Objective: To assess 3D morphological variations and local and systemic biomarker profiles in subjects with a diagnosis of temporomandibular joint osteoarthritis (TMJ OA).Design: Twenty-eight patients with long-term TMJ OA (39.9 +/- 16 years), 12 patients at initial diagnosis of OA (47.4 +/- 16.1 years), and 12 healthy controls (41.8 +/- 12.2 years) were recruited. All patients were female and had cone beam CT scans taken. TMJ arthrocentesis and venipuncture were performed on 12 OA and 12 age-matched healthy controls. Serum and synovial fluid levels of 50 biomarkers of arthritic inflammation were quantified by protein microarrays. Shape Analysis MANCOVA tested statistical correlations between biomarker levels and variations in condylar morphology.Results: Compared with healthy controls, the OA average condyle was significantly smaller in all dimensions except its anterior surface, with areas indicative of bone resorption along the articular surface, particularly in the lateral pole. Synovial fluid levels of ANG, GDF15, TIMP-1, CXCL16, MMP-3 and MMP-7 were significantly correlated with bone apposition of the condylar anterior surface. Serum levels of ENA-78, MMP-3, PAI-1, VE-Cadherin, VEGF, GM-CSF, TGF beta b1, IFN gamma g, TNF alpha a, IL-1 alpha a, and IL-6 were significantly correlated with flattening of the lateral pole. Expression levels of ANG were significantly correlated with the articular morphology in healthy controls.Conclusions: Bone resorption at the articular surface, particularly at the lateral pole was statistically significant at initial diagnosis of TMJ OA. Synovial fluid levels of ANG, GDF15, TIMP-1, CXCL16, MMP-3 and MMP-7 were correlated with bone apposition. Serum levels of ENA-78, MMP-3, PAI-1, VE-Cadherin, VEGF, GM-CSF, TGF beta 1, IFN gamma, TNF alpha, IL-1 alpha, and IL-6 were correlated with bone resorption. Uremic syndrome, arising from kidney malfunction, consists of a set of systemic changes caused by the accumulation of toxic substances to the body. Since, with the advance of medicine, the animals lived more reaching advanced age and entering thus on track of senility, chronic renal disease, became a common complaint in the routine of the ambulatory. This high rate of morbidity generates an increased need for clarification of pathophysiology involved in this disease. The components of the uremic syndrome include water and sodium imbalance, anemia, intolerance to carbohydrate, neurological disorders, disorders of the gastrointestinal tract, osteoarthritis, immunological incompetence and metabolic acidosis. The clinical manifestations occur in isolation or in combination. In most cases canines patients are subject to an assessment when the kidney disease has evolved to the final stage with uremic syndrome and installed already, under these conditions, the prognosis is reserved

Osteoarticular diseases are very frequent in small animals, especially dogs. Osteoarthritis is the most common degenerative arthropathy in cats and dogs. It is one of the most important skeletal disease in small animals and it is divided into two broad classes: secondary and genetics. The secondary is originated from situations that can cause joint instability like as trauma, ligament problems and obesity among others. The gene for osteoarthritis comes from hereditary problems such as hip dysplasia, elbow dysplasia and other diseases that can cause articular instability. The hereditary osteoarticular diseases are important only because they can result in the osteoarthritis. It is important to prevent the joint instability and consequently the clinical signs that affect the quality of life of the animals. The most common clinical signs are intermittent claudication, a decrease in performance, reluctance to jump and climb ladders among others. To prevent the clinical signs or mitigate them, the treatments need to be focused on pain control and especially restoration of the affected joint. The treatments most used are the medical and surgery. Anti-inflammatory and analgesics are used in large scale for pain control and to inhibit inflammatory mediators. Furthermore, medical treatment includes weight reduction, physiotherapy and proper nutrition along with anti-inflammatory and analgesics. Surgical treatments are indicated if there is an inadequate response to medical treatment. Physiotherapy is often used in conjunction with other treatments. Besides the reduction of weight is essential and obesity is unacceptable. Cats also suffer from osteoarthritis, but the diagnosis is rarer than in dogs. The trimming and shoeing are of great significance in the performance and longevity of the athlete horse, and if not done or done incorrectly, cause a variety of limb injuries, which may make unusable the animal for sport, a fact of frequent occurrence, due to the small number of trained professionals in this area. The anatomy of the hoof and its functions should be maintained by maintaining the balance of the same (correct hoof trimming) and, if necessary, proper shoeing, all to avoid the hull balance alterations that lead to osteoarthritis, musculoskeletal disorders, chronic pain at the bead, synovitis, pedal osteitis, navicular disease, in addition to increase tension in the flexor tendons, suspensory ligament and proximal sesamoid causing tendinitis, desmitis and sesamoiditis proximal

Articular cartilage is the structure that coats the bone ends in regions where two bones are articulated, allowing movement. It has inefficient intrinsic and extrinsic mechanisms of repair, usually resulting in fibrocartilage formation after injury. Such repair have lower strength, stiffness and usability features when compared to hyaline cartilage. The mesenchymal stem cells have the potential to regenerate tissue without the production of scar, and because of this feature it is well studied. But to have its maximum chondrogenic potential, it is necessary to use scaffolds and growth factors. Biomaterials play the role of scaffold for the cells allowing them to become attached, grow and produce extracellular matrix, leading to formation of repair with hyaline cartilage. In this sense, the purpose of this study is to provide information on the various studies using cell therapy and / or biomaterials to produce hyaline cartilage

Structured AbstractObjectivesTo investigate the 3D morphological variations in 169 temporomandibular ioint (TMJ) condyles, using novel imaging statistical modeling approaches.Setting and sample populationThe Department of Orthodontics and Pediatric Dentistry at the University of Michigan. Cone beam CT scans were acquired from 69 subjects with long-term TMJ osteoarthritis (OA, mean age 39.115.7years), 15 subjects at initial consult diagnosis of OA (mean age 44.914.8years), and seven healthy controls (mean age 4312.4years).Materials and methods3D surface models of the condyles were constructed, and homologous correspondent points on each model were established. The statistical framework included Direction-Projection-Permutation (DiProPerm) for testing statistical significance of the differences between healthy controls and the OA groups determined by clinical and radiographic diagnoses.ResultsCondylar morphology in OA and healthy subjects varied widely with categorization from mild to severe bone degeneration or overgrowth. DiProPerm statistics supported a significant difference between the healthy control group and the initial diagnosis of OA group (t=6.6, empirical p-value=0.006) and between healthy and long-term diagnosis of OA group (t=7.2, empirical p-value=0). Compared with healthy controls, the average condyle in OA subjects was significantly smaller in all dimensions, except its anterior surface, even in subjects with initial diagnosis of OA.ConclusionThis new statistical modeling of condylar morphology allows the development of more targeted classifications of this condition than previously possible.

Low back pain is considered a major cause of lameness and performance decay, but identify the location of the lesions and quantify the intensity of pain is difficult because often the most obvious clinical sign, in some cases, is not the pain itself, but, low athletic performance. The diagnosis and treatment of this disease are associated with a high cost and have a strong impact on the equine industry, since it results in direct economic losses. The diagnosis of low back pain is made through physical examination and additional tests, including radiography, ultrasound, thermography and blockades. Major injuries causing back pain in horses is the contact or overlap of the spinous processes, supraspinous desmitis, osteoarthritis of the articular processes, vertebral body lesions and myositis. Therapeutic measures used in the treatment of these diseases are systemic inflammatory steroid, local infiltration, acupuncture, chiropractic, physical therapy, changes in training and surgeries.
