The ovariectomized (OVX) rat, a preclinical model for studying postmenopausal bone loss, may also be used to study differences in alveolar bone (AB). The objectives of this study were to quantify the differences in AB following estrogen replacement therapy (ERT), and to investigate the relationship between AB structure and density, and trabecular bone at the femoral neck (FN) and third lumbar vertebral body (LB3). Estrogen treated rats had a higher bone volume fraction (BV/TV) at the AB region (9.8% P < 0.0001), FN (12% P < 0.0001), and LB3 (11.5% P < 0.0001) compared to the OVX group. BV/TV of the AB was positively correlated with the BV/TV at the FN (r = 0.69 P < 0.0001) and the LB3 (r = 0.75 P < 0.0001). The trabecular number (Tb.N), trabecular separation (Tb.Sp), and structure model index (SMI) were also positively correlated (P < 0.05) between the AB and FN (r = 0.42, 0.49, and 0.73, respectfully) and between the AB and LB3 (r = 0.44, 0.63, and 0.69, respectfully). Given the capacity of AB to respond to ERT, future preclinical drug/nutritional intervention studies aimed at improving skeletal health should include the AB as a region of interest (ROI).

Acute alterations in cell volume can substantively modulate subsequent metabolism of substrates. However, how such alterations in skeletal muscle modulate protein metabolism is limited. The purpose of this study was to determine the time dependent influence of extracellular osmotic stress on protein turnover in skeletal muscle cells. L6 cells were incubated in hyperosmotic (HYPER; 425.3 ± 1.8mmol/kg), hypo-osmotic (HYPO; 235.4 ± 1.0mmol/kg) or control (CON; 333.5 ± 1.4mmol/kg) media for 4, 8, 12, or 24hrs. During the final 4hrs, incorporation of L-[ring-3,5-3H]-tyrosine was measured to estimate protein synthesis. Western blotting measured markers of protein synthesis and degradation. No differences were observed in any outcomes except p70S6K phosphorylation whereby HYPO was lower (p<0.05) than CON and HYPER; which remained similar except for a large increase at 8hrs for HYPER. These findings suggest that regardless of duration, extracellular osmotic stress does not significantly affect protein metabolism in L6 cells.

Accelerated life testing (ALT) is widely used to obtain reliability information about a product within a limited time frame. The Cox s proportional hazards (PH) model is often utilized for reliability prediction. My master thesis research focuses on designing accelerated life testing experiments for reliability estimation. We consider multiple step-stress ALT plans with censoring. The optimal stress levels and times of changing the stress levels are investigated. We discuss the optimal designs under three optimality criteria. They are D-, A- and Q-optimal designs. We note that the classical designs are optimal only if the model assumed is correct. Due to the nature of prediction made from ALT experimental data, attained under the stress levels higher than the normal condition, extrapolation is encountered. In such case, the assumed model cannot be tested. Therefore, for possible imprecision in the assumed PH model, the method of construction for robust designs is also explored.

This thesis tested whether cognitive performance during passive heat stress may be affected by changes in cerebrovascular variables as opposed to strictly thermally-induced changes. A pharmacological reduction in cerebral blood flow (CBF) using indomethacin along with a hypocapnia-induced CBF reduction during passive heat stress (Tre ~1.5°C above baseline) were used to investigate any cerebrovascular-mediated changes in cognitive performance. Repeated measures analysis of variance indicated that One-Touch Stockings of Cambridge (OTS) performance was not affected by a significant reduction in CBF during passive heat stress. More specifically, OTS accuracy measures did not change as a result of either a reduction in CBF or increasing passive heat stress. However, it was found that OTS response time indices improved with increasing passive heat stress independent of CBF changes. In conclusion, a significant reduction in CBF does not cause additional changes in performance of an executive functioning task during severe passive heat stress.

Although reductions in cerebral blood flow (CBF) may be implicated in the development of central fatigue during environmental stress, the contribution from hypocapnia-induced reductions in CBF versus reductions in CBF per se has yet to be isolated. The current research program examined the influence of CBF, with and without consequent hypocapnia, on neuromuscular responses during hypoxia and passive heat stress. To this end, neuromuscular responses, as indicated by motor evoked potentials (MEP), maximal M-wave (Mmax) and cortical voluntary activation (cVA) of the flexor carpi radialis muscle during isometric wrist flexion, was assessed in three separate projects: 1) hypocapnia, independent of concomitant reductions in CBF; 2) altered CBF during severe hypoxia and; 3) thermal hyperpnea-mediated reductions in CBF, independent of hypocapnia. All projects employed a custom-built dynamic end-tidal forcing system to control end-tidal PCO2 (PETCO2), independent of the prevailing environmental conditions, and cyclooxygenase inhibition using indomethacin (Indomethacin, 1.2 mg·Kg-1) to selectively reduce CBF (estimated using transcranial Doppler ultrasound) without changes in PETCO2. A primary finding of the present research program is that the excitability of the corticospinal tract is inherently sensitive to changes in PaCO2, as demonstrated by a 12% increase in MEP amplitude in response to moderate hypocapnia. Conversely, CBF mediated reductions in cerebral O2 delivery appear to decrease corticospinal excitability, as indicated by a 51-64% and 4% decrease in MEP amplitude in response to hypoxia and passive heat stress, respectively. The collective evidence from this research program suggests that impaired voluntary activation is associated with reductions in CBF; however, it must be noted that changes in cVA were not linearly correlated with changes in CBF. Therefore, other factors independent of CBF, such as increased perception of effort, distress or discomfort, may have contributed to the reductions in cVA. Despite the functional association between reductions in CBF and hypocapnia, both variables have distinct and independent influence on the neuromuscular system. Therefore, future studies should control or acknowledge the separate mechanistic influence of these two factors.

The purpose of this study was to examine the effects of increased extracellular leucine concentration on protein metabolism in skeletal muscle cells when exposed to 3 different osmotic stresses. L6 skeletal muscle cells were incubated in either a normal or supplemental leucine (1.5mM) medium set to hypo-osmotic (230 ± 10 Osm), iso-osmotic (330 ± 10 Osm) or hyper-osmotic (440 ± 10 Osm) conditions. 3H-tyrosine was used to quantify protein synthesis. Western blotting analysis was performed to determine the activation of mTOR, p70S6k, ubiquitin, actin, and μ-calpain. Hypo-osmotic stress resulted in the greatest increase in protein synthesis rate under the normal-leucine condition while iso-osmotic stress has the greatest increase under the elevated-leucine condition. Elevated-leucine condition had a decreased rate in protein degradation over the normal condition within the ubiquitin proteasome pathway (p<0.05). Leucine and hypo-osmotic stress therefore creates a favourable environment for anabolic events to occur.

Kurzfassung: Der Markt für ökologische Lebensmittel wächst stark. Verbraucher kaufen Produkte aus ökologischem Landbau aus einer Vielzahl von Gründen. Ein Teil dieser Gründe lässt sich nicht auf die Produktqualität zurückführen, sondern beruht auf der Annahme, dass sich der Produktionsprozess des Ökologischen Landbaus hinsichtlich der Schonung von Umweltressourcen, der Nachhaltigkeit der Produktion und sozialen Komponenten vom konventionellen Anbau unterscheidet. Daneben spielt der Wunsch nach einer gesunden Ernährung eine Rolle. Ökologische Lebensmittel können als Vertrauensgüter verstanden werden. Lebensmittelskandale machten in den vergangenen Jahren auch vor ökologischen Lebens¬mitteln nicht Halt. Folgerichtig erschütterte dies das Vertrauen der Verbraucher in ökologische Produkte. Mit steigender Produktion könnte die Gefahr, das weitere solche Ereignisse auftreten, steigen. Daher besteht Bedarf für Methoden, die die ökologische Produktqualität im Sinne einer Authentizitätsprüfung prüfen. Eine solche Prüfung könnte sich auf die Analyse sekundärer Pflanzenstoffe stützen. Diese Gruppe von Pflanzeninhaltsstoffen spielt bei der Diskussion um die besondere Qualität ökologischer Pflanzenprodukte eine große Rolle. Postuliert wird, dass ökologisch angebaute Pflanzen mangels mineralischer Düngung und mangels Schädlingsbekämpfung mit synthetischen Pestiziden einem erhöhten Stress ausgesetzt sind. Dies soll sich in einem höheren Niveau der mit den Selbstverteidigungsmechanismen der Pflanze eng verbundenen sekundären Pflanzenstoffe ausdrücken. Wichtige Untergruppen der sekundären Pflanzenstoffe sind Carotinoide und Polyphenole. An Weizen (Triticum aestivum L. und Triticum durum L.) und Möhre (Daucus carota L.) als für den ökologischen Landbau wichtigen Produkten wurden Messungen der Carotinoid- und Polyphenolkonzentration mit dem Ziel durchgeführt, die potentielle Eignung dieser Pflanzenstoffe als Biomarker zur Authentizitätsprüfung ökologischer Produkte zu evaluieren. Dazu wurden Proben aus ökologischem und konventionellem Anbau (Paarvergleich) untersucht. Diese stammten aus Langzeit-Feldversuchen (Weizen aus dem DOK- und dem MASCOT-Versuch), Feldversuchen und von Betriebspaaren untersucht. Ein generell höheres Niveau sekundärer Pflanzenstoffe in Möhren bzw. Weizen aus ökologischem Anbau gegenüber Proben aus konventionellem Anbau wurde nicht gefunden. Die Carotinoide waren weder bei der Möhre noch beim Weizen zur Authentizitätsprüfung geeignet. Die Konzentration der Carotinoide wurde stark durch die nicht dem Anbau¬verfahren zuzuordnenden Faktoren Klima, Sorte und Standort beeinflusst. Die Luteinkonzentration war das einzige durch das Anbauverfahren systematisch beeinflusste Carotenoid bei Weizen und Möhre. Die Unterschiede der Luteinkonzentration waren aber im Paarvergleich von Proben (ökologischer versus konventioneller Anbau) nicht durchgängig signifikant. Die Eignung von Polyphenolen als potentielles Authentizitätskriterium wurde nur an Möhren geprüft. Im Paarvergleich unterschieden sich die Konzentrationen einzelner Polyphenole signifikant und konsistent über Probenjahre und Standorte, nicht jedoch über Sorten hinweg. Wie bei den Carotinoiden konnte auch hier ein starker Einfluss von Probenjahr, Standort und Sorte gezeigt werden. Trotz der Variation durch diese nicht dem Anbau zuzuordnenden Faktoren war eine korrekte Klassifizierung der Proben nach Anbauverfahren möglich. Dies wurde mittels Diskriminanzanalyse getestet. Die Polyphenole sind daher potentiell als Authentizitätskriterium geeignet.

Competence development is considered a preventive strategy of burnout. At an organizational context some competences could be linked as precursors or consequences. In self-assessment of competence development, students perceive stress tolerance as a priority competence to ameliorate. Moreover employers and recruitment consultants agree that this is a new authentic challenge for organizations. The main reasons of this result are debated, this study should consider the importance of competence development from a holistic point of view. In addition it considers the exploration of the relationship between stress tolerance and competence development, according to Conservation Resources (COR) theory (Hobfoll 1988, 1989, 1998, 2004) where the resource loss is considered the principal component in the stress process

Stressors and traumatic events may contribute to the development of many psychopathologies, PTSD among them. People with this disorder can present significant cognitive impairments and losses in memory, particularly autobiographical memory (AM).This paper aims to present a systematic review of literature considering changes on autobiographical memory in people exposed to potentially traumatic stressors. Therefore, we performed a computerized search in databases PsycINFO, PubMed, Web of Science and Pilots in March 2012.A total of 29 articles were selected and grouped into three categories: studies comparing Trauma X PTSD, studies comparing PTSD X Trauma X Control and comparison studies according to post-traumatic symptoms. Results demonstrate that people with PTSD presents alterations in a larger number of AM components compared to the cases in which PTSD did not developed. In the same way, subjects never exposed to trauma did not demonstrate significant alterations on AM when compared to the other groups. The results of this study allowed concluding that changes of some components of the AM are primarily associated with PTSD, but it was not possible to clarify whether such changes are related to the timely development of the disorder, or if they are also observed in traumatic memories even in the absence of disorder.
