Millions of people worldwide are currently infected with human papillomavirus (HPV), herpes simplex virus (HSV) or human immunodeficiency virus (HIV). For this enormous contingent of people, the search for preventive and therapeutic immunological approaches represents a hope for the eradication of latent infection and/or virus-associated cancer. To date, attempts to develop vaccines against these viruses have been mainly based on a monovalent concept, in which one or more antigens of a virus are incorporated into a vaccine formulation. In the present report, we designed and tested an immunization strategy based on DNA vaccines that simultaneously encode antigens for HIV, HSV and HPV. With this purpose in mind, we tested two bicistronic DNA vaccines (pIRES I and pIRES II) that encode the HPV-16 oncoprotein E7 and the HIV protein p24 both genetically fused to the HSV-1 gD envelope protein. Mice i.m. immunized with the DNA vaccines mounted antigen-specific CD8⁺ T cell responses, including in vivo cytotoxic responses, against the three antigens. Under experimental conditions, the vaccines conferred protective immunity against challenges with a vaccinia virus expressing the HIV-derived protein Gag, an HSV-1 virus strain and implantation of tumor cells expressing the HPV-16 oncoproteins. Altogether, our results show that the concept of a trivalent HIV, HSV, and HPV vaccine capable to induce CD8⁺ T cell-dependent responses is feasible and may aid in the development of preventive and/or therapeutic approaches for the control of diseases associated with these viruses.

Four glycoproteins (gD, gB, gH, and gL) are required for herpes simplex virus (HSV) entry into the cell and for cell-cell fusion in transfected cells. gD serves as the receptor-binding glycoprotein and as the trigger of fusion; the other three glycoproteins execute fusion between the viral envelope and the plasma or endocytic membranes. Little is known on the interaction of gD with gB, gH, and gL. Here, the interactions between herpes simplex virus gD and its nectin1 receptor or between gD, gB, and gH were analyzed by complementation of the N and C portions of split enhanced green fluorescent protein (EGFP) fused to the glycoproteins. Split EGFP complementation was detected between proteins designated gDN + gHC, gDN + gBC, and gHN + gBC + wtgD, both in cells transfected with two or tree glycoproteins and in cells transfected with the four glycoproteins, commited to form syncytia. The in situ assay provides evidence that gD interacts with gH and gB independently one of the other. We further document the interaction between gH and gB. To elucidate which portions of the glycoproteins interact with each other we generated mutants of gD and gB. gD triggers fusion through a specialised domain, named pro-fusion domain (PFD), located C-terminally in the ectodomain. Here, we show that PFD is made of subdomains 1 and 2 (amino acids 260–285 and 285–310) and that each one partially contributed to herpes simplex virus infectivity. Chimeric gB molecules composed of HSV and human herpesvirus 8 (HHV8) sequences failed to reach the cell surface and to complement a gB defective virus. By means of pull down experiments we analyzed the interactions of HSV-HHV8 gB chimeras with gH or gD fused to the strep-tag. The gB sequence between aa residues 219-360 was identified as putative region of interaction with gH or critical to the interaction.

Herpes simplex virus 1 (HSV-1) infects oral epitelial cells, then spreads to the nerve endings and estabilishes latency in sensory ganglia, from where it may, or may not reactivate. Diseases caused by virus reactivation include mild diseases such as muco-cutaneous lesions, and more severe, and even life-threatening encephalitis, or systemic infections affecting diverse organs. Herpes simplex virus represents the most comprehensive example of virus receptor interaction in Herpesviridae family, and the prototype virus encoding multipartite entry genes. In fact, it encodes 11-12 glycoproteins and a number of additional membrane proteins: five of these proteins play key roles in virus entry into subsceptible cells. Thus, glycoprotein B (gB) and glycoprotein C (gC) interact with heparan sulfate proteoglycan to enable initial attachment to cell surfaces. In the next step, in the entry cascade, gD binds a specific surface receptor such as nectin1 or HVEM. The interaction of glycoprotein D with the receptor alters the conformation of gD to enable the activation of gB, glycoprotein H, and glycoprotein L, a trio of glycoproteins that execute the fusion of the viral envelope with the plasma membrane. In this thesis, I described two distinct projects: I. The retargeting of viral tropism for the design of oncolytic Herpesviruses: • capable of infecting cells through the human epitelial growth factor receptor 2 (HER2), overexpressed in highly malignant mammary and ovarian tumors and correlates with a poor prognosis; • detargeted from its natural receptors, HVEM and nectin1. To this end, we inserted a ligand to HER2 in gD. Because HER2 has no natural ligand, the selected ligand was a single chain antibody (scFv) derived from MAb4D5 (monoclonal antibody to HER2), herein designated scHER2. All recombinant viruses were targeted to HER2 receptor, but only two viruses (R-LM113 and R-LM249) were completely detargeted from HVEM and nectin1. To engineer R-LM113, we removed a large portion at the N-terminus of gD (from aa 6 to aa 38) and inserted scHER2 sequence plus 9-aa serine-glycine flexible linker at position 39. On the other hand, to engineer R-LM249, we replaced the Ig-folded core of gD (from aa 61 to aa 218) with scHER2 flanked by Ser-Gly linkers. In summary, these results provide evidence that: i. gD can tolerate an insert almost as big as gD itself; ii. the Ig-like domain of gD can be removed; iii. the large portion at the N-terminus of gD (from aa 6 to aa 38) can be removed without loss of key function; iv. R-LM113 and R-LM249 recombinants are ready to be assayed in animal models of mammary and ovary tumour. This finding and the avaibility of a large number of scFv greatly increase the collection of potential receptors to which HSV can be redirected. II. The production and purification of recombinant truncated form of the heterodimer gHgL. We cloned a stable insect cell line expressing a soluble form of gH in complex with gL under the control of a metalloprotein inducible promoter and purified the heterodimer by means of ONE-STrEP-tag system by IBA. With respect to biological function, the purified heterodimer is capable: • of reacting to antibodies that recognize conformation dependent epitopes and neutralize virion infectivity; • of binding a variety cells at cell surface. No doubt, the availability of biological active purified gHgL heterodimer, in sufficient quantities, will speed up the efforts to solve its crystal structure and makes it feasible to identify more clearly whether gHgL has a cellular partner, and what is the role of this interaction on virus entry.

Recent developments in piston engine technology have increased performance in a very significant way. Diesel turbocharged/turbo compound engines, fuelled by jet fuels, have great performances. The focal point of this thesis is the transformation of the FIAT 1900 jtd diesel common rail engine for the installation on general aviation aircrafts like the CESSNA 172. All considerations about the diesel engine are supported by the studies that have taken place in the laboratories of the II Faculty of Engineering in Forlì. This work, mostly experimental, concerns the transformation of the automotive FIAT 1900 jtd – 4 cylinders – turbocharged – diesel common rail into an aircraft engine. The design philosophy of the aluminium alloy basement of the spark ignition engine have been transferred to the diesel version while the pistons and the head of the FIAT 1900 jtd are kept in the aircraft engine. Different solutions have been examined in this work. A first V 90° cylinders version that can develop up to 300 CV and whose weight is 30 kg, without auxiliaries and turbocharging group. The second version is a development of e original version of the diesel 1900 cc engine with an optimized crankshaft, that employ a special steel, 300M, and that is verified for the aircraft requirements. Another version with an augmented stroke and with a total displacement of 2500 cc has been examined; the result is a 30% engine heavier. The last version proposed is a 1600 cc diesel engine that work at 5000 rpm, with a reduced stroke and capable of more than 200 CV; it was inspired to the Yamaha R1 motorcycle engine. The diesel aircraft engine design keeps the bore of 82 mm, while the stroke is reduced to 64.6 mm, so the engine size is reduced along with weight. The basement weight, in GD AlSi 9 MgMn alloy, is 8,5 kg. Crankshaft, rods and accessories have been redesigned to comply to aircraft standards. The result is that the overall size is increased of only the 8% when referred to the Yamaha engine spark ignition version, while the basement weight increases of 53 %, even if the bore of the diesel version is 11% lager. The original FIAT 1900 jtd piston has been slightly modified with the combustion chamber reworked to the compression ratio of 15:1. The material adopted for the piston is the aluminium alloy A390.0-T5 commonly used in the automotive field. The piston weight is 0,5 kg for the diesel engine. The crankshaft is verified to torsional vibrations according to the Lloyd register of shipping requirements. The 300M special steel crankshaft total weight is of 14,5 kg. The result reached is a very small and light engine that may be certified for general aviation: the engine weight, without the supercharger, air inlet assembly, auxiliary generators and high pressure body, is 44,7 kg and the total engine weight, with enlightened HP pump body and the titanium alloy turbocharger is less than 100 kg, the total displacement is 1365 cm3 and the estimated output power is 220 CV. The direct conversion of automotive piston engine to aircrafts pays too huge weight penalties. In fact the main aircraft requirement is to optimize the power to weight ratio in order to obtain compact and fast engines for aeronautical use: this 1600 common rail diesel engine version demonstrates that these results can be reached.

[EN]We present advances of the meccano method for T-spline modelling and analysis of complex geometries. We consider a planar domain composed by several irregular sub-domains. These sub-regions are defined by their boundaries and can represent different materials. The bivariate T-spline representation of the whole physical domain is constructed from a square. Herpes simplex virus entry into cells requires a multipartite fusion apparatus made of gD, gB and heterodimer gH/gL. gD serves as receptor-binding glycoprotein and trigger of fusion; its ectodomain is organized in a N-terminal domain carrying the receptor-binding sites, and a C-terminal domain carrying the profusion domain, required for fusion but not receptor-binding. gB and gH/gL execute fusion. To understand how the four glycoproteins cross-talk to each other we searched for biochemical defined complexes in infected and transfected cells, and in virions. We report that gD formed complexes with gB in absence of gH/gL, and with gH/gL in absence of gB. Complexes with similar composition were formed in infected and transfected cells. They were also present in virions prior to entry, and did not increase at virus fusion with cell. A panel of gD mutants enabled the preliminary location of part of the binding site in gD to gB to the aa 240-260 portion and downstream, with T306P307 as critical residues, and of the binding site to gH/gL at aa 260-310 portion, with P291P292 as critical residues. The results indicate that gD carries composite independent binding sites for gB and gH/gL, both of which partly located in the profusion domain. The second part of the project dealt with rational design of peptides inhibiting virus entry has been performed. Considering gB and gD, the crystal structure is known, so we designed peptides that dock in the structure or prevent the adoption of the final conformation of target molecule. Considering the other glycoproteins, of which the structure is not known, peptide libraries were analyzed. Among several peptides, some were identified as active, designed on glycoprotein B. Two of them were further analyzed. We identified peptide residues fundamental for the inhibiting activity, suggesting a possible mechanism of action. Furthermore, changing the flexibility of peptides, an increased activity was observed,with an EC50 under 10μM. New approaches will try to demonstrate the direct interaction between these peptides and the target glycoprotein B.

Mit dem System KCo2-xCuxS2 wurde ein neues magnetoresistives System gefunden. Der negative Magnetowiderstand ist mit der Größenordnung von 10 % in 8 Tesla bei 4 K klein im Vergleich zu Mangan-Perowskiten, jedoch eindeutig intrinsisch.Die magnetische Struktur des Thiospinells Fe0.5Cu0.5Cr2S4 konnte durch Neutronenbeugung, Mößbauer-Spektroskopie sowie begleitende Bandstrukturrechnungen aufgeklärt werden. Ein negativen Magnetowiderstand von 5,5 % nahe der Curie-Temperatur in Magnetfeldern von 8 Tesla bei der isostrukturellen eisenreichen Verbindung Fe0.75Cu0.25Cr2S4 wurde gefunden.Die intermetallischen Verbindungen des Gadoliniums weisen alle hohe negative Magnetowiderstände bei TC auf. Sowohl bei GdAl2 als auch bei GdPdP und GdPtP werden Widerstandsabsenkungen in 8 Tesla beobachtet, die bei ~1,5 TC 4 % erreichen und bis zu Temperaturen von 5 K über 6 % liegen. Während der Transportmechanismus in GdAl2 offenbar auf einer direkten Gd-Gd Wechselwirkung beruht, ist bei GdPdP und GdPtP bei tiefen Temperaturen ein nicht eindeutiges Verhalten beobachtbar. Ein Einfluss von Fremdphasen kann jedoch ausgeschlossen werden.Unter den metallreichen Phosphiden hexagonaler Struktur zeigt Fe2P große negative MR-Effekte von 7 % schon bei Raumtemperatur in hohen Feldern. Nahe der ferromagnetischen Ordnung reagiert die Verbindung auf äußere Felder bei niedrigen Feldstärken von weniger als 2 Tesla mit einer Erhöhung der Leitfähigkeit um 10 bis 11 %.

Resonante Laserionisations-Massenspektrometrie an Gadolinium zur Isotopenhäufigkeitsanalyse mit geringsten Mengen Die selektive Spuren- und Ultraspurenanalyse des Erdalkalielements Gadolinium eröffnet eine Vielzahl von Anwendungen in der Biomedizin und Kosmochemie. Zum Erreichen der hohen Anforderungen bezüglich Isotopen- und Isobarenselektivität von S>10^7 sowie Gesamteffizienz von e>10^-6 wurde der Einsatz der resonanten Laserionisations-Massenspektrometrie untersucht. Dazu erfolgte die Weiterentwicklung und Anpassung des existierenden Diodenlaser-Quadrupolmassenspektrometersystems. Durch Ionenflugbahn-Simulationsrechnungen wurde für das Quadrupol-Massenspektrometer die erreichbare Nachbarmassenunterdrückung und Transmission in Abhängigkeit von der Auflösung theoretisch vorhergesagt. Die Werte wurden experimentell bestätigt. Aus der beobachteten Peakstruktur erfolgte die Ableitung einer Methode zur Bestimmung der Energieunschärfe des eingesetzten Ionisationsprozesses. Zum Auffinden eines effizienten dreifach resonanten Anregungsschemas wurden die Isotopieverschiebungen und Hyperfeinstrukturen aller stabilen Gadoliniumisotope in zahlreichen Übergängen für die einfach, zweifach und dreifach resonante Ionisation präzise vermessen. Das aufgenommene Spektrum autoionisierender Resonanzen zeigte etwa 150 bislang unbekannter Zustände mit Resonanzüberhöhungen von bis zu fünf Größenordnungen im Ionisationswirkungsquerschnitt. Die entwickelte Methode der Hyperfeinzustandsselektion ermöglichte die Bestimmung der Drehimpulsquantenzahl J der autoionisierenden Resonanzen. Die analytische Charakterisierung der dreistufig resonanten Ionisation von Gadolinium ergab eine Isotopen- und Isobarenselektivität von S(Isotop)>10^12 und S(Isobar)>10^7. Die mit dem Diodenlasersystem erreichte Nachweiseffizienz von e=1-3x10^-6 mit einer untergrundlimitierten Nachweisgrenze von wenigen 10^9 Atomen Gd-158 erlaubte erste Demonstrationsmessungen an medizinischen Gewebeproben.

L’attuale condizione che caratterizza il settore energetico richiede un necessario processo di riconversione che, oltre a favorire il risparmio energetico, riduca la dipendenza dai combustibili fossili ed accresca l’impiego di fonti energetiche rinnovabili, dando un contributo fondamentale alla riduzione delle emissioni di gas serra come diversi accordi internazionali richiedono. Si rende pertanto necessario accelerare i processi che da alcuni anni stanno favorendo l’utilizzo di energia da fonti rinnovabili. Tra queste, le fonti legate ai processi di trattamento biologico dei reflui stanno avendo un interessante sviluppo. Esistono numerosi processi biologici che consentono la produzione di energia in maniera indiretta, quali ad esempio i processi di digestione anaerobica finalizzati alla produzione di biogas e/o produzione biologica di idrogeno. In tale contesto si inserisce la tecnologia delle Microbial Fuel Cell, che consente la produzione diretta di energia elettrica, finalizzata al recupero energetico inteso al miglioramento dell’efficienza energetica e alla riduzione dei costi d’esercizio di impianti di trattamento biologico dei reflui. Il presente lavoro di Tesi di Dottorato sperimentale, svoltosi in collaborazione al laboratorio PROT.-IDR. della sede ENEA di Bologna, riporta i risultati dell’attività di ricerca condotta su una MFC (Microbial Fuel Cell) a doppio stadio biologico per il trattamento di reflui ad elevato carico organico e produzione continua di energia elettrica. E’ stata provata l’applicabilità della MFC con entrambi i comparti biotici utilizzando elettrodi di grafite non trattata ottenendo, con un carico organico in ingresso di circa 9 gd-1, valori di potenza massima prodotta che si attestano su 74 mWm-2, corrente elettrica massima generata di 175 mAm-2 ad una tensione di 421 mV, ed una conversione di COD in elettricità pari a 1,2 gCODm-2d-1. I risultati sono stati molto positivi per quanto riguarda le prestazioni depurative ottenute dalla MFC. L’efficienza di depurazione misurata ha raggiunto un valore massimo del 98% di rimozione del COD in ingresso, mentre e la concentrazione di azoto ammoniacale nell’effluente raccolto all’uscita del sedimentatore è sempre stata inferiore a 1 mgN-NH4+l-1. Tra gli obiettivi posti all’inizio della sperimentazione si è rivelata di notevole interesse la valutazione del possibile utilizzo della MFC come sistema per il monitoraggio on-line del COD e degli acidi grassi volatili (VFA) prodotti all’interno di un digestore anaerobico, attraverso la definizione di una correlazione tra i dati elettrici registrati in continuo e le concentrazioni di CODanaer e VFA misurate in diversi periodi della sperimentazione. L’analisi DGGE della biomassa catodica ha fornito uno strumento analitico utile allo studio della diversità della comunità microbica sospesa ed adesa al catodo e ha confermato la forte similarità delle specie batteriche riconosciute nei campioni analizzati. In particolare, le bande di sequenziamento ottenute sono affiliate ai gruppi batterici Firmicutes, -Proteobacteria, -Proteobacteria, -Proteobacteria e Bacteroidetes. Da quanto emerso dalla sperimentazione condotta si può pertanto concludere che ad oggi le MFC sono in fase di evoluzione rispetto ai primi prototipi utilizzati per lo studio delle comunità microbiali e per la comprensione dei meccanismi di trasferimento elettronico. Sfruttarne la potenza prodotta in maniera commerciale diviene una grande sfida per il futuro, ed è opinione comune che le prime applicazioni pratiche delle MFC saranno come fonte di recupero energetico per i dispositivi utilizzati per il monitoraggio dell’ambiente e per il trattamento delle acque reflue.

Il lavoro è stato suddiviso in tre macro-aree. Una prima riguardante un'analisi teorica di come funzionano le intrusioni, di quali software vengono utilizzati per compierle, e di come proteggersi (usando i dispositivi che in termine generico si possono riconoscere come i firewall). Una seconda macro-area che analizza un'intrusione avvenuta dall'esterno verso dei server sensibili di una rete LAN. Questa analisi viene condotta sui file catturati dalle due interfacce di rete configurate in modalità promiscua su una sonda presente nella LAN. Le interfacce sono due per potersi interfacciare a due segmenti di LAN aventi due maschere di sotto-rete differenti. L'attacco viene analizzato mediante vari software. Si può infatti definire una terza parte del lavoro, la parte dove vengono analizzati i file catturati dalle due interfacce con i software che prima si occupano di analizzare i dati di contenuto completo, come Wireshark, poi dei software che si occupano di analizzare i dati di sessione che sono stati trattati con Argus, e infine i dati di tipo statistico che sono stati trattati con Ntop. Il penultimo capitolo, quello prima delle conclusioni, invece tratta l'installazione di Nagios, e la sua configurazione per il monitoraggio attraverso plugin dello spazio di disco rimanente su una macchina agent remota, e sui servizi MySql e DNS. Ovviamente Nagios può essere configurato per monitorare ogni tipo di servizio offerto sulla rete.

Oncolytic virotherapy exploits the ability of viruses to infect and kill cells. It is suitable as treatment for tumors that are not accessible by surgery and/or respond poorly to the current therapeutic approach. HSV is a promising oncolytic agent. It has a large genome size able to accommodate large transgenes and some attenuated oncolytic HSVs (oHSV) are already in clinical trials phase I and II. The aim of this thesis was the generation of HSV-1 retargeted to tumor-specific receptors and detargeted from HSV natural receptors, HVEM and Nectin-1. The retargeting was achieved by inserting a specific single chain antibody (scFv) for the tumor receptor selected inside the HSV glycoprotein gD. In this research three tumor receptors were considered: epidermal growth factor receptor 2 (HER2) overexpressed in 25-30% of breast and ovarian cancers and gliomas, prostate specific membrane antigen (PSMA) expressed in prostate carcinomas and in neovascolature of solid tumors; and epidermal growth factor receptor variant III (EGFRvIII). In vivo studies on HER2 retargeted viruses R-LM113 and R-LM249 have demonstrated their high safety profile. For R-LM249 the antitumor efficacy has been highlighted by target-specific inhibition of the growth of human tumors in models of HER2-positive breast and ovarian cancer in nude mice. In a murine model of HER2-positive glioma in nude mice, R-LM113 was able to significantly increase the survival time of treated mice compared to control. Up to now, PSMA and EGFRvIII viruses (R-LM593 and R-LM613) are only characterized in vitro, confirming the specific retargeting to selected targets. This strategy has proved to be generally applicable to a broad spectrum of receptors for which a single chain antibody is available.

La distribuzione delle Terre Rare (REE) nell’ambiente deriva da una serie di processi interagenti all’interno di un sistema dinamico che include il ciclo idrologico, attività geologiche, fisiche – chimiche (weathering), fenomeni atmosferici e attività umane. Queste contribuiscono alla dispersione superficiale di tutti gli elementi e in particolare di quelli in traccia tra cui i lantanidi (La, Ce, Pr, Nd, Pm, Sm, Eu, Gd, Tb, Dy, Tm, Ho, Er, Yb e Lu) che si ritrovano un po’ ovunque in tutte le matrici, frazionate in modo diverso. L’acqua destinata al consumo alimentare è una di queste. Prelevata di solito da acquiferi profondi, superficiali o da pozzi posti in differenti contesti litologici, può essere soggetta a modifiche importanti delle proprietà fisiche-chimiche e microbiologiche. Più acque minerali relative a marche differenti e conservate in bottiglie di vetro e PET messe a confronto, hanno dimostrato che il vetro è in grado di rilasciare numerosi elementi in funzione della tipologia di acqua contenuta, del tipo di vetro (chiaro o colorato) e delle condizioni predisponenti il rilascio (proprietà intrinseche dell’elemento, fattori esterni quali fonti di calore, ecc.). Terre rare come Ce, La, Pr, Nd, Sm, Eu, Er, Dy, Y e Sc possono essere ceduti in modo differenziale nell’acqua delle bottiglie. In più casi l’arricchimento è evidenziato da acque nelle quali è stata aggiunta anidride carbonica, mentre non si notano cambiamenti eccessivi nel corrispettive naturali. Per i contenitori in PET si registra un marcato rilascio di antimonio (Sb), ampiamente usato sotto forma di triossido di antimonio nel ciclo produttivo delle plastiche. Rispetto alle acque minerali, quelle di rubinetto soffrono gli effetti dei trattamenti di potabilizzazione e dell’interazione con il sistema di distribuzione idrico. La distribuzione dei lantanidi è influenzata dalle condizioni redox (particolarmente per Ce, ed Eu) e dalla relativa capacità di essere complessati e, quindi, mobilizzati in maniera differente. Nonostante il forte impatto delle attività umane, i valori anomali di alcuni (Gd, Nd) sono nell’ordine dei ng/L, come del resto per tutti gli altri e non costituiscono un pericolo per la salute umana. Al fattore geogenico si attribuiscono i tenori maggiori (ΣREE >1000ng/L) riscontrati in Sardegna.

The so called material science is an always growing field in modern research. For the development of new materials not only the experimental characterization but also theoretical calculation of the electronic structure plays an important role. A class of compounds that has attracted a great deal of attention in recent years is known as REME compounds. These compounds are often referred to with RE designating rare earth, actinide or an element from group 1 - 4, M representing a late transition metal from groups 8 - 12, and E belonging to groups 13 - 15. There are more than 2000 compounds with 1:1:1 stoichiometry belonging to this class of compounds and they offer a broad variety of different structure types. Although many REME compounds are know to exist, mainly only structure and magnetism has been determined for these compounds. In particular, in the field of electronic and transport properties relatively few efforts have been made. The main focus in this study is on compounds crystallizing in MgAgAs and LiGaGe structure. Both structures can only be found among 18 valence electron compounds. The f electrons are localized and therefor not count as valence electrons. A special focus here was also on the magnetoresistance effects and spintronic properties found among the REME compounds. An examination of the following compounds was made: GdAuE (E = In, Cd, Mg), GdPdSb, GdNiSb, REAuSn (RE = Gd, Er, Tm) and RENiBi (RE = Pr, Sm, Gd - Tm, Lu). The experimental results were compared with theoretic band structure calculations. The first half metallic ferromagnet with LiGaGe structure (GdPdSb) was found. All semiconducting REME compounds with MgAgAs structure show giant magnetoresistance (GMR) at low temperatures. The GMR is related to a metal-insulator transition, and the value of the GMR depends on the value of the spin-orbit coupling. Inhomogeneous DyNiBi samples show a small positive MR at low temperature that depends on the amount of metallic impurities. At higher fields the samples show a negative GMR. Inhomogeneous nonmagnetic LuNiBi samples show no negative GMR, but a large positive MR of 27.5% at room temperature, which is interesting for application.

In the framework of the micro-CHP (Combined Heat and Power) energy systems and the Distributed Generation (GD) concept, an Integrated Energy System (IES) able to meet the energy and thermal requirements of specific users, using different types of fuel to feed several micro-CHP energy sources, with the integration of electric generators of renewable energy sources (RES), electrical and thermal storage systems and the control system was conceived and built. A 5 kWel Polymer Electrolyte Membrane Fuel Cell (PEMFC) has been studied. Using experimental data obtained from various measurement campaign, the electrical and CHP PEMFC system performance have been determinate. The analysis of the effect of the water management of the anodic exhaust at variable FC loads has been carried out, and the purge process programming logic was optimized, leading also to the determination of the optimal flooding times by varying the AC FC power delivered by the cell. Furthermore, the degradation mechanisms of the PEMFC system, in particular due to the flooding of the anodic side, have been assessed using an algorithm that considers the FC like a black box, and it is able to determine the amount of not-reacted H2 and, therefore, the causes which produce that. Using experimental data that cover a two-year time span, the ageing suffered by the FC system has been tested and analyzed.

Le microreti caratterizzate da unità di generazione con Fonti Energetiche Rinnovabili (FER), sono oggetto di crescente interesse in due contesti molto diversi tra loro: l’integrazione della Generazione Diffusa (GD) in reti di distribuzione pubbliche nei paesi sviluppati e l’elettrificazione di zone rurali attualmente non servite dalla rete elettrica. L’introduzione dei sistemi di accumulo nelle microreti rende possibile la compensazione della variabilità delle FER trasformando le microreti in sistemi di produzione che nel complesso risultano completamente regolabili. L’obiettivo del presente lavoro di tesi è dimostrare come le microreti possono svolgere un ruolo fondamentale per lo sviluppo delle infrastrutture elettriche nei paesi in via di sviluppo e nelle economie emergenti ponendo le basi per l’elettrificazione di aree remote e scarsamente popolate ad oggi non connesse alla rete di distribuzione dell’energia.
