Myosins are molecular motors associated with the actin cytoskeleton that participate in the mechanisms of cellular motility. During the development of the nervous system, migration of nerve cells to specific sites, extension of growth cones, and axonal transport are dramatic manifestations of cellular motility. We demonstrate, via immunoblots, the expression of myosin Va during early stages of embryonic development in chicks, extending from the blastocyst period to the beginning of the fetal period. The expression of myosin Va in specific regions and cellular structures of the nervous system during these early stages was determined by immunocytochemistry using a polyclonal antibody. Whole mounts of chick embryos at 24-30-h stages showed intense immunoreactivity of the neural tube in formation along its full extent. Cross-sections at these stages of development showed strong labeling in neuroepithelial cells at the basal and apical regions of the neural tube wall. Embryos at more advanced periods of development (48h and 72 h) showed distinctive immunolabeling of neuroepithelial cells, neuroblasts and their cytoplasmic extensions in the mantle layer of the stratified neural tube wall, and neuroblasts and their cytoplasmic extensions in the internal wall of the optic cup, as well as a striking labeling of cells in the apparent nuclei of cranial nerves and budding fibers. These immunolocalization studies indicate temporal and site-specific expression of myosin Va during chick embryo development, suggesting that myosin Va expression is related to recruitment for specific cellular tasks.

Background: Cancer is the second leading cause of death in Argentina, and there is little knowledge about its incidence. The first study based on population-based cancer registry described spatial incidence and indicated that there existed at least county-level aggregation. The aim of the present work is to model the incidence patterns for the most incidence cancer in Córdoba Province, Argentina, using information from the Córdoba Cancer Registry by performing multilevel mixed model approach to deal with dependence and unobserved heterogeneity coming from the geo-reference cancer occurrence. Methods: Standardized incidence rates (world standard population) (SIR) by sex based on 5-year age groups were calculated for 109 districts nested on 26 counties for the most incidence cancers in Cordoba using 2004 database. A Poisson twolevel random effect model representing unobserved heterogeneity between first level-districts and second level-counties was fitted to assess the spatial distribution of the overall and site specific cancer incidence rates. Results: SIR cancer at Córdoba province shown an average of 263.53±138.34 and 200.45±98.30 for men and women, respectively. Considering the ratio site specific mean SIR to the total mean, breast cancer ratio was 0.25±0.19, prostate cancer ratio was 0.12±0.10 and lower values for lung and colon cancer for both sexes. The Poisson two-level random intercepts model fitted for SIR data distributed with overdispersion shown significant hierarchical structure for the cancer incidence distribution. Conclusions: a strong spatial-nested effect for the cancer incidence in Córdoba was observed and will help to begin the study of the factors associated with it.

Crop yield is influenced by several factors with variability in time and space that are associated with the variations in the plant vigor. This variability allows the identification of management zones and site-specific applications to manage different regions of the field. The purpose of this study was the use of multispectral image for management zones identification and implications of site-specific application in commercial cotton areas. Multispectral airborne images from three years were used to classify a field into three vegetation classes via the Normalized Difference Vegetation Index (NDVI). The NDVI classes were used to verify the potential differences between plant physical measurements and identify management zones. The cotton plant measurements sampled in 8 repetitions of 10 plants at each NDVI class were Stand Count, Plant Height, Total Nodes and Total Bolls. Statistical analysis was performed with treatments arranged in split plot design with Tukey’s Test at 5% of probability. The images were classified into five NDVI classes to evaluate the relationship between cotton plant measurement results and sampling location across the field. The results have demonstrated the possibility of using multispectral image for management zones identification in cotton areas. The image classification into three NDVI classes showed three different zones in the field with similar characteristics for the studied years. Statistical differences were shown for plant height, total nodes and total bolls between low and high NDVI classes for all years. High NDVI classes contained plants with greater height, total nodes and total bolls compared to low NDVI classes. There was no difference in Stand Count between low and high NDVI classes for the three studied years. The final plant stand was the same between all NDVI classes for 2001 and 2003 as it was expected due to the conventional seeding application with the same rate of seeds for the entire field.

Photo identification of individual blue whales during summer and autumn off the northwestern Isla de Chiloé, southern Chile, were collected from marine surveys conducted from 2004 to 2006. Re-sightings of individual whales both within and between years may provide evidence of residency and site fidelity by blue whales in the area. These records further document the importance of the northwestern Isla de Chiloé as a feeding area for blue whales. These records also highlight the necessity of further comparisons with photographic catalogues from other areas in southern Chile, off the northwestern coast of South America and the Pacific coast of Central America to better understand seasonal movements, distribution of individuals along the eastern South Pacific, and their wintering areas.

Chelydra serpentine serpentine (Common Snapping Turtle). Nesting Behavior and Site Selection. On May 31, 2008, we observed a ca. 36 cm CL Chelydra serpentine serpentine in unexpected nesting circumstances. Observations were made from 0908 to 1027 h on clear day with ambient temp ca 24° C. The nest site near Longmont, Colorado, USA (40.1599528°N, 105.1259861°W; WGS84) was 4.3 m from a seasonal ditch running north-south to connect two lakes, one on either side of a busy road. The intriguing aspect of the nest location was that it was immediately adjacent to a heavily traveled concrete sidewalk that bordered a road with a high volume of traffic. The frequent foot traffic on the sidewalk and virtually constant vehicle traffic on the adjacent road did not deter the turtle from the particular nest location, even though virtually identical soils, gradients, and directional aspects were available equidistant from the ditch, but away from constant human activity.

CD33 is a myeloid cell surface marker absent on normal hematopoietic stem cells and normal tissues but present on leukemic blasts in 90% of adult and paediatric acute myeloid leukaemia (AML) cases. By virtue of its expression pattern and its ability to be rapidly internalized after antibody binding, CD33 has become an attractive target for new immunotherapeutic approaches to treat AML. In this study two immunoconjugates were constructed to contain a humanised single-chain fragment variable antibody (scFv) against CD33 in order to create new antibody-derived therapeutics for AML. The first immunoconjugate was developed to provide targeted delivery of siRNAs as death effectors into leukemic cells. To this purpose, a CD33-specific scFv, modified to include a Cys residue at its C-terminal end (scFvCD33-Cys), was coupled through a disulphide bridge to a nona-d-arginine (9R) peptide carrying a free Cys to the N-terminal. The scFvCD33-9R was able to completely bind siRNAs at a protein to nucleic acid ratio of about 10:1, as confirmed by electrophoretic gel mobility-shift assay. The conjugate was unable to efficiently transduce cytotoxic siRNA (siTox) into the human myeloid cell line U937. We observed slight reductions in cell viability, with a reduction of 25% in comparison to the control group only at high concentration of siTox (300 nM). The second immunoconjugate was constructed by coupling the scFvCD33-Cys to the type 1 ribosome inactivating protein Dianthin 30 (DIA30) through a chemical linking The resulting immunotoxin scFvCD33-DIA30 caused the rapid arrest of protein synthesis, inducing apoptosis and leading ultimately to cell death. In vitro dose-dependent cytotoxicity assays demonstrated that scFvCD33-DIA30 was specifically toxic to CD33-positive cell U937. The concentration needed to reach 50 % of maximum killing efficiency (EC50) was approximately 0.3 nM. The pronounced antigen-restricted cytotoxicity of this novel agent makes it a candidate for further evaluation of its therapeutic potential.

It was observed in the ‘80s that the radiation damage on biological systems strongly depends on processes occurring at the microscopic level, involving the elementary constituents of biological cells. Since then, lot of attention has been paid to study elementary processes of photo- and ion-chemistry of isolated organic molecule of biological interest. This work fits in this framework and aims to study the radiation damage mechanisms induced by different types of radiations on simple halogenated biomolecules used as radiosensitizers in radiotherapy. The research is focused on the photofragmentation of halogenated pyrimidine molecules (5Br-pyrimidine, 2Br-pyrimidine and 2Cl-pyrimidine) in the VUV range and on the 12C4+ ion-impact fragmentation of the 5Br-uracil and its homogeneous and hydrated clusters. Although halogen substituted pyrimidines have similar structure to the pyrimidine molecule, their photodissociation dynamics is quite different. These targets have been chosen with the purpose of investigating the effect of the specific halogen atom and site of halogenation on the fragmentation dynamics. Theoretical and experimental studies have highlighted that the site of halogenation and the type of halogen atom, lead either to the preferential breaking of the pyrimidinic ring or to the release of halogen/hydrogen radicals. The two processes can subsequently trigger different mechanisms of biological damage. To understand the effect of the environment on the fragmentation dynamic of the single molecule, the ion-induced fragmentation of homogenous and hydrated clusters of 5Br-uracil have been studied and compared to similar studies on the isolated molecule. The results show that the “protective effect” of the environment on the single molecule hold in the homogeneous clusters, but not in the hydrated clusters, where several hydrated fragments have been observed. This indicates that the presence of water molecules can inhibit some fragmentation channels and promote the keto-enol tautomerization, which is very important in the mutagenesis of the DNA.

Der Fokus dieser Arbeit lag in der Synthese von funktionellen HPMA-Copolymeren, sowohl für die Darstellung definierter Polymer-Antikörper Konjugate, als auch zum effizienten Transport von p-DNA in Polymer-DNA Komplexen (Polyplexe). Nach ausführlicher physikalischer und chemischer Charakterisierung wurden gezielt ihre Wechselwirkungen mit (Immun)-Zellen untersucht und so ihr Potential für die Verwendung in der Tumor-Immuntherapie aufgezeigt.rnFür das gezielte Ansprechen von bestimmten Immunzellen mit Schlüsselfunktionen besitzen monoklonale Antikörper ein großes Potential. Im Rahmen dieser Arbeit gelang die Darstellung definierter Polymer-Antikörper Konjugate über das gezielte Einführen von Thiol-Gruppen an Antikörper und die Synthese eng verteilter, Maleinimid funktionalisierter HPMA-Copolymere. Diese sehr gut definierten, funktionellen HPMA-Copolymere konnten über die Kombination der RAFT-Polymerisation und Reaktivester Polymeren gewonnen werden. Unterschiedliche Polymerstrukturen ermöglichten die Synthese verschiedener Arten von Polymer-Antikörper Konjugaten. Speziell die Untersuchung der verschiedenen Konjugate aus dem für dendritische Zellen spezifischen aDEC-205 Antikörper an Immunzellen aus dem Knochenmark von Mäusen lieferten wertvolle Erkenntnisse über Struktur-Wirkungsbeziehungen und zeigten die Möglichkeit der gezielten Adressierung von Immunzellen mit Schlüsselfunktionen bei der Aktivierung einer (Tumor)-Immunabwehr am Beispiel von dendritischen Zellen. Gleichzeitig erlaubt der Syntheseweg sowohl die gleichzeitige und kontrollierte Einführung auch komplexerer Stimuli am Polymerrückgrat als auch die Verwendung verschiedener Antikörper.rnÜber die Kombination der RAFT-Polymerisation und polymeren Reaktivestern wurde ebenso die Synthese von neuartigen kationisch-hydrophilen Polylysin-b-poly(HPMA) Blockcopolymeren als effiziente Transporter für den komplexen aber wirkungsvollen Wirkstoff p-DNA in Form von Polymer-DNA Komplexen (Polyplexe) realisiert. Da diese Polyplexe gleichzeitig eine Abschirmung der sensitiven p-DNA über eine poly(HPMA)-Korona vermitteln, stellen sie allgemein ein geeignetes Transportmittel für einen therapeutischen Transport von p-DNA dar. Diese Polyplexe sind in der Lage, humane Nierenkarzinomzellen (HEK-293T Zelllinie) zu transfizieren ohne signifikante Zytotoxizität zu zeigen. Darüber hinaus gelang eine große Steigerung der Transfektionseffizienz, ohne eine gleichzeitige Erhöhung der Zytotoxizität, durch die gezielte Einführung von Redox-stimuliresponsiven Disulfid-Gruppen zwischen den einzelnen Blöcken. Diese Polyplexe stellen einen polymeren Vektor zur transkriptionellen Regulierung von Zellen dar, zum Beispiel für die transkriptionelle Aktivierung von dendritischen Zellen, durch die Verwendung speziell dafür modifizierter p-DNA-Konstrukte. rnDurch die Verknüpfung einer ortsspezifischen enzymatischen Kopplung und kupferfreien Cyclooctin-Azid Kupplung gelang die kontrollierte und kovalente Modifizierung von polymeren Mizellen mit aDEC-205 Antikörpern an der hydrophilen poly(HPMA)-Korona. Diese Methode bietet die Möglichkeit der Anbindung der effektiven aber anspruchsvollen Erkennungsstruktur Antikörper an komplexere Polymerstrukturen und andere nano-partikulären Systeme, zum Beispiel an die zuvor genannten Polyplexe, um eine zellspezifische und verbesserte Aufnahme und Prozessierung zu erreichen.rnDiese Studien zeigen somit, sowohl die Möglichkeit der selektiven Addressierung von Immunzellen mit Schlüsselfunktionen wie dendritischer Zellen, als auch die Möglichkeit der transkriptionellen Regulation von Zellen durch Polyplexe. Sie stellen somit einen ersten Schritt zur Herstellung funktioneller, nanopartikulärer Systeme zur Verwendung in der Tumor-Immuntherapie dar. rn

IgE antibodies bind the high-affinity IgE Fc receptor (FcεRI), found primarily on mast cells and basophils, and trigger inflammatory cascades of the allergic response. Inhibitors of IgE-FcεRI binding have been identified and an anti-IgE therapeutic antibody (omalizumab) is used to treat severe allergic asthma. However, preformed IgE-FcεRI complexes that prime cells before allergen exposure dissociate extremely slowly and cannot be disrupted by strictly competitive inhibitors. IgE-Fc conformational flexibility indicated that inhibition could be mediated by allosteric or other non-classical mechanisms. Here we demonstrate that an engineered protein inhibitor, DARPin E2_79 (refs 9, 10, 11), acts through a non-classical inhibition mechanism, not only blocking IgE-FcεRI interactions, but actively stimulating the dissociation of preformed ligand-receptor complexes. The structure of the E2_79-IgE-Fc(3-4) complex predicts the presence of two non-equivalent E2_79 sites in the asymmetric IgE-FcεRI complex, with site 1 distant from the receptor and site 2 exhibiting partial steric overlap. Although the structure is indicative of an allosteric inhibition mechanism, mutational studies and quantitative kinetic modelling indicate that E2_79 acts through a facilitated dissociation mechanism at site 2 alone. These results demonstrate that high-affinity IgE-FcεRI complexes can be actively dissociated to block the allergic response and suggest that protein-protein complexes may be more generally amenable to active disruption by macromolecular inhibitors.

The oxygen consumption (VO2 microL/h/mg) of sham and of traumatized rat brains within 30 min and 6 h after a lateral fluid percussion injury (FPI) was measured with the Cartesian microrespirometer. Brain slices were cut at the plain of injury and site-specific 20-60-microg cores of tissue were transferred to the microrespirometer. In sham brains, the cortical VO2 (CVO2) was 13.78+/-0.64 and the hippocampal VO2 (HPVO2) was 11.20+/-0.58 microL/h/mg (p<0.05). Within 30 min of the injury, the respective values of 16.89+/-0.55 and 14.91+/-0.06 were significantly increased (p<0.05). The combined VO2 (CVO2, HPVO2) of 12.49+/-0.06 microL/h/mg in shams was significantly less than the combined VO2 of 15.90+/-0.59 microL/h/mg at 30 min post FPI (p<0.001). The maximal CVO2 of 19.49+/-1.10 microL/h/mg and the maximal HPVO2 of 15.98+/-0.99 microL/h/mg were both obtained from the ipsilateral side of the injury. Whereas the contralateral cortical value for injured brains was not significantly different from that of the shams, both ipsilateral and contralateral hippocampal values were significantly greater than that of the shams in response to injury (p<0.05). By 6 h postinjury, the combined VO2 had dropped to 10.01+/-0.84 microL/h/mg but was not significantly lower than the sham values. The data indicate that normal CVO2 is greater than normal HPVO2. The FPI produces significant increases in both CVO2 and HPVO2. Also, while the immediate increase in CVO2 appears to be injury-site dependent, that is, regional, the increase in HPVO2 appears to be global.
