estejam exercendo atividades de gestão. De acordo com teóricos como Edgard Morin (2004), em se tratando de educação fundamental, média ou superior os aspectos educacionais podem ser considerados deficitários justamente porque, ao segmentar o conhecimento, eles promovem uma alienação do indivíduo, eliminando sua capacidade criativa e reflexiva. Seria interessante, portanto, que ao avaliar a capacidade cognitiva de uma pessoa, a inteligência a ser mensurada não seja abordada através de um único espectro de conhecimento, mas através de muitos deles. A teoria das Inteligências Múltiplas, desenvolvida por Howard Gardner vem de encontro a essa necessidade, pois de acordo com o autor, a inteligência de um indivíduo deve ser mensurada através de uma gama de nove espectros: Linguística, Musical, Lógico-Matemática, Espacial, Corporal Cinestésica, Interpessoal, Intrapessoal, Naturalista e Existencial. Isto posto, este trabalho aborda uma metodologia computacional para classificação e descoberta de padrões em indivíduos, sejam esses alunos ou profissionais graduados, de uma determinada área. Além da metodologia, foi proposto um estudo de caso, considerando cursos superiores relacionados à área de Computação no Brasil.

The present data publication provides permanent links to original and updated versions of validated data files. The data files include properties of seawater, particulate matter and dissolved matter that were measured from discrete water samples collected with Niskin bottles during the 2009-2013 Tara Oceans expedition. Properties include pigment concentrations from HPLC analysis (10 depths per vertical profile, 25 pigments per depth), the carbonate system (Surface and 400m; pH (total scale), CO2, pCO2, fCO2, HCO3, CO3, Total alkalinity, Total carbon, OmegaAragonite, OmegaCalcite, and dosage Flags), nutrients (10 depths per vertical profile; NO2, PO4, N02/NO3, SI, quality Flags), DOC, CDOM, and dissolved oxygen isotopes. The Service National d'Analyse des Paramètres Océaniques du CO2, at the Université Pierre et Marie Curie, determined CT and AT potentiometrically (Edmond 1970; DOE 1994) on samples preserved according to Dickson et al. (2007). More than 250 vertical profiles of these properties were made across the world ocean. DOC, CDOM and dissolved oxygen isotopes are available only for the Arctic Ocean and Arctic Seas (2013).

Hole 504B in the eastern equatorial Pacific has been the focus of five scientific drilling expeditions since it was first drilled in 1979. During these five legs, a series of temperature logs has been obtained over a time span of almost 8 yr, documenting the geothermal and hydrologic state of the oceanic crust in this region. Immediately following reentry at the onset of ODP Leg 111 operations, a high-resolution temperature probe was lowered into the borehole and a precise record of temperature vs. depth in Hole 504B was recorded down to 1300 mbsf. As was observed during previous legs, the temperature gradient in the upper 400 m was reduced, indicating that downhole flow of cool ocean waters through the casing continued, though at a diminished rate. As subhydrostatic pressures in the upper basement have gradually diminished, the volume of flow has decayed from an estimated 6000-7000 L/hr in late 1979 to about 80 L/hr during Leg 111. At depths below 480 mbsf, a predominantly conductive heat transfer environment enabled the temperature gradient log to be analyzed with respect to lithology on both fine and broad scales. Anomalies in the gradient log in the cased section through the sedimentary column were found to correspond to biostratigraphic age markers and/or sharp changes in sediment composition and texture. Broad variations in temperature gradient within the basement correlated with large-scale porosity trends. Conductive heatflow estimates depict a systematic reduction with depth, ranging from approximately 196 mW/m**2 in the sediments to 120 ± 17 mW/m**2 at 1300 mbsf. Possible causes for this observation were examined from several perspectives, but none was suitably convincing. A fluid instability analysis indicated the likely existence of convection cells within the borehole and substantiated the hypothesis of mixing within the borehole postulated from isotopic and chemical studies of borehole waters. However, such mixing of borehole fluids does not provide an adequate explanation for the heatflow variations, and the disparity between surficial and deep values of heat flow remains unresolved.
