Resumo:
Conventional descriptions place conservation activities between the two poles of active restorative intervention and passive abandonment. This paper proposes that site stewards at mining heritage sites often follow presentation strategies that sit outside this neat dualism. Drawing on material presented in the form of three case studies, this paper identifies the actions these strategies entail and considers the results in terms of an aesthetic of decay. To consolidate the argument, a new overarching term is introduced to describe this strategy: contrived dereliction, in order to foreground its essential features. The paper then outlines the advantages, limitations and requirements of contrived dereliction as a heritage management and presentation practice.

La colonie de Jacques Cartier et de Jean-François de la Rocque de Roberval de 1541-1543 au cap Rouge, près de Québec, est aujourd’hui au coeur d’une patrimonialisation sans précédent. L’intérêt récent pour cette première tentative coloniale française en Amérique du Nord, un pan de géohistoire oublié, s’explique par un changement de discours qui s’est dessiné au début du 21e siècle, coïncidant ainsi avec la mise au jour et la mise en valeur du site archéologique Cartier-Roberval. L’histoire prendra donc une nouvelle tournure, il ne sera plus question de tentative mais plutôt d’une colonie à part entière. Le site Cartier-Roberval se voit aujourd’hui aux premières loges d’un grand mouvement commémorateur qui mise et met en valeur sa singularité. L’étude critique sur les processus de patrimonialisation à l’oeuvre sur le site en question permet de mieux comprendre l’ampleur et le dénouement possible du phénomène qui s’opère actuellement.

The cytokine hormone leptin is a key signalling molecule in many pathways that control physiological functions. Although leptin demonstrates structural conservation in mammals, there is evidence of positive selection in primates, lagomorphs and chiropterans. We previously reported that the leptin genes of the grey and harbour seals (phocids) have significantly diverged from other mammals. Therefore we further investigated the diversification of leptin in phocids, other marine mammals and terrestrial taxa by sequencing the leptin genes of representative species. Phylogenetic reconstruction revealed that leptin diversification was pronounced within the phocid seals with a high dN/dS ratio of 2.8, indicating positive selection. We found significant evidence of positive selection along the branch leading to the phocids, within the phocid clade, but not over the dataset as a whole. Structural predictions indicate that the individual residues under selection are away from the leptin receptor (LEPR) binding site. Predictions of the surface electrostatic potential indicate that phocid seal leptin is notably different to other mammalian leptins, including the otariids. Cloning the grey seal leptin binding domain of LEPR confirmed that this was structurally conserved. These data, viewed in toto, support a hypothesis that phocid leptin divergence is unlikely to have arisen by random mutation. Based upon these phylogenetic and structural assessments, and considering the comparative physiology and varying life histories among species, we postulate that the unique phocid diving behaviour has produced this selection pressure. The Phocidae includes some of the deepest diving species, yet have the least modified lung structure to cope with pressure and volume changes experienced at depth. Therefore, greater surfactant production is required to facilitate rapid lung re-inflation upon surfacing, while maintaining patent airways. We suggest that this additional surfactant requirement is met by the leptin pulmonary surfactant production pathway which normally appears only to function in the mammalian foetus.

The Ythan Estuary in Aberdeenshire is a Site of Special Scientific Interest due to the numerous protected species which incorporate it into their ecosystem. In recent years algae growth in the estuary has been seen to increase, causing concern over the integrity of the estuary as a habitat for wildlife. By examining sediment, river flow, nutrient levels and temperature it is possible to identify the driving factors behind this increased algal growth. Using data gathered in the field as well as historical climate data the contribution of each of these factors to algae growth can be determined.

O nitrogênio e um dos nutrientes mais demandados pelas espécies vegetais, sua presença no solo, sob formas orgânicas ou minerais disponíveis para as plantas, está vinculada à qualidade e quantidade dos resíduos vegetais aportados ao solo. O estudo teve o objetivo de avaliar a influência do cultivo do eucalipto e da acácia na composição das formas orgânicas e inorgânicas de N e, na abundância natural de 15N em um Argissolo Amarelo. Para isso, foram coletadas amostras de solo e serapilheira em monocultivos do Eucalyptus urograndis (clone do Eucalyptus urophylla S. T. Blake x Eucalyptus grandis W. Hill ex Spreng) de ciclo curto (sete anos), sistemas de cultivo de rotação com acácia ( Acacia mangium Willd.) após monocultivo de eucalipto, monocultivo de eucalipto de ciclo longo (24 anos) e mata nativa (Mata Atlântica) como condição original de solo do litoral Norte do Espírito do Santo. Foram avaliados os teores de C orgânico total, N total, N-NH4+, N-NO3-, relação C/N, fracionamento do N orgânico e abundância natural de 15N no solo e serapilheira. Das formas de N-orgânico hidrolisado, o N-amino foi a fração que apresentou maior contribuição (39%), seguida pela fração de N-não identificado (27%), da fração N-amida (18%) e N-hexosamina (15%). O povoamento de acácia promoveu menor abundância natural de 15N e maiores teores de N total e C orgânico no solo e aumentou as formas orgânicas de N-hidrolisado, quando comparado àqueles de eucalipto de ciclo curto. Isso indica o aumento de formas lábeis de N orgânico no solo para as plantas e redução da humificação da matéria orgânica do solo (MOS) de acácia. Nesse sentido, a rotação de cultivos florestais com acácia após eucalipto de ciclo curto contribuiu para o aumento de formas orgânicas no solo, importantes para a nutrição de plantas, por serem potenciais fontes de nutrientes às plantas em curto período de tempo.

O Castelo de Noudar está implantado num íngreme promontório da herdade da Coitadinha, a Noroeste de Barrancos, integrando o atual Parque da Natureza de Noudar. Construído a partir de 1303, foi alvo de diversas transformações ao longo do tempo, para responder às exigências militares de cada época, e chega aos nossos dias num avançado estado de decadência física. A investigação desenvolvida assenta, em grande parte, no desenho: de Noudar a uma escala territorial, como elemento que participa numa rede fortificada complexa; da evolução morfológica e funcional, que nos permite ter noção das várias configurações do castelo ao longo dos séculos; num desenho propositivo que desenvolve uma estratégia para valorização do lugar e do seu património material e imaterial. Hoje é fundamental compreender como as valências do Parque e da fortaleza se podem cruzar? Este trabalho pretende refletir sobre possibilidades de valorização para contrariar a degradação em que a fortaleza se encontra; ABSTRACT: The Noudar Castle is located in a steep promontory in the herdade da Coitadinha, Northeast from Barrancos, part of the Natural Park of Noudar. Its construction began in 1303, and underwent several transformations throughout its existence, adapting to the military advancements thru out the ages. Today, it shows an advanced state of decay. The investigation is focused on the design of Noudar on a territorial scale, taking part in a complex network of fortified systems; its functional and morphological evolution, which allows us to comprehend the configurations of the castle throughout the centuries; and on a proposed design that develops a strategy to enhance and bring value to this place and its material and non-material heritage. Today it's crucial to comprehend how can the values of the Park and fortress intertwine. This thesis reflects on the possibilities of valuing this site, counter acting the decay which has fallen upon it.

The growing concern about the depletion of oil has spurred worldwide interest in finding alternative feedstocks for important petrochemical commodities and fuels. On the one hand, the enormous re-serves found (208 trillion cubic feet proven1), environmental sustainability and lower overall costs point to natural gas as the primary source for energy and chemicals in the near future.2 Nowadays the transformation of methane into useful chemicals and liquid fuels is only feasible via synthesis gas, a mixture of molecular hydrogen and carbon monoxide, that is further transformed to methanol or to hydrocarbons under moderate reaction conditions (150-350 °C and 10-100 bar).3 For a major cost reduction and in order to valorize small natural gas sources, either more efficient "syngas to products" catalysts should be produced or the manner in which methane is initially activated should be changed, ideally by developing catalysts able to directly oxidize methane to interesting products such as methanol. On the other hand, from the point of view of CO2 emissions, the use of the re-maining fossil resources will further contribute to global warming. In this scenario, the development of efficient routes for the transformation of CO2 into useful chemicals and fuels would represent a considerable step forward towards sustainability. Indeed, the environmental and economic incen-tives to develop processes for the conversion of CO2 into fuels and chemicals are great. However, for such conversions to become economically feasible, considerable research is necessary. In this lecture we will summarize our recent efforts into the design of new catalytic systems, based on MOFs and COFs, to address these challenges. Examples include the development of new Fe based FTS catalysts, electrocatalysts for the selective conversion of CO2 into syngas, the development of efficient catalysts for the utilization of formic acid as hydrogen storage vector and the development of new enzyme inspired systems for the direct transformation of methane to methanol under mild reaction conditions. Resumo O estudo realizado no âmbito deste relatório centra-se numa análise do papel do turista na conservação do património natural e cultural, em particular do Parque Nacional da Tijuca, no Rio de Janeiro. Os objetivos definidos para este trabalho são dilucidar o conceito de turismo cultural nas suas inter-relações com o ecoturismo; compreender o estado da arte em relação à intervenção do terceiro setor (ONG associações, fundações) e do turista particular na conservação do património; caracterizar o Parque Nacional da Tijuca; caracterizar a Amigos do Parque; e por fim indagar sobre a predisposição do turista para se associar à Associação dos Amigos do Parque Nacional da Tijuca. Para isso foi concebido um inquérito, realizado pela estagiária aos turistas que visitavam o parque, fazendo assim uma recolha de dados que serviram de base para as conclusões deste trabalho, levando no fim à proposta de algumas estratégias e diretivas a seguir para atrair novos associados, em particular turistas. Os resultados levam a concluir que no geral o turista não está predisposto a contribuir para a conservação e associar-se à Amigos do Parque. A percentagem de turistas que afirmaram estar predispostos a associarem-se foi de apenas 5%, sendo maioritariamente de turistas com idades entre os 25 e os 44 anos, com formação superior e provenientes dos países da América do Sul. O turista foi também questionado quanto à predisposição para fazer uma doação pontual, para a qual a taxa de respostas positiva foi mais elevada com 22%, com a contribuição também de turistas europeus. Apesar destes valores, o número de visitantes anuais do Parque Nacional da Tijuca aumenta a cada ano (prevendo-se que continue a aumentar), sendo que em 2014 foi de 3.086.207 de pessoas, e destes, cerca de 72% foram turistas. É importante conseguir o apoio destes turistas através da associação e através de doação, já que é relevante não só a nível financeiro, como também a nível de internacionalização e reconhecimento da associação e do parque, o que levará a uma maior rede de associados. Palavras-chave: ecoturismo; terceiro setor; conservação; Associação dos Amigos do Parque Nacional da Tijuca vii Abstract The study in this report focuses on an analysis of the tourist role in the conservation of natural and cultural heritage, in particular of the Tijuca National Park in Rio de Janeiro. The goals set for this work are to elucidate the concept of cultural tourism in its interrelations with ecotourism; to understand the state of the art concerning the third sector (NGO associations, foundations) and private tourist in heritage conservation; to characterize the Tijuca National Park; to characterize the Friends of the Park; and finally to inquire about the willingness of the tourist to join the Association of Friends of the Tijuca National Park. For this a survey was designed and conducted by the intern to the tourists visiting the park, thus allowing for a collection of data that formed the basis to the conclusions of this work, leading at the end to the proposal of some strategies and policies to attract new members, particularly tourists. The results lead to the conclusion that as a rule the tourist is not willing to contribute to the conservation and join the Friends of the Park. The percentage of tourists who assumed they were willing to join the association was only 5%, and mostly tourists aged 25 and 44, with higher education, and from the South American countries. Tourists were also asked about their willingness for giving donations, for which the rate of positive responses was higher 22%, with the contribution of European tourists. Despite these numbers, the number of annual visitors to the Tijuca National Park increases every year (and it is expected to continue to increase). In 2014 ir received 3,086,207 visitors, and of these, about 72% were tourists. It is important to get the support of these tourists through association and through donation, since it is relevant not only financially, but also as a tool of the internationalization and recognition of the association and the park, which will lead to a larger network of associates. In today’s world heritage worldwide are at the risk not only because of natural process of decay and destruction but also by social change like urbanization, globalization and homogenization of cultures. With these emerging problems, the heritage conservation discourse also has reached to a new dimension including broader range of concepts like tangible heritage, intangible heritage, community participation, indigenous knowledge and many more. Even with the changing scenario in the international context about the heritage conservation, Nepal’s heritage conservation still focus on monuments, sites and buildings. In add to that the conservation practices are still top-down approach and community involvements are limited only in plans. While numerous intangible heritages like masking dances chariot processions, festivals and rituals, which form an integral part of the daily social life of people are still being continued and managed by the community and its people, without with out serious attention form the government. In Kathmandu Valley these heritages has been maintained with the traditional social association of people known as “Guthi” which has been continuing since 5th Century. Most of the tangible and intangible heritages have survived for centuries because of this unique association of people. Among the numerous festivals of the Kathmandu Valley, the festival Yenya Punhi was chosen as a case for this study, which is also a major festival of Kathmandu. This festival is the perfect example for the study as its celebrated in the city that is the most urbanized city of Nepal with the challenges of the every modern city like social changes and urbanization. Despite modern challenges Guthi still plays a major role in the heritage conservation in Kathmandu Valley. Now there are some interventions of the various formal institutions. So this study will be focusing on the management, continuity and problems of the festival along with Nepal’s position in terms of intangible heritage conservation. The problem of Kathmandu and Yenya Punhi festival is the problem of every country in the similar situation so with this case study it can be a good example for finding solutions of the similar problem not only the other festivals within Nepal but also elsewhere in the world; Resumo: Conexão de Património: Festival Yenya Punhi um caminho de fortalecimento de identidade: A experiência de Catmandu Nos dias de hoje, os patrimónios mundiais encontram-se em risco, não só devido ao processo natural de degradação e destruição, mas também pelas mudanças sociais, tais como a urbanização, globalização e homogeneização de culturas. Com o emergir destes problemas, o discurso de conservação de Património atingiu também uma nova dimensão, incluíndo uma área mais abrangente de conceitos, como por exemplo, património material, património imaterial, participação da comunidade, conhecimento indígena, entre outros. Mesmo com este cenário de mudança no contexto mundial de conservação do património, a preservação do património do Nepal continua a focar-se em monumentos, sítios e edíficios. A acrescentar a isso, as práticas de conservação ainda têm uma abordagem descendente e os envolvimentos da comunidade são limitados por planificações. Enquanto que os numerosos patrimónios imateriais como danças com máscaras, procissões, festivais e rituais, os quais formam uma parte integral da vida diária social das pessoas que as continuam e as gerem em comunidade, sem uma atenção séria por parte do governo. No Vale de Catmandu, este património tem sido mantido pela associação tradicional de pessoas conhecidas como ''Guthi'' desde o século V. A maior parte destes patrimónios materiais e imateriais tem sobrevivido durante séculos graças a esta associação única de pessoas. Entre os numerosos festivais do Vale de Catmandu, o festival Yenya Puhni foi escolhido para este estudo, pois é também um grande festival em Catmandu. Este festival é o exemplo perfeito para este estudo, pois é celebrado na cidade mais urbanizada do Nepal, com os desafios das cidades modernas tais como mudanças sociais e urbanização. Apesar dos desafios da modernização, os ''Guthi'' ainda desempenham um papel importante na preservação do património do Vale de Catmandu. Agora, existem algumas intervenções de várias instituições formais Então, este estudo irá focar-se na gestão, continuidade e problemas do festival, juntamente com a posição do Nepal em termos de conservação de património imaterial. O problema de Catmandu e do festival Yenya Punhi é o problema de todos os países em situação semelhante então, este estudo pode ser um bom exemplo para encontrar soluções de problemas parecidos, não só em outros festivais no Nepal mas também para qualquer parte do mundo.

Natural stone has been a popular and reliable building material throughout history appearing in many historic monuments and in more recent buildings. Research into the intrinsic properties of specific stones is important because it gives us a greater understanding of the factors that limit and act on them. This can help prevent serious problems from occurring in our buildings bringing both esthetic benefits and financial savings. To this end, the main objective of this research has been to study the influence of the fabric and the mineral composition of two types of sandstone on their durability. The first is a red continental sandstone from the Buntsandstein Age called “Molinaza Roja”, which is quarried in Montoro (Cordoba). The second is quarried in Ronda (Malaga) and is sold under the trade name of “Arenisca Ronda”. It is a light pink-whitish calcarenite deposited during the Late Tortonian to Late Messinian. We characterized their petrological and petrophysical properties by studying their rock fabrics, porous systems and mechanical properties. In order to obtain a complete vision of the behavior of their rock fabrics, we also carried out two decay tests, the salt crystallization and the freeze–thaw tests. We then measured the effects on the textures of the altered samples during and after the decay tests and we evaluated the changes in the porous system. By comparing the results between intact and altered samples, we found that Arenisca Ronda is less durable because it has a high quantity of expandable clays (smectites) and a high percentage of pores in the 0.1–1 μm range, in which the pressure produced by salt crystallization is strongest. In Molinaza Roja the decay agents caused significant sanding due to loss of cohesion between the clasts, especially during the salt crystallization test. In both stones, the anisotropies (oriented textures) have an important role in their hydric and dynamic behavior and also affect their mechanical properties (especially in the compression resistance). No changes in color were detected.

À travers l’augmentation des désastres dits « naturels » au cours de la dernière décennie, des populations se sont retrouvées soudainement sans maison, sans endroit où loger. L’absence d’endroit où loger amènera donc les populations affectées à se déplacer temporairement et parfois de façon permanente. Cette étude s’intéresse à un cas spécifique de relocalisation dans un site organisé, Corail-Cesselesse, créé quelques mois après le tremblement de terre dévastateur de janvier 2010, en Haïti. Initialement occupé par des ménages provenant surtout des quartiers de Delmas et Port-au-Prince et qui s’étaient réfugiés sur le vaste terrain de golf de Pétionville après avoir perdu leurs habitations, le site de Corail est ainsi étudié de façon descriptive et comparative pour évaluer l’évolution de la vulnérabilité des ménages qui y vivent. Pour ce faire, une revue du concept portant sur la vulnérabilité et la gestion des risques est nécessaire pour y dégager les indicateurs clés servant à l’analyse de l’évolution des états précédant et succédant à une catastrophe naturelle. En particulier, une approche combinant trois méthodes ralliant le qualitatif et quantitatif est utile pour conduire cette évaluation. À travers des questionnaires, des données géospatiales et d’entrevues auprès de professionnels en aménagement dans les pays en développement, on analyse dans quelle mesure la vulnérabilité sociale a évolué. On constate que la prise de décision sur la création de Corail a négligé plusieurs dimensions sociales nécessaires pour permettre aux familles de se rétablir d’un aléa d’une telle amplitude.

Chloroperoxidase (CPO) is a heme-containing glycoprotein secreted by the marine fungus Caldariomyces fumago. Chloroperoxidase contains one ferriprotoporphyrin IX prosthetic group per molecule and catalyzes a variety of reactions, such as halogenation, peroxidation and epoxidation. The versatile catalytic activities of CPO coupled with the increasing demands for chiral synthesis have attracted an escalating interest in understanding the mechanistic and structural properties of this enzyme. In order to better understand the mechanisms of CPO-catalyzed enantioselective reactions and to fine-tune the catalytic properties of chloroperoxidase, asparagine 74 (N74) located in the narrow substrate access channel of CPO was replaced by a bulky, nonpolar valine and a polar glutamine using site-directed mutagenesis. The CPO N74 mutants displayed significantly enhanced activity toward nonpolar substrates compared to wild-type CPO as a result of changes in space and polarity of the heme distal environment. More interestingly, N74 mutants showed dramatically decreased chlorination and catalase activity but significantly enhanced epoxidation activity as a consequence of improved kinetic perfection introduced by the mutation as reflected by the favorable changes in kcat and kcat/KM of these reactions. It is also noted that the N74V mutant is capable of decomposing cyanide, the most notorious poison for many hemoproteins, as judged by the unique binding behavior of N74V with potassium cyanide. Histidine 105 (H105) was replaced by a nonpolar amino acid alanine using site-directed mutagenesis. The CPO H105 mutant (H105A) displayed dramatically decreased chlorination and catalase activity possibly because of the decreased polarity in the heme distal environment and loss of the hydrogen bonds between histidine 105 and glutamic acid 183. However, significantly increased enantioselectivity was observed for the epoxidation of bulky styrene derivatives. Furthermore, my study provides strong evidence for the proposed histidine/cysteine ligand switch in chloroperoxidase, providing experimental support for the structure of the 420-nm absorption maximum for a number of carbon monoxide complexes of heme-thiolate proteins. For the NMR study, [dCPO(heme)] was produced using 90% deuterated growth medium with excess heme precursors and [dCPO(Phe)] was grown in the same highly deuterated medium that had been supplemented with excess natural phenylalanine. To make complete heme proton assignments, NMR spectroscopy has been performed for high-resolution structural characterization of [dCPO(heme)] and [dCPO(Phe)] to achieve unambiguous and complete heme proton assignments, which also allows important amino acids close to the heme active center to be determined.
