The development of an ultrasensitive biosensor for the low-cost and on-site detection of pathogenic DNA could transform detection capabilities within food safety, environmental monitoring and clinical diagnosis. Herein, we present an innovative approach exploiting endonuclease-controlled aggregation of plasmonic gold nanoparticles (AuNPs) for label-free and ultrasensitive detection of bacterial DNA. The method utilizes RNA-functionalized AuNPs which form DNA-RNA heteroduplex structures through specific hybridization with target DNA. Once formed, the DNA-RNA heteroduplex is susceptible to RNAse H enzymatic cleavage of the RNA probe, allowing the target DNA to liberate and hybridize with another RNA probe. This continuously happens until all of the RNA probes are cleaved, leaving the nanoparticles unprotected and thus aggregated upon exposure to a high electrolytic medium. The assay is ultrasensitive, allowing the detection of target DNA at femtomolar level by simple spectroscopic analysis (40.7 fM and 2.45 fM as measured by UV-vis and dynamic light scattering (DLS), respectively). The target DNA spiked food matrix (chicken meat) is also successfully detected at a concentration of 1.2 pM (by UV-vis) or 18.0 fM (by DLS). In addition to the ultra-high sensitivity, the total analysis time of the assay is less than 3 hours, thus demonstrating its practicality for food analysis.

[EN]This paper describes in detail a real-time multiple face detection system for video streams. The system adds to the good performance provided by a window shift approach, the combination of different cues available in video streams due to temporal coherence. The results achieved by this combined solution outperform the basic face detector obtaining a 98% success rate for around 27000 images, providing additionally eye detection and a relation between the successive detections in time by means of detection threads.

[EN]This paper describes a face detection system which goes beyond traditional approaches normally designed for still images. First the video stream context is considered to apply the detector, and therefore, the resulting system is designed taking into consideration a main feature available in a video stream, i.e. temporal coherence. The resulting system builds a feature based model for each detected face, and searches them using various model information in the next frame. The results achieved for video stream processing outperform Rowley-Kanade's and Viola-Jones' solutions providing eye and face data in a reduced time with a notable correct detection rate.

[EN]Vision-based applications designed for humanmachine interaction require fast and accurate hand detection. However, previous works on this field assume different constraints, like a limitation in the number of detected gestures, because hands are highly complex objects to locate. This paper presents an approach which changes the detection target without limiting the number of detected gestures. Using a cascade classifier we detect hands based on their wrists. With this approach, we introduce two main contributions: (1) a reliable segmentation, independently of the gesture being made and (2) a training phase faster than previous cascade classifier based methods. The paper includes experimental evaluations with different video streams that illustrate the efficiency and suitability for perceptual interfaces.

[EN]This paper describes a real-time approach for face detection and selection of frontal views, for further processing. Typically, face detection papers provide results for a set of single images but the problem of face detection in video streams rarely is tackled. Instead of performing an exhaustive search for every video stream frame a set of opportunistic ideas applied in a cascade fashion and based on temporal and spatial coherence provide promising results in real-time.

[EN]Facial image processing is becoming widespread in human-computer applications, despite its complexity. High-level processes such as face recognition or gender determination rely on low-level routines that must e ectively detect and normalize the faces that appear in the input image. In this paper, a face detection and normalization system is described. The approach taken is based on a cascade of fast, weak classi ers that together try to determine whether a frontal face is present in the image.

[EN]This paper describes an approach for detection of frontal faces in real time (20-35Hz) for further processing. This approach makes use of a combination of previous detection tracking and color for selecting interest areas. On those areas, later facial features such as eyes, nose and mouth are searched based on geometric tests, appearance veri cation, temporal and spatial coherence. The system makes use of very simple techniques applied in a cascade approach, combined and coordinated with temporal information for improving performance. This module is a component of a complete system designed for detection, tracking and identi cation of individuals [1].

[EN]In this paper, a basic conceptual architecture aimed at the design of Computer Vision System is qualitatively described. The proposed architecture addresses the design of vision systems in a modular fashion using modules with three distinct units or components: a processing network or diagnostics unit, a control unit and a communications unit. The control of the system at the modules level is designed based on a Discrete Events Model. This basic methodology has been used to design a realtime active vision system for detection, tracking and recognition of people. It is made up of three functional modules aimed at the detection, tracking, recognition of moving individuals plus a supervision module.

[EN]Enabling natural human-robot interaction using computer vision based applications requires fast and accurate hand detection. However, previous works in this field assume different constraints, like a limitation in the number of detected gestures, because hands are highly complex objects difficult to locate. This paper presents an approach which integrates temporal coherence cues and hand detection based on wrists using a cascade classifier. With this approach, we introduce three main contributions: (1) a transparent initialization mechanism without user participation for segmenting hands independently of their gesture, (2) a larger number of detected gestures as well as a faster training phase than previous cascade classifier based methods and (3) near real-time performance for hand pose detection in video streams.

Les courriels Spams (courriels indésirables ou pourriels) imposent des coûts annuels extrêmement lourds en termes de temps, d’espace de stockage et d’argent aux utilisateurs privés et aux entreprises. Afin de lutter efficacement contre le problème des spams, il ne suffit pas d’arrêter les messages de spam qui sont livrés à la boîte de réception de l’utilisateur. Il est obligatoire, soit d’essayer de trouver et de persécuter les spammeurs qui, généralement, se cachent derrière des réseaux complexes de dispositifs infectés, ou d’analyser le comportement des spammeurs afin de trouver des stratégies de défense appropriées. Cependant, une telle tâche est difficile en raison des techniques de camouflage, ce qui nécessite une analyse manuelle des spams corrélés pour trouver les spammeurs. Pour faciliter une telle analyse, qui doit être effectuée sur de grandes quantités des courriels non classés, nous proposons une méthodologie de regroupement catégorique, nommé CCTree, permettant de diviser un grand volume de spams en des campagnes, et ce, en se basant sur leur similarité structurale. Nous montrons l’efficacité et l’efficience de notre algorithme de clustering proposé par plusieurs expériences. Ensuite, une approche d’auto-apprentissage est proposée pour étiqueter les campagnes de spam en se basant sur le but des spammeur, par exemple, phishing. Les campagnes de spam marquées sont utilisées afin de former un classificateur, qui peut être appliqué dans la classification des nouveaux courriels de spam. En outre, les campagnes marquées, avec un ensemble de quatre autres critères de classement, sont ordonnées selon les priorités des enquêteurs. Finalement, une structure basée sur le semiring est proposée pour la représentation abstraite de CCTree. Le schéma abstrait de CCTree, nommé CCTree terme, est appliqué pour formaliser la parallélisation du CCTree. Grâce à un certain nombre d’analyses mathématiques et de résultats expérimentaux, nous montrons l’efficience et l’efficacité du cadre proposé.

Puccinia psidii (Myrtle rust) is an emerging pathogen that has a wide host range in the Myrtaceae family; it continues to show an increase in geographic range and is considered to be a significant threat to Myrtaceae plants worldwide. In this study, we describe the development and validation of three novel real-time polymerase reaction (qPCR) assays using ribosomal DNA and β-tubulin gene sequences to detect P. psidii. All qPCR assays were able to detect P. psidii DNA extracted from urediniospores and from infected plants, including asymptomatic leaf tissues. Depending on the gene target, qPCR was able to detect down to 0.011 pg of P. psidii DNA. The most optimum qPCR assay was shown to be highly specific, repeatable, and reproducible following testing using different qPCR reagents and real-time PCR platforms in different laboratories. In addition, a duplex qPCR assay was developed to allow coamplification of the cytochrome oxidase gene from host plants for use as an internal PCR control. The most optimum qPCR assay proved to be faster and more sensitive than the previously published nested PCR assay and will be particularly useful for high-throughput testing and to detect P. psidii at the early stages of infection, before the development of sporulating rust pustules.

The present article reflects the progress of an ongoing master’s dissertation on language engineering. The main goal of the work here described, is to infer a programmer’s profile through the analysis of his source code. After such analysis the programmer shall be placed on a scale that characterizes him on his language abilities. There are several potential applications for such profiling, namely, the evaluation of a programmer’s skills and proficiency on a given language or the continuous evaluation of a student’s progress on a programming course. Throughout the course of this project and as a proof of concept, a tool that allows the automatic profiling of a Java programmer is under development. This tool is also introduced in the paper and its preliminary outcomes are discussed.

When performing Particle Image Velocimetry (PIV) measurements in complex fluid flows with moving interfaces and a two-phase flow, it is necessary to develop a mask to remove non-physical measurements. This is the case when studying, for example, the complex bubble sweep-down phenomenon observed in oceanographic research vessels. Indeed, in such a configuration, the presence of an unsteady free surface, of a solid–liquid interface and of bubbles in the PIV frame, leads to generate numerous laser reflections and therefore spurious velocity vectors. In this note, an image masking process is developed to successively identify the boundaries of the ship and the free surface interface. As the presence of the solid hull surface induces laser reflections, the hull edge contours are simply detected in the first PIV frame and dynamically estimated for consecutive ones. As for the unsteady surface determination, a specific process is implemented like the following: i) the edge detection of the gradient magnitude in the PIV frame, ii) the extraction of the particles by filtering high-intensity large areas related to the bubbles and/or hull reflections, iii) the extraction of the rough region containing these particles and their reflections, iv) the removal of these reflections. The unsteady surface is finally obtained with a fifth-order polynomial interpolation. The resulted free surface is successfully validated from the Fourier analysis and by visualizing selected PIV images containing numerous spurious high intensity areas. This paper demonstrates how this data analysis process leads to PIV images database without reflections and an automatic detection of both the free surface and the rigid body. An application of this new mask is finally detailed, allowing a preliminary analysis of the hydrodynamic flow.

Forensic speaker comparison exams have complex characteristics, demanding a long time for manual analysis. A method for automatic recognition of vowels, providing feature extraction for acoustic analysis is proposed, aiming to contribute as a support tool in these exams. The proposal is based in formant measurements by LPC (Linear Predictive Coding), selectively by fundamental frequency detection, zero crossing rate, bandwidth and continuity, with the clustering being done by the k-means method. Experiments using samples from three different databases have shown promising results, in which the regions corresponding to five of the Brasilian Portuguese vowels were successfully located, providing visualization of a speaker’s vocal tract behavior, as well as the detection of segments corresponding to target vowels.
